Prediction from Linear Regression Equation for Nitrogen Content Measurement in Bentgrasses leaves Using Near Infrared Reflectance Spectroscopy (근적외선 분광분석기를 이용한 잔디 생체잎의 질소 함량 측정을 위한 검량식 개발)

  • Near Infrared Reflectance Spectroscopy(NIRS) is a quick, accurate, and non-destructive method to measure multiple nutrient components in plant leaves. This study was to acquire a liner regression equation by evaluating the nutrient contents of 'CY2' creeping bentgrass rapidly and accurately using NIRS. In particular, nitrogen fertility is a primary element to keep maintaining good quality of turfgrass. Nitrogen, moisture, carbohydrate, and starch were assessed and analyzed from 'CY2' creeping bentgrass clippings. A linear regression equation was obtained from accessing NIRS values from NIR spectrophotometer(NIR system, Model XDS, XM-1100 series, FOSS, Sweden) programmed with WinISI III project manager v1.50e and ISIscan(R) (Infrasoft International) and calibrated with laboratory values via chemical analysis from an authorized institute. The equation was formulated as MPLS(modified partial least squares) analyzing laboratory values and mathematically pre-treated spectra. The accuracy of the acquired equation was confirmed with SEP(standard error of prediction), which indicated as correlation coefficient($r^2$) and prediction error of sample unacquainted, followed by the verification of model equation of real values and these monitoring results. As results of monitoring, $r^2$ of nitrogen, moisture, and carbohydrate in 'CY2' creeping bentgrass was 0.840, 0.904, and 0.944, respectively. SEP was 0.066, 1.868, and 0.601, respectively. After outlier treatment, $r^2$ was 0.892, 0.925, and 0.971, while SEP was 0.052, 1.577, and 0.394, respectively, which totally showed a high correlation. However, $r^2$ of starch was 0.464, which appeared a low correlation. Thereof, the verified equation appearing higher $r^2$ of nitrogen, moisture, and carbohydrate showed its higher accuracy of prediction model, which finally could be put into practical use for turf management system.

Therapeutic Effects and Adverse Drug Reactions are Affected by Icotinib Exposure and CYP2C19 and EGFR Genotypes in Chinese Non-Small Cell Lung Cancer Patients

  • Background: The aim of this study was to evaluate how CYP2C19 affects icotinib and metabolite' exposure, and to determine whether the exposure and EGFR genotype influences survival time, tumor metastasis and adverse drug reactions. Materials and Methods: 274 NSCLC patients who accepted 125mg icotinib/t.i.d. were chosen from a phase III study. Blood samples were obtained in $672^{nd}$ ($4^{th}$ week) and $1,680^{th}$ hours ($10^{th}$ week), and plasma was used to quantify the concentration of icotinib and blood cells were sampled to check the genotypes. Clinical data were also collected at the same time, including EGFR genotypes. Plasma concentrations were assessed by HPLC-MS/MS and genotype by sequencing. All data were analyzed through SPSS 17.0 and SAS 9.2. Results: CYP 2C19 genotypes affected bio-transformation from icotinib to M24 and M26, especially in poor-metabolisers. Higher icotinib concentrations (>1000 ng/mL) not only increased patient PFS and OS but also reduced tumor metastasis. Patients with mutant EGFR experienced a higher median PFS and OS (234 and 627 days), especially those with the 19del genotype demonstrating higher PR ratio. Patients who suffered grade II skin toxicity had a higher icotinib exposure than those with grade I skin toxicity or no adverse effects. Liver toxic reactions might occur in patients with greater M20 and M23 plasma concentrations. Conclusions: CYP2C19 polymorphisms significantly affect icotinib, M24 and M26 exposure. Patients with mutant EGFR genotype and higher icotinib concentration might have increased PFS and OS and lower tumor metastasis. Liver ADR events and serious skin effects might be respectively induced by greater M20, M23 and icotinib concentrations.

