  • The effect of growth and the carbon sources including the molar ratio of fructose to glucose was studied for the maximization of inosine-5'-monophosphate (5'-IMP) production from Brebibacterium ammoniagenes D-21530. According to experimental results, fructose was more efficient to 5'-IMP accumulation than glucose, while the latter was better for the cell growth than the former. To synchronously use glucose and fructose as carbon source is to optimally control the cell growth and maximum production of 5'-IMP without change of other conditions. The optimal weight percent of fructose to sum of glucose and fructose was 20~40%, and the productivity improvement over the utilization of fructose was about 40%. And also the optimality of purine base such as adenine and guanine were considered. The optimal concentrations of adenine and guanine were near 50㎎/l.

Effects of Dietary Nucleotide Supplementation on the Growth Performance, Feed Utilization, Hematological Parameters and Innate Immunity in Red Seabream Pagrus major (사료 내 Nucleotides 첨가가 참돔P(agrus major)의 성장, 사료효율, 혈액성상 및 비특이적 면역반응에 미치는 영향)

  • Two studies were conducted to investigate the effects of dietary nucleotides (NT) on the growth performance, hematological parameters, and innate immune responses of red seabream Pagrus major. In Expt I, six experimental diets were formulated: a control, four that contained each NT at a level of 0.15% (inosine monophosphate, IMP; adenosine monophosphate, AMP; guanosine monophosphate, GMP; and uridine monophosphate, UMP), and one with a 1:1:1:1 mixture of NTs (IMP, AMP, GMP, and UMP). In Expt II, five experimental diets were formulated that contained 0, 0.1, 0.2, 0.4, and 1.0% IMP (commercial product). Triplicate groups of juvenile (initial body weight 33.1 g) and growing (initial body weight 120 g) red seabream were fed one of the experimental diets to apparent satiation for 8 weeks in Expt I and 12 weeks in Expt II. In Expt I, fish fed diets with NT had higher growth performance than the control group. The nitroblue tetrazolium and lysozyme activities were higher in fish fed the mixed-NT diet, and lowest in the control group. In Expt II, the final body weight and feed utilization of fish fed the 0.1% IMP diet were significantly higher than those of fish fed the control or 1.0% IMP diets. Diet palatability was improved significantly when 0.1% IMP was added. The lysozyme activity was higher in fish fed diets with 0.4-1.0% than in the control group. These results suggest that supplementation of 0.15% IMP and Mixed-NTs in diet can enhance the growth and immune responses in juvenile red seabream. The optimum IMP level appears to be 0.2% in practical feed formulation for growing red seabream.

A Study on an Efficient Method to Evaluate Intermodulation Product (혼변조적의 효과적인 산출 방식에 관한 연구)

  • In this paper, we present analysis results of various intermodulation products (IMPs) evaluation methods previously proposed for nonlinear systems. The results indicate that out of these methods, Fuenzalida and Simbo's method is the best for efficient evaluation of IMPs. Also, we present a detailed mathematical analysis of IMP and show how to apply the equations related to IMP to actual program implementations with examples. In this paper, we newly introduce three methods to reduce the IMP calculation time and improve the accuracy of the outputs :1) TWTA curve fitting method by LS (Least Square), 2) IMP evaluation technique which is based on a look-up table 3) Gaussian spectral shaping for PSK signal instead of convolution. The IMP evaluation results obtained by the new approaches introduced in this paper resulted in a good match with the published outputs in the open literature and showed improved performance especially in the TWTA curve .

Phenotypic and Genetic Parameters for Inosine Acid in Relation to Carcass and Meat Quality Traits in Pigs

  • A total of 135 F2 finishing pigs (65 barrows and 69 gilts) from resource population (Large White${\times}$Meishan) were slaughtered at about 87.8 kg BW. Contents of inosine acid (IMP) and carnine (HR) in muscle were assayed by HLPC and genetic parameters for IMP content and HR content were estimated using full sibs model. There was significant sex effect on IMP content(p<0.05), $3.561{\pm}0.077mg/g$ for gilt and $3.287{\pm}0.085mg/g$ for barrow. Heritability estimates for IMP and HR content were 0.127 and 0.357, respectively. The phenotypic correlation between IMP content and HR was 0.335, pH (A) 0.024, water lose rate (WLR) -0.069, intramuscular fat (IMF) -0.214, average marbling score (MARB) -0.143, average backfat measurements (AVBF) -0.084 and average color value (CV) -0.156, respectively. The result indicated that inosine acid content in meat might be retained or slightly improved by reducing backfat depth in pig breeding.

