왕쥐똥나무잎(Ligustrum ovalifolium H.) 추출물의 세포독성을 살펴보기 위하여 RAW264.7 대식세포를 이용하여 세포의 생존율을 살펴본 결과 물 추출물 및 에탄올 추출물 모두 0.2 mg/mL의 농도까지 전혀 독성을 나타내지 않았다. 또한 왕쥐똥나무잎 추출물의 항염증 효과를 LPS에 의해 활성화된 RAW264.7 대식세포에서의 NO 생성억제 및 ROS 소거능과 염증관련 단백질 발현의 변화를 통하여 확인하였다. RAW264.7 대식세포에 LPS를 처리한 결과 NO의 함량이 11 ${\mu}M$ 수준으로 증가하였으나, 왕쥐똥나무잎 에탄올 추출물(0.05, 0.1, 0.2 mg/mL)을 처리하였을 때 NO의 함량이 7.03, 6.74, 6.64 ${\mu}M$로 농도 의존적으로 감소하였다. 왕쥐똥 나무잎 추출물이 LPS를 처리하여 생성되는 활성산소종에 미치는 영향을 확인한 결과, LPS를 처리한 대조군은 ROS가 36.55%로 증가하였으나, 왕쥐똥나무잎 에탄올 추출물(0.05, 0.1, 0.2 mg/mL)을 처리한 군은 세포내 활성산소종을 농도 의존적(23.86, 8.55, 5.48%)으로 감소시켰다. 또한 왕쥐똥나무잎 에탄올 추출물은 NO 생성과 연관 있는 iNOS 단백질의 발현을 농도 의존적으로 저해하였으며 이는 NO 생성 억제가 iNOS의 발현저해를 경유한 것으로 사료된다. 또한 다수의 항염증 약물들의 작용기전이 되는 COX-2의 생성억제를 살펴본 결과 왕쥐똥나무잎 에탄올 추출물은 LPS에 의해 발현되는 COX-2 단백질의 발현을 유의성 있게 억제하였음을 확인할 수 있었다. 이상의 결과를 요약하면 왕쥐똥나무잎 추출물이 LPS로 유도된 RAW264.7 대식세포내 활성산소종(ROS)과 산화질소 라디칼(NO)을 억제함으로써 염증을 억제하는 것으로 보이며, 이는 선행연구에서 나타난 왕쥐똥나무잎 추출물의 높은 라디칼 소거능 및 항산화능과 관련이 있는 것으로 판단된다. 또한 염증과 관련된 iNOS, COX-2 발현을 저해함으로써 왕쥐똥나무잎 추출물이 염증억제 효과를 나타내는 것으로 사료된다. 따라서 본 연구는 항염증 물질의 연구에 기초 자료로 활용이 가능할 것으로 기대된다. 또한 염증과 관련된 cytokine 및 단백질 발현 메커니즘에 대한 추가적인 연구가 필요할 것으로 판단된다.
본 연구에서는 염생식물을 이용한 생리활성 소재 개발 연구의 일환으로 가는갯능쟁이의 용매 추출물 및 분획물을 제조하여 LPS로 염증을 유도한 마우스 유래 macrophages에서의 NO 생성 및 염증관련인자의 발현에 미치는 영향을 검토하여 가는갯능쟁이의 항염증 소재로서의 이용 가능성을 검토하였다. 가는갯능쟁이는 methylene chloride와 methanol을 이용하여 제조한 조추출물(crude extract, 48.3 mg)을 용매극성에 따라 단계적으로 분획하여 n-hexane 층(11.5 mg), 85% aqueous methanol 수용액층(85% aq. MeOH, 11.3 mg), n-butanol 층(n-BuOH, 11.2 mg)과 water 층(12.3 mg)을 얻었다. 가는갯능쟁이의 조추출물 및 분획물의 RAW264.7 대식세포에 대한 독성 여부를 측정하여 세포에 대한 독성이 나타나지 않는 처리 농도에서 추출물 및 분획물의 항염증 활성을 확인하였다. 조추출물은 LPS로 유도된 NO 생성을 유의적으로 억제하는 효과를 나타내었으며, 주요 염증 유발인자인 iNOS와 COX-2의 발현 또한 효과적으로 억제하였다. 분획물의 NO 생성 억제효과는 85% aq. MeOH> n-BuOH >n-hexane > $H_2O$의 순으로 나타났으며, 세포내 전사수준에서 염증관련 유전자의 발현 억제율은 $H_2O$ 분획 처리군에 비해 85% aq. MeOH, n-BuOH 및 n-hexane 분획 처리군에서 높게 나타났다. 이상의 결과로부터 항염증 활성이 가장 높게 나타난 85% aq. MeOH 분획층으로 부터 염증억제 효과가 있는 유효 성분의 분리가 기대된다.
