Analysis of Dissociation Pathway of HET-s Prion Using Steered Pulling Simulation

  • Prion is a group of the proteins known for its infection mechanisms of Creutzfeldt-Jakob disease (CJD) and other diseases. Solved structures and proven biological roles of fungal prions add tremendous potential to conducting computational simulations. Our research focuses on the binding dynamics of HET-s(218-289), one of the heterokaryon fungal prion originated from Podospora anserina, by calculating the binding free energy using umbrella sampling at 300 K. The binding free energy calculated was $-54.5kcal\;mol^{-1}$, relatively similar to the binding energy of other amyloid fibrils. The simulation result suggests the thermodynamic properties of ${\beta}$-solenoid of HET-s prion and its similarity in dissociation pathways compared to amyloids.

Poly A tail을 결핍한 Simian virus 40 spliced RNA의 세포내 분포 (The abundant presence of nonpolyadenylated SV40 late 19S spliced RNA in the nucleus of monkey cell)

  • We have examined the structures and cellular distributions of the SV40 late RNAs present in monkey cells at late times after infection. One particular RNA species, spliced at residue 373(373-RNA), was found to be as abundant as the major late 16S RNAs. This result was unexpected since previous reports showed that the molecular ratio of the 373-spliced 19S RNA to 16S RNA is approximately 0.1 among either cytoplasmic polyadenylated or polysomal viral RNAs. Both sedimentation and electrophoretic analysis indicated that the 373-RNA was approximately 16S to 19S in size. Therefore, it was not a splicing intermediate or the product of premature termination of transcription within the late leader region. Whereas most SV40 late 16S RNA is polyadenylated and located in the cytoplasm, the majority of 373-RNA was found to lack poly A, and be located in the nucleus.

  • We have shown in Part I [1] of this study that medical needle-stick injuries are causing serious health problems to healthcare personnel and other professionals that require the attention of healthcare and textile researchers to develop new protective systems. Responding to such need, a needle force measurement device that is capable of measuring dynamic forces experienced by medical needles during needle penetration through protective articles was developed and described in part I. This paper reports the results of evaluation of protective woven fabrics from high performance fibers and standard and bifid medical needles using the force measurement system. The woven fabrics varied in cover factor, number of layers, and orientation angle. Standard and bifid needles with different gap widths were used to evaluate the resistance of the fabric to needle penetration.

Inhibitory Effects of Streptomyces sp. MBTH32 Metabolites on Sortase A and Sortase A-Mediated Cell Clumping of Staphylococcus aureus to Fibrinogen

  • Sortase A (SrtA), a type of transpeptidase responsible for anchoring surface proteins to the peptidoglycan cell wall, is important in the virulence of gram-positive bacteria. Three compounds were isolated from marine-derived Streptomyces sp. MBTH32 using various chromatography techniques. The structures of these compounds were determined based on spectroscopic data and comparisons with previously reported data. Among the metabolites tested, lumichrome showed strong inhibitory activity against Staphylococcus aureus SrtA without affecting cell viability. The results of cell clumping activity assessment suggest the potential for using this compound to treat S. aureus infection by inhibiting SrtA activity.

  • A minimally invasive procedure refers to a procedure that minimizes tissue damage due to incision. By limiting the size of the incision, it can be expected to reduce the time taken for wound healing, reduce pain, and reduce the risk of infection. Recently, studies have been attempted to dissect the structures of the neuromusculoskeletal system using ultrasound-guided minimally invasive technique. Among those, dissecting thread may be utilized in several musculoskeletal diseases, including carpal tunnel syndrome, cubital tunnel syndrome, trigger finger, and selective crural fasciectomy. In this brief review, the use of dissecting thread in treating neuromusculoskeletal diseases are described.

Computational approaches for molecular characterization and structure-based functional elucidation of a hypothetical protein from Mycobacterium tuberculosis

  • Adaptation of infections and hosts has resulted in several metabolic mechanisms adopted by intracellular pathogens to combat the defense responses and the lack of fuel during infection. Human tuberculosis caused by Mycobacterium tuberculosis (MTB) is the world's first cause of mortality tied to a single disease. This study aims to characterize and anticipate potential antigen characteristics for promising vaccine candidates for the hypothetical protein of MTB through computational strategies. The protein is associated with the catalyzation of dithiol oxidation and/or disulfide reduction because of the protein's anticipated disulfide oxidoreductase properties. This investigation analyzed the protein's physicochemical characteristics, protein-protein interactions, subcellular locations, anticipated active sites, secondary and tertiary structures, allergenicity, antigenicity, and toxicity properties. The protein has significant active amino acid residues with no allergenicity, elevated antigenicity, and no toxicity.

