천연식물자원 지모와 황백피 혼합 수추출물의 안전성평가 (Evaluation of Genotoxicity and 14-day Repeat Dose Toxicity of Water Extract of the Mixture of Natural Plants, Anemarrhena and Phellodendron)

  • The safety of a new natural plant composition (ADP) was assessed on the genotoxicity study and 14-day repeat dose toxicity study. ADP contains a mixed water extract obtained from the mixture of Phellodendron cortex (Phellodendron amurense) and Anemarrhena rhizoma (Anemarrhena asphodeloides), and poses the contractile properties mediated by alpha-adrenoceptor of the prostate and urethra as well as antioxidant and anti-inflammatory properties. In order to evaluate genetic safety, in vivo micronucleus test was performed in ICR mice orally administered with three dose levels of 1250, 2500, 5000 mg/kg body weight, and vehicle and positive control. In the 14 days study, Sprague-Dawley rats were treated with ADP at the dose levels of 500, 1000, 2000 mg/kg once a day, and clinical signs, body weights, hematology, serum biochemistry, necropsy findings and organ weights were monitored and examined. In experimental results, ADP treatment, compared with vehicle control, did not induce the micronucleated erythrocytes from mouse bone marrow. In the 14 days study, any significant and toxicological differences in all measurements of parameters were not observed in ADP treatment groups of animals, compared with vehicle treatment. The No-Observed-Adverse-Effect-Level (NOAEL) of ADP in the 14 days study was determined to be greater than 2000 mg/kg/day in both sexes.

비만치료를 목적으로 마테(Yerba Mate, Ilex paraguariensis)를 함유한 복합투여물의 효과 및 부작용에 대한 고찰 (A Review of Combination Effects and Adverse Effects of Yerba Mate (Ilex paraguariensis) on the Treatment of Obesity)

  • Objectives : The aim of this study is to review the combination effects and adverse effects of Yerba Mate (Ilex paraguariensis) on the treatment of obesity. Methods : Four databases were searched, including Pubmed, National assembly library, RISS4u, and NDSL, for studies published before March 31, 2016 using the keywords 'yerba mate', 'Ilex paraguariensis', and those related to 'obesity'. Clinical trials, and both in vivo and in vitro studies were classified according to the following categories: publication year, nationality, types and composition of test compounds, combination effects, adverse effects, and safety. Results : A total of 16 articles (14 clinical trials, one in vivo, one in vitro) were included in the analysis. Combination effects were reported from six YGD (Yerba Mate, Guarana, Damiana, Zotrim), five Meltdown, one Shred-matrix, one DBX (DymaBurn Xtreme), one XSIII research study, and adverse effects were reported from two YGD (Zotrim), four Meltdown, one Shred-matrix and one MetEF research study. YGD and Shred-matrix increased satiety while YGD and MetEF had adverse gastrointestinal effects potentially due to guarana. Meltdown and Shred-matrix increased energy expenditure and fat oxidation and had the adverse effects of increased heart rate and blood pressure due to the thermogenic effect of caffeine and yohimbine. DBX increased resting energy expenditure but no difference was observed in respiratory exchange ratio. XSIII reduced body weight gain and food intake more than each of the extracts did on their own. Conclusion : Taken together, the combination effects and adverse effects differed according to the composition of test compounds and the ratio of ingredients. Multicomponent approaches should be used for treating multifactorial diseases such as obesity. As a general conclusion, there is a place for a combination of herbal components including yerba mate, based on the principle of Kun-Shin-Choa-Sa for the treatment of obesity.

