The pathological hallmark of rheumatoid arthritis (RA) is a synovial pannus that comprises proliferating and invasive fibroblast-like synoviocytes, infiltrating inflammatory cells, and an associated neoangiogenic response. Animal models have been established to study these pathological features of human RA. Spontaneous and induced animal models of RA primarily reflect inflammatory aspects of the disease. Among various induced animal models, collagen-induced arthritis (CIA) and collagen antibody-induced arthritis (CAIA) models are widely used to study the pathogenesis of RA. Improved transplantation techniques for severe combined immunodeficiency (SCID) mouse models of RA can be used to evaluate the effectiveness of potential therapeutics in human tissues and cells. This review provides basic information on various animal models of RA, including CIA and CAIA. In addition, we describe a SCID mouse coimplantation model that can measure the long-distance migration of human RA synoviocytes and cartilage destruction induced by these cells.
Candida albicans, a polymorphic fungus, causes systemic and local infections. Recent reports show that the fungus is a main etiological agent for the arthritis. For trea tment, antifungal drugs and/or rheumatoid drugs are used, but resistance and side effects limit application of the drugs. In search of new sources for treatment of the fungal arthritis, we choose Egb 761 (extract of Ginkgo leaves 761), one of the most popular over-the-counter herbal medicines. The Egb 761 contains two major ingredients such as terpene and flavonoid. In the present study, we examined if the terpene portion of Egb 761 had anti-inflammatory activity against C.albicans-caused arthritis. The terpene was extracted with combination of methanol and water from the Egb 761, followed by gel-permeation chromatography. Presence of terpene was determined by the Salkowski colorimetric method and HPLC analysis. For an animal model of inflammation induction, mice were given an emulsion form of C.albicans cell wall mixed with Complete Freund's Adjuvant (CFA) by footpad-injection. Results showed that intraperitoneal administration of the water-soluble portion that contained terpene and flavonoid reduced the inflammation. Whereas the terpene had anti-inflammatory activity, flavonoid portion had no such activity, For determination of possible mechanism of the activity, the terpene seemed to be suppression of nitric oxide (NO) production from LPS-treated macrophages. Taken together the Ginkgo terpene may have anti-inflammatory effect against C.albicans-caused arthritis, possibly by blocking NO production.
To evaluate effect of CRSST on inhibiting the occurrence of arthritis, we performed the experiments including production of inflammatory cytokine and immunoglobin in collagen induced arthritis model. The results were obtained as follows. CRSST extract shows any cytotoxicity effect on mouse lung fibroblast cells at dose of 400 ㎍/㎖. CRSST group shows inhibitory effect on arthritis incidence than control group for six weeks. Arthritis index of CRSST group reduces from 4 weeks (75±17.4%) to 6 weeks (33.3±10.0%) compared with control group. In CRSST group, production of cytokines which shows suppressive effect on inflammation (IL-4, IL-10 ) are increased and which promotes inflammation (TNF-α, INF-γ) are decreased in blood. In CRSST group, production of immunogloblin (IgG2b, IgG3 and IgM) is reduced compared with control group, and rate of CD4+ and CD3+ T cell is lower in joint and higher in lymph node compared with control group. From above results it could be accepted that CRSST shows anti-arthritis effect via immune system especially through the controlling the inflammatory cytokines and immunoglobins. CRSST could be usefully applied for the prevention and treatment of RA. And also is expected to be clinically helpful on the treatment of RA through modification.
The fruit of Terminalia chebula Retzius has been used as a panacea in India and Southeast Asia but its biological activities have not been fully elucidated. Here we report anti-arthritic and analgesic effect of NDI10218, a standardized ethanol extract of Terminalia chebula, on collagen-induced arthritis and acetic acid-induced writhing model, respectively. Arthritis was induced in DBA/1J mice by immunizing bovine type II collagen and mice were treated with NDI10218 daily for 5 weeks after the onset of the disease. NDI10218 reduced the arthritis index and blocked the synovial hyperplasia in a dose-dependent manner. The serum levels of pro-inflammatory cytokines TNF-${\alpha}$, IL-6, and IL-$1{\beta}$ were significantly reduced in mice treated with NDI10218. Production of the inflammatory IL-17, but not immunosuppressive IL-10, was also inhibited in splenocytes isolated from NDI10218-treated arthritis mice. Administration of NDI10218 markedly decreased the number of T cell subpopulations in the regional lymph nodes of the arthritis mice. Finally, NDI10218 reduced the number of abdominal contractions in acetic acid-induced writhing model, suggesting an analgesic effect of this extract. Taken together, these results suggest that NDI10218 can be a new therapeutic candidate for the treatment of rheumatoid arthritis.
