식물에서도 동물에서와 같이 RGD(Arg-Gly-Asp) sequence를 매개로 원형질막과 extracellular matrix(ECM)가 연결되어 있다는 사실이 알려지고 있으므로 RGD sequence가 식물세포의 생장, 분화 등의 여러 가지 생리적인 현상에 관여하리라는 가능성을 생각할 수 있다. 그래서 RGD sequence가 당근(Daucus carota L.) 배양세포에서 에틸렌 생성에 미치는 영향을 조사하였다. RGD sequence를 포함하는 합성 peptide는 당근 배양세포에서 에틸렌의 생성을 촉진시켰다. RGD peptide는 Indole-3-acetic acid(IAA)와 함께 처리한 경우 에틸렌 생성을 더욱 촉진시켰으며, ACC synthase의 활성도 증가시켰다. RGD sequence가 에틸렌 생성을 촉진시킬 때 어떠한 특이성이 있는가를 확인하기 위해 이와 유사한 RGE(Arg-Gly-Glu) peptide, RGK(Arg-Gly-Lys) peptide를 각각 처리하여 보았다. RGE peptide는 RGD peptide와 마찬가지로 에틸렌 생성을 촉진시켰으나 RGK peptide는 에틸렌 생성에 영향을 미치지 않았다. 따라서 RGD peptide의 에틸렌 생성 촉진작용은 RGD dequence를 포함하는 RGD peptide가 ACC synthase의 활성을 증가시켜서 일어나는 현상이라 생각되며, 또한 특정한 아미노산 서열을 요구하는 특이적 반응이라고 생각된다.
경작지 근권토양에서 식물생장촉진물질인 auxin라 항진균물질을 동시에 생산하는 농업용 미생물제제로 사용할 PGPR 균주를 선발하고 동정하였다. 경북 경산 일대의 다수확 밭토양의 근권토양에서 균주를 선발하고 그 중 auxin 생산성이 높은 2균주 K36, N21를 분리하였다. 이들 균주를 동정한 결과 K36은 Pantoea agglomerans이고 N21은 Pseudomonans fulva로 동정되었다. 선발된 균주의 식물생장촉진능을 in vivo 녹두발아촉진검사법으로 확인하였으며, 식물병원진균에 대한 길항력을 확인하였다. 배양조건에 따른 auxin 생산성을 조사한 결과 K36 균주는 $pH\;7.5,\;35^{\circ}C$, 37시간에서 최대로 나타났고, N21균주는 $pH\;7.5,\;35^{\circ}C$, 20시간에서 최대값을 나타내었다.
뽕나무의 화아의 기내배양을 실시할 경우 배지종류 및 생장조절물질의 농도가 각 기관의 생장 및 분화에 미치는 영향을 조사한 결과를 요약하면 다음과 같다. 1. Murashige and Skoog 배지는 Greshoff and Doy 및 Wolter and Skoog 배지에 비하여 자웅화아의 생장에 양호한 결과를 나타내었으며 자화의 성장은 2.0ppm BAP 농도에서 양호하였다. 2. 자화 및 웅화의 화아를 가지고 있는 삽수를 7일간 삽목하여 발아시킨 후 배양한 구가 삽목을 하지 않고 직접 치상한 구보다 화아의 발육이 양호하였다. 3. 고농도 Kinetin과 저농도 IAA를 혼용 첨가한 M.S. 배지에서 생장이 양호하여 발화되었고 건전한 주두 및 약을 얻을 수 있었다.
연령을 달리하는 인삼토양 내 근권세균의 다양성과 이들 세균이 갖고 있는 식물생장에 유용한 특성에 대하여 조사하였다. 인삼 근권으로부터 총 15개 속에 포함되는 143 균주를 분리하였다. 인삼 연령에 따른 세균 군집의 변화가 있었으나, 분리 균주의 58%가 Firmicutes에 속하였다. 특히 Bacillus는 연령에 관계없이 가장 우점하는 속으로 나타났다. 이들 분리 균주 중 30개의 균주를 선발하여 세포벽 분해효소 분비, 인돌아세트산 생성, siderophore 생성, 인 가용화, 식물병원성 곰팡이의 저해능 등에 대하여 평가한 결과, 다수의 균주가 식물생장을 촉진할 수 있는 기능적 특성을 갖고 있었다. 이 중에서도 모든 조사 항목에 대하여 양성 반응을 보인 B. subtilis, B. amyloliquefaciens, B. velezensis, B. licheniformis 균주는 향후 인삼 경작을 위한 생장 촉진 미생물제제로 활용가치가 높을 것으로 기대된다.

