Avian coccidiosis is a very important disease of poultry industry. In recent years, many researchers studied for the immunology of avian coccidiosis. Coccidial vaccines have been used at some poultry farms in many countries. In order to evaluate the anticoccidial effects of coccidial vaccine to chicken treated with anticoccidial drugs, 4 week-old chicken were administered anticoccidial drugs and challenged with avian coccidia at 2 weeks after vaccination of coccidia. The body weight gains, lesion scores, oocyst indices and anticoccidial indices of chicken were investigated at the time of vaccination, challenge and necropsy. The body weight gains and feed conversion rates of groups immunized with coccidial vaccine and treated with anticoccidial drugs were moderately higher and lower than those of groups treated with anticoccidial drugs, respectively. The lesion score and oocyst indices of all groups immunized with coccidial vaccine and/or treated with anticoccidial drugs were milder than those of infected control groups. The anticoccidial indices of groups immunized with coccidial vaccine and treated with anticoccidial drugs were moderately higher and lower than those of groups. treated with anticoccidial drugs. But all of the investigated items were not statistically significant.

The infectious bursal disease (IBD) is a highly contagious and acute poultry disease caused by Birnavirus. However, the vaccination is the only disease prevention, but several factors impeded vaccine development. Thus, a need for time to develop a novel technique for managing and treating respiratory diseases in poultry birds. Passive immunization is a hope and a possible alternative used in birds to meet this need. The current research attempted to produce egg yolk-based polyclonal antibodies against the IBD virus. The benefits of IgY include ease of extraction, lack of reaction with mammalian Fc receptors, and low production cost. Commercial layers were immunized with inactivated IBD virus subcutaneously according to the treatment regimen. The eggs were gathered daily, and yolk antibodies were extracted with the ammonium sulfate precipitation technique. The use of an indirect hemagglutination test demonstrated that IgY was IBD-specific. Until the end of the experiment, the specific IgY immunoglobulins did not lose activity when stored at 4℃. The specific immunoglobulin (IgY) treated challenged birds were demonstrated 92% recovery in comparison to the control group. The study implies that the IBDV specific IgY is an easily prepared and rich source of antibodies and offers an alternative therapeutic agent to cure IBD-infected birds.

To reduce the economic impact and control Low pathogenic avian influenza (LPAI), vaccination with inactivated vaccine has been considered in this country. We tried to develop inactivated vaccine with reassorted H9N3 AI virus which has different type of neuraminidase compare to those of field AI virus. Before reassorted vaccine was produced, we confirm the virus as master seed by limiting dilution, RT-PCR and sequencing method. Also, we evaluate the biological characteristics of the virus to find out the possibility of prevention against field infection of AI virus. Finally, we evaluate the safety and efficacy of the vaccine made of reassorted AI virus in the specific pathogen free (SPF) chickens. After limiting dilution, we choose RV7CE4 as a vaccine candidate and compare the gene sequence of this vaccine strain to those of AI05GA which is parents strain. Compared to amino acid sequences of specific gene of AI05GA and RV7CE4, exhibited a high degree of amino acid sequence homology. In the safety and efficacy test, there were no specific clinical signs or mortality. Reassorted H9N3 viruses were reisolated in cloaca swab on 5 days post inoculation. In the vaccine study, once or twice vaccination was performed and challenged with H9N2 field virus (01310). Vaccine has no adverse effect on birds and formed good immune capability which reduce viral shedding in the birds infected with 01310. Based on the above result, we developed reassorted H9N3 vaccine which will efficiently prevent the low pathogenic AIV (H9N2) infection in the poultry farms.
