A Study on the Improvement Methods for Hybrid Sorghum Seeds Production

  • Sorghum (Sorghum bicolor L.) is a major cereal grain crop of the world and It can be cultivated under high temperature and dry condition with high adaptability to the adverse environment. It is usually eaten with rice in korea and most of domestic sorghum cultivars have been developed by pure line selection and cross breeding. In prior studies, Hybrid cultivar (F1 seed) was known to improve 30-40% in yield and resist to disease and pest than pure line varieties due to hybrid vigor. Therefore, study on the hybrid sorghum is continuously necessary. In korea, hybrid sorghum cultivar have been developed, so we need to find methods seed production technology for supply and commercialization of hybrid seed cultivar. Female inbred line(A-line) and male inbred line(R-line) are needed for hybrid seed production. This study was carried out to investigate growth characteristics and yield of hybrid sorghum according to the seedling period and planting rate between female inbred line(YSA1) and male inbred line (Sodamchal, Miryang 19). When the flowering period of two inbred groups in the process of hybrid seed production is coincides, hybrid seed production is increase. The female inbred line and male inbred lines were cultivated at Daegu in 2021. Two inbred lines were sown at intervals of 3 days and 7days and were evaluated flowering period. As a result, the flowering period of the female inbred line and male inbred lines were matched at the 7days interval. To find out optimal planting rate, two inbred lines were cultivated under different plating rate (4:2, 5:2, 6:2). Yield of YSA1/Sodamchal(F1) was the highest 130(kg/10a) in the 4:2 ratio and yield of YSA1/Miryang 19 was the highest quantity of 139(kg/10a) in the 6:2 ratio. The results of this study could be helpful for hybrid Sorghum seeds production.

국내 육성 주요 옥수수 자식계통 및 교잡종의 종실 성분 특성 (Chemical Composition of Seed from Inbred Lines and Hybrids of Maize Recently Developed in Korea)

  • 옥수수 우량 교잡종 육성을 위한 자료로 활용하고자 국내 육성 옥수수 자식계통, $F_1$ 종실 및 $F_2$ 종실의 이화학적 특성과 지방산 및 아미노산 조성을 각각 분석 검토하였다. 1. $F_2$ 종실의 단백질 함량이 9.0%로 자식계통 11.4%와 $F_1$ 종실 10.9%에 비하여 유의하게 낮은 것으로 나타났다. 지방 함량은 $F_2$ 종실이 자식계통과 $F_1$ 종실보다 높았다. 회분은 자식계통, $F_1$ 종실, $F_2$ 종실간 조성변이에 차이가 없었다. 2. 자식계통, $F_1$ 종실, $F_2$ 종실 모두 linoleic acid의 조성비가 가장 높고 oleic acid, palmitic acid, stearic acid, linolenic acid의 순으로 지방산 조성비가 높은 것으로 나타났다. $F_2$ 종실의 포화지방산의 함량은 17.6%로 $F_1$ 종실과는 비슷하였으나, 자식계통보다는 낮았다. $F_2$ 종실의 불포화지방산의 함량은 82.4%로 $F_1$ 종실과는 비슷하였으나, 자식계통보다는 높았다. 3. $F_2$ 종실의 leucine의 함량이 자식계통 및 $F_1$ 종실보다 낮았다. 반면 $F_2$ 종실의 valine, serine, threonine, cysteine, aspartic acid, lysine, arginine, histidine 등의 함량이 자식계통 및 $F_1$ 종실보다 높았다. Alanine 등 나머지 아미노산은 자식계통, $F_1$ 종실, $F_2$ 종실간 조성변이가 비슷한 경향이었다. 4. 산성아미노산인 MMA(monoamino monocarboxylic acid)가 자식계통, $F_1$ 종실, $F_2$ 종실에서 그 조성 비율이 가장 높았으며 산성 아미노산인 DMA(diamino monocarboxylic acid), 방향족 아미노산인 AAA(aromatic amino acid) 순으로 조성비가 높았다.

튀김옥수수 자식계통들에 대한 형태적 특성 (Analysis of Morphological Characteristics Among Popcorn Inbred Lines)

  • 본 연구는 강원도농업기술원 옥수수연구소에서 튀김옥수수 품종개발을 위하여 육성한 79개의 자식계통들에 대하여 총 13개의 양적 및 질적 형질들을 이용하여 형태적 변이 연구를 수행하였다. 3개의 질적 형질 중에서 웅수색(QL1)은 연한 자주색을 나타내는 계통(46계통)들이 가장 많았고, 자수색(QL2)과 줄기색(QL3)에서는 각각 녹색을 나타내는 계통(55계통, 75계통)들이 가장 많았다. 반면에 10개의 양적형질 중에서 간장(QN1)은 $174.2{\pm}34.9$ cm, 착수고(QN2)는 $103.4{\pm}24.7$ cm, 이삭장(QN3)은 $9.4{\pm}3.0$ cm, 착립장(QN4)은 $8.4{\pm}2.6$ cm, 이삭경(QN5)은 $24.9{\pm}7.7$ mm, 이삭열수(QN6)는 $14.0{\pm}2.3$ 열, 이삭중(QN7)은 $36.5{\pm}26.0$ g, 종실중(QN8)은 $30.9{\pm}19.3$ g, 백립중(QN9)은 $10.4{\pm}3.8$ g, 발아율(QN10)은 평균 $95.3{\pm}8.1%$를 각각 나타내었다. 분석에 이용된 자식계통들 중에서 5개의 자식계통(PS0-001, PS0-003, PS1-002, PS1-003, PS2-009)들은 조사된 7개의 수량 관련 형질들 중 5개 이상의 형질들에서 비교적 높은 경향이었다. 주성분 분석은 분석에 이용된 13개의 양적 및 질적 형질들 중에서 자수색(QL2), 이삭장(QN3), 착립장(QN4), 이삭경(QN5), 이삭중(QN7), 종실중(QN8), 백립중(QN9)은 제 1 주 성분에서 양의 방향에 크게 기여하였고, 반면에 웅수색(QL1), 줄기색(QL3), 착수고(QN2) 그리고 이삭열수(QN6)는 음의 방향에 크게 기여하였다. 제 2 주성분에서는 간장(QN1), 착수고(QN2) 그리고 종실중(QN8)은 양의 방향에 크게 기여하였고, 반면에 웅수색(QL1), 자수색(QL2), 이삭장(QN3), 이삭열수(QN6) 그리고 100립중(QN9)은 음의 방향에 기여하였다.

