시설원예(施設園藝) 작물(作物)에서 토착(土着) VA균근균(菌根菌)에 관한 연구(硏究) -III. 토착(土着) VA 균근균(菌根菌) 접종효과 (Studies on the Indigenous Vesicular-Arbuscular Mycorrhizal Fungi(VAMF) in Horticultural Crops Grown Under Greenhouse -III.   • 공시작물(供試作物) 오이, 토마토, 가지, 고추 및 멜론의 육묘단계(育苗段階)를 포함(包含)한 초기생육(初期生育)에 토착(土着) VA균근균(菌根菌)의 접종효과를 구명(究明)하기 위해 수행(遂行)한 실험결과(實驗結果)는 다음과 같다. 건물중(乾物重), 초장(草長), 엽면적(葉面積) 등에서 공시(供試)된 5개 작물(作物) 모두 VA균근균(菌根菌)의 접종효과를 보였는데 오이의 경우 엽면적(葉面積)에서 유의성(有意性)있는 차이(差異)를 보이고, 초장(草長)은 정식(定植) 후인 출아(出芽) 후 6주부터 고도(高度)의 유의성(有意性)이 인정(認定)되었다. 토마토의 경우는 엽면적(葉面積)과 초장(草長)에서 전(全) 조사기간(調査時期)에 토착(土着) VA균근균(菌根菌) 접종처리(接種處理)에서 지상부(地上部) 건물중(乾物重)에 대한 VA균근균(菌根菌) 접종효과는 정식(定植) 후에 유의성(有意性)있는 차이(差異)가 인정(認定)되었다. 멜론의 초장(草長)과 지상부(地上部) 건물중(乾物重)에서 전(全) 조사기간(調査期間)에 유의성(有意性)있는 VA균근균(菌根菌) 접종효과를 보이고 인정(認定)되었고 뿌리 신선중(新鮮重)에서도 출아(出芽) 후 4주부터 고도(高度)의 유의성(有意性)이 인정(認定)되었다. 가지의 경우에 지상부(地上部) 건물중(乾物重)의 경시적(經時的) 조사결과(調査結果) 출아(出芽) 후 4주부터 유의성(有意性)있는 VA균근균(菌根菌) 접종효과가 인정(認定)되었으며, 초장(草長)의 경우도 이식(移植) 후에 유의성(有意性)있는 차이(差異)를 나타냈으며, 엽면적(葉面積)과 지상부(地上部) 신성중(新鮮重)은 출아(出芽)후 8주, 12주 조사시(調査時)에 유의성(有意性)이 인정(認定)되었다. 고추의 엽면적(葉面積)에서 VA균근균(菌根菌) 접종효과가 초기(初期)부터 크게 나타났고 지상부(地上部) 건물중(乾物重) 및 신선중(新鮮重)도 출아(出芽) 후 4주를 제외(除外)한 전(全) 조사기간(調査時期)에 유의성(有意性)있는 차이(差異)를 나타냈다. 공시(供試)된 작물(作物)의 VA균근균(菌根菌) 감염률(感染率)은 생율(生育)의 진전(進展)에 따라 증가(增加)되었으며, 고추를 제외(除外)한 나머지 작물(作物)에서 이식(移植) 후 1주경에는 떨어졌으나 출아(出芽) 후 8주의 조사시(調査時)에는 다시 회복(回復)되어 출아(出芽)후 12주에 50% 내외(內外)의 감염률(感染率)을 보였고, 출아(出芽) 후 12주의 공시작물(供試作物) 근권(根圈)중 포자밀도(胞子密度)는 건토(乾土) 50g당 $72.7{\pm}26.3{\sim}100{\pm}10.3$개의 수준(水準)이었으며 오이와 토마토에서 높은 경향(傾向)이었다. 지상부(地上部) 식물체(植物體)의 무기성분(無機成分) 분석결과(分析結果)에서 전(全) 질소(窒素)는 VA균근균(菌根菌) 접종처리(接種處理)에서 낮은 함량(含量)을 보였고, 인산(燐酸)은 토마토와 고추에서, 가리(加里)는 오이와 가지에서, 석회(石灰)는 토마토, 고토(苦土)는 토마토와 멜론에서 가장 높은 함량(含量)을 나타냈으며, 미량질소(微量窒素)인 철(鐵), 아연(亞鉛), 망간 및 구리 등의 함량(含量)에서도 VA균근균(菌根菌) 접종효과가 인정(認定)되었지만 가지에서는 오히려 낮은 흡수량(吸收量)을 보였다.