본 연구는 제주에 자생하고 있는 제주아그배의 생리활성, 즉 항산화 효과와 항염 효과를 알아보고자 수행하였다. 제주아그배의 항산화 활성을 측정하고자 비교 분석한 결과, 제주아그배 에탄을 추출물 (DPPH 소거활성 $IC_{50}$ 값, $16.1\;{\mu}g$/mL; NO 소거활성 $IC_{50}$ 값 $901.2\;{\mu}g$/mL; superoxide 소거활성 및 Xanthine oxidase 억제 활성 $IC_{50}$ 값 $42.5\;{\mu}g$/mL, $2.7\;{\mu}g$/mL)은 좋은 항산화 활성을 보였고, 그 분획물 중에서는 에틸아세테이트 분획물이 항산화 활성 실험 모두에서 높은 소거활성을 나타내었다. 따라서 항산화 활성이 좋은 제주아그배의 추출물의 항염증 효과를 탐색하기 위하여 대식 세포 모델을 이용하였다. LPS로 자극한 마우스 대식세포인 RAW 264.7 세포에서 NO 활성과 세포독성간의 상대적 비교지수인 선택지수를 이용하여 탐색한 결과 에탄을 추출물과 에틸아세테이트 분획물이 2.9와 12.2로 가장 높은 상대적 NO 생성 억제 활성을 보였다. 그리고 NO 생성 억제율에 대한 $IC_{50}$ 값은 각각 $16.7\;{\mu}g$/mL, $4.1\;{\mu}g$/mL으로 나타났다. 이런 결과를 기초로 하여 제주아그배의 에탄올 추출물의 NO 감소가 iNOS 단백질 감소에 의한 것임을 확인한 결과, 시료를 처리한 RAW 264.7 세포에서 iNOS 단백질이 농도 의존적으로 감소하였다. 또한 iNOS 단백질의 억제가 mRNA의 감소로 인한 것인지 조사하고자, RT-PCR을 통해 iNOS의 mRNA 발현을 조사한 결과, 단백질과 마찬가지로 농도 의존적인 mRNA 발현 감소를 볼 수 있었다. 제주아그배의 에탄을 추출물을 농도 별로 처리하였을 때 유의적으로 COX-2 단백질과 mRNA 발현이 농도 의존적으로 억제하였다. 분획물 중에서는 NO 활성과 세포 독성간의 상대적 비교지수인 선택지수가 높았던 에틸아세테이트 분획물이 에탄을 추출물보다도 NO, iNOS, COX-2의 억제 정도가 더 좋은 경향을 나타내었다.
