Improvement of Decarboxylating Agar Medium for Screening Biogenic Amine-Producting Bacteria in Kimchi

  • A modification of decarboxylating agar medium as described by niven was performed to improve the detection method of biogenic amine-producing bacteria and to eliminate the false-positive. A total of 120 bacterial strains isolated from kimchi were used to evaluate different dicarboxylating agar media and for screening biogenic amines. Potential false-positives ranged from approximately 66 to 79% of the strains tested in the already well-known media. In our improved medium, none of the 120 strains showed the potential false-positives. There was a good agreement (81.7%-87.5%) between the results obtained by the improved medium and by HPLC analysis. Consequently, this medium was greatly improved in screening biogenic amine-producing bacteria and discarding false-positives. Of the 120 kimchi isolates, 14.2, 18.3, 37.5, and 0.8% were found by HPLC to be the producers of histamine, tyramine, putrescine (as a form of spermine), and cadaverine, respectively. The proportion of biogenic amine producer during kimchi fermentation increased to a maximum at an immature period and decreased thereafter.

Development of an improved selective media for differentiation of emetic and diarrheal type Bacillus cereus

  • The objective of this study was to develop a differential medium with improved selectivity for the isolation of Bacillus cereus. Mannitol egg yolk polymyxin medium supplemented with D-galactose allowed the differentiation of diarrheal- and emetic-type B. cereus through pH monitoring. The pH of the medium decreased significantly when incubating the emetic-type B. cereus, whereas the pH change was not significant when incubating the diarrheal-type. The addition of pH indicators, such as methyl red and phenol red, to the medium allowed visual differentiation between diarrheal- and emetic-type B. cereus. A solid agar medium was also developed by optimizing the concentrations of medium components such as monosaccharides, agar, egg yolk enrichment, pH indicators, and antibiotics. This study indicates the possibility of applying selective media for the differentiation of diarrheal- and emetic-type B. cereus.

  • The traditional mechanical manometer is improved to develop more convenient and precise manometer in continuous measurement of the gas pressure in medium-low vacuum range (1x10$^{-1}$ -10 Torr.) Glass (solid) is used as a detector material of the improved manometer. Using the strain gauge adhered to thin glass board, mechanical strain corresponding to variation of pressure in measurement system is converted into quantity of electricity, and the quantity of electricity is amplified. Experiments have also shown that the improved manometer have more advantages in reproducibility, measured sensibility, and responsible velocity than taditional one.

  • TQM has been applied to the industry area regardless of the size of company. BNQA model should be identically applied to large companies and small & medium-sized company. However, there are many evidences that TQM model should be modified according to the size of company. In this study, we analyzed the practice of TQM in Korean small & medium-sized Company. Also, we tried to give some suggestions for improving the competitiveness of the small & medium-sized company. The result of this study is that leadership, work system, education and strategy development should be improved in domestic companies, and work system, education and strategy deployment should be improved in foreign companies.

  • The influences of pretreatment medium, the addition of fresh medium, and the size of culture plate on the production of embryos were investigated in isolated microspore culture of hot pepper (Capsicum annuum L.). Among the media used for heat shock pretreatment ($32{\pm}1{^{\circ}C}$), high frequency embryo production was obtained when the sucrosestarvation medium A was used. On the other hand, neither 0.37 M mannitol solution nor NLNS medium supplemented with sucrose was not efficient for embryo production. The addition of culture medium to pretreatment media considerably decreased the embryo production even though embryo development proceeded further. The embryo production was not improved by the addition of fresh medium after 2 or 3 weeks from starting culture. Increase in the size of the culture plate from $3.5{\times}1.0$ cm to $6.0{\times}1.5$ cm improved embryo quality. These results will provide valuable information for developing an efficient microspore culture system of hot pepper for high frequency embryo production.

