표고 봉형 톱밥재배시 배양중 침공처리가 '자담'의 발이 및 자실체 생산에 미치는 영향 (Cultural characteristics and fruiting body productivity of new Lentinula edodes cultivar 'Jadam' in rod-type sawdust cultivation)

  • 표고 신품종 '자담'의 자실체 안정생산을 위해 침공처리가 균사배양 및 자실체 생산성에 미치는 영향을 조사하였다. 침공 직경과 깊이가 크고, 침공수가 많을수록 중량감 모율과 색차는 증가하고 명도는 감소하였다. 종균 접종 후 배지온도는 배양 33일에, CO2농도는 배양 19일에 최고치에 도달하였고, O2농도는 CO2 와 반대의 경향으로 배양 14일에 최저치를 나타냈다. 침공 직경과 깊이가 크고 침공수가 많을수록 배지 품온과 O2농도는 상승하였고, CO2농도는 낮아졌다. 수량성은 5×70 mm-30 처리구에서 644.7 g으로 다른처리구에 비해 가장 높은 수량을 보여 자담의 적정한 침공방법으로 선발하였다.

느타리버섯의 액체종균 배양 조건과 생육 특성 (Culture conditions of liquid spawn and the growth characteristics of Pleurotus ostreatus)

  • 느타리버섯의 생산성 향상을 위하여 액체종균 배양액의 조건을 설정하고자 하였다. Potato dextrose 배양액에서 균체 생장을 위한 최적 pH는 6이었다. 대두박 0.3%와 설탕 및 포도당 함량을 달리한 15가지의 배양액('A'~'O')은 $22^{\circ}C$ 및 pH 6의 조건에서 14일동안 배양되었을 때 설탕과 포도당이 각각 함유된 배양액에서 흡광도는 증가되었다. 특히 5~20%의 포도당만 함유된 배양액('C', 'D', E' 및 'F')은 배양기간의 경과에 따라 흡광도가 증가되는 경향이었다. 배양액의 건조 균체량은 20%이하의 설탕이나 포도당이 단독으로 함유된 배양액에서 감소되었으나, 30%의 설탕('A') 또는 포도당 ('B')이 각각 함유된 배양액에서 건조 균체량은 증가되었다. 1100 mL의 톱밥배지 병재배 시 액체종균의 최적 접종량은 15~20 mL이었다. 고체종균 및 액체종균으로 느타리버섯을 재배하여 조직감을 비교한 결과 고체종균에 비해 액체종균으로 재배된 버섯에서 우수한 경향이었다.

Latex 응집반응을 이용한 동물의 톡소플라즈마병 진단액 개발에 관한 연구 (Development of antigen for the microplate latex agglutination test on toxoplasmosis in animals)

  • This study was conducted to develop a sensitized latex-antigen for serodiagnosis of toxoplasmosis in animals. Tachyzoites of T gondii(RH-strain) harvested from mouse peritoneal cavity were purified through the filtraton of polycarbonate membrane(pore size, $3.0{{\mu}m}$, Costar Co.) and disrupted by ultrasonicator. The tachyzoite suspension was ultracentrifuged for 30 min at $60,000{\times}g(4{^{\circ}C})$ and the supernatant was used as a water-lysate antigen. Polystyrene latex particles of $0.8{{\mu}m}$ in diameter(Sigma) were used for the preparation of sensitized latex-antigen suspension. The several parameters including the preparation conditions, incubation buffer. serum dilution buffer and stability of agglutination reactions were evaluated and the results obtained were summarized as follows : 1. The antigen consisting of a water-lysate of T gondii tachyzoites was adsorbed onto polystyrene latex particles of $0.8{{\mu}m}$ in diameter by adding a latex suspension to an equal volume of diluted antigen solution and by incubating the mixture at $37{^{\circ}C}$ under different conditions. 2. The optimum incubation buffer used for the antigen sensitization was 0.1M Tris-HCl buffer(pH 8.0). 3. The optimum serum dilution buffer used for the latex agglutination test was 0.1M Tris-HCl-NaCl buffer(pH 7.4) containing 300 mM NaCl. But 0.1M Tris-HCl-NaCl buffer(pH 7.4) containing 300-600 mM NaCl, 0.5% BSA and 0.01% Tween-20 improved the agglutination pattems and cleared the background of microplate well without the effects on L.A titer. 4. The time required for antigen sensitization was 40 and 60 min in incubation buffer(pH 8.0) at $37{^{\circ}C}$. But the optimun time for antigen sensitization was min at $37{^{\circ}C}$. 5. The optimun quantity of antigen absorbed on latex particles for proper agglutination was the range of 20 to $32{\mu}g$ of latex particles. 6. The optimun concentration of the latex-antigen suspension for the proper agglutination reaction was determined as 0.2%(w/v). 7. The specificity, rapidity and simplicity of the latex-particle agglutination test suggested that it might be adaptable to large scale serum screening.