The aim of this study was to isolate and characterize lead (Pb)-solubilizing bacteria from heavy metal-contaminated mine soils and to evaluate their inoculation effects on the growth and Pb absorption of Brassica juncea. The isolates were also evaluated for their plant growth-promoting characteristics as well as heavy metal and salt tolerance. A total of 171 Pb-tolerant isolates were identified, of which only 15 bacterial strains were able to produce clear haloes in solid medium containing PbO or $PbCO_3$, indicating Pb solubilization. All of these 15 strains were also able to dissolve the Pb minerals in a liquid medium, which was accompanied by significant decreases in pH values of the medium. Based on 16S rRNA gene sequence analysis, the Pb-solubilizing strains belonged to genera Bacillus, Paenibacillus, Brevibacterium, and Staphylococcus. A majority of the Pb-solubilizing strains were able to produce indole acetic acid and siderophores to different extents. Two of the Pb-solubilizing isolates were able to solubilize inorganic phosphate as well. Some of the strains displayed tolerance to different heavy metals and to salt stress and were able to grow in a wide pH range. Inoculation with two selected Pb-solubilizing and plant growth-promoting strains, (i.e., Brevibacterium frigoritolerans YSP40 and Bacillus paralicheniformis YSP151) and their consortium enhanced the growth and Pb uptake of B. juncea plants grown in a metal-contaminated soil. The bacterial strains isolated in this study are promising candidates to develop novel microbe-assisted phytoremediation strategies for metal-contaminated soils.
Bacteria were isolated from roots of sugarcane varieties grown in the fields of Punjab. They were identified by using API20E/NE bacterial identification kits and from sequences of 16S rRNA and amplicons of the cpn60 gene. The majority of bacteria were found to belong to the genera of Enterobacter, Pseudomonas, and Klebsiella, but members of genera Azospirillum, Rhizobium, Rahnella, Delftia, Caulobacter, Pannonibacter, Xanthomonas, and Stenotrophomonas were also found. The community, however, was dominated by members of the Pseudomonadaceae and Enterobacteriaceae, as representatives of these genera were found in samples from every variety and location examined. All isolates were tested for the presence of five enzymes and seven factors known to be associated with plant growth promotion. Ten isolates showed lipase activity and eight were positive for protease activity. Cellulase, chitinase, and pectinase were not detected in any strain. Nine strains showed nitrogen fixing ability (acetylene reduction assay) and 26 were capable of solubilizing phosphate. In the presence of 100 mg/l tryptophan, all strains except one produced indole acetic acid in the growth medium. All isolates were positive for ACC deaminase activity. Six strains produced homoserine lactones and three produced HCN and hexamate type siderophores. One isolate was capable of inhibiting the growth of 24 pathogenic fungal strains of Colletotrichum, Fusarium, Pythium, and Rhizoctonia spp. In tests of their abilities to grow under a range of temperature, pH, and NaCl concentrations, all isolates grew well on plates with 3% NaCl and most of them grew well at 4 to $41^{\circ}C$ and at pH 11.
Cold-adapted plant growth-promoting bacteria (PGPB) with multiple functions are an important resource for microbial fertilizers with low-temperature application. In this study, culturable cold-adapted PGPB strains with nitrogen fixation and phosphorus solubilization abilities were isolated. They were screened from root and rhizosphere of four dominant grass species in nondegraded alpine grasslands of the Qilian Mountains, China. Their other growth-promoting characteristics, including secretion of indole-3-acetic acid (IAA), production of siderophores and ACC deaminase, and antifungal activity, were further studied by qualitative and quantitative methods. In addition, whether the PGPB strains could still exert plant growth-promoting activity at 4℃ was verified. The results showed that 67 isolates could maintain one or more growth-promoting traits at 4℃, and these isolates were defined as cold-adapted PGPB. They were divided into 8 genera by 16S rRNA gene sequencing and phylogenetic analysis, of which Pseudomonas (64.2%) and Serratia (13.4%) were the common dominant genera, and a few specific genera varied among the plant species. A test-tube culture showed that inoculation of Elymus nutans seedlings with cold-adapted PGPB possessing different functional characteristics had a significant growth-promoting effect under controlled low-temperature conditions, including the development of the roots and aboveground parts. Pearson correlation analysis revealed that different growth-promoting characteristics made different contributions to the development of the roots and aboveground parts. These cold-adapted PGPB can be used as excellent strain resources suitable for the near-natural restoration of degraded alpine grasslands or agriculture stock production in cold areas.

