In order to investigate the antitumor effect by Dangguihwalhyultang after B-l6 cells were transplanted in C57BL/6 mice, and the immune responses in mice induced by methotrexate, the extract of Dangguihwalhyultang was orally administered to the ICR mice. Experimental studies were performed for measurance of metastasis, cell cytotoxicity in vitro, life extention, weight of cancer, natural killer cell activity. productivity of interleukin-2. The results were summarized as follows: 1. Mean survival time in Dangguihwalhyultang-treated group was prolonged, as compared with control group(14.63%) significantly(P<0.05). 2. Inhibition of metastasis in Dangguihwalhyultang-treated group was higher than control group with significance on 14th day(P<0.05). 3. On the weight of solid tumor. Dangguihwalhyultang-treated group was less than control group with significance(P<0.05). 4. On the MTT assay. Dangguihwalhyultang concentration inhibited cell viability was $368.8{\mu}g/well$. 5. Natural killer cell activity in Dangguihwalhyultang-treated group was significantly increased on 100:1, 50:1 E/T(effect cell/target cell) ratio(P<0.05). 6. Production of interleukin-2 in Dangguihwalhyultang-treated group was significantly increased(P<0.05).

This study was conducted to evaluate the effect of additives (antioxidants for free radicals reduction) in cigarette filter treated with various antioxidants (three types of proanthocyanidins and ascorbic acid) and various concentrations of ascorbic acid and loaded with activated carbon on the delivery of free radicals of mainstream smoke (MS) by ESR. Also, we analyzed Hoffmann's analytes andscavenger activity according to the storage time and in vitro cytotoxicity. The analysis of spin number of vapor and particulate phase free radicals in MS are decreased to $14{\sim}24\;%$ and $16{\sim}40\;%$, respectively. As aresult of antioxidant potential for inactivity of vapor and particulate phase free radicals, natural antioxidants were more effective than ascorbic acid. Based on the result of the analysis of Hoffmann's analytes for various antioxidantstreated cigarette filters during the smoking, cigarette filter treated with ascorbic acid showed the lower amount of the deliveries of hydroquinone, isoprene and quinoline in MS than those treated with the other antioxidants. In the significant t-test on the difference of the cytotoxicity among the various antioxidants treated-cigarette filters, there are no significant differences at the 95 % confidence level. Those results indicated that the antioxidants were useful for reducing free radicals in MS because of the fast reaction between antioxidant and free radicals.

This study was carried out to prove the effects of CBTLC on the $5{\alpha}$-reductase inhibition, the sterilizing power for Propionibactrium acnes, the cytotoxicity of human monocyte, the inhibition for prosta-glandins($PGE_2$), Interleukins(IL-$1{\beta}$) and TNF-${\alpha}$ in inflammation, and the size of Hamster ear sebaceous gland concerned with Acnes. The result were obtained as follows : 1. On the $5{\alpha}$-reductase inhibition of CBTLC in vitro, An undiluted solution of CBTLC was $71{\%}$ inhibition on $5{\alpha}$-reductase and $\frac{1}{10}$ diluted solution of CBTLC was $20{\%}$ inhibition on $5{\alpha}$-reductase. 2. On the sterilizing power for Propionibactrium acnes concerned in Acnes, an undiluted solution and $\frac{1}{10}$ diluted solution of CBTLC formed 12mm clear zone diameters. 3. CBTLC was showed the cytotoxicity of human monocyte in $0.08{\%}$ and $0.12{\%}$ of CBTLC. 4. $0.01{\%}$ and $0.04{\%}$ of CBTLC inhibited the production of prostaglandins($PGE_2$) in the human monocyte stimulated with P. acnes LPS. 5. $0.08{\%}$ and downward of GMHBS inhibited the production of interleukins(lL-$1{\beta}$) in the human monocyte stimulated with P. acnes LPS. 6. $0.08{\%}$ and $0.12{\%}$ of GMHBS inhihited the production of TNF-${\alpha}$ in the human monocyte stimulated with P. acnes LPS. 7. As the antiandrogenic compound, CBTLC was used in hamster ears with topical application. CBTLC was diminished slightly on the size of hamster ears sebaceous gland.