  • Sometimes we have two groups of inbred lines and there are only interest in gca comparisons within group($p_{1}$) and group($p_{2}$) and not between two groups. For example, suppose there two Lab, each of the 2 Labs have obtained the best lines. For this purpose we now give a method of constructing block designs for diallel cross experiments and we will explain how to calculate efficiency. Then we show the efficiencies in the table.

국내 서양종꿀벌 순계의 형태적 특징 (Morphometric Characterization of Honey Bee, Apis mellifera Linnaeus, Inbred Lines in Korea)

  • 이탈리안벌인 A, C, F계통과 코카시안벌인 D, V계통을 2005년부터 2007년까지 국내에서 수집하였다. 수집한 계통은 육종을 위해 격리된 섬에서 근친교배를 통해 순계로 분리하였다. 이 연구는 꿀, 로열젤리 다수확계통 선발에 있어 개체군 선발과 육종 효율을 높이기 위해 수행되었다. 23 개의 형태학적 특성을 평가하고 두 아종의 기존 데이터와 비교한 결과, 이탈리안벌 순계계통은 코카시안벌 순계 계통과 달리8개의 특성이 기존의 이탈리안벌과 유사해 더 많은 특성이 보존되고 있음을 알 수 있었다. 또한 국내에서 유지되고 있는 순계들은 타 지역의 동일 계통과 차이를 보여 분리된 순계의 형태적인 특징이 확인되었다.

  • A class of block designs for general combining ability comparisons within two groups of inbred lines in diallel crosses is given. These block designs are constructed by using balanced block designs obtained by cyclically developing a single initial block. Also, the efficiencies of block designs are tabulated for number of lines 26 or less.

  • Giemsa C-banded mitotic chromosome prepatations from White Leghorn, New Hampshire and Rhode Island Red inbred lines were compared for frequency of C-band regions on individual chromosomes. Except for autosomes 3, 6, 8 and 9 and W sex chromosomes, C-banding was extremely variable in other macrochromosomes. No divergence for C-band difference between homologous chromosomes of these lines was detected. Approximately 75% of the mitotic metaphase microchromosomes have recognizable C-band regions with the current technique.

Comparative Gene Expression Analysis of Seed Development in Waxy and Dent Corn (Zea mays L.)

  • We used Illumina/HiSeq sequencing for analysis of gene expression profiling among four maize seed types (dent, CM3 and CM6; waxy, CM5 and CM19) at 10 DAP (days after pollination). A total of 88,993,000 (CM3), 103,817,340 (CM6), 103,139,640 (CM5), and 66,978,958 (CM19) sequence reads were generated with read lengths of about 0.9, 1.0, 1.0, and 0.7 billion bp, respectively. We obtained 69.1 (CM3), 71.0 (CM6), 71.2 (CM5), and 71.8% (CM19) high quality reads from the raw data and compared them with reference RNA sequences in a public DB (NCBI). It was revealed that mapped reads were 58%, 63%, 62%, and 62% of the EST reference in CM3, CM6, CM5 and CM19, respectively; and more than 51,000 genes were expressed based on RPKM criteria (over 0.25 value) in each CM3, CM6, CM5, and CM19 inbred line. In differentially expressed gene (DEG) analysis, we found that 3,527 genes were differentially expressed by at least two-fold with 1,709 upregulated in the two waxy inbred lines and 1,818 upregulated in the two dent inbred lines. We also detected genes for the sucrose and starch biosynthesis pathways based on BINs, and different expression patterns between waxy and dent inbred lines were shown for the gene set for starch synthesis, such as sh2, bt2, du1, wx1, and ae1. Although some genes were more expressed in dent lines, most genes for starch synthesis were much expressed in waxy lines. Especially, there was greater expression of the sus2 gene in both waxy lines compared with the dent lines.

선발과정에서의 세대별 QTL 좌위 고정에 관한 연구 (The Response of QTL in Generation during Selection)

  • The objective of this study was to determine the response of QTL in each generation during selection to develop inbred lines. The simulation program was written in Fortran. Magnitude of QTL effects, base population size, number of QTL assigned to population, and the allelic frequency for the positive allele at each major QTL were highly associated with number of generations to fixation of QTLs during selection. Populations with larger QTL effects and larger base population size had more individuals with fixed QTL. However, a smaller number of QTL assigned to population had a higher fraction of individuals with fixed QTL at each generation compared with more populations with QTL. This simulation study will help to design biological experiments for detection of QTL-marker association using inbred population and to determine optimum number of lines with fixed QTL during inbred line development. To complement this study, additional simulation should be need with abundant replicates, more various population sizes, magnitude of QTL effects, and recombination between markers and QTLs.