Optimizing the composition of the medium for the viable cells of Bifidobacterium animalis subsp. lactis JNU306 using response surface methodology

  • This research improved the growth potential of Bifidobacterium animalis subsp lactis strain JNU306, a commercial medium that is appropriate for large-scale production, in yeast extract, soy peptone, glucose, L-cysteine, and ferrous sulfate. Response surface methodology (RSM) was used to optimize the components of this medium, using a central composite design and subsequent analyses. A second-order polynomial regression model, which was fitted to the data at first, significantly lacked fitness. Thus, through further analyses, the model with linear and quadratic terms plus two-way, three-way, and four-way interactions was selected as the final model. Through this model, the optimized medium composition was found as 2.8791% yeast extract, 2.8030% peptone soy, 0.6196% glucose, 0.2823% L-cysteine, and 0.0055% ferrous sulfate, w/v. This optimized medium ensured that the maximum biomass was no lower than the biomass from the commonly used blood-liver (BL) medium. The application of RSM improved the biomass production of this strain in a more cost-effective way by creating an optimum medium. This result shows that B. animalis subsp lactis JNU306 may be used as a commercial starter culture in manufacturing probiotics, including dairy products.

Simultaneous Improvement of Formaldehyde Emission and Adhesion of Medium-Density Fiberboard Bonded with Low-Molar Ratio Urea-Formaldehyde Resins Modified with Nanoclay

  • In wood-based composite panels, low-molar ratio (LMR) urea-formaldehyde (UF) resins usually result in reduced formaldehyde emission (FE) at the expense of poor adhesion. However, the FE and adhesion of medium-density fiberboard (MDF) bonded with LMR UF resins were both improved in this study. The modified LMR UF resins with transition metal ion-modified bentonite (TMI-BNT) nanoclay simultaneously improved the FE and adhesion of MDF panels. The modified LMR UF resins with 5% TMI-BNT resulted in a 37.1% FE reduction and 102.6% increase in the internal bonding (IB) strength of MDF panels. Furthermore, thickness swelling and water absorption also significantly decreased to 13.0% and 24.9%, respectively. These results imply that TMI-BNT modification of LMR UF resins could enhance the formation of a three-dimensional network rather than crystalline domains, resulting in improved cohesion.

  • 한국인의 장내 균총 조사를 하던 중 Bifidobacterium 선택배지로 개발된 Mitsuoka의 BS배지에서 상당수의 Bifidobacterium이 사멸되는 것으로 나타났다. 이에 보다 개량된 Bifidobacterium의 선택배지를 조제하기 위하여 한국인들로부터 Bifidobacterium들을 비선택 배지및 기존의 선택배지에서 분리한 뒤 동정하였다. 분리 동정된 균주들에 대하여 항생제와 대사저해제에 대한 감수성 조사 및 특이 기질검사를 하여 선택배지 조제의 기초 자료로 활용하여 Bifidobacterium이 선택적으로 자랄 수 있는 배지로 TP배지를 고안하였다 TP배지는 상용되는 Mitsuoka의 BS배지보다 Bifidobacterium 선택성이 우수하였고 colony 성장속도도 빨랐다.

  • This paper proposes an improved modulation scheme for the medium voltage modular multi-level converter (MMC), which operates in the nearest level control and applies in the medium voltage direct current (MVDC) system. In the proposed modulation scheme, the offset (neutral-to-zero output) voltage is adjusted, with the phase voltage magnitude, thereby maintaining a constant value with N+1 level in the controllable modulation index (MI) range. In order to confirm the proposed scheme's validity, computer simulations for the 22.9 kV - 25 MVA MMC were performed with PSCAD/EMTDC, as well as hardware experiments for the 380 V - 10 kVA MMC. The proposed modulation scheme offers to build a constant pole voltage regardless of the MI value, and to build a phase voltage with improved total harmonic distortion (THD).

채널예약기능을 부가한 개선된 CSMA/CD 프로토콜 (An Improved CSMA/CD with Channel Reservation Facility)

  • 본 논문에서는 단일의 전송매체를 여러 스테이션이 사용하기 위한 효율적인 매체접속제어 방식을 제안하였다. 매체접속제어 방식은 크게 예약방식과 충돌 허용방식으로 구분되며 네트워크 환경에 따라 각각의 방식이 효율성에서 장점을 지닌다. 제안된 매체접속제어 방식은 충돌 허용방식인 CSMA/CD를 기본으로 하며 충돌현상을 지연하여 채널을 예약하는 기능을 부가하였다. 따라서 충돌이 발생하면 예약과정을 거쳐 채널에는 예약에 성공한 스테이션만 경쟁에 참여하게 되어 매체접속 확률을 높이게 된다. 제안된 프로토콜은 충돌의 횟수가 감소하여 전체적으로 채널의 활용도가 향상되는 것을 시뮬레이션으로 확인하였다.