Background: Fluralaner is a novel drug belonging to the isoxazoline class that acts on external parasites of domestic animals. It is used systemically via drinking water, especially against red poultry mite in layer chickens. Fluralaner is frequently used in layers infected with D. gallinae. However, no study to date has investigated the effects of feed intake and water hardness. Objectives: This study aimed to investigate the effects of variable water hardness and feed intake on the pharmacokinetic profile of fluralaner. Methods: Layer chickens were divided into four groups (n = 8): fed + purified water (Group 1), feed restricted + purified water (Group 2), feed restricted + hard water (Group 3), and feed restricted + soft water (Group 4). After administering a single dose of the drug with drinking water, the blood samples were collected for 21 days. Fluralaner concentrations in plasma samples were determined by liquid chromatography/tandem mass spectrometry. The maximum plasma concentration (Cmax), time to reach maximum plasma concentration (tmax), area under the concentration-time curve values (AUC0-21d), half-life (t1/2), and other pharmacokinetic parameters were calculated. Results: Although the highest maximum plasma concentration (Cmax) was determined in Group 1 (fed + purified water), no statistically significant difference was found in the Cmax, tmax, t1/2, MRT0-inf_obs, Vz/Fobs, and Cl/F_obs parameters between the experimental groups. Conclusions: It was concluded that the feed intake or water hardness did not change the pharmacokinetic profile of fluralaner in layer chickens. Therefore, fluralaner could be used before or after feeding with the varying water hardness in poultry industry.

This study was conducted to identify the location characteristics of infected farms in the areas where livestock diseases were clustered(southern Gyeonggi-do and Chungcheong-do), analyze the probability of disease occurrence in poultry farms, find out the areas corresponding to the conditions, and use them as the basis for prevention of livestock diseases, selection of discriminatory prevention zones, and establishment of prevention strategies and as the basic data for complementary measures. The increase of one poultry farm within a radius of 3-kilometers increases the risk of HPAI infection by 10.9% compared to the previous situation. The increase of 1m in distance from major roads with two lanes or more reduces the probability of HPAI infection by 0.001% compared to the previous situation. If the distance of the poultry farm located with 15 kilometers from a major migratory bird habitat increases by 15 to 30 kilometers, the chance of infection with HPAI is reduced by 46.0%. And if the distance of the same poultry farm increase by more than 30 kilometers, the chances of HPAI infection are reduced by 88.5%. Based on the results of logistic regression, the predicted probability was generated and the actual area of the location condition with 'more than 15 poultry farms within 3km a radius of, within 1km distance from major roads, and within 30km distance from major migratory birds habitat was determined and the infection rate was measured. It is expected that the results of this study will be used as basic data for preparing the data and supplementary measures when the quarantine authorities establish discriminatory quarantine areas and prevention strategies, such as preventive measures for the target areas and farms, or control of vehicles, by identifying the areas where livestock diseases are likely to occur in the region.

Newcastle disease (ND) is a highly contagious infection of poultry, Two pathology-based techniques, in situ RT-PCR and in situ hybridization (ISH) were applied to formalin-fixed, paraffin-embedded tissues from chickens naturally infected with velogenic ND virus (VNDV). Two pairs of primers and a probe for ISH and in situ RT-PCR, respectively, were selected from highly conserved region of matrix gene of NDV. The ISH experiment was carried out using MicroProbe$^{TM}$ capillary action system within 2 hours. In situ RT-PCR was performed using MicroProbe$^{TM}$ capillary action system and GeneAmp In Situ PCR system. With ISH and in situ RT-PCR, viral nucleic acid was detected in the central nervous system of chickens from infected with neurotropic velogenic Newcastle disease virus (NVNDV), whereas viral nucleic acid was detected in various organs or tissues of chickens from infected with viscerotropic velogenic Newcastle disease virus (VVNDV). In the NVND group, positive signals were characteristically defined in the cytoplasm of neuron, vascular endothelial cells, and perivascular mononuclear macrophages in the central nervous system. One of NVND group, chicken from one farm exhibited positive signals in the bronchial epithelium. The VVND group chickens showed positive reaction in the macrophages, vascular endothelium, and bronchiolar epithelium. Markedly, viral nucleic acid was detected in the macrophages of morphologically normal tissues which were peripheral or located in distant areas from lesions. The central nervous system of chickens infected with VVND virus had positive signals in the vascular endothelial cell, perivascular mononuclear macrophages and some neuron. The number and intensity of the positive cells by in situ RT-PCR were more and stronger, respectively, in comparison with those by ISH. Particularly, positive reaction was detected in macrophages infiltrating in cardiac muscle by in situ RT-PCR, but not obtained by ISH. Therefore, these results demonstrated that ISH is a rapid diagnostic method for detection of NDV and in situ RT-PCR can be used as an efficient method for detection of low viral load infection or subclinical viral infection of NDV.