A Floristic Study of Mt. Myeonsan·Myobong (Taebaek-si, Samcheok-si, Bonghwa-gun) in Korea (면산·묘봉(태백시, 삼척시, 봉화군)의 식물상)

  • This study was carried out to elucidate the distribution of vascular plants and their usefulness of Mt. Myeonsan (1,245 m) and Myobong (1,168 m) in Gangwon-do and Gyeongsangbuk-do. The vascular plants that were collected 15 times from April 2012 to October 2013 consisted a total of 551 taxa; 96 families, 314 genera, 489 species, 3 subspecies, 54 varieties and 6 forms. The plants that are specially noteworthy are 17 taxa of Korean endemic plants, 2 taxa of Critical Endangered Species (CR), 3 taxa of Endangered Species (EN) and 12 taxa of Vulnerable Species (VU) in rare plants as categorized by the Korean Forest Service. Furthermore, V, IV, III degrees of floristic regional indicator plants as categorized by the Korean Ministry of Environment included 2 taxa, 22 taxa and 26 taxa, respectively. Among them, edible, pasture, medicinal, ornamental, timber, dye, fiber, industrial and unknown usefulness plants included 215 taxa, 184 taxa, 163 taxa, 59 taxa, 17 taxa, 6 taxa, 5 taxa, 5 taxa and 138 taxa, respectively. In addition, 25 taxa of naturalized plants were observed.

A Clinical Analysis of Major Salivary Gland Tumors-104 Cases (주타액선종양 104례에 대한 분석)

  • Backgroud and Objectives: Major salivary gland tumor is a rare disease with a largely unknown origin. And also corresponding to the diversity of histopathologic characteristics is an equally wide distribution of clinical behavior that makes treatment decisions difficult. The aim of this study is to analysis the clinical experience of our major salivary gland tumor and to suggest a guidline of management. Materials and Methods : We studied clinical manifestations and treatment results of 104 patients who were diagnosed as major salivary gland tumors. Tumor site, patient age and sex of the patients were analysed. Histopathologic diagnosis, treatment modality, lymph nodes metastasis, recurrence rate and 5-year survival rate were studied on malignant tumors. Results: 1) 48 patients were male and 56 patients were female. 2) Benign tumors were 79 cases. They consisted of 57 cases(72%) of parotid tumor, 22 cases(27%) of submandibular tumor. And pleomorphic adenoma was the most common benign tumor. 3) Malignant tumors were 25 cases and were consisted of 19 cases(76%) of parotid tumor, 4 cases(l6%) of submandibular tumor and 2 cases(8%) of sublingual gland tumor with high-grade mucoepidermoid carcinoma as the most common cause. 4) In the malignant tumors, the number of stage I, II tumors was 14 and that of stage III, IV tumors was 11. Neck node metastasis was noted in 8 cases. 5) 8 cases of malignant tumor were died and all of them had high-grade malignancy. And they were dead before 17 months from the time of diagnosis. 6) Mean survival duration in the malignant salivary gland tumor was 56 months. 7) High-grade malignancy had a 5-year survival rate of 57.9%. A 5-year survival rate of TI, T2 patients was 92.9% and that of T3, T4 patients was 36.4%. That of patients who had neck node metastasis was 37.5% and that of those who had tree neck was 82.4%. Conclusion: The results of this study suggest that the survival rate is dependent on the tumor size, lymph node matastasis and histologic grade. And also the early diagnosis and radical surgery result in the increased survival rate and are effective for prevention of the tumor recurrance.

Hardness Correction Algorithm Applicable to Korea as Related to Aquatic Toxicity Variation for Heavy Metals (국내 적용가능한 중금속 수서독성에 대한 경도보정 알고리즘 연구)

  • Water hardness is a significant parameter as related to aquatic toxicity of some heavy metals. Hardness dependent metals include cadmium, copper, chromium (III), nickel, lead, zinc and silver. Developed countries have applied the hardness correction procedure to derive the water quality criteria for protecting the aquatic organisms. In the present study, we investigated the hardness correction algorithms used in United States of America, European Union, Australia/New Zealand, and Canada, and analyzed the details associated with those algorithms. Toxicity values of heavy metals were definitely different after hardness correction for all of algorithms analyzed. We found that the hardness corrected toxicity values followed by the algorithms of USA and Australia/New Zealand were very similar or same, however they were slightly different for cadmium at the hardness less than 30 mg $CaCO_3\;L^{-1}$. Among the hardness correction algorithms studied, the algorithm used in Australia/New Zealand appears to be a good choice to apply in Korean situation due to its simplicity compared to the algorithm of USA.

