Embryonic Development of Siberian Sturgeon Acipenser baerii under Hatchery Conditions: An Image Guide with Embryological Descriptions

  • Normal embryonic development at a constant temperature ($18^{\circ}C$) has been described for the Siberian sturgeon Acipenser baerii (Acipenseriformes). Hormone-induced spawning and artificial insemination were performed to prepare embryonic batches for embryologic examination. After insemination, early cleavages of the Siberian sturgeon embryos continued for 7 h post-fertilization (HPF), showing the typical pattern of uneven holoblastic cleavage. Blastulation and gastrulation began at 9 HPF and 19 HPF, respectively. Epiboly formation (2/3 covered) was observed at 25 HPF during gastrulation. Neurulation was initiated with the formation of a slit-like neural groove from the blastopore at 33 HPF. During neurulation, the primary embryonic kidney (pronephros) and s-shaped heart developed. The embryos underwent progressive differentiation, which is typical of Acipenseriform species. A mass hatching was observed at 130 HPF, and the average total length of the hatched prolarvae was 10.5 mm. The hatched prolarvae possessed a typical pigment plug (yolk plug). The results of this study are valuable not only as a reference guide for the artificial propagation of Siberian sturgeon in hatcheries but also as the basis for the derivation of developmental gene expression assays for this species.

  • Gonad maturation of Manila clam, Ruditapes philippinarum was induced in this study using a recirculation system over 8 weeks in early spring. Clams used in the experiment were collected in $15^{th}$ April 2010 from the west coast of Korea, as the surface water temperature remained $11^{\circ}C$. To induce gametogenesis and subsequent maturation seawater temperature was elevated $1^{\circ}C$ per day over 10 days to reach $20^{\circ}C$. For the experiment, clams were raised in 120 L quadrangle tank maintained with re-circulated seawater system over 57 days. Water quality parameters including the water temperature, salinity dissolved oxygen, ammonium ion and nitrate levels in the tanks were monitored daily. Mixture of concentrated microalgae including Tetraselmis, Isochrysis, Pavlova and Thalassiosira weissflogii was supplied to clams twice a day, and quantity of the daily ration was adjusted as 3% of clam body dry weight. Histology was applied to examine gonad maturation. Daily monitoring of the water quality parameters indicated that the recirculation system supplied suitable environment to Manila clam; the nitrogenous components stayed below toxic levels (< 0.2 mg/L). At the beginning of the study, clams were mostly in early developing stage. As the seawater temperature reached $20^{\circ}C$, 10 days after the experiment, 20% of clams reached late development at 12 days. First ripe clams were observed at 42 days and 40% of clams were in ripe and ready for spawning at the end of study, 57 days after the experiment. In this study, gametogenesis of Manila clam was successfully induced by elevating water temperature and supplying commercially produced microalgae in a recirculation tank system.

  • 인간의 태반성 성선자극호르몬(HCG) 또는 성선자극호르몬-방출호르몬 유도체(GnRH-A)와 pimozide를 이용하여 산란시기의 암컷황복에서 인공 산란을 유도하였다. 호르몬 또는 호르몬유도체의 효과는 배란 후 인공수정시켜 수정율과 배체형성율 및 면역방사측정법으로 혈액의 plasma 내 성선자극호르몬(GTH) 양을 측정하여 결정하였다. 수정율과 배체형성율이 가장 좋은 HCG 농도는 어체중 kg 당 1,000 IU이었다. 또한 GnRH-A ($10\;{\mu}g/kg$)와 pimozide를 동시에 처리했을 때 수정율과 배체형성율이 좋은 pimozide의 농도는 5 mg/kg이었다. Pimozide(1, 5 mg/kg)를 단독으로 처리한 어류에서는 plasma 내 GTH양에 영향을 줄 수 없었지만, GnRH-A와 동시에 처리했을 때 GTH양은 현저히 증가되었다. GnRH-A와 pimozide 및 dopamine을 동시에 처리했을 때 혈액의 plasma 내 GTH양은 크게 감소되었다. 이상의 결과는 산란시기 또는 이시기이외에 황복의 뇌로부터 여러 가지 성선자극호르몬과 성선자극억제호르몬을 분비됨을 시사해 준다.