Changes of ATP Related Compounds of Large Anchovy During Drying and Storage (대멸치의 건조 및 저장 중 ATP 관련화합물의 변화)

  • This study was conducted to examine the changes of contents of ATP related compounds during drying of large anchovy and storage of dried large anchovy (DLA) according to storage temperature and package method. The total content of ATP related compounds of raw large anchovy was $36.0{\mu}mole/g$ and the major ATP related compounds were consisted of IMP and hypoxanthine. The IMP content of DLA was the highest in $20^{\circ}C$ cold-air drying, and the breakdown of IMP was progressed rapidly in $60^{\circ}C$ air drying, followed by $40^{\circ}C$ air drying, sun drying, and $20^{\circ}C$ cold-air drying. During storage of DLA, ATP was not detected while ADP and AMP was detected in a very small amount, and the changes of ATP related compounds were coincided with the changes of contents of IMP, inosine and hypoxanthine. The changes of ATP related compounds with different package method did not show distinct differences, while with different storage temperature showed clear difference. The content of IMP was over $8.88{\mu}mole/g$ on 60 days at $-20^{\circ}C$, while were over $0.83 {\mu}mole/g$ and$ 0.202 {\mu}mole/g$ on 16 days at$ 5^{\circ}C$ and$ 25^{\circ}C$, respectively. These results suggest that the breakdown of IMP depends on storage temperature and frozen storage affects good quality of DLA during storage.

  • This study was designed to determine the changes of ATP-related compounds, especially the concentration of IMP, and compares the relationship between IMP and flavor of pork loin chops during storage as three different storage temperatures (include $4^{\circ}C$ CF and PF). Pork loin chops were kept under $4^{\circ}C$ $-1.5{\pm}0.5^{\circ}C$ (control freezing storage) and $-3{\pm}0.5^{\circ}C$ (partially freezing storage). The changes of TBA values, ATP-related compounds, pH values, in CF stored samples were higher than PF stored samples, but it had lower TBA values than $4^{\circ}C$ storage. The IMP concentration reached their peak after 2 days in $4^{\circ}C$, 5 days in CF and 7 days in PF storage, and the ATP, ADP, AMP contents of the loin chops showed minimum, respectively. Flavor of meat sensory score for $4^{\circ}C$ stored samples were more intense (p<0.05) than CF and PF samples on day 2 of storage. However, after storage for 5 days, flavor scores for CF samples were more intense (p<0.05) than $4^{\circ}C$ and PF samples. Flavor scores for PF samples were more intense (p<0.05) than $4^{\circ}C$ and CF on day 7 of storage. As the meat with the peak of IMP contents was most preferred, it was considered that the content of IMP was related to the flavor of meat and that CF, PF had influence on the IMP content.

  • In this paper, the degradation of nucleotides and their related compounds in hag-fish muscle during roasting was studied. The results showed that IMP was dominant in fresh hag-fish showing 63% of total nucleotides while the contents of ATP, ADP, AMP, inosine and hypoxanthine were low. The ATP tended to degrade rapidly during roasting, but 80% of IMP remained and ATP, ADP and AMP were also entirely disappeared. In consideration of flavor quality, it was consumed that roasting is an effective preparation method of hag-fish, as far as IMP is concerned.

