청상보하환(淸上補下丸)이 흑색종(黑色腫)의 폐전이(肺轉移) 억제(抑制) 및 면역조절작용(免疫調節作用)에 미치는 영향(影響) (Antitumor and Immunomodulatory Effects of Chungsangbohahwan on Murine Melanoma-induced Lung Metastasis)

  • 1. 연구배경 최근 놀라운 과학기술의 발전에 힘입어 난치병이라고 생각되어 오던 질환들이 하나씩 정복되어 가고 있으나 평균수명의 연장, 생활수준 향상으로 인한 고지방식, 스트레스 및 과학기술발전의 부작용 등을 이유로 새로운 질환들이 나타나고 있는 실정이다. 종양도 이런 대표적인 질환중 하나로 최근 경제적, 시간적으로 많은 투자를 해왔으나 아직까지 그 발생원인이나 성장기전 등이 자세히 밝혀지지 않았다. 이에 본 연구는 효후(哮吼)를 치료하는 청상보하환(淸上補下丸)을 이용하여 흑색종의 폐전이 억제능과 이에 의한 면역증강에 관하여 연구하고자 하였다. 2. 연구결과 B-16 세포를 이용한 C57BL/6계 마우스에 있어서의 폐전이 억제능을 측정한 결과 청상보하환(淸上補下丸) 투여군이 대조군에 비해 검액처리 7일후에는 52.70%, 14일후에는 19.43%의 유의성있는 억제율을 보였으며, 청상보하환(淸上補下丸)의 B-16 세포에 대한 직접적인 항암작용을 알아 보기 위한 시험관내 세포독성 실험에서는 청상보하환(淸上補下丸)의 용량이 $5{\mu}g/well$, $2.5{\mu}g/well$, $1.25{\mu}g/well$, and $0.625{\mu}g/well$인 경우 유의성있는 효과를 얻을수 있었으며 생존율에 의한 $IC_{50}$$2.17{\mu}g/well$이었다. NK-activity를 측정한 결과 청상보하환(淸上補下丸) 투여군이 작동세포와 표적세포의 비율이 100:1, 50:1의 경우에 있어서 $72.63{\pm}5.17%$, $26.94{\pm}2.43%$를 보여 대조군에 비하여 각각 31.12%, 43.68% 유의성있는 증가를 보였다. IL-2의 생산능을 측정한 결과 청상보하환(淸上補下丸) 투여군은 $1392.00{\pm}22.31pg/ml$을 보여 대조군에 비해 4.04%의 유의성 있는 증가를 보였다.

4계통 재래종 닭고기의 화학적 특성 및 면역활성 (Comparison of Chemical Composition and Immune-enhancing Activity of the Four Lines of Korean Native Chickens)

  • 본 연구는 4종의 재래닭인 연산오계, 현인흑계, 횡성약닭, 황봉의 일반성분, 카르노신, 안세린 함량 및 면역활성을 비교하기 위하여 실시하였고, 대조구로 백색 레그혼을 사용하였다. 시험용 닭을 모두 동일한 조건에서 13주간 사육하여 도계한 후 화학적 성분조사를 실시하였다. 일반성분 분석에서는 4종의 재래닭이 백색레그혼에 비하여 조지방 함량이 높았으며, 콜레스테롤 함량은 현인흑계가 다른 품종에 비하여 높았고 횡성약닭이 유의적으로 낮았다(p<0.05). 면역증진 효능을 측정하기 위하여 대식세포를 이용하여 실험한 결과는 재래닭 4종의 가슴육에서만 대식세포 증식 및 NO 억제 효능이 있었다. 대식세포 탐식작용은 현인흑계와 횡성약닭가슴육에서 유의적으로 증가되었고, 염증 관련 사이토카인(IL-4, IL-10, $IFN-{\gamma}$) 유전자 발현은 백색레그혼에 비하여 4종의 재래닭에서 억제효과가 컸다. 결론적으로 4종류의 재래닭이 백색레그혼에 비하여 면역증진 효능이 높았으며, 특히 가슴육에서 효능이 있었다. 재래닭 품종 간에도 일반성분 및 면역조절능에 차이가 있는 것으로 나타났다.