A Study on Expansion Possibility of Treatment Capacity in Public Livestock Manure Treatment Plant Integrated Individual Farmhouses (개별농가와 연계한 가축분뇨 공공처리시설의 처리용량 확대 가능성에 관한 연구)

  • The objective of this study is to predict the expansion possibility of treatment capacity in public livestock manure treatment plant (PLMTP) integrated individual farmhouses. According to the treatment efficiency and cost reducing effect, expansion possibility was examined using three cases; (i) decrease of influent concentration from 20,000 mg/L BOD to 1,000 mg/L BOD, (ii) maintenance of low concentration influent with minimum revising existing facilities (BIOSUF) and (iii) maintenance of low concentration influent without revising existing facilities (liquid corrosion method, LCM). In BIOSUF, the treatment capacity increased from 130 ton/day to 300 ton/day. Also, LCM resulted in expansion of treatment capacity from 210 ton/day to 250 ton/day while that of designed concentration influent decreased from 210 ton/day to 190 ton/day. The treatment costs were 14,674 won/ton and 9,929 won/ton for BIOSUF and LCM, respectively. After some revisions, it will be changed to 7,221 won/ton and 8,277 won/ton. Therefore, it must be considered that the livestock manure treats to low concentration and flows into PLMTP for the efficient operation and reducing treatment cost.

Flora of Vascular Plants in Mueuido (Incheon), Korea (무의도(인천광역시)의 관속식물상)

  • The objective of the study was to investigate vascular plants of Mueuido in Incheon from April to October, 2011. The investigation was carried out for mountains (Mt. Horyonggok and Mt. Guksa), coastal areas and annexed islands (Silmido and Somueuido). Vascular plants were found to have 338 taxa, including 90 families, 235 genera, 297 species, 5 subspecies, 32 varieties and 4 forms. Rare and endangered plants designated by Korea Forest Service had 5 taxa including Utricularia bifida, U. vulgaris var. japonica, U. racemosa, Ottelia alismoides, and Arisaema heterophyllum. The floristic regional indicator plants found in this area had 3 taxa of grade V and 5 taxa of grade III. Naturalized plants consisted of 20 taxa that made up 8.3% of the total vascular plants in this area.

Application of Differential Expression of Genetic Profiles in Brain Tumors with Variable [$^{18}F$]-fluorodeoxyglucose Uptake

  • [ $^{18}F$ ]-fluorodeoxyglucose (FDG) uptake on positron emission tomography (PET) scan has been found to reflect tumor aggressiveness and prognosis in various types of cancer. In this study, the gene expression profiles of glial tumors were evaluated to determine whether glial tumors with high $^{18}F$-FDG uptake have more aggressive biological potential than with low uptake. Surgical specimens were obtained from the 12 patients with glial tumors (4 males and 8 females, age range 42-68 years). The tumor samples were divided into two groups based on the $^{18}F$-FDG uptake PET scan findings: high $^{18}F$-FDG uptake (n=4) and low $^{18}F$-FDG uptake (n=8). The pathological tumor grade was closely correlated with the $^{18}F$-FDG uptake pattern: Glial tumors with high $^{18}F$-FDG uptake were pathologically Edmondson-Steiner grade III, while those with low uptake were grade II. The total RNA was extracted from the frozen tissues of all glial tumors (n=12), and adjacent non-cancerous tissue (n=3). The gene expression profiles were evaluated using cDNA microarray. The glial tumors with high $^{18}F$-FDG uptake showed increase expression of 15 genes compared to those with low uptake (P<0.005). Nine genes were down-regulated. Gene expression is closely related to cell survival, cell-to-cell adhesion or cell spreading; therefore, glial tumors with high $^{18}F$-FDG uptake appear to have more aggressive biological properties than those with low uptake.

