  • 비료제품에는 의도적 혹은 비의도적으로 농약으로 등록된 indole-3-acetic acid (IAA) 및 indole-3-butyric acid (IBA)성분에 대한 잔류 가능성이 꾸준히 제기되고 있으나, 생장조정제가 아닌 비료제품에서 mg/L 수준의 옥신류 잔류분석법이 마련되어 있지 않았다. 따라서, 본 연구에서는 생체시료에 미량 잔류하는 IAA 및 IBA 분석에 사용되어 온 liquid chromatography-mass/mass spectrometry (LC-MS/MS)와 gas chromatography-MS 기기분석법을 활용하여 농축 액상비료제품에 적용할 수 있는지 조사하였으며, 액상 비료제품에 적용 가능한 정밀기기 분석법을 개발하고자 하였다. 수용액 상태의 시료에서 식물 생장호르몬인 IAA와 IBA를 가장 손쉽게 정제할 수 있는 방법으로 hydrophile-lipophile balance (HLB) solid phase extraction를 활용하였으나, 제한된 조건에서 LC-MS/MS를 통한 정량분석은 적합한 회수율을 확보하지 못하였고, 정성분석만 가능함을 확인하였다. 반면, 비료제품 250배 희석액을 사용하여 HLB 정제, trimethylsilyl chloride을 이용한 methylation을 통한 GC-MS 분석에서는 검출한계 1.4 mg/L과 93-107%의 회수율로 액상 비료제품에서 IAA와 IBA 정량분석법을 확립할 수 있었다.

  • High activity of acidic ethylacetate extract from the culture supernatant of ginseng growth-promoting bacterium Pseudomonas fluorescens KGPP 207 and its fractions were demonstrated through wheat coleoptile bioassay. The following auxins and auxin-like compounds were identified in these fractions by combined gas chromatography-mass spectrometry: indole-3-acetic acid, indole-3-acetic acid methyl and ethyl ester, indole-3-butyric acid, indole-3-lactic acid and its methyl ester, indole-3-propionic acid, indole-3-pyruvic acid, p-hydroxyphenyl acetic acid, p-hydroxyphenyl acetic acid methyl and ethyl ester, phenyl acetic acid and its methyl ester. The bacterium KGPP 207 belongs to the strain of P. fluorescens which produces plant growth regulators and its beneficial effect on the ginseng growth may be due to the formation of the identified compounds.

Plant Hormones Promote Growth in Lichen-Forming Fungi

  • The effect of plant hormones on the growth of lichen-forming fungi (LFF) was evaluated. The use of 2,3,5-triiodobenzoic acid and indole-3-butyric acid resulted in a 99% and 57% increase in dry weight of the lichen-forming fungus Nephromopsis ornata. The results suggest that some plant hormones can be used as inducers or stimulators of LFF growth for large-scale culture.

  • Effects of topical application of 100, 200 and 300ng/$m\ell$ of 3-indole butyric acid (3IBA) on larval parameters were studied in polyvoltine Pure Mysore breed of B. mori. Each concentration was administered independently, at 36h to III, IV & V instars, at 48h to IV & V instars, and at 72h to V instar larvae. Of the various dosages used, the repeated applications of loons of 3-IBA at 36h to III, IV & V instars resulted in a significant increase in larval weight, silk gland weight, cocoon shell weight and fecundity and a significant decrease in larval duration and cocooning percentage when compared with the corresponding parameters of the untreated controls.

  • 간이 녹지삽목시설의 실용성과 발근촉진제 IBA의 발근촉진효과를 조사하기 위하여 북부 highbush blueberry 3품종 'Duke', 'Elizabeth', 'Elliot'를 공시하여 IBA 100ppm, 300ppm, 500ppm, 1,000ppm의 2시간 처리와 3,000ppm 20초 침지처리를 실시한 후 발근특성을 조사하였다. 분산분석에 의한 요인분석 결과, 발근율과 근계지수에 품종효과가 있었으며 발근율, 최대근장, 근계지수에서 IBA의 처리효과도 관찰되었다. 특히 근계지수에서 고도의 유의성 있는 처리효과가 있었다. 그러나 두 요인의 상호작용은 없었다. 무처리구에서 'Duke'의 발근율은 45%, 'Elizabeth'와 'Elliot'는 각각 90%와 85%로 품종간 차이가 있었으며 3품종 모두 고농도 20초 처리와 저농도 2시간 처리구에 의하여 발근이 촉진되었다. 그러나 무처리의 발근율이 낮았던 'Duke'의 발근촉진효과가 다른 두 품종에 비해 현저하였다.