Radioprotective role of amifostine on osteointegration of titanium implants in the tibia of rats

  • Background: Titanium is the most widely used metal for bone integration, especially for cancer patients receiving ionizing radiation. This study aimed to investigate the amifostine administration that would reduce the effects of radiation on bone healing and osseointegration in rat models. Objectives: It is aimed that the application of amifostine in rats receiving radiotherapy treatment will reduce the negative effects of ionizing radiation on the bone. Methods: Thirty-five adult male Wistar rats were randomly divided into one healthy and four experimental groups. In three consecutive days, two experimental groups of rats (AMF-RT-IMP and RT-IMP) were exposed to radiation (15 Gy/3 fractions of 5 Gy each). Then the titanium implants were inserted into the left tibia. Before the radiotherapy process, a 200 mg/kg dose of amifostine (AMF) was administered to the rats in the AMF-IMP and AMF-RT-IMP groups. Twenty-eight days after the screw implant, all rats were sacrificed, and their blood samples and tibia bones were collected for analysis. Results: The results indicated an accelerated bone formation and a more rapid healing process in the screw implants in the AMF-IMP, AMF-RT-IMP, and AMF-RT groups than in the RT-IMP group. Also, bone-implant contact area measurement and inflammation decreased with amifostine treatment in the implants subjected to irradiation (p < 0.05). Conclusions: The results obtained in the present study suggested that amifostine prevents the losses of bone minerals, bone integrity, and implant position from ionizing-radiation when given before exposure.

Effect of Temperature and Inosine Monophosphate on Spore Germination of Artificially Inoculated Bacillus cereus in Traditional Korean Fermented Sauces (전통 장류를 이용한 소스제품에 접종된 Bacillus cereus spore의 발아에 미치는 온도 및 inosine monophosphate 첨가 효과)

  • The effects of temperature and inosine monophosphate (IMP) on the germination of artificially inoculated Bacillus cereus spores in ganjang, doenjang, and gochujang sauces were investigated. The pH and salt concentration of the sauces were as follows: ganjang (4.84, 7.45%), doenjang (4.26, 5.07%), and gochujang (3.54, 3.24%), respectively. Treatment at $40^{\circ}C$ or $80^{\circ}C$ resulted in a B. cereus growth of 0.8 log CFU/g in ganjang sauce; this concentration increased to 2.25 log CFU/g and 1.21 log CFU/g, respectively, when 10 mM IMP was added. Compared with that for ganjang sauce, the germination effect was comparatively lower for doenjang sauce by 10 mM IMP+$40^{\circ}C$ treatment (0.46 log CFU/g) and 10 mM IMP+$80^{\circ}C$ treatment (0.85 log CFU/g). However, no germination effect was noted for gochujang sauce by any treatment in this study. Therefore, the results suggested that pH is an important factor affecting spore germination in sauces.

Detection of the Carbapenem Resistance Gene in Gram-negative Rod Bacteria Isolated from Clinical Specimens (임상검체에서 분리된 그람음성막대균으로부터 카바페넴 내성 유전자 검출)

  • Carbapenem-resistant Enterobacteriaceae (CRE) poses an increasing public health threat and has limited treatment options with high associated mortality. Genotypes of carbapenemase that threaten public health (blaKPC, blaNDM, blaIMP, and blaVIM) and blaOXA-48-like genes were detected by phenotypic and molecular diagnosis, and related gene distribution patterns were investigated. Phenotypic testing using the modified Hodge test confirmed positivity in all 41 strains examined, and carbapenemase inhibitory testing using meropenem+phenyl boronic acid or meropenem+EDTA confirmed positivity in 18 and 8 strains, respectively. Polymerase chain reaction revealed the presence of amplification products in 28 strains of blaKPC, 25 strains of blaNDM, 5 strains of blaIMP, 1 strain of blaVIM, and 13 blaOXA-48-like strains. In addition, 7 strains of blaKPC+blaNDM, 1 strain of blaKPC+blaIMP, 1 strain of blaNDM+blaOXA-48-like, 1 strain of blaNDM+blaVIM, 4 strains of blaKPC+blaNDM+blaIMP, and 4 strains of blaKPC+blaNDM+blaOXA-48-like were identified. Melting curve analysis using real-time PCR was wholly consistent with PCR results. The study shows genetic identification of highly specific CRE by real-time PCR could be used to provide early diagnoses and infection control, improve surveillance, and prevent the transmission of CRE.