  • Perhaps the most common type of reproductive dysfunction in captive fish is failure of females to undergo final oocyte maturation and thus to ovulate and spawn. The success of aquaculture could therefore be improved by developing techniques to enhance natural spawning, artificial maturation, and/or to induce ovulation in farmed fish. This study aimed to investigate the effects of prostaglandin $E_2$ ($PGE_2$) and prostaglandin $F_{2{\alpha}}$ ($PGF_{2{\alpha}}$) on in vitro oocyte maturation (germinal vesicle breakdown, GVBD) and ovulation in the marine fish Chasmichthys dolichognathus. Post-vitellogenic follicles (0.80-0.94 mm diameter oocytes) were incubated with $PGE_2$ or $PGF_{2{\alpha}}$ at concentrations of 5, 50, or 500 ng/mL for 24 hours. A significant increase in GVBD was seen in 0.84 mm and 0.94 mm oocytes incubated with 50 ng/mL $PGE_2$ compared with the control. There was no significant increase in GVBD in any of the other experimental conditions (5 or 500 ng/mL $PGE_2$ or 5, 50, or 500 ng/mL $PGF_{2{\alpha}}$). Neither of the prostaglandins induced ovulation at the concentrations tested.These results suggest that GVBD was induced by incubation with 50 ng/mL $PGE_2$.

인위적 성숙 유도된 뱀장어 Anguilla japonica의 자연산란에 의한 난발생과 Pre-leptocephalus 자어의 발달 (Development of the Eggs and Pre-Leptocephalus Larvae by Natural Spawning of Artificially-Matured Japanese Eel, Anguilla japonica)

  • 극동산 뱀장어의 자연 수정란을 이용하여 $23{\pm}0.5^{\circ}C$의 여과 해수가 공급되는 실내수조에서 난발생과 pre-leptocephalus 자어발달을 관찰하였다. 친어 관리에 있어서 암컷은 연어 뇌하수체 추출물(20 mg/fish)과 수컷은 HCG (1 IU/g BW)를 매주 복강 주사하여 인위적인 성성숙 및 자연산란을 유도하였다. 수정란은 직경 1.0 mm 정도의 분리 부성란으로, 전형적인 반상분할(discoidal cleavage)을 통해 분할이 이루어졌다. 부화는 $23^{\circ}C$의 여과 해수에서 수정 후 38시간부터 개시되었다. 부화 직후의 자어는 전장 약 3.0 mm, 근절수는 평균 42개였다. 부화자어의 입과 항문은 부화 후 4.5일째에 열렸고, 난황은 부화 후 6.5 일째에 거의 흡수되었다. 자어는 14.5일 동안 생존하였고, 이때 전장 $5.87{\pm}0.25mm$, 근절은 98개였다. 그러나 수정란과 prelaptocephalus 자어발생의 형태학적 특징은 자연수정과 건식법에 의한 인공수정과 큰 차이점은 없었다.

  • 좀수수치 Kichulchoia brevifasciata의 산란기 특징 및 초기생활사 연구는 생물학적 특성 및 복원학적 기초자료를 확보하기 위하여 실시하였다. 산란기인 2011년 6월부터 7월까지 전라북도 고훙군 금산면에서 족대를 이용하여 채집한 후 산란기 특징을 분석하였고, 초기생활사는 성숙한 개체에 Ovaprim을 0.5 mL/kg 주사하여 성숙란을 얻은 후 건식법으로 인공수정 시켜 진행하였다. 성숙한 암컷은 전장은 46~76mm이고 GSI는 $9.6{\pm}3.77%$였고, 수컷은 전장 42~52mm이고 GSI는 $3.5{\pm}1.04%$였다. 성비는 0.1로 수컷이 매우 적었으며, 특별한 이차성징은 관찰되지 않았다. 포란수는 $69{\pm}35.3$개였으며, 산란된 성숙란은 담황색의 분리침성란으로 난경 $1.46{\pm}0.07mm$였다. 수정난은 수온 $25^{\circ}C$에서 66시간 후에 부화하였으며, 부화 직후 전기자어의 크기는 전장 $5.5{\pm}0.07mm$였다. 부화 후 6일째에는 전장 $9.0{\pm}0.29mm$로 난황을 모두 흡수하여 후기자어로 이행하였으며, 부화 후 17일째에는 전장 $12.6{\pm}0.24mm$로 모든 지느러미 기조가 정수가 되어 치어기로 이행하였다. 부화 후 80일째에는 전장 $33.0{\pm}2.19mm$로 체형과 체측반문이 비교적 성어와 유사하였다.

