Purification of Rotavirus Infection-Inhibitory Protein from Bifidobacterium breve K-110

  • The inhibitory activity of fifty Bifidobacteria toward the infectivity of a rotavirus, which is the predominant cause of sporadic diarrhea in infants and young children, was investigated, and Bifidobacterium breve K-110 was found to have the most potent inhibitory activity. Accordingly, the rotavirus infection-inhibitory protein was purified, and its molecular weight was determined to be 76 kDa by SDS-PAGE. It was heat-labile and its 50% inhibitory concentration ($IC_{50}$) was 0.045 $\mu g/ml$.

  • The root nodules and Glycine max were classified as determinate nodule based on their morphological characteristics, and isolated endosymbiont as a Bradyrhizobium based on its growth rate and single subpolar flagellum. The isolate was similar to B. japonicum USDA110 in utilization of carbon source, growth at 38.deg.C and 2% NaCl, production of $H_{2}$S and especially in the restriction endonuclease digestion pattern of symbiotic genes, allowing them to be placed in sTI group together. The former, however, grew better than the later in broad pH range from 5.0 to 9.5. Infectivity and effectivity of the isolate were confirmed by inoculation of soybean seedlings with the isolates. Characteristics of the reisolated endosymbiont from induced root nodules were identical to those of the first isolate. From these results, it was confirmed that Bradyrhizobium strain isolated from the root nodules of Glycine max was a real symbiont, and was named B. japonicum SNU001.

Exploration of the Binding Mode of Indole Derivatives as Potent HIV-1 Inhibitors Using Molecular Docking Simulations

  • The HIV-1 envelope glycoprotein gp120 plays a vital role in the entry of the virus into the host cells. The crucial role of the glycoprotein suggests gp120 as potential drug target for the future antiviral therapies. Identification of the binding mode of small drug like compounds has been an important goal in drug design. In the current study we attempt to propose binding mode of indole derivatives in the binding pocket of gp120. These derivatives are reported to inhibit HIV-1 by acting as attachment inhibitors that bind to gp120 and prevent the gp120-CD4 interaction and thus inhibit the infectivity of HIV-1. To elucidate the molecular basis of the small molecules interactions to inhibit the glycoprotein function we employed the molecular docking simulation approach. This study provides insights to elucidate the binding pattern of indole-based gp120 inhibitors and may help in the rational design of novel HIV-1 inhibitors with improved potency.

Galleria mellonella 유충을 이용한 곤충병원성 선충의 배양 조건 (Culture Condition of Entomopathogenic Nematodes Using Galleria mellonella Larva)

  • A simple method for the in vivo production of third-stage infective juveniles(IJs) of Steinernema glaseri was developed. Using Galleria mellonella larvae, only IJs can be rapidly generated inadequate quantities for field application. The nematode inoculation concentration and incubation temperature were critically important. The most effective temperature for infectivity of Steinernema glaseri IJs to Galleria mellonella larvae was 33$^\circ C$. However, the total number of menatodes harvested at 25$^\circ C$ about 66,000 IJs per larva was significantly greater than those at other temperatures. The optimal inoculation number of nematodes was 60 to 80 nematodes per host larva. The higher nematode inoculation concentration of 100 IJs per larva caused a rapid decrease in the total number of IJs harvested. As the inoculation medium pH increased, the number of IJs harvested increased and reached about 110,000 IJs per larva at pH 9.0. The pathogenicity of IJs decreased y increasing the salt concentration in the medium.

Isolation and physiological characterization of a novel virus infecting Stephanopyxis palmeriana (Bacillariophyta)

  • Recent studies have shown the importance of viruses as potential sources of plankton mortality, which affect primary production and biogeochemical functions of their hosts. Here, we report basic characteristics of a novel virus (Stephanopyxis palmeriana virus: SpalV) that causes lysis of a culture of the diatom S. palmeriana, which was isolated in Jaran Bay, Korea, in August 2008. SpalV is a round-shaped viral particle ~25-30 nm in diameter that propagates in its host's cytoplasm. In addition, it shows species-specific infectivity among the tested diatom species. The burst size and latent period are estimated to be roughly 92 infectious units $cell^{-1}$ and <80 h, respectively.

