Analysis of Fatty Acyl Groups of Diacyl Galactolipid Molecular Species by HPLC/ESI-MS with In-source Fragmentation

  • The structures of molecular species of galactolipids, such as monogalactosyl diacylglycerol (MGDG) and digalactosyl diacylglycerol (DGDG), isolated from wheat flour have been investigated using negative-ion electrospray ionization (ESI) mass spectrometry interfaced with high performance liquid chromatography (HPLC). According to the result of HPLC analysis, MGDG and DGDG were found to consist of mixtures of five and four molecular species, respectively. The galactolipids have been also analyzed to determine their fatty acid compositions, using HPLC/ESI-MS combined with in-source (or cone voltage) fragmentation. HPLC/ ESI-MS is very useful for one-step analysis of mixtures of galactolipids with a small sample quantity. Especially, the carboxylate anions produced in in-source fragmentations of the negative-ion of each component separated by HPLC provide valuable information on the composition of its fatty acyl chains.

Fragmentation Behavior Studies of Chalcones Employing Direct Analysis in Real Time (DART)

  • Chalcones are naturally occurring, biologically active molecules generating interest from a wide range of research applications including synthetic methodology development, biological activity investigation and studying fragmentation patterns. In this article, a series of chalcones has been synthesized and their fragmentation behavior was studied using modern ambient ionization technique Direct Analysis in Real Time (DART). DART ion source connected with an ion trap mass spectrometer was used for the fragmentation of various substituted chalcones. The chalcones were introduced to the DART source using a glass capillary without sample preparation step. All the chalcones showed prominent molecular ion peaks $[M]^{{\cdot}+}$ corresponding to the structures. Multistage mass spectral data $MS^n$ ($MS^2$ and $MS^3$) were collected for all the chalcones studied. The chalcones with substitutions at 3, 4 or 5 positions gave product ion peaks with the loss of a phenyl radical ($Ph^{\cdot}$) by radical initiated ${\alpha}$-cleavage, while substitution at 2 position of chalcone in the A-ring gave a product ion peak with the loss of substituted styryl radical (PhCH = $CH^{\cdot}$). In case of the chalcones with the substituent at 4 positions in A and B rings gave both types of fragmentation patterns. In conclusion, chalcones can be easily characterized using modern DART interface in very short time and efficiently without any cumbersome sample pretreatment.

  • Fiber fracture is one of the dominant failure phenomena to determine total mechanical properties in composites. Fiber fracture locations were measured by optical microscopic method and acoustic emission (AE) as functions of matrix toughness and surface treatment by the electrodeposition (ED), and then two methods were compared. Two AE sensors were attached on the epoxy specimen and fiber fracture signals were detected with elapsed time. The interfacial shear stress (IFSS) was measured using tensile fragmentation test and AE system. In ED-treated case, the number of the fiber fracture measured by an optical method and AE was more than that of the untreated case. The signal number measured by AE were rather smaller than the number of fragments measured by optical method, since some fiber fracture signals were lost while AE detection. However, one-to-one correspondence between the x-position location by AE and real break positions by optical method was generally established well. The fiber break source location using AE can be a valuable method to measure IFSS for semi- or nontransparent matrix composites nondestructively (NDT).

Fragmentation Management Method for 6LoWPAN (6LoWPAN에서 단편화 관리 기법)

  • 6LoWPAN is IPv6 packets transmission technology at Sensor network over the IEEE 802.15.4 Standard MAC and Physical layer. Adaptation layer between IP layer and MAC layer performs fragmentation and reassembly of packet for transmit IPv6 packets. RFC4944, IETF 6LoWPAN WG standard document define packet fragmentation and reassembly. In this paper, we propose the IRM(Immediate Retransmission Method) and SRM(Selective Retransmission Method) to manage packet fragmentation and reassembly at 6LoWPAN. Each time destination receives a fragmented packet, it sends Ack message to the source node on IRM. However, on SRM, the destination node receives all fragmented packet, it sends Ack message or Nak message to the source node. In this case, Nak message include the dropped packet number. To compare the performance of the proposed schemes, we develop a simulator using C++. The result of simulation shows the proposed schemes provider better performance than RFC4944 standard scheme.

