Estimation of the incubation period of P. vivax malaria in Korea from 2006 to 2008 (2006년-2008년 삼일열 말라리아환자의 잠복기 연구)

  • Based on the detailed travel history of cases from 2006 to 2008 who reside in non-malarious areas, statistical estimates of the incubation periods were obtained. The data suggest that cases fall into two categories with short- and long-term incubation periods, respectively. 72 and 25 cases successfully met our criteria for inferring the durations of short- and long-term incubation periods. The mean short- and long-term incubation periods were estimated to be 25.42 days and 328.6 days weeks, respectively.

  • One thousand and eighty eggs of Japanese quail (Coturnix coturnix Japonica) were set into the incubator maintaining 36 treatment groups (3 egg weight groups ${\times}3$ seasons ${\times}4$ preincubation holding periods) to evaluate their hatchability performances. Holding periods had significant (p<0.05) effect on hatchability and the seasons showed significant (p<0.05) effect on chick weight. All the parameters (except fertility) were significantly (p<0.01) influenced by the egg weight. None of the parameters maintained regular trend with egg weight and pre-incubation holding periods. Significant interactions were not observed on any of the parameters (except fertility) studied. The egg weight maintained significant (p<0.05) negative correlation with fertility and positive correlation with chick weight. Eggs of medium weight (9.10 to 10.00 cm) could be hatched satisfactorily between 4 and 7 days of pre-incubation holding periods in any season of the year.

The Relationship Between Egg Incubation Period and Temperature in Several Species of Plecoptera

  • Egg incubation periods of 14 species of plecoptera were examined at $10^{\circ}\;{\sim}\;11^{\circ}C$, $15^{\circ}\;{\sim}\;16^{\circ}C$, $20^{\circ}C$, and $23^{\circ}C$ under dark conditions. The total effective temperature (TET) was calculated by multiplying mean egg incubation days and water temperature of incubation periods. The relation between the TET and the incubation temperature was used to compare the life cycle of respective species. Perlodid species had higher TET values with a positive relation to incubation temperature than those of other species. Perlid species had low TET values in the 14 species with negative to variable relation, and Chloroperlid species showed variable to positive relations to incubation temperature. These results suggest that the relation between the TET and the water temperature reflected on their habitat of respective species.

  • Ontogeny, distributions and relative frequencies of somatostatin-immunoreactive cells were investigated in the proventriculus of the chicken embryos with incubation periods. Samples were taken from 10 groups(10 days of incubation to hatching) and studied by immunohistochemical methods. The findings were as follows. Somatostatin-immunoreactive cells were observed from 12 days of incubation in the proventricular glands and after that increased with incubation periods. The first observation time of these cells in the epithelium were at 15 days of incubation in the basal portion but in 16 and 17 days of incubation, no immunoreactive cells were observed in the epithelium but after that a few immunoreactive cells were observed in the basal portion and gastric gland regions. The shapes of these cells were spherical to spindle in the proventricular glands and spherical to round in the epithelium and gastric gland.

Adverse effects of pesticide/metabolites on boar spermatozoa

  • The metabolites of agrichemicals, such as organophosphorus pesticides, are known to be more hazardous than their parent pesticides. 3,5,6-trichloro-2-pyridinol (TCP) is a major degradation product of chlorpyrifos, one of the organophosphate insecticides widely used in agriculture. In vivo or in vitro exposure to chlorpyrifos has been known to interfere with male reproductive functions, leading to reduced fertility in mammals. Therefore, this study was performed to examine the changes in the fertilization competence of boar spermatozoa exposed to TCP. Sperm samples were subjected to varying concentrations of TCP (10, 50, 100, 200 µM) and different periods of incubation. Sperm motility, motion kinematics, viability, acrosome integrity, intracellular reactive oxygen species (ROS) production, and gene expression levels (ODf2, ZPBP2, AKAP3 and AKAP4) were evaluated after exposure of the sperm to TCP. A significant dose-dependent reduction in motility was observed in sperm samples incubated with TCP compared to the controls after both incubation periods. Sperm viability was significantly decreased in samples incubated with 50, 100, and 200 µM TCP in both incubation periods. A significantly lower percentage of normal acrosomes and gene expression levels were observed in sperm samples exposed to 50, 100, and 200 µM TCP after both incubation periods, compared to the controls. There was a significant increase in the ROS production in spermatozoa incubated with 100 - 200 µM TCP after both incubation periods. Consequently, the direct exposure of boar spermatozoa to TCP interferes with sperm functions and leads to decreased fertilization. In order to identify and address the various causes of reproductive decline, the impact of chemical metabolites needs to be discussed in depth.