Objective: This study was carried out to investigate the effects of Gami-Shengmayuipung-tang on acne. Methods : The effects of Gami-Shengmayuipung-tang (SYTSRG) on acne were measured by the $5\alpha-reductase$ inhibition, the sterilizing power for Propionibactrium acnes, the cytotoxicity of human monocyte, the inhibition for prostaglandins$(PGE_2)$, interleukins $(IL-l\beta)$ and $TNF-\alpha$ in inflammation, and the size of the hamtster ear sebaceous gland related to P. acnes. Results: On the $5\alpha-reductase$ inhibition of SYTSRG in vitro, an undiluted solution of SYTSRG showed $80\%$ inhibition on $5\alpha-reductase$ and 1/10 diluted solution of SYTSRG showed $40\%$ inhibition on $5\alpha-reductase$. On the sterilizing power for Propionibactrium acnes related acne, an undiluted solution and 1/10 diluted solution of SYTSRG formed $12\beta{\AE}$ clear zone diameters. SYTSRG did not show cytotoxicity of human monocyte. Concentrations of $0.01\%\;and\;0.04\%\;and\;0.08\%$ of SYTSRG inhibited the production of prostaglandins $(PGE_2)$ in the human monocyte stimulated with P. acnes LPS. $0.08\%$ and less of SYTSRG inhibited the production of interleukins $(IL-l\beta)$ in the human monocyte stimulated with P. acnes LPS. Concentrations of $0.04\%,\;0.08\%\;and\;0.12\%$ of SYTSRG inhibited the production of $TNF-\alpha$ in the human monocyte stimulated with P. acnes LPS. As the antiandrogenic compound, SYTSRG was used in hamster ears with topical application. SYTSRG diminished the size of the hamster ear sebaceous gland in males, but not in females. Conclusion: The present data suggest that SYTSRG may affect the primary stage of inflammation of acne.
Statement of problem : Resin cements were used widely on all ceramic crowns, but the influence of resin cements on biocells was not understood clearly. Purpose : This study was investigated to evaluate the biocompatibility of resin cements for all-ceramic crowns. Material and Method : The resin cements used in this study were Panavia F (Kuraray Co., Ltd. Japan), Variolink II (Vivadent Ets., Schann / Liechtenstein), and Bistite II (Bistite dual cure resin cement-clear Tokuyama Soda Co. Japan). The viability of normal human oral keratocytes, gingival fibroblast, and gingival fibroblast immortalized by Human Papilloma virus 16 was measured in vitro for evaluation of cytotoxicity on resin cements, and the response of pulp tissue was analyzed and evaluated with light microscope after application of cements at cutting edge of incisors. Results : The normal human oral keratocytes was the most sensitive to toxicity of resin cement, and toxicity of cements was higher in Bistite II than in Variolink II. The cell viability of immortalized gingival fibroblast did not affected by type of cement and cultivation period, but there was a tendency that cytotoxicity in Bistite II was higher than in Variolink II. The cell viability of gingival fibroblast was similar to that of immortalized gingival fibroblast regardless of cement type, but Bistite II showed more toxic than others after 5 days cultivation. The responses of pulp tissue according to cement type were similar after 2 days cultivation, but revealed high toxicity in Bistite II after 10 days cultivation. Conclusion : Variolink II was more biocompatible than any other resin cements used in this study.