녹두(Phaseolus aureus L.) 급여가 당뇨 유발 흰쥐의 혈당 및 지질성분 개선에 미치는 영향 (Effects of Mung Bean (Phaseolus aureus L.) on Blood Glucose and Lipid Composition Improvement in Streptozotocin-induced Diabetic Rats)

  • 녹두(Phaseolus aureus L.)가 당뇨 유발 흰쥐에 있어서 혈당 및 지질성분, 단백질, 전해질 농도 등에 미치는 영향을 확인하기 위하여 기본 식이를 섭취시킨 BD군, 기본 식이군에 5% 녹두 급여군(BM군), 당뇨 유발 실험군(BS군)과 BS군에 5% 녹두를 섭취시킨 실험군(SM군)으로 나누어 실험을 수행하였다. 혈청 지질성분(총 콜레스테롤, LDL-콜레스테롤, 중성지질, 인지질, 유리 콜레스테롤, 콜레스테롤 에스테르), 혈당 및 유리지방산(non esterified fatty acid, NEFA)의 농도와 동맥경화지수(atherosclerotic index, AI), 심혈관 위험지수(cardiac risk factor, CRF)는 당뇨 유발군에서 여타 실험군과 비교하였을 때 유의적인 차이를 보이며 증가를 나타내었고(p<0.05), 5% 녹두 급여군에서 감소되는 것으로 나타났다. 당뇨유발군인 BS군에 비해 5% 녹두를 첨가시킨 SM군에서 고밀도 지단백 콜레스테롤(high density lipoprotein-cholesterol, HDL-콜레스테롤) 및 총 콜레스테롤에 대한 HDL-콜레스테롤의 농도 비는 유의적인 차이를 보이며 증가를 나타내었다(p<0.05). 혈청 단백질 농도에 있어서, 당뇨 유발군(BS군)에 5% 녹두를 급여(SM군)한 흰쥐의 혈청 알부민(albumin) 농도와 알부민/글로불린 비(albumin/globulin ratio, A/G 비)는 증가 하는 것으로 관찰되었다. 나트륨(Na) 및 염소(Cl)는 SM군에서 BS군 보다 농도가 감소된 것으로 확인하였다. 이상의 결과, 5% 녹두는 흰쥐의 혈당 및 지질대사 기능 개선에 도움이 되는 것으로 판단된다.

Korean solar salts reduce obesity and alter its related markers in diet-induced obese mice

  • BACKGROUND/OBJECTIVE: The aim of this experiments was to show anti-obesity effects of Korean solar salt from different salt fields in diet-induced obese mice. MATERIALS/METHODS: Diet-induced obesity (DIO) was induced by a high-fat diet (HFD; 45% cal from fat) in C57BL/6J mice for eight weeks. The mice were fed with the designated diets (chow diet for Normal, HFD for Control, 0.47%-salt-mixed HFD for purified salt (PS), Guerande solar salt from France (SS-G), solar salt from Y salt field (SS-Y), solar salts from T salt field (SS-T) and S salt field (SS-S)) for another eight weeks. We checked body weight, food efficiency ratio (FER) and tissue weights (liver and epididymal adipose tissue (EAT)), and observed serum concentrations of triacylglycerol (TG), total cholesterol (TC), leptin and insulin. We also evaluated gene expressions of adipogenic / lipogenic mRNAs of $C/EBP{\alpha}$, $PPAR{\gamma}$ and FAS and beta-oxidation-related factors ($PPAR{\alpha}$ and CPT-1) in liver and EAT. The mineral composition of salt samples were analyzed using inductively coupled plasma optical emission spectrometry (ICP-OES). RESULTS: SS-T and SS-S significantly reduced body weight gain, FER, and weight of EAT compared to control and other samples (P < 0.05). SS-T and SS-S also significantly decreased serum levels of TG, TC, leptin and insulin (P < 0.05). SS-T and SS-S suppressed expressions of adipogenic / lipogenic mRNAs in liver and EAT, while promoting expression of beta-oxidation-related factors. The lowest sodium concentration was observed in SS-T ($30.30{\pm}0.59%$), and the lowest sodium-to-potassium (Na/K) ratio was found in SS-S (17.81). CONCLUSIONS: Our study shows that well-processed Korean solar salt may have anti-obesity effects in vivo, probably owing to its differences in mineral composition and other components, presumably resulting from the manufacturing processes. Further research is needed into the mechanism and to explore optimal manufacturing processes.