Effect of Ribitol and Plant Hormones on Aposymbiotical Growth of the Lichenforming Fungi of Ramalina farinacea and Ramalina fastigiata

  • This study was aimed at evaluating the growth promoting effect of symbiotic algal polyol (ribitol) and plant hormones on the lichen-forming fungi (LFF), Ramalina farinacea (CH050010 and 40403) and Ramalina fastigiata. The addition of ribitol to basal (malt-yeast extract) medium enhanced the relative growth rates of all three LFF. R. farinacea (CH050010), R. farinacea (40403) and R. fastigiata (H06127) showed 35.3%, 29.0% and 29.3% higher growth rates, respectively, compared to the control. IBA (indole-3-butyric acid) and TIBA (2,3,5-tridobenzoic acid) also increased growth rates of the LFF by 34 to 64% and 7 to 28%, respectively, compared to the control. The combination of ribitol with IBA or TIBA synergistically increased the growth of all LFF. For example, ribitol and IBA treatments increased growth rates of R. farinacea (CH050010), R. farinacea (40403) and R. fastigiata (H06127) by 79.4%, 40.3% and 72.8% in, respectively, compared to those grown on the basal medium. The stimulating effect of ribitol and IBA on the LFF growth induced vertical development of the fungal mass in culture. We suggest that lichen-forming fungal growth of Ramalina lichens can be stimulated aposymbiotically by supplementing polyols and plant hormones to the basal medium in the mass production of lichen secondary metabolites under large scale culture conditions.

자색비유황세균 Rhodopseudomonas faecalis의 식물생장촉진능 (Plant Growth Promotion by Purple Nonsulfur Rhodopseudomonas faecalis Strains)

  • 광합성 자색비유황세균으로 동정된 균주들을 논 퇴적토와 분뇨처리장 슬러지로부터 분리하였고 그들의 식물생장촉진능을 조사하였다. 이 균주들의 배양액에서 대표적인 식물호르몬인 indole-3-acetic acid (IAA)와 indole-3-butyric acid (IBA) 및 5'-aminolevulinic acid (ALA)의 생성을 측정하였다. 분리 균주 중 Rhodopseudomonas faecalis D15가 modified Biebl and Pfennig 배지에서 IAA는 769.8 ${\mu}g$/mg protein, IBA는 1323 ${\mu}g$/mg protein 그리고 ALA는 7.4 mM/mg protein의 가장 높은 생성률을 나타내었으며, R. faecalis C9는 20.8 ${\mu}g$/mg protein의 가장 높은 gibberellin 생성률을 나타내었다. 이 균주들을 토양에 파종한 토마토 종자에 접종하고 15일 후에 자라 난 유묘의 뿌리 길이와 건조중량은 비접종 대조군보다 더 컸으며 특히 C9 균주 처리 시료는 건조중량이 대조군에 비해 119.4% 증가하였다. 이 자색비유황세균 분리균주는 식물생장 촉진을 위한 친환경적 생물비료로 사용할 수 있을 것이다.

Induction of in vitro root tubers in Holostemma annulare (Roxb.) K. Schum. for the production of bioactive metabolites

  • Holostemma annulare (Family Asclepiadaceae) is an invaluable vulnerable medicinal plant; the root tubers are used in Ayurveda medicine and by folk healers to treat various ailments. In this study, Schenk and Hildebrandt medium fortified with the cytokinins 6-benzyl adenine, kinetin, and auxins, including indole 3-butyric acid, indole 3-acetic acid, α-naphthaleneacetic acid, and 2,4-dichlorophenoxyacetic acid, were checked for their efficiency on root tuber induction from different explants. Adventitious root tubers were more successfully induced from in vitro leaf segments and shoots when cultured in Schenk and Hildebrandt medium supplemented with 0.5 mg/l of α-naphthaleneacetic acid. In addition, preliminary phytochemical analysis of in vitro root tubers and identification of different secondary metabolites were conducted. Thin layer chromatography and high performance thin layer chromatography analysis of the crude methanolic extracts of the in vitro root tuber identified the presence of lupeol, a bioactive triterpene. Adventitious root tuber induction offers a novel method for the in vitro production of bioactive metabolites that can be scaled up by bioreactors, thus ensuring the conservation and sustainable utilization of H. annulare. The study warrants further scale-up production and pharmacological investigation that can be extended for pharmaceutical needs.

In vitro propagation of Bambusa nutans Wall. ex Munro through axillary shoot proliferation

  • This communication describes for the first time an efficient and reproducible protocol for large-scale multiplication of Bambusa nutans. Nodal segments collected from field-grown clumps and cultured on Murashige and Skoog (MS) medium supplemented with $4.4{\mu}M$ benzylaminopurine (BA) and $2.32{\mu}M$ kinetin (Kin) gelled with 0.2% gelrite yielded 80% aseptic cultures with 100% bud-break. The in vitro-formed shoots obtained after bud-break were successfully multiplied in MS liquid medium supplemented with $13.2{\mu}M$ BA, $2.32{\mu}M$ Kin, and $0.98{\mu}M$ indole-3-butyric acid (IBA). Sub-culturing of shoots every 3 weeks on fresh multiplication medium yielded a consistent proliferation rate of 3.5-fold. Shoot clusters containing three to five shoots were successfully rooted with 100% success on half-strength MS liquid medium supplemented with $9.8{\mu}M$ IBA, $2.85{\mu}M$ indole-3-acetic acid (IAA), $2.68{\mu}M$ naphthaleneacetic acid (NAA), and 3% sucrose. Plantlets grown in vitro were acclimatized and subsequently transferred to the field. Inter-simple sequence repeat analysis has confirmed the genetic uniformity of the tissue-cultured plants up to 27 passages.