Exposure of chlorpyrifos impairs the normal function of boar spermatozoa

  • The misuse of pesticides has resulted in environmental pollution, which directly or indirectly affects all life on earth. Chlorpyrifos is a chlorinated organophosphorus pesticide that is commonly used in agriculture. The aim of this study was to investigate the effects of chlorpyrifos on the fertilization function of boar spermatozoa. Sperm samples from boars were subjected to varying concentrations of chlorpyrifos from 10 to 200 µM for two incubation periods, 30 min or 2 hrs. The boar spermatozoa were then evaluated for motility, motion kinematics, viability, acrosome integrity, chromatin stability, and generation of intracellular reactive oxygen species (ROS). There was a significant percentage reduction in sperm motility and motion kinematic parameters after both incubation periods (p < 0.05). The proportion of viable spermatozoa decreased after incubation for 30 min and 2 hrs in a dose-dependent manner (p < 0.05). A significantly lower percentage of normal acrosomes was observed in spermatozoa exposed to 200 µM chlorpyrifos over both incubation periods, compared to the controls. The damage to sperm DNA was significantly higher when the exposure time to chlorpyrifos was longer. There was a significant increase in the ROS levels in spermatozoa incubated with chlorpyrifos for 2 hrs (p < 0.05). From the results of the present study, it is concluded that direct exposure of boar spermatozoa to chlorpyrifos altered boar sperm characteristics, suggesting potential toxicity that may affect the male reproductive function.

  • To evaluate the effect of spermatozoa culture on glycosidase activity of frozen-thawed spermatozoa in human, the spermatozoa were treated experimentally and assayed for activities of ${\alpha}$-L-fucosidase, ${\alpha}$-D-mannosidase, ${\beta}$-D-galactosidase and N-acetyl-${\beta}$-D-glucosaminidase (${\beta}$-GlcNAc'ase). The ${\beta}$-GlcNAc'ase activity was at least two-folds higher than other glycosidases regardless of spermatozoa incubation (p<0.05). The spermatozoa motility was decreased with incubation periods, but no effects by different glycosidases on the changes of spermatozoa motility during the various periods of incubation. In all glycosidases, the spermatozoa-zona binding rates in spermatozoa without incubation were higher than in spermatozoa incubated for 2 h (p<0.05). ${\beta}$-GlcNAc'ase is present mainly in the plasma membrane of spermatozoa frozen-thawed in human. It was also shown that the glycosidase activity was increased in all glycosidases in spite of lower sperm-zona binding by spermatozoa incubation.

  • This study was carried out to develop the biochemical pulping method to enhance energy saving and decrease the capital cost through the soda pulping under atmospheric pressure (100%). Nonwood substrates, rice straw, were pretreated by white-rot fungi, Ganoderma Iucidum. It has acquired several basic data that can be applied in bio-soda pulping. The results of this study were as follow. Without any nutrients or with glucose, N and glucose+N the weight losses of rice straws inoculated by Canoderma Iucidum were 8.5~29.8%, 9.3~32% 11.8~30.1% and 11.8~24.4% respectively for 5, 10, 15, 20, 25 and 30 days incubation. The more extending incubation periods, the more increasing weight losses. The yield of untreated rice saw was 54.8% after pulping. When any nutrients was not added or glucose, N and glucose+N were added for the pretreatment, the total yields were ranged 50.6~38.5%, 48.6~34.4%, 47.2~38.4% and 49.5~42.6% respectively for 5, 10, 15, 20, 25 and 30 days incubation. The yields were gradually decreased based on extending of incubation. The physical properties of rice straw soda pulps without fungal treatment, the density, breaking length, burst index, tear index and folding endurance were 0.24 g/$\textrm{cm}^3$, 2.32 km, 0.91 kPa $m^2$/g, 56.7 mN.$m^2$/g and 35 times, respectively. After pretreatment without any nutrients or with glucose, N and glucose+N as nutrients the density was 0.27~0.30 g/$\textrm{cm}^3$, the breaking length 3.14~5.25 km, burst index 1.42~2.78 kPa.$\textrm{m}^2$/g, tear index45.8~64.5 mN.$\textrm{m}^2$/g and folding endurance 47~288 times at all incubating periods when pulping was done. The physical properties were increased with the increasing incubation periods. However, when glucose+N was added, the physical properties were shown superior results each incubating duration.