Ellagic acid plays a protective role against UV-B-induced oxidative stress by up-regulating antioxidant components in human dermal fibroblasts

  • Ellagic acid (EA), an antioxidant polyphenolic constituent of plant origin, has been reported to possess diverse pharmacological properties, including anti-inflammatory, anti-tumor and immunomodulatory activities. This work aimed to clarify the skin anti-photoaging properties of EA in human dermal fibroblasts. The skin anti-photoaging activity was evaluated by analyzing the reactive oxygen species (ROS), matrix metalloproteinase-2 (MMP-2), total glutathione (GSH) and superoxide dismutase (SOD) activity levels as well as cell viability in dermal fibroblasts under UV-B irradiation. When fibroblasts were exposed to EA prior to UV-B irradiation, EA suppressed UV-B-induced ROS and proMMP-2 elevation. However, EA restored total GSH and SOD activity levels diminished in fibroblasts under UV-B irradiation. EA had an up-regulating activity on the UV-B-reduced Nrf2 levels in fibroblasts. EA, at the concentrations used, was unable to interfere with cell viabilities in both non-irradiated and irradiated fibroblasts. In human dermal fibroblasts, EA plays a defensive role against UV-B-induced oxidative stress possibly through an Nrf2-dependent pathway, indicating that this compound has potential skin antiphotoaging properties.

Effects of the Antitumor Component, F-D-P, Isolated from Elfvingia applanata on the Immune Response

  • An antitumor component, F-D-P, was purified from the hot water extract of the carpophores of Elfvingia applanata by precipitation with ethanol, dialysis, and passage through a column of DEAE-cellulose ion exchange. F-D-P inhibited the growth of Sarcoma 180 in mice showing the tumor inhibition ratio of 88.3% in doses of 20 mg/kg for ten days. Chemical analysis of F-D-P showed that it was composed of polysaccharide(65.3%) and protein(6.5%0, and that the monosaccharides consisting of the polysaccharide was glucose(89.1%) and mannose(10.9%). The immunomodulatory activities of F-D-P were explored by determining its effect on the proliferation of the whole and subpopulations of lymphocytes, and on the generation of natural killer(NK) cell activity in vitro. F-D-P was mitogenic to total lymphocytes and B cells, but not to purified T cells, even in the presence of accessory cells. F-D-P did not increase NK cell activity when added to cultures of resting lymphocytes. From these results, it is clear that F-D-P modulates primarily the humoral immune responeses.

Inhibition of TNF-α-Mediated NF-κB Transcriptional Activity by Dammarane-Type Ginsenosides from Steamed Flower Buds of Panax ginseng in HepG2 and SK-Hep1 Cells

  • Panax ginseng is a medicinal herb that is used worldwide. Its medicinal effects are primarily attributable to ginsenosides located in the root, leaf, seed, and flower. The flower buds of Panax ginseng (FBPG) are rich in various bioactive ginsenosides, which exert immunomodulatory and anti-inflammatory activities. The aim of the present study was to assess the effect of 18 ginsenosides isolated from steamed FBPG on the transcriptional activity of NF-${\kappa}B$ and the expression of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)-stimulated target genes in liver-derived cell lines. Noticeably, the ginsenosides $Rk_3$ and $Rs_4$ exerted the strongest activity, inhibiting NF-${\kappa}B$ in a dose-dependent manner. SF and $Rg_6$ also showed moderately inhibitory effects. Furthermore, these four compounds inhibited the TNF-${\alpha}$-induced expression of IL8, CXCL1, iNOS, and ICAM1 genes. Consequently, ginsenosides purified from steamed FBPG have therapeutic potential in TNF-${\alpha}$-mediated diseases such as chronic hepatic inflammation.