Nutrient Requirements of Exercising Swamp Buffalo, Bubalus bubalis. II. Details of Work Energy of Cows and Its Relation to Heart Rate

  • Four young swamp buffalo cows of similar age ranging in body weight (W) between 280 to 380 kg and trained for doing physical exercise were used in two consecutive experiments, each using a latin square design, to determine energy expenditure for draught. The experiments consisted of field trials using 4 levels of work load, i.e. no work as control and loads amounting 450 to 500 Newton (N) continuous traction for respectively 1, 2 and 3 h daily for 14 consecutive days for experiment 1, and no work, traction loads equaling 5, 10 and 15% of W for 3 h daily for 14 days for experiment 2. Heart rate during rest and exercise was monitored using PE-3000 HR monitor. Cows were fed only king grass (Penisetum purpuroides) ad libitum and were subjected to balance trials. Body composition was estimated in vivo by the body density method and daily energy expenditure (EE) was calculated from ME minus RE. RE was calculated from the changes in body-protein and -fat measured before and immediately after the 14 d experimental period assuming an energy equivalent of 39.32 MJ/kg fat and 20.07 MJ/kg protein. $E_{exercise}$ ($EE_{work}\;-\;EE_{resting}$), which was the energy spent for doing the traction during 1, 2 and 3 h was 7.13, 15.45 and 19.90 MJ, respectively. $EE_{work}$ for the 1 h treatment group was 39.75 MJ/d equivalent to 1.30 times $EE_{resting}$. The values for the 2 and 3 h treatment groups were 1.75 and 1.86 times resting energy requirement, respectively. Absolute efficiency of work in all exercise trials of experiment 2 was around 27.28%. The increases of daily $E_{exercise}$ values were correlated to elevation of heart rate (HR) according to the equation $E_{exercise}=(0.270HR^{0.363}\;-\;1)$ MJ, while draught force related to heart rate according to the equation DF (N)=6.66 HR - 361.62. Blood glucose and triglyceride levels were gradually elevated with time during the course of exercise. Mean values of blood glucose were 91.7, 115.0 and 116.2 mg/dl for cows after 1, 2 and 3 h pulling loads at 15% W respectively as compared to 88.2 mg/dl prior to work. In the same order and treatment, mean blood triglyceride concentrations were 13.5, 13.3 and 14.8 mg/dl, and 11.5 mg/dl for control. For blood lactate, the values were 1.68, 1.63 and 1.66 mM, and 0.80 mM for control. Glucose was used as the major source of energy during the initial phase of exercise, but for prolonged work, fat will replace carbohydrate as the main substrate. Accumulation of lactate persisted for some time at the end of the exercise trials.

항생제 대체제로서 Dicarboxylic Acid 급여가 in vitro 반추위 발효성상, 착유우의 유량 및 유성분에 미치는 영향 (Effects of Dicarboxylic Acid as an Alternative to Antibiotic on in vitro Rumen Parameters, Milk yield and Milk Compositions in Lactating Cows)