Bio-Soda Pulping of Rice Straw with Pleurotus cornucopiae under Atmospheric Pressure

Under the conditions without any nutrients or with glucose, N and glucose + N, the weight losses of rice straws inoculated by Pleurotus cornucopiae were 12.1∼32.6 %, 12.0∼26.3 %, 13.0∼25.4 % and 15.3∼24.7 % for 5, 10, 15, 20, 25 and 30 days incubation periods respectively. The more the fungal incubation was extended, the more the weight losses were gained. The yield of untreated rice straw was 54.8 % after pulping. When any nutrients was not added or glucose, N and glucose + N were added for the pretreatment, the total yields were ranged to 57.3∼42.9 %, 51.0∼43.3 %, 51.7∼43.9 % and 52.1∼46.1 % for 5 different incubation periods respectively. The yields were gradually decreased based on the extending of the incubation periods. The physical properties of the rice straw soda pulp without fungal treatment, the density, breaking length, burst index, tear index and folding endurance were 0.24g/㎤, 2.32 Km, 0.91 kPaㆍ$m^2$/g, 46.7 mNㆍ$m^2$/g and 21 times, respectively. In the case of pretreatment without any nutrients or with glucose, N and glucose + N as nutrients, the density was 0.24g/㎤, the breaking length was 3.30∼6.46 Km, the burst index was 1.36∼3.01 kPaㆍ$m^2$/g, the tear index was 33.0∼57.0 mNㆍ$m^2$/g and the folding endurance was 14∼381 times at most incubating periods, when pulping was done. The physical properties were increased as the incubation duration was extended. Especially, when N and glucose + N were added, the physical properties showed superior results during each incubation period.

Differential Expression of Th1- and Th2- Type Cytokines in Peripheral Blood Mononuclear Cells of Murrah Buffalo (Bubalus Bubalis) on TLR2 Induction by B. Subtilis Peptidoglycan

  • Peripheral blood mononuclear cells (PBMCs) discriminate microbial pathogens and induce T-cell responses of appropriate effector phenotype accordingly. Toll-like receptors (TLRs), in part, mediate this microbial recognition and differentiation while the development of T-cell effector functions critically depends on the release of Th1- or Th2- type cytokines. In the present study, buffalo PBMCs were stimulated under in vitro culture conditions by Bacillus subtilis cell wall petidoglycan, a TLR2 ligand, in a dose- and time- dependent manner. The expression of TLR2 as well as the subsequent differential induction of the Th1 and Th2 type cytokines was measured. Stimulation was analyzed across five doses of peptidoglycan ($10{\mu}g/ml$, $20{\mu}g/ml$, $30{\mu}g/ml$, $40{\mu}g/ml$ and $50{\mu}g/ml$) for 3 h, 12 h, 24 h and 36 h incubation periods. We observed the induction of TLR2 expression in a dose- and time-dependent manner and the peptidoglycan induced tolerance beyond $30{\mu}g/ml$ dose at all incubation periods. The correlation between peptidoglycan stimulation and TLR2 induction was found positive at all doses and for all incubation periods. Increased production of all the cytokines was observed at low doses for 3 h incubation, but the expression of IL-4 was relatively higher than IL-12 at the higher antigen doses, indicating tailoring towards Th2 response. At 12 h incubation, there was a pronounced decrease in IL-4 and IL-10 expression relative to IL-12 in a dose- dependent manner, indicating skewing to Th1 polarization. The expression of IL-12 was highest for all doses across all the incubation intervals at 24 h incubation, indicating Th1 polarization. The relative expression of TNF-${\alpha}$ and IFN-${\gamma}$ was also higher while that of IL-4 and IL-10 showed a decrease. For 36 h incubation, at low doses, relative increase in the expression of IL-4 and IL-10 was observed which decreased at higher doses, as did the expression of all other cytokines. The exhaustion of cytokine production at 36 h indicated that PBMCs became refractory to further stimulation. It can be concluded from this study that the cytokine response to sPGN initially was of Th2 type which skews, more pronouncedly, to Th1 type with time till the cells become refractory to further stimulation.
