The Distribution of Indicator Microorganisms and Identification of Antibiotic Resistant Strains in Domestic Animal Feces (가축 분변 유래 지표미생물 분포 및 항생제 내성 균주의 동정)

  • Objectives: To estimate the microbial contaminant load discharged from livestock farms, we randomly selected livestock farmers of cattle, swine, and fowl and collected bacterial strains from domestic animals' feces and compost samples. Recently, as multi-antibiotic-resistant bacteria and super bacteria showing resistance to a variety of antibiotics have been reported one after another, the ecological and health hazard of antibiotic-resistant bacteria is emerging as an important issue. Methods: Monitored indicator microorganism constituents were totak coliform (TC), fecal coliform (FC), and aerobic bacteria. The multi-antibiotic-resistant bacteria were identified from investigated indicator microorganisms by 16S rRNA sequencing. Results: By microbiological analysis, the largest population of aerobic bacteria ($1.5{\times}10^5$ CFU/g) was found in cattle fecal compost, and total coliforms ($1.1{\times}10^7$ CFU/g) and fecal coliforms ($1.0{\times}10^5$ CFU/g) were found primarily in swine fecal compost, while the lowest population was found in fowl fecal compost. Among the 67 strains separated from aerobic bacteria, five strains expressing high antibiotic resistance were selected in each sample. We found the multi-antibiotic resistant strains to be Shigella boydii, Staphylococcus lentus, Acinetobacter sp. and Brevibacterium luteolum. Conclusions: These results suggest that increasing numbers of multi-antibiotic-resistant bacteria in the environment have a close relation to the reckless use of antibiotics with livestock.

Lab-on-a-Chip for Monitoring the Quality of Raw Milk

  • A lab-on-a-chip (LoC) was designed for simultaneous monitoring of microorganisms, antibiotic residues, somatic cells, and pH in raw milk. The LoC was fabricated from polydimethylsiloxane (PDMS) using microelectromechanical system (MEMS) technology, which consisted of two parts; a protein array and microchannel. The protein array was fabricated by immobilizing five types of antibodies corresponding to two microorganisms, two antibiotic residues, and somatic cells. A sol-gel film was deposited on a glass substrate to immobilize the antibodies. The target analytes in raw milk could be bound with the corresponding antibody by an immunoreaction, and the antigen-antibody complex was detected using fluorescence microscopy. SNARF-dextran was used as a pH indicator, and the SNARF-entrapped hydrogel was attached to the microchannel in the chip. After injecting the milk sample into the channel, the pH was measured by monitoring the change in fluorescence intensity by fluorescence microscopy. The on-chip simultaneous assay of two microorganisms (E. coli O157:H7 and Streptococcus agalactiae), two antibiotic residues (penicillin G and dihydrostreptomycin), and neutrophils was successfully accomplished using the proposed LoC system.

Photoreactivation Study of Wastewater Treatment Effluent Disinfected by UV-disinfection for Water Reuse (용수재이용을 위한 하수처리 유출수의 UV 소독 후 광회복 조사)

  • Photoreactivation of microorganism following UV-disinfection is one of the research topics of interest in assessing the UV-disinfection performance. Apparent photoreactivation was examined under fluorescent lamp and solar radiation as well as in darkness. Total coliform, fecal coliform, and Escherichia coli were used as indicator microorganisms, and their concentration was monitored with time after UV-disinfection. Under the darkness, their initial concentration of 10∼30 MPN/100 mL increased to the level of 100 MPN/100 mL after 24 hours, which implied that part of damaged microorganisms by UV-disinfection might be repairable with time. Under the fluorescent lamp, photoreactivation was more apparent that their concentration increased up to 1,000 MPN/100 mL which might significantly impair the water uses specially in reuse of reclaimed wastewater. However, their concentration further decreased down to below 2 MPN/100 mL under the solar radiation primarily due to additional disinfection by solar radiation rather than photoreactivation. Samples not disinfected by UV-disinfection also demonstrated substantial decrease of their concentration under solar radiation from about 5,000 MPN/100 mL to less than 30 MPN/100 mL in 24 hours. But direct reuse of effluent without disinfection is not recommended because natural decay by solar radiation may take time and be affected by climatic conditions. The result suggests that photoreactivation of pathogenic microorganisms may not be concerned in agricultural reuse of reclaimed wastewater because solar radiation may provide further disinfection after UV-disinfection.