Wood Decay Fungi in South Korea: Polypores from Seoul

  • In Seoul, a majority of plant communities have undergone significant changes over the last few decades; however, how wood decay fungi have responded and adapted to the changes in vegetation remains unknown. Through an ongoing investigation of Korean indigenous fungi, ca. 300 specimens with poroid basidiocarp were collected in Seoul during 2008~2012. Morphological examination and molecular analysis using the internal transcribed spacer and nuclear large subunit ribosomal DNA region sequences helped identify 38 species belonging to 28 genera, 10 families, and 5 orders in this area. Among them, three polypores, Abundisporus pubertatis, Coriolopsis strumosa, and Perenniporia maackiae were found to be new to South Korea.

친환경 제제로부터 식물병원균에 대한 길항 미생물의 선발 (Selection of Antagonistic Microorganisms against Plant Pathogens from Eco-friendly Formulations)

  • BACKGROUND: Some microorganisms extant in nature have ability to suppress various plant pathogens, and also can promote plant growth. Thus microorganisms are such great source of antimicrobial agents to develop antagonistic microorganism production and eco-friendly crop management. We isolated the microorganisms in various eco-friendly formulations. The suppressive abilities against plant pathogens have been characterized in vitro level. METHODS AND RESULTS: The indigenous microorganisms have been isolated from Cooked rice, Black sugar, Rice Bran, and Red clay using dilution plating method. Population of bacteria and fungi were above 107 in the all formulations. We isolated and pure cultured the microorganisms based on morphological characteristics. Three major plant pathogens (Fusarium oxysporum, Rhizoctonia solani, Phytophthora capsici) have been used to select antagonistic microorganisms. Total 20 bacteria and 9 fungi showed the pathogen growth suppression ability in vitro condition. The selected microorganisms were identified by ITS sequence similarity. CONCLUSION: All tested eco-friendly formulations contained high-density of the microorganisms. Among the isolated microorganisms, Bacillus spp. and Streptomyces spp. showed the most effective antifungal activity against the plant pathogens such as F. oxysporum, R. solani, and P. capsici. Among the selected fungi Trichoderma sp. demonstrated antifungal activity. Our results suggest that the currently adapted eco-friendly formulations might useful for sustain agricultural system.

생물방제균 Pseduomonas fluorescens 2112의 선발과 고추역병균에 대한 항진균성 길항작용 (Selection and Antifungal Activity of Antagonistic Bacterium Pseudomonas sp. 2112 against Red-Pepper Rotting Phytophthora capsici)

  • In order to select multifunctional powerful antagonistic biocontrol agent against red-pepper rotting fungi Phytophthora capsici, we isolated an indigenous antagonistic bacterium which produces antifungal substances and siderophores from a local soil of Kyongju, Korea. The isolated strain was identified as Pseudomonas fluorescens biotype F. The antibiotic produced from P. fluorescens 2112 inhibited hyphae growth and the zoospore germination of Phytophthora capsici. The favorable carbon, nitrogen source and salts for the production of antibiotic from P. fluorescens 2112 were glycerol, beef extract and LiCi at 1.0%, 0.5% and 5 mM, respectively. And antagonistic activity of P. fluorescens 2112 was confirmed against P. capsici in vivo.

Phylogenetic Status of an Unrecorded Species of Curvularia, C. spicifera, Based on Current Classification System of Curvularia and Bipolaris Group Using Multi Loci

  • A seed-borne fungus, Curvularia sp. EML-KWD01, was isolated from an indigenous wheat seed by standard blotter method. This fungus was characterized based on the morphological characteristics and molecular phylogenetic analysis. Phylogenetic status of the fungus was determined using sequences of three loci: rDNA internal transcribed spacer, large ribosomal subunit, and glyceraldehyde 3-phosphate dehydrogenase gene. Multi loci sequencing analysis revealed that this fungus was Curvularia spicifera within Curvularia group 2 of family Pleosporaceae.