변형 체내 표지법에 의한 적혈구 표지시 결합효율에 영향을 미치는 인자 평가 (The Evaluation of Factors Which Influence Binding Efficiency of Modified in Vivo Erythrocyte Labeling Technique)

  • 목적: 변형 체내 표지법으로 적혈구 표지시 영향을 미칠 수 있는 인자에는 수혈, 환자에게 투여된 약제, 염화주석의 양, 발생기 내부성장 시간, tinning 시간, 배양시간, 혈액 양, 항응고제의 종류, 채취한 혈액을 배양시 rotating invertor 사용 유무 등 매우 다양하다. 저자들은 tinning 시간, 혈액 양, 채취한 혈액을 배양시 rotating invertor 사용 유무, 배양시간, 항응고제의 종류에 따라 변형 체내 표지법으로 적혈구 표지시 높은 결합효율을 유지시키기 위한 최상의 조건을 알아보고자 하였다. 대상 및 방법: 2003년 3월부터 2004년 2월까지 과거에 빈혈, 혈액응고 질환, 출혈성 질환 등의 질병이 없는 자발적인 지원자 30명(남:여=19:11, 연령: $25{\pm}2$세)을 대상으로 하였다. Tinning시간, 혈액 양, 배양시간과 항응고제의 종류에 대한 실험에서는 각각의 인자마다 15명을 대상으로 45개씩, 모두 180개의 혈액 샘플을 얻었고, rotating invertor 사용유무에 대한 실험에서는 15명을 대상으로 30개를 얻어 총 210개의 혈액 샘플을 얻었다. 채취 한 혈액샘플은 원심분리기에서 상층액과 하층액으로 분리시켜 각각의 결합효율을 계산한 후 SPSS 10.0 프로그램을 이용하여 paired T-test와 one way ANOVA통계 검정을 실시하였다. 결과: Tinning 시간에 따른 결합효율은 5분일 때 $45{\pm}23%$, 20분일 때 $97{\pm}8%$, 35분일 때 $98{\pm}6%$로 20분과 35분에서 유의하게 높은 결합효율을 보였다(p<0.001). 혈액 양에 따른 결합효율은 1 mL일 때 $73{\pm}32%$, 3 mL일 때 $91{\pm}10%$, 5 mL일 때 $96{\pm}7%$로 3 mL와 5 mL에서 유의하게 높은 결합효율을 보였다(p<0.05). Rotating invertor를 사용한 경우 결합효율은 $96{\pm}7%$로 사용하지 않은 경우의 $87{\pm}18%$에 비하여 높았으나 통계학적으로 유의한 차이는 없었다(p>0.05). 배양시간에 따른 결합효율은 10분일 때 $96{\pm}7%$, 25분일 때 $95{\pm}12%$, 40분일 때 $98{\pm}3%$로 통계학적으로 유의한 차이는 없었다(p>0.05). 항응고제 종류에 따른 결합효율은 헤파린을 사용한 경우 $89{\pm}20%$, CPDA를 사용한 경우 $97{\pm}6%$, ACD를 사용한 경우 $98{\pm}4%$로 CPDA와ACD를 사용한 경우에 유의하게 높은 결합효율을 보였다(p<0.001). 결론: 변형 체내 표지법으로 적혈구를 표지시 우수한 결합효율을 유지하기 위해서는 채취하는 혈액의 양은 3 mL 이상, 배양시간은 10분 이상(10분-40분), 항응고제는 ACD나 CPDA tinning 시간은 20분 이상(20-35분)을 유지하고, 가능한 rotating invertor를 사용하는 것이 좋을 것으로 생각된다.

