The Hsp90 chaperone machinery: from structure to drug development

  • Hsp90, an evolutionarily conserved molecular chaperone, is involved in the folding, stabilization, activation, and assembly of a wide range of 'client' proteins, thus playing a central role in many biological processes. Especially, several oncoproteins act as Hsp90 client proteins and tumor cells require higher Hsp90 activity than normal cells to maintain their malignancy. For this reason, Hsp90 has emerged as a promising target for anti-cancer drug development. It is still largely unknown how Hsp90 can recognize structurally unrelated client proteins. However, recent progress in structural studies on Hsp90 and its interaction with various co-chaperones has broadened our knowledge of how the Hsp90 ATPase activity, which is essential for its chaperone function, is regulated and coupled with the conformational changes of Hsp90 dimer. This review focuses on the roles of various Hsp90 co-chaperones in the regulation of the Hsp90 ATPase cycle, as well as in the selection of client proteins. In addition, the current development of Hsp90 inhibitors based on the structural information will be discussed.

Secretory Production of Human Leptin in Bacillus subtilis

  • Human leptin is identified as a 16kDa (146 amino acids) protein secreted from adipocytes which influences body weight homeostasis. In order to produce active leptin, the human obese gene coding for leptin was expressed in Bacillus subtilis WB600 strain which is deficient in six extracellular proteases. The recombinant leptin was produced in a culture supernatant, and in a culture supernatant, it was contained as high as 48% for total proteins. After simple purification steps, which consisted of ammonium sulfate precipitation and anion-exchange column chromatography, 2.3 mg of leptin with a purity greater than 95% was obtained from the 0.51 culture with the recovery yield of 38.3%. The purified leptin showed the correct folding structure with one disulfied bond.

A Study on Excavated Costumes of the Go′s of Jang Taek (광주출토 장택고씨 복식유물에 관한 연구)

  • This paper studied the excavated costumes of the Go's of Jang Taek that had been found in Kwangju metropolitan city in 1986. The excavated costumes mainly show Po(포) including Chopni(첩리), Dapho(답호), Danryung(단령), Jikryung (직령). In addition, there are some kinds of trousers, hat, korean socks. The meanings of excavated costumes of the Go's of Jang Taek are the followings: 1. They offer the important research materials to studying of general dress. 2. They offer actual proof materials of 15-16 century in opposition to depending on documentary records. 3. They offer the useful informations on the form, textile, dyeing because of good condition. 4. They offer the variety of Po that many persons weared at that times. 5. They offer the practical and rational construction by folding pleats and needlework. 6. They did not quilted padded clothes in comparison with other dress at that times. 7. They offer the important materials on the trousers. In the near future, we will study excavated costumes on dyeing, textile structure, trousers formation and so on.

Changes in Structural and Functional Responses of Bacterial Communities under Different Levels of Long-Term Compost Application in Paddy Soils

  • Soils amended for long-term with high levels of compost demonstrated greater abundance of bacterial members of the phylum Bacteroidetes whereas a decreasing trend in the relative abundance of phylum Acidobacteria was noted with increasing levels of compost. Metabolic profiles predicted by PICRUSt demonstrated differences in functional responses of the bacterial community according to the treatments. Soils amended with lower compost levels were characterized by abundance of genes encoding enzymes contributing to membrane transport and cell growth whereas genes encoding enzymes related to protein folding and transcription were enriched in soils amended with high levels of compost. Thus, the results of the current study provide extensive evidence of the influence of different compost levels on bacterial diversity and community structure in paddy soils.

  • The purpose of this study is to analyse the inclinations and expressions in contemporary architecture. Specially, we call this tendency and architectural movements as architectural metamorphosis. Metamorphosis in architecture present the core of the change of Forms and spirits in a change of outward shape and terrestrial identity. As in Ovid's extended dramatic poem of change and transformation, Metamorphoses, all Souls are deathless, and migrates from one form to another. Like these stories in Metamorphoses, Ovid tells the soul never dies, but leaps one form to anther, and can take any shape. So the architectural form, transformation and deformation in contemporary architecture means architectural sensations and cognitions can even approach the soul of form and shape under the transformation. The expressions and design strategies of metamorphosis in comtemporary architecture reveal continuous and sequential formations of space, linear structure with force and vector, rhythmical wavement and folding surface, lively wiggly flows of volumns and objects, and so on. Such qualities came from the periodical needs; separation of structure and surface, poly-surfacial movement, poly-sensual expression and experience, dematerialization and the dematerialized space, formless of non-formal architecture, digital architecture. Architecture of Metamorphosis is the ways and the needs of our period to overcome the static limits prohibits the liberal thoughts, to find the ways toward the opportunities and diversities and to unlock the imaginaire of the contemporary architecture.