  • Amanita (Agaricales, Basidiomycota) is one of the most well-known genera composed of poisonous mushrooms. This genus of almost 500 species is distributed worldwide. Approximately 240 macrofungi were collected through an ongoing survey of indigenous fungi of Mt. Jeombong in Inje County, Korea in 2014. Among these specimens, 25 were identified as members of Amanita using macroscopic features. Specimens were identified to the species level by microscopic features and molecular sequence analyses of the internal transcribed spacer and large subunit of nuclear ribosomal RNA. We molecularly identified 13 Amanita species, with seven species matching previously recorded species, four species (A. caesareoides, A. griseoturcosa, A. imazekii, and A. sepiacea) new to Korea, and two unknown species.

Three Unrecorded Penicillium Species from Marine Environments in South Korea

  • Penicillium is the most common fungal genus in various terrestrial and marine environments. The number of new and unrecorded Penicillium species from various substrates and habitats are continuously increasing. As part of a project to discover indigenous fungi in South Korea, three unrecorded Penicillium species were isolated from mudflats and seaweeds. Based on morphological characteristics and phylogenetic analyses of β-tubulin and calmodulin loci, they were identified as P. amaliae, P. infrabuccalum, and P. manginii. Here, we provide a detailed morphological description and sequence information of these previously unrecorded species.

Six New Recorded Species of Macrofungi on Gayasan National Park in Korea

  • The fungi play important roles in maintaining the balance and homeostasis of natural ecosystems. We investigated fungal specimens indigenous to the Gayasan National Park located in the deep inlands of southeastern Korea from 2017-2020. Six fungal species in the Korean macromycota-Chiua olivaceoreticulata, Entoloma conchatum, Galerina sulciceps, Hebeloma radicosoides, Spongiporus gloeoporus, and Tricholoma sinoacerbum-were identified based on morphological characteristics and rDNA sequences. The six fungal speices were newly revealed on the Korean Peninsula, and it supports that the continuous investigaton is the best way to realize the mycosis.

Mycorrhization of Quercus acutissima with Tuber borchii and Tuber melanosporum

  • Truffles are ectomycorrhizal fungi that belong to the genus Tuber. They exhibit symbiotic relationships, particularly with oak (Quercus spp.) and hazel (Corylus spp.) trees. We performed an inoculation using a spore suspension to synthesize mycorrhizae between European truffles, Tuber borchii and Tuber melanosporum, and an indigenous oak species, Quercus acutissima. This resulted in the formation of mycorrhizae within 2 months after inoculation. Despite having the same host plant, differences in features were observed between Tuber species, including color and mantle type. These results indicate that Q. acutissima is a suitable host plant for truffle cultivation in Korea and provide a better understanding of the mycorrhization of T. borchii and T. melanosporum.

Newly Recorded Macrofungi from Taebaeksan National Park in Korea

  • Mt. Taebaeksan extends from Gangwon-do Province (Taebaek-si, Youngwon-gu, and Jeongseon-gun) to Gyeongsangbuk-do Province (Bongwha-gun), South Korea. Indigenous fungi present in the park were investigated between 2019 and 2022. All collected specimens were identified to the species level based on their morphological characteristics and molecular phylogenetic analysis using sequences from the internal transcribed spacer (ITS) and large subunit (LSU) of ribosomal DNA. Among them, 17 species-Cyanosporus bifarius, Dacryobolus angiospermarum, Entoloma sericeum, Flammulina rossica, Fuscopostia leucomallella, Homophron helvolescens, Hygrophorus queletii, Hymenochaete huangshanensis, Inocybe albodiscoides, Lactarius fulvihirtipes, Lepiota ignivolvata, Physisporinus eminens, Ramaria gracilis, Russula albolutea, Russula cremicolor, Stropharia lignicola, and Tengioboletus subglutinosus-were newly recorded macromycota in Korea.
