Effect of Apple Dietary Fiber on the in vitro Growth of Intestinal Bacteria (사과의 식이섬유질이 장내세균의 in vitro 생육에 미치는 영향)

  • Various fractions of apple fibers such as crude pulp, total dietary fiber, soluble dietary fiber, and insoluble dietary fiber were prepared and added to the proteose peptone-yeast extract-fildes (PYF) media to see their effects on the growth of type cultures of intestinal bacteria. Most microbes tested in this experiment grew well in PYF media with the soluble dietary fiber of apple than with the insoluble dietary fiber. Especially Bifidobacterium species such as B. adolescentis, B. animalis, B. infantis, B. longum, B. thermophilum showed higher growth in PYF media containing the soluble dietary fiber than other fiber fractions. However, pectin-added media didn't promote the growth of most microbes used in the experiment. In the in vitro mixed culture using rat feces as starter, the addition of the soluble dietary fiber or pectin to the basal medium showed larger proportion of Bifidobacterium species in total bacteria than that of glucose.

  • This research is about the pulsatile pump system utilized in the perfusion bioreactor for the in vitro human tissue culture. A pulsatile pump system which can be applied to the culture of the vascular tissues including blood vessel is developed by using the idea of human heart's blood pumping into organs as followings: culture chamber, a pressurizing device which generates laminar pulsatile flow by controlling the x-sectional area of the culture media delivering tubing, a compliance chamber which supplies the pressuring device with a constant pressure, and a peristaltic pump which circulates the culture media in a circuit ranging from the culture chamber to the compliance chamber. The developed pulsatile pump system shows that a physiology of the human heart's blood pumping including pulsatile pressure waveform of systolic-diastolic pressure is well represented. Not only time domain but also frequency domain characteristics of pulsatile pump system which are necessary for the vascular tissue culture such as pulsatile pressure waveform's shape, the frequency, and the magnitude can be easily generated and manipulated by using the proposed system.

Micropropagation of Echinosophora koreensis Nakai, a Korean Endemic Species in Danger, Using Axillary Buds

  • To establish in vitro axillary bud culture conditions of Echinosophora koreensis Nakai, one of Korean endemic endangered species famous for beautiful flowers, we tested the influence of plant growth regulators (PGRs) in shooting and rooting stage from in vitro plants. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the media induced 2.5 to 3 shoots per bud during 4 weeks of culture. And media including 0.5 mg L-1 thidiazuron (TDZ) produced 3 to 4 shoots per bud. However, zeatin and isopentenyl adenine (2-ip) were not successful to increase shoot number, and the combination treatments of BA with other PGRs were also not effective. Shoots were smaller than 2 cm in length, in most of the treatments. In rooting, naphthalene acetic acid (NAA) treatments in the range of 0.5 to 4.0 mg L-1 appeared to increase rooting rate by 10% to 60% approximately when compared with the control but roots developed with callus clusters. Indole butyric acid (IBA) addition had little effect on rooting (below 10%), while some roots were longer than in NAA treatments and some shoots were longer on high IBA concentrations (4.0 to 8.0 mg L-1). It is suggested that micropropagation is a highly applicable and promising to multiplication and conservation of rare and endangered endemic species.

In Vitro Plant Regeneration from Stolen Node Explant in Eremochloa Ophiuroides (Munro) Hack

  • In vitro shoot regeneration and multiple shoot induction has been obtained from the stolen node explants in Eremochloa ophiuroides (Munro) Hack. The highest number of shoots ($10.66{\pm}0.21$) was observed from initial explants after one month culture duration on Murashige and Skoog (MS) medium containing 6-benzyladenine (BA: 0.5 mg/l). First generation shoot was excised and sub-cultured on the same fresh media for further multiplication of shoots. An enhanced number of second round shoots ($15.33{\pm}0.21$) was obtained compared to the initial culture media containing BA (0.5 mg/l). The number of shoots/stolon node was higher among all the concentrations of BA than kinetin (KN). In vitro regenerated shoots were successfully rooted in the phytohormone free MS medium. Plantlets generated with roots were transferred to pots containing compound mixture of soil and kept in green house conditions. Acclimatized plants showed 100% survival rate with normal morphology in green house conditions. The present study demonstrates the effect of explant and different plant growth regulators towards in vitro response in E. ophiuroides. Moreover, the study reveals the effect of cytokinin on induction of shoot number per stolen node explant in E. ophiuroides.