Induction of Deletion Mutation for the Enzymatic Domain in the Shigatoxin2e A Subunit Gene of Esherichila coli O139 Isolates and Expression of Mutated Protein (분리 대장균 O139의 Shigatoxin2e A 유전자의 효소 활성부에 대한 결손변이 유발 및 변이 단백질의 발현)

  • This study was done to produce a mutated protein inactivated cytotoxicity of Shigatoxin 2e (Stx2e) of E.coli O139 isolates by deletional mutagenesis of Stx2e A subunit gene encoding active-site cleft of enzymatic domain in ST2e holotoxin. Cytotoxicity of the toxoid expressed from the mutant Stx2e gene was compared with wild type Stx2e for development of vaccine candidate. A recombinant plasmid pED18 containing Stx2e gene ot E.coli O139 isolates was used to generate mutation plasmid. Deletion mutagenesis was conducted for Stx2e A subunit gene encoding enzymatically active domain by polymerase chain reaction (PCR) using ot designed primer to induce deletional mutation. DNA sequence analysis was confirmed that the pentamer (Typ 202- Ser 206) that lies within the proposed active-site cleft in the second region was completely deleted. A DNA fragment of 1.1 kb that encode the new mutant Stx2eA gene was inserted into plasmid pRSET vector digested with EcoRV-Hind III and named pEDSET The PEDSET was transformed in E. coli for expression of mutant protein and the protein was confirmed by SDS-PACE and Western-blotting. The protein expressed by the mutant was tested to confirm the reduction of cytotoxic activities on Vero cell using microcytotoxicity assay compared with wild type Stx2e, the cytotoxicity of deletional mutant protein was at least reduced by 3,000-fold on Vero cell.

The Protective Effects of Different Mycotoxin Adsorbents against Blood and Liver Pathological Changes Induced by Mold-contaminated Feed in Broilers

  • An experiment was conducted to determine the effects of different mycotoxin adsorbents including esterified glucomannan (EGM), hydrated sodium calcium aluminosilicate (HSCAS) and compound mycotoxin adsorbent (CMA) on performance, blood parameters, and liver pathological changes in broilers fed mold-contaminated feed. Two hundred and forty 10-day-old broilers were randomly assigned to one of the five dietary treatments including: i) control diet; ii) mold-contaminated diet; iii) moldcontaminated diet+0.05% EGM; iv) mold-contaminated diet+0.2% HSCAS; v) mold-contaminated diet+0.1% CMA. At 35-days-old, blood and liver tissue samples were collected for analysis. 0.1% CMA improved ADG and ADFI during 10-42 d compared to the moldcontaminated group (p<0.05). The mold-contaminated diet increased total white blood cell (WBC) number, haemoglobin (Hgb) concentration, hematocrit (Hct) level, serum aspartate aminotransferase (AST) and ${\gamma}$-glutamyl transferase (GGT) activities, and decreased red blood cell (RBC) number and serum globulin (GLB) and urea nitrogen (BUN) concentrations (p<0.05). The three mycotoxin adsorbents alleviated the alteration of RBC, WBC, Hgb and AST caused by the mold-contaminated diet. Furthermore, 0.1% CMA increased GLB concentration and decreased Hct level and GGT activity (p<0.05). Liver superoxide dismutase (SOD) activity was reduced, and myeloperoxidase (MPO) activity was increased by the mold-contaminated diet (p<0.05). Both EGM and HSCAS prevented the increase of MPO activity (p<0.05). Liver lesion, including severe vacuolar degeneration of hepatocytes, was observed in chicks fed the mold-contaminated diet. 0.05% EGM prevented these effects except for biliary hyperplasia and mild vacuolar degeneration. 0.2% HSCAS showed medium vacuolar degeneration of hepatocytes. Liver of broilers fed 0.1% CMA revealed a mild vacuolar degeneration. These results indicate that a mold-contaminated diet results in adverse effects on blood parameters and liver morphology. 0.05% EGM and 0.2% HSCAS partially alleviated the adverse effects. However, 0.1% CMA almost completely ameliorated the adverse effects.