  • 본 연구는 항생제 대체제로서 Dicarboxylic acid가 in vitro 반추위 발효성상, 착유우의 유량 및 유성분에 미치는 영향을 조사하기 위한 목적으로 실시하였다. 반추위 발효성상을 조사하기 위하여 batch culture 방법을 이용하였으며, Dicarboxylic acid의 급여가 착유우의 유량 및 유성분에 미치는 영향을 조사하기 위하여 평균체중 $649{\pm}19 kg$으로 일일 산유량 $38{\pm}3.95kg$, 유지방 $3.22{\pm}0.23%$, 산 차 $2.5{\pm}1.43$차인 홀스타인 착유우 10두를 산 차 및 산유량 등을 고려하여 시험사료에 따른 2개 군(대조구 5두, Dicarboxylic acid 급여구 5두)으로 나누어 실험을 실시하였다. Dicarboxylic acid는 사료급여량의 2%를 첨가하여 1일 2회 오전과 오후 착유 시 급여하였다. Dicarboxylic acid는 반추위내 pH에 긍정적인 영향을 주어 건물소화율이 유의적으로 증가하였다. 따라서 Dicarboxylic acid는 반추위 발효환경에 긍정적인 영향을 미치는 것으로 판단된다. 건물소화율은 Dicarboxylic acid를 단독 급여할 때 가장 높았다. 또한 Dicarboxylic acid를 추가 급여하면 반추위 내 총 VFA 함량이 증가하는 경향을 나타내었다. Acetic acid 농도는 Dicarboxylic acid 단독 처리구에서 높게 생성되는 경향을 나타내었다. 착유우의 유량은 Dicarboxylic acid를 급여한 처리구에서 대조구 대비 약 14%의 유량이 증가하였다. Dicarboxylic acid는 유성분 및 MUN에는 영향을 미치지 않는 것으로 판단된다. 따라서 항생제 대체제로서 Dicarboxylic acid 첨가는 반추위 내 발효환경을 안정화하여 건물소화율을 향상시키고 착유우의 유량을 높일 수 있을 것으로 판단된다.

Effects of Lactic Acid Bacteria, Storage Temperature and Period on Fermentation Characteristics, and in vitro Ruminal Digestibility of a Total Mixed Ration

  • This study evaluated the effect of lactic acid bacteria (LAB, a mixture of Enterococcus faecium and Lactobacillus plantarum) supplementation, the storage temperature, and storage period on the fermentation characteristics and in vitro ruminal digestibility of a total mixed ration (TMR). The TMR was prepared into two groups, namely, CON (control TMR without the LAB) and ML (supplementing a mixture of E. faecium and L. plantarum in the ratio of 1% and 2% (v/w), respectively). Both groups were divided and stored at 4℃ or 25℃ for 3, 7, and 14 d fermentation periods. Supplementing LAB to the TMR did not affect the chemical composition of TMR except for the lactate and acetate concentration. Storage temperatures affected (p<0.05) the chemical composition of the TMR, including pH, lactate, and acetate contents. The chemical composition of TMR was also affected (p<0.05) by the storage period. During in vitro rumen fermentation study, the ML treatment showed lower (p<0.05) dry matter digestibility at 24 h incubation with a higher pH compared to the CON. There was no difference in the in vitro dry matter digestibility (IVDMD) of TMR between the CON and ML treatment however, at 24 h, ML treatment showed lower (p<0.05) IVDMD with a higher pH compared to the CON. The effects of storage temperature and period on IVDMD were not apparent at 24 h incubation. In an in vivo study using Holstein steers, supplementing LAB to the basal TMR for 60 d did not differ in the final body weight and average daily gain. Likewise, the fecal microbiota did not differ between CON and ML. However, the TMR used for the present study did include a commercial yeast in CON, whereas ML did not; therefore, results were, to some extent, compromised in examining the effect of LAB. In conclusion, storage temperature and period significantly affected the TMR quality, increasing acetate and lactate concentration. However, the actual effects of LAB supplementation were equivocal.

산 완충용액의 pH가 인공치근우식의 형성에 미치는 영향 (THE INFLUENCE OF pH ON THE FORMATION OF ARTIFICIAL ROOT CARIES IN ACID BUFFER SOLUTION)

  • Recently root caries is reported as an increasing oral disease Many researchers studied in vivo and in vitro enamel lesions but due to its peculiar structure and different chemical composition compared to the enamel, little effort has been made to root caries study. The purpose of this study is to compare and to evaluate the effect of pH on the progression of artificial root caries lesion. Under the constant degree of saturation, the specimens were divided into 4 groups(pH 4.3, 5.0, 5.5, 6.0). Each group was immersed in acid buffer solution for 1, 3, 5, 7 days and examined using the polarizing microscope. The results were as follows: 1. The body of the lesion in the dentin showed higher degree of positive birefringence compared to sound dentin. 2. The rate of progression of the lesion slowed as the pH increased. 3. In all groups, the lesion progressed rapidly at the initiation or the experiment but increased gradually as time elapsed. In conclusion, the study shows that the pH in the acid buffer solution influenced the rate of progression of the lesion in artificial root caries. Compared to the previous enamel study, the initial pattern of the lesion progressed rapidly and as time elapsed increased gradually.