  • 인터넷이 지속적으로 성장과 발전을 거듭해가고 있는 이면에는 이를 악용하기 위한 다양한 인터넷 공격들이 발생하고 있다. 초기 인터넷 환경에서는 공격자가 역량과시 및 취미 등으로 인터넷 환경을 악용한 공격이 존재하였지만, 금전적인 이득을 목적으로 각종범죄와 연관된 체계적으로 복잡한 공격들이 발생하고 있다. 최근 들어서 바이러스나 윔과 같은 구조가 단순한 소스 멀티타깃(one source multi-target)의 형태가 존재하였지만, 멀티소스 싱글타깃(multi-source single target)의 형태를 갖는 APT(Advanced Persistent Threat, 지속적인 지능형 공격)으로 사용자들로 하여금 막대한 피해를 입히고 있다. 그러므로 본 논문에서는 Agent 및 관리 시스템은 악성코드 감염을 사전에 예방하는 기능을 고도화하여 사용자의 부정행위를 통한 자료유출을 감시할 수 방지 시스템을 설계 및 구현하였다. 성능평가에서는 감사데이터 생성 여부, 무결성 침해 발생 시 탐지 여부, 정상트래픽 오탐 여부, 프로세스 탐지 및 차단 기능 설정, Agent 정책 적용 가능여부에 대해서 기능을 분석하였다.

  • 섬유화성 종격동염은 종격동 조직외 과다한 섬유화로 인한 종격동 구조물의 압박과 폐쇄로 비롯된 임상증상을 특정으로 하는 드문 질환으로 정확한 병태생리는 알지 못하나 육아종성 감염질환에서 비롯되는 것이 가장 중요한 원인이라고 알려져 있다. 미국의 일부 지방에서는 Histoplasmosis가 원인이 되는 증례가 많이 보고되어 있으나 결핵이 원인이 된 경우는 비교적 적다. 저자들은 결핵이 그 원인으로 밝혀진 섬유화성 종격동염 2예를 경험하였기에 문헌 고찰과 함께 보고하는 바이다.

Bovine Genome-wide Association Study for Genetic Elements to Resist the Infection of Foot-and-mouth Disease in the Field

  • Foot-and-mouth disease (FMD) is a highly contagious disease affecting cloven-hoofed animals and causes severe economic loss and devastating effect on international trade of animal or animal products. Since FMD outbreaks have recently occurred in some Asian countries, it is important to understand the relationship between diverse immunogenomic structures of host animals and the immunity to foot-and-mouth disease virus (FMDV). We performed genome wide association study based on high-density bovine single nucleotide polymorphism (SNP) chip for identifying FMD resistant loci in Holstein cattle. Among 624532 SNP after quality control, we found that 11 SNPs on 3 chromosomes (chr17, 22, and 15) were significantly associated with the trait at the p.adjust <0.05 after PERMORY test. Most significantly associated SNPs were located on chromosome 17, around the genes Myosin XVIIIB and Seizure related 6 homolog (mouse)-like, which were associated with lung cancer. Based on the known function of the genes nearby the significant SNPs, the FMD resistant animals might have ability to improve their innate immune response to FMDV infection.

Orientia tsutsugamushi의 유핵세포내 감염능 분석 및 기전 (Infectivity of Orientia tsutsugamushi to Various Eukaryotic Cells and Their Cellular Invasion Mechanism)

  • Orientia tsutsugamushi is obligate intracellular bacterium that grows within the cytoplasm of the eukaryotic host cells. Therefore capability of the attachment, entry into the host cell and intracellular survival should be critical process for oriential infection. In this study we investigated the cellular invasion mechanism of Orientia tsutsugamushi and the role of transmembrane heparan sulfate proteoglycan, which binds diverse components at the cellular microenvironment and is implicated as host cell receptors for a variety of microbial pathogens. First of all Orientia tsutsugamushi can invade a wide range of nonprofessional phagocytic cells including fibroblast, epithelial cells and endothelial cells of various host species, including Band T lymphocytes. Thus, it was postulated that the attachment of O. tsutsugamushi requires the recognition of ubiquitous surface structures of many kinds of host cells. Treatments with heparan sulfate and heparin inhibited the infection of Orientia tsutsugamushi in dose-dependent manner for L cell, mouse fibroblast, whereas other glycosaminoglycans such as chondroitin sulfate had no effect. Collectively, these findings provide strong evidence that initial interaction with heparan sulfate proteoglycan is required for the oriential invasion into host cells.