백련차 분말을 대체한 우리밀 식빵의 품질 특성 (Quality Characteristics of Domestic Wheat White Bread with Substituted Nelumbo nucifera G. Tea Powder)

  • In this study, domestic wheat white breads were prepared with the substitution of 1.5, 3.0, 4.5, and 6.0% Nelumbo nucifera G. tea powder(NNTP). The samples and a control were then compared with regard to quality characteristics, including pH, total titratable acidity, fermentation power of dough expansion, specific volume, baking loss, moisture content, color, textural characteristics, external and internal surface appearances, and sensory qualities in order to determine the optimal ratio of NNTP in the formulation. As the NNTP contents increased, the pH of dough and bread, baking loss, and lightness decreased, whereas the total titratable acidity of dough and bread increased. The fermentation power of dough expansion increased with increasing incubation time. The NNTP samples evidenced significantly higher specific volume, greenness, yellowness, resilience, and crumb consistency than were observed in the control group. However, hardness and fracturability evidenced the opposite effect. The water content and uniformity of the crumb pores were highest at a substitution level of 1.5%, and were lowest at a level of 4.5%. Crumb color, flavor, and delicious taste decreased with increasing NNTP contents, whereas lotus leaf flavor, astringency, bitterness, and off-flavor increased. Density of the crumb pore and crumb springiness were not significantly different among the samples. Softness, chewiness, and overall acceptability were maximal with the 1.5% substitution, and were minimal in the 6.0% group. In conclusion, our results demonstrate that $1.5{\sim}3.0%$ NNTP may prove quite useful as a substitute for domestic wheat flour in the production of white bread, and may evidence favorable nutritional and functional properties.

영가철 및 산업폐기물을 활용한 비소오염토양의 안정화 효과조사 (Investigation of Stabilization Effect on Arsenic Contamination Soils using Zerovalent Iron and Industrial by-products)

  • In order to investigate stabilization effect on As-contaminated soils treated by zero-valent iron(ZVI) and industrial by-products, batch tests and column tests were carried out with As-contaminated soils collected from farmland around the abandoned mine site. In batch tests, ZVI and industrial by-products(blast furnace slag, steel refining slag and oyster shell powder) were used as treatment materials to reduce As. Industrial by-products were mixed with As-contaminated soils, in the ratio of 1%, 3%, 5% and 7% on the weight base of dried soil. After incubation, all samples showed the reduction of As concentration and it was expected that ZVI and steel refining slag were effective treatment materials to remove As among treatment materials used in batch test. In column tests, columns were made by acrylic with the dimension of diameter=10cm, height=100cm, thickness=1cm and these columns were filled with untreated soils and treated soils mixed with ZVI and steel refining slag(mixing ratio=3%). Distilled water was discharged into the columns with the velocity of 1 pore-volume/day. During test, pH, EC, Eh and As concentration were measured in the regular term(1 pore-volume). As a result, ZVI and steel refining slag were shown 93%, 62% reduction of As concentration respectively by comparison with untreated soils. Therefore, if ZVI and steel refining slag are used as treatment materials in As-contaminated soils, it is expected that the As concentration in soils is reduced effectively.

Optimal Culture Conditions for the Shoot Growth of Freesia 'Shiny Gold' during Bioreactor Culture

  • For rapid production of freesia 'Shiny Gold' shoots by using a bioreactor, several culture conditions were investigated. Young shoots (< 1 cm) obtained from freesia corm section in vitro were used as plant materials for this experiment. As a basic experimental environment, 20 young shoots were inoculated into a 5 L balloon type bubble reactor which contained 1 L 1/2 strength MS medium supplemented with 30 g sucrose (3%), and the aeration was 0.1 vvm (vessel volumes per minute). The bioreactors were placed in a growth room with $23^{\circ}C$ temperature, 60% relative humidity and $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ light condition (16 h/8 h, day/night). The concentrations of MS media were set with 1/4, 1/2, 1 strength, medium volume 10, 20, 40%, sucrose concentration 3, 6, 9%, and aeration 0.1, 0.2, 0.4 vvm. After 4 weeks of cultivation, the growth indexes including the fresh and dry weight, and plant height were evaluated. At the same time, the consumption, pH, and EC of medium were estimated 4 weeks after incubating. The best results were achieved when 40 young shoots were incubated in a bioreactor in which 1 L of 1/2 strength MS medium supplemented with 6% sucrose was used for the rapid production of freesia shoots. The shoots were 17 cm in plant height and 1.0 g in fresh weight only 4 weeks after incubation which could be a good plant material suitable for corm enlargement in vitro. No correlation was observed between the growth of freesia shoots and the consumption, pH or EC of medium.