Fangchinoline (FAN) is a bisbenzylisoquinoline alkaloid that is widely known for its anti-tumor properties. The goal of this study is to examine the effects of FAN on arthritis and the possible pathways it acts on. Human fibroblast-like synovial cells (FLS), carrageenan/kaolin arthritis rat model (C/K), and collagen-induced arthritis (CIA) mice model were used to establish the efficiency of FAN in arthritis. Human FLS cells were treated with FAN (1, 2.5, 5, 10 µM) 1 h before IL-1β (10 ng/mL) stimulation. Cell viability, reactive oxygen species measurement, and western blot analysis of inflammatory mediators and the MAPK and NF-κB pathways were performed. In the animal models, after induction of arthritis, the rodents were given 10 and 30 mg/kg of FAN orally 1 h before conducting behavioral experiments such as weight distribution ratio, knee thickness measurement, squeaking score, body weight measurement, paw volume measurement, and arthritis index measurement. Rodent knee joints were also analyzed histologically through H&E staining and safranin staining. FAN decreased the production of inflammatory cytokines and ROS in human FLS cells as well as the phosphorylation of the MAPK pathway and NF-κB pathway in human FLS cells. The behavioral parameters in the C/K rat model and CIA mouse model and inflammatory signs in the histological analysis were found to be ameliorated in FAN-treated groups. Cartilage degradation in CIA mice knee joints were shown to have been suppressed by FAN. These findings suggest that fangchinoline has the potential to be a therapeutic source for the treatment of rheumatoid arthritis.

This study will be a source of good information for the researchers working in the area of drug designing and molecular Pharmacology especially in anti-inflammatory and anti arthritic medicines for target based therapy.
The systematic approach to pharmacologic treatment is typically to begin with the safest, simplest, and most conservative measures. It has been realized that the more rapidly inflammation is under control, the less likely it is that there will be permanent sequelae. Nonsteroidal anti-inflammatory drugs (NSAIDs) are the mainstay of initial treatment for inflammation. In addition, the slow-acting antirheumatic drugs (SAARDs) and disease-modifying antirheumatic drugs (DMARDs) have efficacy of anti-inflammatory action in children with chronic arthritis. New therapeutic modalities for inflammation, such as etanercept and infliximab, promise even further improvements in the risk/benefit ratio of treatment. It is not typically possible at the onset of the disease to predict which children will recover and which will go on to have unremitting disease with lingering disability or enter adulthood with serious functional impairment. Therefore, the initial therapeutic approach must be vigorous in all children.
The balance between osteoblast-dependent bone formation and osteoclast-dependent bone resorption maintains bone homeostasis. In inflammatory conditions, this balance shifts toward bone resorption, causing osteolytic bone lesions observed in rheumatoid arthritis and periodontitis. A recently discovered family of cytokine IL-17 is widely reported to mediate diverse inflammatory processes. During the last decade, novel roles for IL-17 in skeletal homeostasis have been discovered indicating the potential importance of this cytokine in bone metabolism. This review will summarize and discuss the involvement of IL-17 during bone homeostasis in both physiologic and pathologic conditions. A better understanding of the role of IL-17 in skeletal systems warrants an advance in bone biology, as well as development of therapeutic strategies against bone-lytic diseases, such as rheumatoid arthritis and periodontitis.
The aim of this study was to investigate the anti-inflammatory and anti-oxidant activity of Ligularia fischeri leaf extract on adjuvant induced arthritis in experimental mice. The oral administration of the L. fischeri leaf extract (LF), at doses of 100 and 200 mg/kg body weight once a day for 3 weeks, significantly reduced hindpaw swelling and the production of inflammatory cytokines (tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$, and IL-6). Treatment with LF (100 mg/kg) also decreased the serum levels of triglyceride and low density lipoprotein(LDL)-cholesterol, and increased high density lipoprotein(HDL)-cholesterol contents compared with those of a control group. The induction of arthritis significantly increased oxidized proteins such as protein carbonyl, advanced oxidation protein products, and advanced glycation end-products in the lung, heart, and brain. Treatment with LF for 3 weeks reduced the levels of oxidized proteins. These results suggest that L. fischeri extract might be beneficial in the treatment of chronic inflammatory disorders.
This study was peformed to investigate possible mechanisms underlying possible effect of Coicis Semen composition (CSC) on inflammatory diseases using in vivo model of RA in the mice. Results are summarized as follows. In production of inflammatory cytokines, INF-${\gamma}$ in the spleen and IL-6 in the serum were decreased by CSC treatment. TNF-${\alpha}$ in serum was significantly decreased, IL-4 in the spleen was significantly increased by CSC treatment. In production of rheumatoid factors, IgM and IgG were significantly decreased by CSC treatment. The present data suggest that CSC treatment can improve pathological damage by CIA. So we expect that CSC should be used as a effective drugs for not only rheumatoid arthritis but also another autoimmune disease. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.