Background: Panax ginseng is one of the most important medicinal plants and is usually harvested after 5 to 6 years of cultivation in Korea. Heavy metal (HM) exposure is a type of abiotic stress that can induce oxidative stress and decrease the quality of the ginseng crop. Siderophore-producing rhizobacteria (SPR) may be capable of bioremediating HM contamination. Methods: Several isolates from ginseng rhizosphere were evaluated by in vitro screening of their plant growth-promoting traits and HM resistance. Subsequently, in planta (pot tests) and in vitro (medium tests) were designed to investigate the SPR ability to reduce oxidative stress and enhance HM resistance in P. ginseng inoculated with the SPR candidate. Results: In vitro tests revealed that the siderophore-producing Mesorhizobium panacihumi DCY119T had higher HM resistance than the other tested isolates and was selected as the SPR candidate. In the planta experiments, 2-year-old ginseng seedlings exposed to 25 mL (500 mM) Fe solution had lower biomass and higher reactive oxygen species level than control seedlings. In contrast, seedlings treated with 108 CFU/mL DCY119T for 10 minutes had higher biomass and higher levels of antioxidant genes and nonenzymatic antioxidant chemicals than untreated seedlings. When Fe concentration in the medium was increased, DCY119T can produce siderophores and scavenge reactive oxygen species to reduce Fe toxicity in addition to providing indole-3-acetic acid to promote seedling growth, thereby conferring inoculated ginseng with HM resistance. Conclusions: It was confirmed that SPR DCY119T can potentially be used for bioremediation of HM contamination.

Though information exists regarding the pathogenesis of the shot-hole disease (SH) in flowering cherry (FC), there has been a lack of research focusing on SH management. Therefore, here, we investigated the inhibitory activities of antagonistic bacteria against SH pathogens both in vitro and in vivo as well as their biochemical characteristics and bioactive compounds. Two biosurfactant-producing bacterial antagonists, identified as Bacillus velezensis strains JCK-1618 and JCK-1696, exhibited the best effects against the growth of both bacterial and fungal SH pathogens in vitro through their cell-free culture filtrates (CFCFs). These two strains also strongly inhibited the growth of the pathogens via the action of their antimicrobial diffusible compounds and antimicrobial volatile organic compounds (VOCs). Crude enzymes, solvent extracts, and biosurfactants of the two strains exhibited antimicrobial activities. Liquid chromatography/electrospray ionization time-of-flight mass spectrometric analysis of the partially purified active fractions revealed that the two antagonists produced three cyclic lipopeptides, including iturin A, fengycin A, and surfactin, and a polyketide, oxydifficidin. In a detached leaf assay, pre-treatment and co-treatment of FC leaves with the CFCFs led to a large reduction in the severity of the leaf spots caused by Epicoccum tobaicum and Bukholderia contaminans, respectively. In addition, the two antagonists produced indole-3-acetic acid, siderophore, and a series of hydrolytic enzymes, along with the formation of a substantial biofilm. To our knowledge, this is the first report of the antimicrobial activities of the diffusible compounds and VOCs of B. velezensis against the SH pathogens and their efficiency in the biocontrol of SH.
The effects of gibberilin ($GA_3$) on levels of endogenous indole-3-acetic acid (IAA) and zeatin in both fresh and stratified American ginseng (Panax quinquefolium) seeds were investigated. In our first experiment, the fresh seeds were stratified after soaked in 80 ppd $GA_3$ solution for 24 hours. We found that the IAA concentration in embryo increased by 50.7% and 82.1% respectively at the 120th day and the 188th day of stratification, and the zeatin concentration also increased by 3.8% and 51.6% respectively. In our second experiment, we treated the seeds after 134 days stratification with 80 ppm GA3 for 24 hours and then continued to stratify them. We found that the IAA concentration in embryo increased by 32.9% and 17.7% respectively at the 164th day and the 208th day of stratification while zeatin concentration increased by 22.7% and 30.6% respectively In our another experiment, we studied the effects of $GA_3$, abscislc acid (ABA) and GA, plus ABA on germination rate of seeds treated with these plant hormones during stratification. We found that when the stratified seeds whose ratio of embryo had reached 75% were treated with 80 ppm GA3 for 24 hours and then were allowed to be stratified for another 88 days, the weight and length of embryo (p < 0.05), and germination rate (p < 0.01) increased. In contrast, the 25 ppm ABA treated with for 24 hours was found to Inhibit the growth of embryo (p < 0.05) and reduce the germination rate (p < 0.05) . The experiment of combination treatment of $GA_3$ and ABA showed that $GA_3$ could relieve the inhibitory effects of the ABA on the development of the seeds.