본 연구의 목적은 블루베리 잎의 용매별 추출물에 따른 약리활성에 대한 검증 및 효능 평가이다. 블루베리 잎의 항산화, 항염증에 대한 효과를 확인하였다. 염증 반응은 자극이 가해지면 histamine, serotonin, prostaglandin과 같은 혈관 활성물질에 의해 혈관 투과성이 증대되어 염증을 유발하고 cytokine, free radical, lysosomal enzyme 등 다양한 매개 인자가 관여한다. 자극에 의한 macrophage cell의 염증반응은 tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), interleukin-6(IL-6), interleukin-$1{\beta}$(IL-$1{\beta}$)와 같은 pro-inflammatory cytokine의 발현이 유도되고, inducible nitric oxide synthase(i-NOS)와 cyclooxygenase-2(COX-2)에 영향을 받는 유전자의 발현을 자극하게 되어 nitric oxide(NO) 및 $PGE_2$등의 염증 인자가 생성된다. 이에 따라 블루베리 잎 추출물의 항염증에 대한 연구를 위해 이에 영향을 주는 인자인 i-NOS, COX-2의 단백질 발현억제 작용을 확인 하였다. 그 결과 BLA > BLE > BLW 순서로 높은 효능을 확인 할 수 있었다. 가장 효과가 좋은 BLA 처리군에서 $PGE_2$ 분비량 및 다양한 염증성 인자의 mRNA 발현량을 확인하였다. 측정 결과, BLA($100{\mu}g/mL$)는 $PGE_2$ 분비량을 85.3% 억제하였으며 i-NOS, COX-2, IL-$1{\beta}$, IL-6, TNF-${\alpha}$ 단백질과 mRNA 발현이 각각 86.8%, 85.7%, 62.7%, 77%, 32.2% 억제되는 효과를 확인할 수 있었다. 결론적으로 블루베리 70% 아세톤 추출물(BLA)의 항염증 효과가 가장 높았으며, 블루베리 잎은 세포의 mRNA 및 단백질 수준에서 염증인자들의 억제를 통해 대식세포에서 항염증 효과가 명백히 확인되었다. 더 나아가, nuclear factor kappa B(NF-${\kappa}B$), signal transducer and activator of transcription(STAT-1), mitogen-activated protein kinases(MAPKs) 등의 세포 내 염증관련 중간기전을 연구해볼 가치가 있다고 사료된다.
본 연구에서는 탈지된 갈색거저리 유충으로부터 효율적인 단백질 추출법 확립을 위한 최적 pH 조건을 조사하였다. 단백질 추출 pH 조건은 7부터 12까지 사용하였으며, 단백질 침전 pH는 2, 4, 그리고 6을 사용하였다. 이러한 조건에서 추출한 단백질의 함량, 갈변도 및 회수량을 분석한 결과 갈색거저리 유충 단백질의 최적 추출 조건은 단백질의 갈변도를 최소화 할 수 있으며 32.4%의 회수율을 나타내는 용출 pH 9.0와 침전 pH 4.0로 결정하였다. 다음으로 알칼리 추출법으로 추출한 갈색거저리 유충 단백질의 항염증 효능을 검토하였다. 갈색거저리 유충 단백질이 LPS에 의해 활성화 되는 대식세포의 염증반응 개선효과를 확인하기 위하여 염증매개 인자인 nitric oxide(NO)의 생성 및 iNOS와 COX-2 단백질의 발현량을 측정하였다. 그 결과 갈색거저리 유충 단백질은 세포독성 없이 NO의 생성을 억제 하였으며, MAPK 와 $NF-{\kappa}B$ 신호전달 억제를 통하여 iNOS와 COX-2 단백질의 발현량을 감소시켰다. 이러한 연구결과는 갈색거저리 유충 단백질이 항염증 치료제 및 단백질 보충제 개발을 위한 잠재적 가능성을 가지고 있는 것으로 사료된다.
최근 국내 기후 변화로 인해 열대 과일 재배가 가능해지게 되었다. 특히 대표적으로 제주도에서 애플망고의 재배가 활발히 이루어지고 있으며 이로 인해 소비자들의 수요가 증가하고 있다. 그러나 애플망고 과육을 제외한 껍질과 같은 부산물은 활용도가 떨어져 폐기되고 있는 실정이다. 이에 본 연구에서는 부산물로서 폐기되어지는 애플망고 껍질을 활용하기 위해 이를 이용하여 항산화 및 항염증 효과를 검증하고자 열수 및 70% 에탄올로 추출하여 실험을 진행하였다. 먼저 애플망고 껍질의 열수 및 70% 에탄올 추출물의 총 폴리페놀 함량을 확인하였으며, 그 결과 66.08 ± 0.12 mg TAE/100 g, 100.13 ± 0.23 mg TAE/100 g의 함량을 나타내었다. 항산화 활성을 측정하기 위해 전자공여능과 2,2'- azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) 라디칼 소거능을 측정하였다. 전자공여능 측정결과, 1,000 ㎍/ml 농도에서 열수 추출물은 86.89%가 나타났고, 70% 에탄올 추출물은 94.06%를 확인하였다. ABTS 라디칼 소거능을 측정 결과로 1,000 ㎍/ml 농도에서 열수 추출물은 80.62% 를 나타내었고, 70% 에탄올 추출물은 98.66%의 소거능을 확인하였다. 이로 인해 애플망고 껍질 추출물의 항산화 활성은 대조군인 비타민 C와 유사한 효과를 가진 것을 확인하였다. 애플망고 껍질 추출물의 세포 생존율을 측정한 결과, 대식세포인 RAW 264.7 세포에서는 500 ㎍/ml 농도에서 99% 이상의 세포 생존율을 확인하였다. 이후 세포실험은 세포생존율이 99% 이상인 농도구간을 설정하여 진행하였다. 염증성 매개 물질로 알려진 nitric oxide (NO)에 대한 생성 억제 효과를 확인한 결과, 애플망고 껍질의 열수 추출물은 46%의 억제 효과가 나타났고, 70% 에탄올 추출물은 40%의 억제 효과를 확인하여 애플망고 추출물의 NO 생성량 저해 효과를 가진 것을 확인하였다. 염증 매개 인자의 억제 효과를 확인하기 위해 RAW 264.7 세포를 이용하여 iNOS와 COX-2의 단백질 및 mRNA 발현량을 측정하였다. 그 결과, LPS 단독 처리군에 비해 애플망고 껍질 추출물을 처리하였을 때 iNOS와 COX-2의 단백질 발현량은 최종 농도인 500 ㎍/ml에서 열수 추출물은 12%, 97% 억제하는 것을 확인하였고, 70% 에탄올 추출물은 62%, 91% 억제하는 것을 확인하였다. 또한 LPS 단독 처리군에 비해 iNOS와 COX-2의 mRNA 발현량 결과, 500 ㎍/ml 농도에서 열수 추출물은 3%, 33%의 억제효과를 확인하였고, 70% 에탄올 추출물은 29%, 36%의 억제효과가 나타나는 것을 확인하였다. 본 연구 결과들을 토대로 애플망고 껍질이 항산화 및 항염증 활성을 갖고 있는 것을 검증하여 천연 소재로서의 활용 가치가 높을 것으로 사료된다.