A study on the fast prediction of the fragmentation zone using artificial neural network when a blasting occurs around a tunnel (인공신경망을 이용한 터널 주변 폭파 시 파쇄영역의 빠른 예측에 관한 연구)

  • When collapse occurs due to explosion near a tunnel, fragmentation zone should be comprehended quickly to recover the function of the tunnel itself. In this study, a method to interpret explosion behavior and predict the fragmentation zone fast. For this purpose, the various 3D-meshes were generated using SolidWorks and explosion analyses were carried out using AUTODYN. The influence of explosion variables such as source location on fragmentation volume were examined by performing sensitivity analyses. Also, a training database for an artificial neural network analysis had been established and the optimal training model was selected, and the predicted results for fragmentation volume and radius were verified. The suggested method had demonstrated that it could be effective for the fast prediction of fragmentation zone.

A pheromone mutant of Schizosaccharomyces pombe displays nucleolar fragmentation

  • Stresses and nutritional starvation are two main external signals for the induction of sex pheromones in the fission yeast Schizosaccharomyces pombe. In an attempt to identify the components involved in transduction of starvation signals, we screened 135 temperature-sensitive (ts) mutants and isolated 6 mutants that induced the pheromone even in the presence of a nitrogen source. These mutants exhibited two distict induction phenotypes: pheromone induction at restrictive but not at permissive temperatures; and pheromone induction at both permissive and restrictive temperatures. The times required for the maximum pheromone induction at the restrictive temperature differed slightly in each mutant. In addition to the pheromone induction phenotype, the ts243 and ts304 mutants exhibited cell-division-cycle defects. The ts304 mutant cells showed an abnormal cytoplasmic DAPI staining pattern. The nucleolus of this mutant seemed to be fragmented, a phenomenon which is typically observed in aged yeast cells. The result of our genetic analysis indicated that the pheromone induction mutants belonged to 6 separate complementation groups. We designated these mutants pws1 to pws6.

Enhancing mechanism of the saccharification of uncooked starch in an agitated bead reaction system (무증자전분의 분쇄마찰매체에 의한 효소당화촉진 Mechanism의 규명)

  • In an agitated bead reaction system, the enzymatic saccharification of uncooked starch was substantially enhanced. The enhancement mechanism was investigated front the view of the structural aspect of starch. The mechanical impact caused by the movement of the attrition-milling media resulted neither the destruction of microcrystalline structure nor the fragmentation of starch granule. instead, the most distinct phenomenon was the swelling of starch granule up to about 2.5 times, and the swelling mechanism was not similar with that caused by cooking. However, in the case of the enzyme addition in the attrition coupled reaction system, the swollen starch was easily fragmented into the large number of small particles by the synergistic action of the enzyme and milling-media. The exposed surface area of the fragmented particles plays the major role in enhancing the saccharification. The saccharification rate was quite different depending on the source of starch, the reason was discussed in terms of the granular structure of uncooked starches.

Antimicrobial and Anticancer Activity of Korean Traditional Soy Sauce and Paste with Chopi (초피첨가 전통장류의 항균 및 항암활성)

  • The fruits of Zanthoxylum piperitum are known as having various physiology vitality, and the abstraction ingredient of the pericarp is also known as having strong antibiotic activities against various bacteria. Therefore, this study was carried out to estimate the effect of physiology vitality when the abstraction ingredient of Z. piperitum was added in soy sauce(Chopi-kanjang) and soybean paste(Chopi-doenjang). For the antibiotic activity against the pathogens of sitotoxism such as Staphylococcus aureus, Salmonella typhimurium, Vibrio parahemolyticus, Escherichia coli 0157:H7, the extracts of the Chopi-kanjang was added 1%, 2%, 4% pericarp of Z. piperitum in the manufacturing process of soy source. According to the results, the growth of E. coli 0157:H7 and V. parahemolyticus were respectively inhibited as 70% and 50% by the Chopi-kanjang added 2% of the ingredient. For the antibiotic effects of the aforementioned Chopi-kanjang against Sal. typhimurium and Sta. aureus, the growth of those pathogens was also inhibited between 40% and 60% according to the manufacturing period of Chopi-kanjang. It was confirmed that the antibiotic activity using the mixture of the abstraction ingredient and Chopi-doenjang was lower than those of Chopi-kanjang. In order to estimate the anticancer activity using by caspase-3 activity, the mixture of the abstraction ingredient of the pericarp of Z. piperitum and Chopi-kanjang was treated to leukemia cells. According to the results, the activities of caspase-3 using the mixture added 1%, 2% and 4% of the abstraction ingredient were respectively increased as much as 4, 12, 15 times comparing with the control which was treated with the soy source only. It could be that the mixture of the abstraction ingredient of the pericarp of Z. piperitum and soy source induced apoptosis, and the mixture of the abstraction ingredient and soybean paste had no effect on the activity of caspase-3. In order to find out the death of the aforementioned cells caused by necrosis or apoptosis, DNA fragmentation in the cell was examined. U-937 cells showed apoptotic DNA fragmentation in the incubation with Chopi-kanjang extract.