Study on Immunostimulating Activity of Macrophage Treated with Purified Polysaccharides from Liquid Culture and Fruiting Body of Lentinus edodes

  • Lentinus edodes is a well-known edible and medicinal mushroom used in Oriental cultures. Recently, L. edodes has attracted a lot of attention owing to its antifungal activity, antibacterial activity, antiviral activity, hepatoprotective effect, antitumor activities, and immunomodulatory and cytotoxic effects. In this study, the water-soluble crude polysaccharides, CPF and CPB, which were obtained from the fruiting body and culture cell-free broth of L. edodes by hot-water extraction and ethanol precipitation, were fractionated by DEAE cellulose and Sepharose CL-6B column chromatography, resulting in six polysaccharide fractions, CPFN-G-I, CPFN-G-II, CPFN-G-III, CPFA-G, CPBN-G, and CPBA-G Among these fractions, CPFN-G-I, CPBN-G, and CPBA-G were shown to stimulate the functional activation of macrophages including NO production, cytokine expression, and phagocytosis.

누에번데기 및 누에애벌레 밀리타리스동충하초(Cordyceps militaris) 열수추출물의 투여가 고형암이 유발된 마우스의 종양성장 억제 및 면역기능에 미치는 영향 (Tumor Growth Inhibitory and Immunomodulatory Activities of Cordyceps Militaris Water Extracts in ICR Mice Bearing Sarcoma-180 Solid Tumor)

  • 본 연구에서는 누에번데기 또는 누에애벌레를 기주로 하여 인공 재배된 밀리타리스동충하초의 고형암 성장억제 및 면역조절 기능을 평가하였다. 이를 위해 sarcoma-180 세포주를 겨드랑이에 피하 이식받은 ICR 마우스를 대상으로 밀리타리스동충하초 또는 유효성 분으로 알려진 cordycepin을 10일간 복강투여한 후 고형암 성 장저지능, 면역관련 장기의 무게변화, 비장 내 면역세포수의 변화, 그리고 비장세포에 의한 interleukin-2(IL-2) 생성능을 측정하여 다음과 같은 결론을 얻었다. 겨드랑이에 고형암을 이식받은 마우스에게 50 mg/kg(CMP50) 또는 100 mg/kg(CMP100)의 누에번데기 밀리타리스동충하초 추출물을 10일간 투여한 결과 생리식염수를 투여한 대조군에 비해 3주 후 고형암 성장이 47.3% 및 57.6% 유의하게 억제되었다(p<0.05). 50 mg/kg(CML50) 및 100 mg/kg(CML100)의 누에애벌레 밀리타리스동충하초를 투여받은 경우 35.5% 및 37.1%, 그리고 1 mg/kg 및 2 mg/kg의 cordycepin을 투여받은 군의 경우 26.1%와 29.8%의 고형암 성장 억제효과가 관찰되었다(p<0.05). 고형암을 이식받은 후 누에번데기 또는 누에애벌레 밀리타리스동충하초를 투여받은 마우스는 대조군에 비해 흉선무게가 38∼44% 유의하게 증가하였고(p<0.05), 비장내 CD4+ 및 CD8+ T-세포수, NK-세포수(63∼110% 증가), 그리고 비장세포에 의한 IL-2 생성능(33∼51%증가)이 모두 유의하게 증가하였다(p<0.05). 이상의 결과를 종합해 볼 때, 밀리타리스동충하초 열수추출물은 고형암의 성장을 억제하는 효과가 있으며, 이와 같은 밀리타리스동충하초 추출물의 항암활성은 면역기능 증강효과와 연관이 있을 것으로 생각된다. 아울러 밀리타리스동충하초의 고형암 성장억제 및 면역조절 기능은 cordycepin 단일물질에 비해 더 우수하였다.