Water Quality and Correlation Analysis Between Water Quality Parameters in the Hwaong Watershed (화옹호 유입하천의 수질현황 및 수질항목간의 상관관계)

  • Most projects of tideland reclamation with dike construction produce estuarine reservoirs, which may result in water quality problems due to blocking of natural flow of stream water to the sea. External loadings to the reservoirs through tributaries are major concerns in a concerned water quality management. The water quality of a reservoir is greatly influenced by watershed drainage, and accurate estimation of pollutant is indispensable for in the reservoir management. Concentrations of the microorganisms in stream water and conventional parameters were monitored in the 13 water quality monitoring sites located in a rural watershed of Hwaong estuarine reservoir. The indicator of microorganisms showed strong correlation between them, and regression equations with $R^2\geq0.70$ may be used fur estimating one from other microorganisms. The relationships between water quality parameters obtained in this study may be used to infer one unknown pollutant concentrations from the measured pollutant loadings. This methodology could be applied to other areas where the watershed characteristics are not significantly different from the study area. High concentrations of nitrogen was observed in water quality monitoring sites affected by urban land uses and numbers of livestock in wet day as well as dry day, due to the influent of diffuse sources.

Monitoring of norovirus and indicator microorganisms from agricultural products and environmental samples in Korea (한국에서의 농산물 및 환경시료에서 노로바이러스와 위생지표세균의 모니터링)

  • Norovirus causes frequent epidemic viral gastroenteritis in Korea. The team for the control of noroviral foodborne outbreaks (NOROTECL) executed a project to trace the cause of norovirus contamination in agricultural products and environmental samples to reduce norovirus outbreaks in Korea. Between January and November in 2015, the contaminations by norovirus and indicator microorganisms such as coliforms, Escherichia coil and male specific coliphage (MSC) were examined in 80 agricultural products, 80 soil samples, 78 human feces samples, 3 animal feces samples, 80 agricultural water samples and 80 river water samples. Semi-nested PCR and DNA sequencing revealed 18 genogroup I and 3 genogroup II noroviruses in a total of 18 samples. These noroviruses were validated by real-time (RT)-PCR analysis. For indicator microorganisms, coliform and E. coli were respectively detected in agricultural products (68, 1%), soils (88, 7%), human feces (44, 12.8%), animal feces (67, 67%), agricultural waters (74, 30%) and river waters (96, 51%). The MSC results revealed 14 positive samples.

  • A device to measure the temperature difference between the supernatant and the sediment blanket in the course of SV30 measurement in processing of activated sludge process. The temperature elevation in the sludge sediment represent the metabolic heat production by the microorganisms and can be an indicator for the capability of waste treatment. The utilities of the device for the analysis of activated sludge process were demonstrated in this report.

Evaluation of Characteristics of Microorganisms Isolated from Public Drinking Water Facilities in Gwangju City (광주지역 먹는물 공동시설의 미생물 특성 및 분포조사)

  • Objective: This study was performed to detect indicator bacteria in drinking spring water samples in Gwangju City and to identify their genus using the VITEK-II system. Methods: The subjects were ten drinking spring water sites in Gwangju. Samples of spring water were taken every month from September 2019 to August 2020. We analyzed for the indicator bacteria Yersinia and microorganisms isolated from the spring water. Result: According to the research results on indicator bacteria, general bacteria in st1-st7 with sterilization facilities in the spring and summer were investigated in the range of 0-2 CFU/mL and 0-12 CFU/mL. In st9, where a sterilization facility was not installed, the most general bacteria were detected (160 CFU/mL). Total coliform and fecal coliform showed unsatisfied rates of 16.7 and 11.1% in spring and 14.7 and 11.8% in summer, respectively. The unsatisfied rates of total coliform for the designated and non-designated spring water facilities were 3.8 and 47.1%, respectively, and for the fecal coliform group they were 2.5 and 35.3%. The difference was confirmed according to the presence of a sterilization facility. Yersinia spp. was not detected in all drinking spring water. Forty-one strains in 25 species were isolated from ten sites. The results classified as major dominant species are Pseudomonas spp. 14.6%, Pantoea spp. 9.8%, Serratia spp. 9.8%, Acinetobacter spp. 9.8%, Citrobacter spp. 7.3%, Bordetella spp. 7.3%, Delftia spp. 4.9%, and Enterobacter spp. 4.9%. Conclusions: Based on the result that various species derived from fecal pollution and artificial pollutants were detected in the non-specified public spring water facilities that many people use, the facilities need institutional complements such as continuous management or complete shutdowns.