The comparative immunomodulatory effects of ${\beta}$-glucans isolated from mushroom fungi (Coriolus versicol), yeast (Saccharomyces cerevisiae) and bacteria (Agrobacterium) on the major functions of macrophages were evaluated. As parameters of macrophage functions, we examined tumoricidal activity, phagocytosis, nitric oxide (NO) production, and the induction of inducible NO synthetase (iNOS) in RAW264.7 cells, following treatments with ${\beta}$-glucans from the three different sources. The results indicated that all ${\beta}$-glucan treatments significantly induced tumoricidal activity in the RAW264.7 cells, with a remarkable effect shown by the beta-glucan from Agrobacterium at a concentration of $10{\mu}g/mL$. There was also a significant increase in iNOS-NO system activity in macrophages treated with ${\beta}$-glucans extracted from yeast; however, iNOS-NO system activity was not markedly changed by the treatment of ${\beta}$-glucans from C. versicolor mushroom fungi or Agrobacterium. Furthermore, the ${\beta}$-glucans from C. versicolor had a significant phagocytotic effect at concentrations of 1, 10, and $100{\mu}g/mL$. Taken together, the present data suggest that these ${\beta}$-glucans, isolated from three different sources, have different effects on macrophage function, and therefore, may have different clinical uses in different for various types of diseases.
To establish the anti-inflammatory activity of the total flavonoid fraction of the root barks of Broussonetia papyrifera (EBP) and a new formula, the ethanol extract of the root barks of B. papyrifera was fractionated with ethylacetate, yielding the hydrophobic prenylated flavonoid-enriched fraction. EBP and the ethanol extract of the whole Lonicera japonica (ELJ) plant were then mixed at a ratio of 1:1 (w/w) to give a new preparation (BL) in the hope of obtaining an optimal formula with a higher anti-inflammatory activity. Evaluation of the effects of these preparations on A23187-treated rat basophilic leukemia (RBL-1) cells revealed that EBP potently inhibited 5-lipoxygenase (5-LOX), while ELJ showed weak inhibition. Additionally, the mixture (BL) clearly showed stronger inhibitory effects against 5-LOX than either preparation alone. These preparations also inhibited cyclooxygenase-2-catalyzed $PGE_2$ and inducible nitric oxide (NO) synthase-catalyzed NO production by lipopolysaccharide-treated RAW 264.7 cells. When tested against arachidonic acid-induced mouse ear edema, EBP showed strong inhibitory activity at doses of 5-200 mg/kg when administered orally, but BL had obviously stronger inhibitory effects. When tested against ${\lambda}$-carrageenan-induced paw edema in mice, BL showed a potent and synergistic anti-inflammatory effect. In addition, in the acetic acid-induced writhing test, BL was found to have strong analgesic activity at 50-400 mg/kg. Taken together, these results indicate that each of these preparations exert anti-inflammatory activity in vitro and in vivo. In particular, BL showed stronger anti-inflammatory activity than EBP, and these anti-inflammatory effects were partially related to the inhibition of eicosanoid and NO production. BL may be useful for the treatment of human inflammatory disorders.