갈근탕(葛根湯)이 면역조절작용에 미치는 영향 (Effect of Gal-Geun-Tang on Antigen-Specific Immune Response)

  • Objectives : Gal-Geun-Tang (GT) has been described from SANGHAN in Korean traditional medicine and known to act against cold, fever, hypertension, and nasal catarrh. However, little has yet been learned about the effect of GT on immune function. In the current study, in vitro and in vivo immunomodulatory activity of GT (water extract) was investigated.Methods : Water extract of GT induced in vitro proliferation of spleen cells and significantly increased their proliferative responses during anti-CD3 activation. Using purified splenic T and B cells, it was revealed that GT has a mitogenic activity to B cells and promotes their proliferation induced by lipopolysaccharide, whereas T cell proliferation was not triggered and GT was rather inhibitory to T cell activation caused by anti-CD3 antibody. In the presence of antigen presenting cells (APC), GT addition resulted in a significant increase of IFNγ and IL-4, but not IL-2, production. However, addition of high concentration (1,000㎍/㎖) of GT led to a marked reduction in T cell cytokine production and under such condition, GT facilitated apoptosis of T cells when examined by flow cytometry with propidium iodide staining.Results : In vivo immunomdulation of GT was also investigated using a mouse model. Following keyhole limpet hemocyanin (KLH) immunization, GT (1 ㎎/day) was orally administered for 9 days. Cell numbers in thymus, spleen and peripheral blood were not altered by GT administration, indicating that such dose is not immunotoxic. Cell numbers in draining lymph nodes (LN) and ex vivo Ag-specific proliferation of LN cells were significantly elevated by GT administration. However, any preferential stimulation of T or B and CD4+ or CD8+ T cell subpopulations was not observed in a flow cytometric analysis of LN cells. This result shows that GT does not promote in vivo B cell proliferation while GT enhances Ag-specific proliferation of LN cells, unlike what was observed in vitro.Conclusions : For a further understanding of in vivo immunomodulatory activity of GT, ex vivo cytokine production of LN cells obtained from KLH-immunized mice was evaluated. Ag-specific IFNγ production was significantly higher in GT-treated mice when compared to PBS-treated control mice. In contrast, IL-4 production in GT-treated group was comparable to control group unlike to in vitro data. In addition, GT administration did not result in any significant differences in serum levels of Ig (IgM, IgG1 and IgG2a) between GT-treated and control groups. Taken together, these data strongly support that GT promotes immune response, more profoundly type 1 helper T cell (Th1) activity and GT may be applicable for treatment of intracellular parasite infection such as viral diseases.

The Differential Immunomodulating Effects of Levan and DFA-IV on Macrophage Function

  • Di-D-fructose-2,6':6,2'-dianhydride (DFA-IV) is a disaccharide consisting of two fructose residues that are prepared from levan by levan fructotransferase. Levan is a homopolysaccharide composed of D-fructofuranosyl residues joined by $\beta$-(2,6) and $\beta$-(2,1) linkages. We compared the immunomodulatory effects of levan with DFA-IV. Tumoricidal activity, phagocytosis and nitric oxide (NO) production were examined in levan- and DFA-IV-treated RAW264.7 cells. The NO production, tumoricidal and phagocytic activities were significantly increased in both treated cells. The results indicate that levan has significantly greater effects on tumoricidal activity than DFA-IV at low concentrations (1 ${\mu}g/mL$) and its effect on NO production shows a similar pattern. These results suggest that tumoricidal activity induced by both samples is mediated by NO production.

Metformin Suppresses MHC-Restricted Antigen Presentation by Inhibiting Co-Stimulatory Factors and MHC Molecules in APCs

  • Metformin is widely used for T2D therapy but its cellular mechanism of action is undefined. Recent studies on the mechanism of metformin in T2D have demonstrated involvement of the immune system. Current immunotherapies focus on the potential of immunomodulatory strategies for the treatment of T2D. In this study, we examined the effects of metformin on the antigen-presenting function of antigen-presenting cells (APCs). Metformin decreased both MHC class I and class II-restricted presentation of OVA and suppressed the expression of both MHC molecules and co-stimulatory factors such as CD54, CD80, and CD86 in DCs, but did not affect the phagocytic activity toward exogenous OVA. The class II-restricted OVA presentation-regulating activity of metformin was also confirmed using mice that had been injected with metformin followed by soluble OVA. These results provide an understanding of the mechanisms of the T cell response-regulating activity of metformin through the inhibition of MHC-restricted antigen presentation in relation to its actions on APCs.