Effects of Media Replacement and Environmental Control for in vitro Microtuber Production of Yam (Dioscorea opposita Thunb.) (둥근마(Dioscorea opposita Thunb.) 기내 소괴경 생산을 위한 배양환경과 배지의 교체효과)

  • To figure out optimum culturing condition for $in$ $vitro$ yam ($Dioscorea$ $opposita$ Thunb.) production, various media components and environmental controls were evaluated, effective temperature, light condition and timing of the liquid media replacement for multiplication of yam in liquid culture were determined in this study. There was no visible difference detected for $15^{\circ}C$ and $25^{\circ}C$ temperature conditions. At $25^{\circ}C$, continuous light condition was more effective compared to 16 hour light/ 8 hour dark condition. Effect of media replacement was tested, and approximately 5 more microtubers obtained and 70% of increase in weight was detected when media replacement was performed. Timing for media replacement was tested at 2, 4, 6, 8, and 10 weeks after initial culture for 12 weeks. Considering both number and weight of microtubers, replacement of media 6 weeks after initial culture was most effective. In terms of component of media, significant increase in weight of microtubers was observed in MS media containing sucrose $60g{\cdot}L^{-1}$. In summary, the most effective condition for $in$ $vitro$ propagation of chinese yam is replacing medium 6 weeks after initial inoculation with MS medium containing sucrose $60g{\cdot}L^{-1}$ in continuous light condition.

Establishment and long-term culture of the cell lines derived from gonad tissues of Siberian sturgeon (Acipenser baerii)

  • To culture germline stem cells in vitro, establishment of the cell lines that can be used as the feeder cells is a prerequisite. In this study, we tried to establish gonad-derived cell lines in Siberian sturgeon (Acipenser baerii). Five 1-year-old A. baerii were used as a donor of gonad tissues, and gonad-dissociated cells were cultured in vitro. Subsequently, determination of growth conditions, long-term culture, characterization, and cryopreservation of the cell lines were also conducted. Five gonad-derived cell lines were stably established and cultured continuously over at least the 73th passage and 402 culture days under the media containing 20 % fetal bovine serum at $28^{\circ}C$. All cell lines consisted of two main cell types based on morphology even if the ratio of the two cell types was different depending on cell lines. Despite long-term culture, all cell lines maintained diploid DNA contents and expression of several genes that are known to express in the A. baerii gonad. After freezing and thawing of the cell lines, post-thaw cell viabilities between 57.6 and 92.9 % depending on cell lines were indentified, suggesting that stable cryopreservation is possible. The results and the cell lines established in this study will contribute to the development of an in vitro system for A. baerii germline stem cell culture.

Silver nitrate and silver-thiosulphate mitigates callus and leaf abscission during Shisham clonal micro-propagation

  • Basal callus formation and leaf abscission is a problem in clonal micropropagation. We have described an in vitro clonal propagation protocol of Dalbergia sissoo Roxb (shisham) 'FRI-14' in which AgNO3 played important role not only in mitigating problem of leaf abscission and basal callus, but also improved shoot induction and multiplication. Best induction and shoot multiplication was obtained on MS media with 1.5 mg/l 6-BAP and 10 mg/l AgNO3 and half-strength MS media with 0.5 mg/l 6-BAP, 2 mg/l AgNO3 and 50 mg/l Adenine sulphate whereas best ex vitro rooting was obtained with 200 mg/l IBA in pulse treatment.