  • Steinernematid와 heterorhabditid 선충은 쥐며느리에 침입하여 치사시킬수는 있었지만 효과는 없었다. Steinernema carpocapsae 포천 strain과 S. glaseri 동래 strain이 S. carpocapse All strain과 Heterorohabditis bacteriophora보다 효과가 있었으며, 병원성 발현에서 선충의 접종농도가 기주의 밀도보다 중요한 요인이었다.

Effects of gamma-irradiation on the infectivity and chromosome aberration of Clonorchis sinensis

  • Effects of gamma irradiation on the worm survival and chromosomal aberration of Clonorchis sinensis were studied. The metacercariae irradiated with various amounts of gamma radiation (ranging from 5 Gy to 50 Gy) were fed to rats, and the effects were compared with those of non-irradiated controls. Recovery rates of adult worms in irradiated groups were reduced gradually as increasing of the irradiation doses. No worm was recovered from rats which were fed with 50 Gy irradiated metacercariae. The chromosome number was 2n = 56 in all worms from all experimental groups. However, the groups irradiated with 20 Gy, 25 Gy or 30 Gy showed variations in the chromosome number, depending on different cells in the same individual. Radiation doses used in this study did not appear to induce chromosome aberrations, however, irradiation with 30 Gy showed slightly reduced chromosome size.

  • Listeria monocytogenes is a facultative bacterium that cause severe clinical disease including meningoencephalitis, septicaemia, and abortion in pregnant women, newborn infants, the debilitated elderly or immunocompromised people. In preliminary experiments on murine listeriosis we noticed suppurative gastritis in mice infected with L. monocytogenes by the intragastric route. The aims of the present study were ⅰ) to describe the histopathology of the experimentally listeria-induced gastroenteritis ⅱ) to investigate the influence of bacterial strain and laboratory mouse strain on infectivity and on the severity of the infection; [3] to examine possible effects of preliminary intragastric administration of sodium bicarbonate. 소 코로나바이러스에 대한 단크론항체 생산과 특성 (Production and characterization of monoclonal antibody against bovine coronavirus)

  • Eight monoclonal antibodies(MAbs) against bovine coronavirus(BCV) were produced and characterized. Three MAbs(1G9, 4H12, 5C1) specific to the S glycoprotein and two HE glycoprotein-specific MAbs(2A5, 5G4) were found to neutralize the BCV in fluorescence focus neutralization(FFN) test. Two HE-specific MAbs from the neutralizing MAbs inhibited the hemagglutinating activity of the BCV. None of the N protein-specific MAbs(1C1, 5A12, 6H1) neutralized the virus infectivity. Bovine coronavirus and mouse hepatitis virus, which belong to group II coronaviruses, were differentiated from other groups of coronaviruses(porcine transmissible gastroenteritis virus, porcine epidemic diarrhea virus, canine coronavirus) by all MAbs in fluorescence antibody test(FA), but not in FFN test.

The first report of Trypanosoma sp. (Sarcomastigophora: Kinetoplastida) infecting catfish, Silurus asotus (Siluriformes: Siluridae) from Hyungsangang (River), Korea

  • Trypanosoma sp. (Sarcomastigophora: Kinetoplastida) was found in the blood of the Korean catfish, Silurus asotus, for the first time in Korea. The morphological characteristics of Trypanosoma sp. in the present study were similar with those of T. carassii, T. tincae and T. danilewskyi. However, the free flagellum length of Trypanosoma sp. was obviously shorter than that of those species. The species identification was reserved until elucidating the pleomor-phism according to the phase of infection and the cross infectivity among fish species.