Assignment of ¹H and $^{13}C$ Nuclear Magnetic Resonances of Ganglioside $G_{A1}$

  • Investigation of the structures of the gangliosides has proven to be very important in the understanding of their biological roles such as regulation of differentiation and growth of cells. We used nuclear magnetic resonance spectros-copy in order to investigate the structure of GA1. In order to do this, the assignment of spectra is a prerequisite. Since GA1 does not have polar sialic acid, the spectral overlap is severe. In order to solve this problem, we use 2D NMR spectroscopy and heteronuclear 1H/13C correlated spectroscopy in this study. Here, we report the complete assignment of the proton and the carbon spectra of the GA1 in DMSO-d6-D20 (98:2, v/v). These assignments will be useful for interpreting 1H and 13C NMR data from uncharacterized oligosaccharides and for determining the linkage position, the number of sugar rings, and the sequence of new ganglioside. Amide proton in ring Ⅲ shows many interring nOes and has intramolecular hydrogen bonding. This appears to be an important factor in tertiary folding of GA1. Based on this assignment, determination of three dimensional structure of GA1 will be carried out. Studies on the conformational properties of GA1 may lead to a better understanding of the molecular basis of its functions.

A Comparative Study on the Style of Clothing of Goguryeo and Xianbei Tribe - Focused on Northern Wei Period - (고구려와 선비족 의복 형태 비교 연구 - 북위시기를 중심으로 -)

  • The purpose of this study is to compare the style of clothing by type of the Goguryeo and Xianbei Tribe based on the examination of the literature centered on the historical relationship between Goguryeo and Xianbei Tribe. The research results are as follows. First, it was found that both the apparels of Goguryeo and Xianbei Tribe had hemp cloth with the V-neckline with the blocked front part and hemp cloth with the open front that is worn by overlapping the straight collars of the upper garment. Both men and women wore the garment of the straight line collar that is worn by folding towards the left or right, or upper garment of the round shaped collar. Women wore an upper garment with short sleeves on top of the upper garment of Korean traditional clothes while men wore pants for the lower garment, and women wore skirt and apron with triangular cloth decoration on top of the skirt. Second, Goguryeo Anak Tomb No. 3 mural painting(year 357) shows that apparel worn by the lady who is the wife of the owner of the tomb is similar to the apparel worn by the lady in the lacquer painting for folding screen of the Sima Jinlong Tomb ancient painting on the woman of chaste reputation in terms of the form and structure.

Analysis of Furniture Market in General Merchandise Stores (대형마트에서 판매되는 가구 시장분석)

  • This study intended to explore trends of furniture sold in general merchandise stores such as Lotte mart, E-mart, GSmart, Home ever, and Home plus, which are run by conglomerates in Korea. Through internet, related books and papers, interviews with the mart-related people, styles, items, prices, manufacturers, and materials of furniture well sold in the marts were researched and analyzed qualitatively. As the result of the analysis, the furniture expressed the scale to be able to put in the cart of the marts, knock-down and folding structure to be easy to disassemble, pack, and move, low prices below one hundred thousand won, the light materials like plastic, aluminum and the more MDF than hard wood. Each item was made of PB reflecting lower prices of 10 to 20 percentage for the consumer.

Organic Geometry in Isabel Toledo's Collections (이사벨 톨리도 컬렉션에 나타난 유기적 기하학)

  • This study examines the organic geometry in Isabel Toledo's collections in terms of the practicality of American sportswear tradition. This study conducts literary survey combined with case analysis of Toledo's works from her debut collection in 1985 to the recent ones. The organic geometry in Toledo's designs refers to the conversion of two-dimensional garment patterns into three-dimensional garment forms with the body as a medium, which is classified into the following categories in this study. First, 'fluidity' describes Toledo's highly fluid jersey dresses which maintain consistent structures by patchwork draping and suspension technique. Second, 'reductionist structure' illustrates that simple geometric shapes such as circles and squares disappear as soon as worn on the body. Third, 'origami construction' explains folding two-dimensional fabrics into three-dimensional forms, which causes the outlines of the body to appear abstract. Toledo's designs deliver the tradition of American sportswear through the organic geometry of garment construction. Toledo's works are authentic American in the aspects that they are functional and modern; they satisfy the practical needs, prioritize the movements of wearers, pursue multi-functions, and their ornamental elements are accompanied by the construction of garments. Isabel Toledo presents designs drawing on her unwavering aesthetics while continuously developing and experimenting creative ways of garment construction.

Effect of A-Zeolite on the Crystallization Behavior of In-situ Polymerized Poly(ethylene terephthalate) (PET) Nanocomposites

  • The crystallization behavior and fine structure of poly(ethylene terephthalate) (PET)/A-zeolite nanocomposites were assessed via differential scanning calorimetry (DSC) and time-resolved small-angle X-ray scattering (TR-SAXS). The Avrami exponent increased from 3.5 to approximately 4.5 with increasing A-zeolite contents, thereby indicating a change in crystal growth formation. The rate constant, k, evidenced an increasing trend with increases in A-zeolite contents. The SAXS data revealed morphological changes occurring during isothermal crystallization. As the zeolite content increased, the long period and amorphous region size also increased. It has been suggested that, since PET molecules passed through the zeolite pores, some of them are rejected into the amorphous region, thereby resulting in increased amorphous region size and increased long period, respectively. In addition, as PET chains piercing into A-zeolite pores cannot precipitate perfect crystal folding, imperfect crystals begin to melt at an earlier temperature, as was revealed by the SAXS profiles obtained during heating. However, the spherulite size was reduced with increasing nanofiller content, because impingement between adjacent spherulites in the nanocomposite occurs earlier than that of homo PET, due to the increase in nucleating sites.