XML Fragmentation for Resource-Efficient Query Processing over XML Fragment Stream (자원 효율적인 XML 조각 스트림 질의 처리를 위한 XML 분할)

  • In realizing ubiquitous computing, techniques of efficiently using the limited resource at client such as mobile devices are required. With a mobile device with limited amount of memory, the techniques of XML stream query processing should be employed to process queries over a large volume of XML data. Recently, several techniques were proposed which fragment XML documents into XML fragments and stream them for query processing at client. During query processing, there could be great difference in resource usage (query processing time and memory usage) depending on how the source XML documents are fragmented. As such, an efficient fragmentation technique is needed. In this paper, we propose an XML fragmentation technique whereby resource efficiency in query processing at client could be enhanced. For this, we first present a cost model of query processing over XML fragment stream. Then, we propose an algorithm for resource-efficient XML fragmentation. Through implementation and experiments, we showed that our fragmentation technique outperformed previous techniques both in processing time and memory usage. The contribution of this paper is to have made the techniques of query processing over XML fragment stream more feasible for practical use.

Evaluation of Water Quality Impacts of Forest Fragmentation at Doam-Dam Watershed using GIS-based Modeling System (GIS 기반의 모형을 이용한 도암댐 유역의 산림 파편화에 따른 수(水)환경 영향 평가)

  • The water quality impacts of forest fragmentation at the Doam-dam watershed were evaluated in this study. For this ends, the watershed scale model, Soil and Water Assessment Tool (SWAT) model was utilized. To exclude the effects of different magnitude and patterns in weather, the same weather data of 1985 was used because of significant differences in precipitation in year 1985 and 2000. The water quality impacts of forest fragmentation were analyzed temporarily and spatially because of its nature. The flow rates for Winter and Spring has increased with forest fragmentations by $8,366m^3/month$ and $72,763m^3/month$ in the S1 subwatershed, experiencing the most forest fragmentation within the Doam-dam watershed. For Summer and Fall, the flow rate has increased by $149,901m^3/month$ and $107,109m^3/month$, respectively. It is believed that increased flow rates contributed significant amounts of soil erosion and diffused nonpoint source pollutants into the receiving water bodies. With the forest fragmentation in the S1 watershed, the average sediment concentration values for Winter and Spring increased by 5.448mg/L and 13.354mg/L, respectively. It is believed that the agricultural area, which were forest before the forest fragmentation, are responsible for increased soil erosion and sediment yield during the spring thaw and snow melts. For Spring and Fall, the sediment concentration values increased by 20.680mg/L and 24.680mg/L, respectively. Compared with Winter and Spring, the increased precipitation during Summer and Fall contributed more soil erosion and increased sediment concentration value in the stream. Based on the results obtained from the analysis performed in this study, the stream flow and sediment concentration values has increased with forest fragmentation within the S1 subwatershed. These increased flow and soil erosion could contribute the eutrophication in the receiving water bodies. This results show that natural functionalities of the forest, such as flood control, soil erosion protection, and water quality improvement, can be easily lost with on-going forest fragmentation within the watershed. Thus, the minimize the negative impacts of forest fragmentation, comprehensive land use planning at watershed scale needs to be developed and implemented based on the results obtained in this research.