다양한 곡류의 Sourdough를 첨가한 식빵의 품질특성 (Quality Characteristics of Bread Containing Sourdough Using Various Grain Flours)

  • The principal objective of this study was to evaluate the effects of the addition of sourdough to various grain flours in bread, specifically, in regards to the physicochemical characteristics of bread dough, sensory evaluation, and bread storage. As the incubation time of sourdough increased, the total titratable acidity increased. Viable yeast counts of sourdough increased consistently until the third day, while lactic acid bacteria counts increased until the second day. The weight of breads containing sourdough made with rye flour, strong flour, and Korean wheat flour were higher than that of the control. However, the height, volume, and specific volume of control were higher than those of the groups with sourdough made with various grain flours. The pH of breads containing sourdough was lower than that of the control, while the total titratable acidity and moisture content were higher than those of the control. In analyzing the visible mold colony during the five days of storage at $30^{\circ}C$, mold growth in breads containing sourdough made of Korean wheat flour, barely flour, and rye flour was retarded. In the color measurement, the L values of the control and bread containing sourdough made with barley flour were higher than that of the other groups after five days. The a value of bread containing sourdough made of rye flour was higher, and the b values of breads containing sourdough made of Korean wheat flour, barley flour and rye flour were higher than those of the other groups after five days. The hardness of breads containing sourdough increased as storage time increased, where as breads containing sourdough made of Korean wheat flour, Korean whole wheat flour, and rye flour revealed no significant differences with control group. Sensory evaluation scores in terms of after swallowing, taste, and overall preference of bread containing sourdough made of Korean wheat flour was higher than those of the control group.

헤미셀룰라아제를 첨가한 보리잎차 식빵의 품질 특성 (Quality Characteristics of Barley Leaves Tea White Bread with Hemicellulase)

  • Barley leaves tea white bread were prepared by the addition of 0.005, 0.010, 0.015, and 0.020% hemicellulase to flour of the basic formulation. The experiments and control were then compared in terms of quality characteristics, including pH, total titratable acidity, fermentation power of dough expansion, specific volume, baking loss, moisture content, color, textural characteristics, internal surface appearances, and sensory qualities in order to determine the optimal ratio of hemicellulase in the formulation. There were no significant differences in pH and total titratable acidity of dough and bread among the experiments. Fermentation power of dough expansion were increased as incubation time increased. Bread made by the addition of hemicellulase had significantly higher specific volume than the control group. However, lightness and yellowness showed the reverse effect. Greenness was not significantly different among the samples. Baking loss was the highest at the 0.020% addition level, and moisture content was maximal with the 0.010% addition, while the lowest in the control bread samples. As hemicellulase contents increased, harness and fracturability decreased. Resilience was maximal with the 0.015% addition, and was minimal in the 0.005% group. In the sensory evaluation, color, flavor, softness, overall acceptability, barley leaves flavor, delicious taste, astringency, bitterness, and off-flavor were not significantly different among the samples. In coclusion, the results indicate that adding 0.010% hemicellulase in barley leaves tea white bread is optimal for quality and provides a product with reasonably high overall acceptability.

In Vitro Development of Porcine Parthenogenetic Embryos under the Oil-free Culture System

  • Optimization of the preimplantation mammalian embryo culture condition was widely focused on refining medium composition under the name of chemically defined media. However, recent research revealed that the alteration of physical environment can be a crucial factor to a successful embryo development. In this study, under the same embryo density, a novel culture device named oil-free micro tube culture (MTC) system was evaluated using porcine parthenogenetic embryos. The activated oocytes were placed into the 0.2 ml thin-wall flat cap PCR tube and cultured to the blastocyst stage. As a preliminary step, embryo density and culture medium volume were optimized under a standard drop culture system. The optimal embryo density range for in vitro culture was 0.5 embryos per ${\mu}l$ in $20\;{\mu}l$ drop (20.5%) and 1.0 embryos per ${\mu}l$ in $10\;{\mu}l$ drop (20.6%). Based on these results, we compared drop culture system and 'MTC' system in terms of the developmental rate to the blastocyst stage. In $20\;{\mu}l$ medium volume, the 'MTC' system showed similar blastocyst formation rate when compared with drop culture system (20.2% versus 20.5%, respectively) while the 'MTC' system showed lower blastocyst formation rate than drop culture system in $10\;{\mu}l$ one (12.7% versus 20.0%, respectively). Therefore the $20\;{\mu}l$ MTC system may be an alternative incubation system for short-distance embryo transport without carrying the $CO_2$ incubator and this provides novel embryo culture device to clinical veterinary embryologists.