A total of 1050 layer and broiler chickens from 63 flocks of 21 poultry farms in South-Kyeonggi area, aged from 1 to 18 weeks, were investigated for the prevalence of Cryptosporidium baileyi infection from May 1992 to March 1993. The results have shown that 44 chickens(4.19%) were infected with C.(baileyi during the period of investigation) Fecal samples were treated by using Sheather's flotating technique and were examined under the light and phase contrast microscopy and then stained by Kinyoun method. Cryptosporidium oocysts were found in 35 out of 650 broilers(5.4%) and 9 out of 400 layer chickens(2.3%) aged mainly from 2 to 12 weeks. The regional infection rate were 4.7% in Pyeong-taek, 5.1% in An-sung and 2.3% in Yong-in, respectively. The average size of isolated oocyst was about $5.23{\times}4.92{\mu}m$ and the oocysts were orally inoculated into 2-day-old SPF chickens for the histological examination of oocyst in bursa of Fabricius. The study has concluded that C. baileyi infects chickens and oocysts were isolated in South-Kyeonggi area.
Aspergillosis, caused by infection with Aspergillus fumigatus and less commonly by other Aspergillus species, is a prevalent and costly respiratory disease of poultry, In a flock of chicks, the number of birds, 4∼5 months old, had become gradually emaciated and subsequently died. Gross necropsy revealed multiple granulomatous masses on the abdominal serosa. The masses, 4∼15 mm in size, were attached on mainly intestinal wall. Also, the smaller masses in size were on mesentery and pancreas. However, only a few small white nodules were scattered throughout liver and lung in few samples. Microscopically, the mass were granulomatous with a central area of necrosis containing numerous septate, branched fungal hypae consistent with Aspergillus sp. These were surrounded by macrophages, giant cells, lymphocytes and fibrous tissues. Nodular lesions of liver and lung were seemed to spread hemotogenously from intestine and the possible route of infection was speculated by oral. This report is a Aspergillus-induced granuloma limited to the serosa of abdominal cavity, especially of intestinal wall.
The clinical and histopathological observation of leucocytozoonosis was carried out during July, 1982 at a poultry compound in Gyeongbuk province and the results were summarized as follows: 1. Leucocytozoonosis was observed in 36,900 of 59,900 chickens (61.6%) and the mortality rate was 18.3% (6,760 of 36,900 chickens infected). 2. Clinical findings were anorexia, dyspnea, paleness of comb, greenish diarrhea and reduction of egg production. 3. Gross lesions were yellowish white spots scattered on parenchymal organs and swelling of liver and spleen. In addition, petechiation and ecchymosis of small intestine, egg yolk and subcutaneous tissues were observed. 4. Histological findings were infiltration of mononuclear cells in liver, heart and lung, appearance of megaloschizonts in heart, liver, proventriculus and pancreas, proliferation of foreign body giant cells in heart, and hyperemia and swelling of all parenchymal organs.
Streptomyces (S.) fradiae is a microbe with broad-spectrum antimicrobial activity, isolated from soil. In the present study, antibacterial effects of S. fradiaea against Salmonella (S.) gallinarum was determined. S. fradiae inhibited growing of S. gallinarum in Luria-Bertani media agar. Moreover, ingestion of S. fradiae markedly inhibited mortality of chickens experimentally infected with S. gallinarum. There is no side effect by S. fradiaeon, in safety of chickens and antibiotic material residues in chicken meat. Taken together, S. fradiae have the antibacterial effects against S. gallinarum. Therefore, we concluded that S. fradiae might be a good microbial candidate for treatment or control of fowl typhoid in chickens.