BACKGROUND/OBJECTIVES: Alzheimer's disease is common age-related neurodegenerative condition characterized by amyloid beta (Aβ) accumulation that leads cognitive impairment. In the present study, we investigated the protective effect of paeoniflorin (PF) against Aβ-induced neuroinflammation and the underlying mechanism in C6 glial cells. MATERIALS/METHODS: C6 glial cells were treated with PF and Aβ25-35, and cell viability, nitric oxide (NO) production, and pro-inflammatory cytokine release were measured. Furthermore, the mechanism underlying the effect of PF on inflammatory responses and Aβ degradation was determined by Western blot. RESULTS: Aβ25-35 significantly reduced cell viability, but this reduction was prevented by the pretreatment with PF. In addition, PF significantly inhibited Aβ25-35-induced NO production in C6 glial cells. The secretion of interleukin (IL)-6, IL-1β, and tumor necrosis factor-alpha was also significantly reduced by PF. Further mechanistic studies indicated that PF suppressed the production of these pro-inflammatory cytokines by regulating the nuclear factor-kappa B (NF-κB) pathway. The protein levels of inducible NO synthase and cyclooxygenase-2 were downregulated and phosphorylation of NF-κB was blocked by PF. However, PF elevated the protein expression of inhibitor kappa B-alpha and those of Aβ degrading enzymes, insulin degrading enzyme and neprilysin. CONCLUSIONS: These findings indicate that PF exerts protective effects against Aβ-mediated neuroinflammation by inhibiting NF-κB signaling, and these effects were associated with the enhanced activity of Aβ degradation enzymes.
Objectives : Cheongnoimyungshin-hwan (CNMSH) is a Herbal compound prescription that is composed mainly of herbal medicines such as Ginseng Radix Alba, Angelicae Gigantis Radix, Dioscoreae Rhizoma, Longan Arillus and cornus cervi parvum, and for the purpose of improving memory and preventing dementia. Methods : In this study, it was investigated whether CNMSH could suppress inflammatory response and oxidative stress in the lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. As a result, CNMSH decreased expression of inducible nitric oxide (NO) synthase and cyclooxygenase-2, and also inhibited production of NO, prostaglandin E2. Results : This effect was associated with the suppression of the expression of p65, one of the nuclear factor-kappaB ($NF-{\kappa}B$) subunits, and increased expression of $I{\kappa}B-{\alpha}$, inhibit the $NF-{\kappa}B$ transcription factor. In addition, CNMSH significantly blocked intracellular reactive oxygen species accumulation in response to LPS stimulation. Furthermore, CNMSH increased expression of nuclear factor erythroid 2-related factor (Nrf)-2 activation and heme oxygenase (HO)-1. Conclusions : Therefore, it has been shown anti-inflammatory and antioxidant effects by inhibiting the expression and production of inflammatory mediators in LPS-stimulated macrophages, and is associated with ROS generation and is activated by Nrf2/HO-1 signaling pathway.

③ 당 사이트는 회원으로부터 제기되는 의견이나 불만이 정당하다고 객관적으로 인정될 경우에는 적절한 절차를

제 11 조 (회원의 의무)
① 이용자는 회원가입 신청 또는 회원정보 변경 시 실명으로 모든 사항을 사실에 근거하여 작성하여야 하며,
허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.
② 당 사이트가 관계법령 및 개인정보 보호정책에 의거하여 그 책임을 지는 경우를 제외하고 회원에게 부여된
ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