조피볼락의 사료원으로 갈색거저리의 영양적 가치 및 소화율 평가 (Nutritional Value and Digestibility of Tenebrio molitor As a Feed Ingredient for Rockfish (Sebastes schlegeli))

  • Tenebrio molitor larvae, also known as yellow mealworms (MW), are rich in protein and lipid and can serve as a potential alternative protein and energy source in commercial aquafeeds. Therefore, this study attempts to evaluate the effects of different drying methods on the nutritional value of MW meal. For this, live MW were cold-anaesthetized before being subjected to three different types of drying methods, including freeze-drying, oven-drying at $60^{\circ}C$ and air-drying at room temperature for three days, and compared for proximate composition and energy content. An in-vivo digestibility test was then conducted to evaluate the nutrient digestibility of MW meal in diets for rockfish, Sebastes schlegeli. A test diet was prepared by mixing the MW meal with a reference diet (Ref) in a 30:70 ratio with chromium oxide as an inert marker at the inclusion level of 0.5%. Rockfish with mean body weight of 150 g were stocked into a fecal collection system equipped with fiberglass tanks of 400 L capacity. Each group of fish was fed one of the experimental diets to apparent satiation for 4 weeks. The results of the proximate analysis showed that drying methods had no significant effect on crude protein, crude lipid, ash and energy contents of MW. Despite being a rich source of protein and lipid, MW meal was deficient in certain amino acids, particularly methionine, and highly unsaturated fatty acids, particularly 22:6n-3 (DHA) and 20:5n-3 (EPA). MW meal showed high digestibility values for protein (93%), lipid (97%) and energy (88%). These results may indicate that MW meal is a nutritious and acceptable feed ingredient, with comparable digestibility values to conventional animal and plant feedstuffs such as fish meal and soybean meal, in practical diet for rockfish at grower stage.

Effects of coffee intake on airway hypersensitivity and immunomodulation: an in vivo murine study

  • BACKGROUND/OBJECTIVES: Coffee is a complex chemical mixture, with caffeine being the most well-known bioactive substance. The immunomodulatory and anti-inflammatory properties of coffee and caffeine impact health in various aspects, including the respiratory system. The objective is to investigate the effects of coffee and caffeine on airway hyperresponsiveness and allergic reactions, as well as to analyze and compare associated cytokine profiles. MATERIALS/METHODS: BALB/c mice were intraperitoneally sensitized with ovalbumin (OVA) and given OVA inhalation to induce airway hypersensitivity. Two weeks after sensitization, they were intragastrically gavaged with coffee or caffeine, both containing 0.3125 mg caffeine, daily for 4 weeks. Control mice were fed with double-distilled water. Serum OVA-specific antibody levels were measured beforehand and 5 weeks after the first gavage. Airway hyperresponsiveness was detected by whole body plethysmography after gavage. Cytokine levels of bronchoalveolar lavage and cultured splenocytes were analyzed. RESULTS: Coffee effectively suppressed T helper 2-mediated specific antibody response. Airway responsiveness was reduced in mice treated with either coffee or caffeine. Compared to the control, coffee significantly reduced OVA-specific immunoglobulin (Ig) G, IgG1 and IgE antibody responses (P < 0.05). Caffeine also attenuated specific IgG and IgG1 levels, though IgE level was unaffected. Coffee significantly reduced interleukin (IL)-4 and increased IL-10 concentration in spleen cells and bronchoalveolar lavage fluid (P < 0.05). CONCLUSIONS: Coffee effectively attenuated airway hyperresponsiveness and systemic allergic responses induced by OVA food allergen in mice. As a complex composition of bioactive substances, coffee displayed enhanced immunomodulatory and anti-inflammatory effects than caffeine.