Objectives: Whether the ultra-highly-diluted remedies used in homeopathy can effectively bring about modulations of gene expressions through acetylation/deacetylation of histones has not been explored. Therefore, in this study, we pointedly checked if the homeopathically-diluted anti-cancer remedy Condurango 30C (ethanolic extract of Gonolobus condurango diluted $10^{-60}$ times) was capable of arresting the cell cycles in cervical cancer cells HeLa by triggering an epigenetic modification through modulation of the activity of the key enzyme histone deacetylase 2 vis-a-vis the succussed alcohol (placebo) control. Methods: We checked the activity of different signal proteins (like $p21^{WAF}$, p53, Akt, STAT3) related to deacetylation, cell growth and differentiation by western blotting and analyzed cell-cycle arrest, if any, by fluorescence activated cell sorting. After viability assays had been performed with Condurango 30C and with a placebo, the activities of histone de-acetylase (HDAC) enzymes 1 and 2 were measured colorimetrically. Results: While Condurango 30C induced cytotoxicity in HeLa cells in vitro and reduced HDAC2 activity quite strikingly, it apparently did not alter the HDAC1 enzyme; the placebo had no or negligible cytotoxicity against HeLa cells and could not alter either the HDAC 1 or 2 activity. Data on $p21^{WAF}$, p53, Akt, and STAT3 activities and a cell-cycle analysis revealed a reduction in DNA synthesis and G1-phase cell-cycle arrest when Condurango 30C was used at a 2% dose. Conclusion: Condurango 30C appeared to trigger key epigenetic events of gene modulation in effectively combating cancer cells, which the placebo was unable to do.

This study attempted to investigate the possibility of the use of Morinda citrifolia (MC) as a cosmetic ingredient from its physiological activities such as antioxidant activity, cytotoxicity and anti-aging effect. MC is a tropical plant that has been used as traditional polynesian foods and medicines for over two thousand years. It has been reported that this shrub can improve antimicrobial, anti-cancer and anti-inflammatory effects and strengthen an immune system. The in vitro antioxidant activity of MC was performed to see the DPPH scavenging activity by measuring total polyphenol content and total flavonoid content. As a result, a lack of any cytotoxicity was confirmed in human dermal fibroblasts (HDF) cell. When MC extract at a concentration of over $50{\sim}100{\mu}g/mL$ was added, MMP-1 expression considerably diminished. In an in vivo test, in addition, cream containing MC extract was prepared and applied to a total of 22 women in their 30 ~ 50s in ages in the morning and in the evening for four weeks. Changes in keratin, melanin index, pore, skin color and wrinkles under the naked eyes were then comparatively measured. Keratin levels slightly increased in the control group but decreased in the experimental group. In addition, wrinkles diminished in the experimental group. This study found that MC extract controls many MMP-1 related mechanisms with great potential for use as a natural ingredient of anti-aging cosmetics.
All fifteen flavonoids (1-15) have been isolated from the roots of Sophora flavescens (Leguminosae) as active principles of the cytotoxic property toward human tumor cell lines such as A549, SK-OV-3, SK-Mel-2, XF498 and HCT15, in vitro. A series of new diarylureas and diarylamides possessing 1,3,4-triarylpyrazole scaffold was synthesized and their in vitro antiproliferative activities against A375P human melanoma cell line and NCI-60 cell line panel were tested. Compounds 9, 11, 12, 14, and 17-21 showed superior potency against A375P to Sorafenib. Over the NCI-60 cancer cell line panel, compound 14 possessing a methoxy group, amide linker, and 4-chloro-3-(trifluoromethyl)phenyl terminal ring showed the highest potency and broad-spectrum anticancer activity. Compound 13 showed high selectivity towards leukemia subpanel over other cancer types.
Two known isoquinoline alkaloids, (+)-chelidonine (1) and (-)-stylopine (2), were isolated from $CHCl_3$-soluble fraction of whole plants of Chelidonium majus L. var. asiaticum, and their structures were identified by spectroscopic methods and X-ray crystallographic analysis. Two isolates (1 and 2) were examined for their in vitro cytotoxic activities against five human cancer cell lines including DU-145 (prostate), MCF (breast), A549 (lung), HePG2 (liver), and HT-29 (colon) by sulphorhodamine B (SRB) assay.