말똥성게 (Hemicentrotus pulcherrimus)의 수정 및 배 발생률을 이용한 해양배출 폐기물의 독성평가 (Toxicity Assessment of Ocean Dumping Wastes Using Fertilization and Embryo Development Rates in the Sea Urchin (Hemicentrotus pulcherrimus))

  • Toxicity assessment of ocean dumping wastes (dye waste, urban sewage, food waste) were examined in the fertilization and embryo development rates of the Sea Urchin, Hemicentrotus pulcherrimus. Spawning was induced by injecting 1 mL of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min the collection of both gametes. The fertilization and embryo development rates test were performed for 10 min and 64 h after fertilization, respectively. The fertilization and embryo development rates in the control condition (not including ocean dumping wastes sludge elutriate) were greater than 90%, but suddenly decreased with increasing of ocean dumping waste sludge elutriate concentrations. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste ($EC_{50}$=4.37; $EC_{50}$=1.76), urban sewage ($EC_{50}$=5.79; $EC_{50}$=2.00) and food waste ($EC_{50}$=7.68; $EC_{50}$=2.16), respectively. The NOEC (<3.13) and LOEC (3.13) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest that biological assay using the fertilization and embryo development rates of H. pulcherrimus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

염분과 표준 독성물질(Cu, Cd)이 둥근성게(Strongylocentrotus nudus)의 수정 및 배 발생률에 미치는 영향 (Effects of Salinity and Standard Toxic Metals (Cu, Cd) on Fertilization and Embryo Development Rates in the Sea Urchin (Strongylocentrotus nudus))

  • Effects of salinity and standard toxic metals on fertilization and embryo development rates were investigated in the sea urchin, Strongylocentrotus nudus. Spawning was induced by injecting 1ml of 0.5 M KCl into the coelomic cavity. The fertilization and embryo development rates were below 20% when salinity was 25 psu or lower, but were above 90% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition (not including Cu and Cd) were greater than 90%, but decreased with a high negative correlation (r) of 0.89 and 0.91 with the increasing of Cu and Cd concentrations, respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of S. nudus are between 30 and 35 psu, and the biological assays of fertilization and embryo development rates using S. nudus are useful methods for the ecological toxicity test of marine pollution elements.

호르몬 처리에 의한 범가자미, Verasper variegatus 의 성숙과 배란유도 (Effectof of Hormonal Treatment of Induced Maturation and Ovulation in the Spotted Halibut, Verapser variegatus)

  • 사육수조내 자연산란이 이루어지지 않는 범가자미 암컷을 대상으로 호르몬 처리에 의안 인위적 성숙.배란 유도 실험이 이루어졌다. 산란시기에 범가자미 체중 kg당 HCG 265~678 IU, 17${\alpha}$ 20${\beta}$ OHP 0.5~1.0${\mu}$g 농도의 복강주사와 LHRHa 63~81 ${\mu}$g의 펠렛을 투여한 결과, HCG농도 약 300 IU가 범가자미의 최정성숙과 배란유도에 효과적인 것으로 나타났다. 17${\alpga}$ 20${\beta}$ OHP와 대조군에서는 난의 성숙이 거의 진행되지 않았으며, LHRHa 펠렛 실험군에서도 난의 최종성숙과 배란이 일어나지 않았다. 따라서 범가자미 암컷의 경우 현재 많이 사용되고 있는 LHRHa 펠렛 투여보다도 저농도의 HCG (dir 300 IU/kg 체중)o 주사가 난의 최종성숙과 배란에 더 효과적인 것으로 나타났다.

The fertilization rate was below 30% when salinity was 20 psu and lower, but was almost above 90% when salinity was 25 psu and higher. The embryo development rate was below 60% when salinity was 25 psu and lower, but was above 80% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition(not including Cu and Cd) were greater than 90%, but decreased a high negative correlation with the increasing of Cu(r=-0.80, r=-0.78) and Cd(r=-0.90, r=-0.82) concentrations, respectively. The fertilization and embryo development rates were significantly inhibited in the addition of Cu($EC_{50}$=17.30 ppb, $EC_{50}$=10.32 ppb) and Cd($EC_{50}$=364.57 ppb, $EC_{50}$=244.04 ppb), respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of H. pulcherrimus are above 25 psu and 30 psu, respectively, and the biological assays of fertilization and embryo development rates using H. pulcherrimus are useful methods for the ecological toxicity test of marine pollution elements.