Analysis of Microorganisms and Antibiotic Resistance in Organic Dairy Farm (유기낙농가 사육환경 중 미생물 및 항생제 내성 분석)

  • Recently, antimicrobial resistance of microorganisms has been a major concern of its relation to food safety and national health, therefore, customer's needs to organic animal food is still increasing. In this study, we reviewed the usage of antimicrobials in animal farms and antimicrobial resistance of microorganisms isolated from organic dairy farm environments. The isolates from dairy farms were Acinetobacter sp., A. lwoffi, A. johnsonii, A. towneri, Aerococcus viridans, Aeromonas media, A. veronii, Bacillus pseudofirmus, B. pumilus, B. licheniformis, Corynebacterium glutamicum, Escherichia coli, Enterococcus faecium, Lysinibacillus fusiformis, Paenibacillus illinoisensis, Staphylococcus epidermidis, S. hominis, Streptococcus equinus, S. lutetiensis, and Saccharomyces cerevisiae. Indicator microorganisms isolated from organic dairy farms were tested for susceptibility to 20 types of antimicrobials. E. coli (ATCC 25922) isolated from dairy farm fence showed resistance to 8 types of antimicrobials, such as oxacillin, penicillin, vancomycin, etc., and E. faecium isolated from feces showed resistance to 9 types of antimicrobials, such as cephalothin, oxacillin, streptomycin, etc., respectively. However, these results showed less antimicrobial resistance compare with customary dairy farm.

The Efficacy of Water Purification and Distribution of Ammonia Oxidizing Bacteria in Shihwa Constructed Wetland (시화호 인공습지의 수질정화 및 암모니아 산화균의 분포 연구)

  • Water quality and the distribution of ammonia oxidizing bacteria were characterized in constructed wetland of Shihwa lake. Both physico-chemical parameters and fecal indicator microorganisms including total coliforms, E.coli, Enterococcus spp. were measured. In addition, denaturant gradient gel electrophoresis (DGGE) was carried out after PCR amplification of amoA gene from input, output, and wetland sites of the Banwol, Donghwa, and Samhwa stream in Shihwa lake area. Physico-chemical parameters were in proper range for typical nitrifying bacteria to grow and perform their biological activities. Average concentrations of fecal indicator microorganisms of wetland samples were lower than those of input sites. These results suggested that microbial water quality improved by the process of constructed wetland. According to phylogenetic information obtained from DGGE from study sites, distribution of nitrifying bacteria from each of input, output, and wetland were generally distinctive one another. In addition, distribution of nitrifying bacteria between Banwol and Donghwa streams showed higher similarity (52.6%) than this of Samhwa stream (15.2%). These results indicated that characteristics of ammonia oxidizing bacteria in Samhwa were unique in comparison with those of Banwol and Donghwa stream.

Identification and Partial Characterization of Lacticin SA72, a Bacteriocin Produced by Lactococcus lactis SA72 Isolated from Jeot-gal

  • Strain SA72 was isolated from Jeot-gal and identified as producer of a bacteriocin, which showed some bactericidal activity against Lactobacillus delbrueckii ATCC 4797. Strain SA72 was tentatively identified as Lactococcus lactis according to the AOI test. Lactococcus lactis SA72 showed a broad spectrum of microorganisms, tested by the modified deferred method. The activity of lacticion SA72, named tentatively as a bacteriocin produced by Lactococcus lactis SA72, was detected during the mid-lon growth phase, reached a maximum during the early stationary phase, and then declined after the late stationary phase. Lacticin SA72 also showed a relatively broad spectrum of activity against non-pathogenic and pathogenic microorganisms when assessed by the spot-on-lawn method. Its anitimicrobial activity on sensitive indicator cells disappeared completely by protease XIV treatment. The inhibitory activity of lacticin SA72 remained after treatment for 15 min at $121^{\circ}C$, 문 was stable in a pH range of 2.0 to 9.0 and all organic solvents examined. It demonstrated a typical bactericidal mode of inhibition against Lactobacillus delbrueckii ATCC 4797. The apparent molecular mass of lacticin SA72 was in the region of 3-3.5 kDa, determined by SDS-PAGE.
