Inflammation appears to have a major role in the development of atherosclerosis. Cyclooxygenase-2 (COX-2) is involved in the inflammatory response via the generation of prostanoids that, in turn, are involved in the production of matrix metalloproteinases (MMPs). This study hypothesized that a vascular infection with cytomegalovirus (CMV) may induce a chronic inflammatory reaction and activated inflammatory cells may express inflammatory mediators such as cyclooxygenase-2 (COX-2) and matrix metalloproteinases-9 (MMP-9). To confirm the hypothesis, the immunohistochemical stains for CMV late antigen, COX-2, MMP-9, macrophage, and T-lymphocyte were performed on CMV-infected atherosclerotic lesions. The immunoreactivity for COX-2 and MMP-9 was evident in all cases of atherosclerosis along with plaques, mainly in macrophages/foamy cells, intimal and medial smooth muscle cells, and endothelial cells of the intima. Within the intima, the increased immunoreactivity for COX-2 and MMP-9 was colocalized to the area stained with CMV late antigen. Sections from control specimens showed no immunoreactivity for CMV late antigen, COX-2 and MMP-9. These data seem to support the hypothesis that CMV may participate in a pathogenetic mechanism for atherogenesis or progression of atherosclerosis.
The regional distribution and relative frequency of endocrine cells in the alimentary tract of the snake, Rhabdophis tigrinus tigrinus, were investigated by immunohistochemical method using 7 antisera. Chromogranin (Cg)-, glucagon-, somatostatin-, gastrin/cholecystokinin (Gas/CCK)-, serotonin-, bovine pancreatic polypeptide (BPP)-immunoreactive cells were identified in this study. Cg-immunoreactive cells were detected throughout the alimentary tract including the esophagus, with predominant frequency in the pylorus. Numerous immunoreactive cells were observed from the esophagus to the pylorus but a few cells were detected in the large intestine. Glucagon-immunoreactive cells were observed from the proximal portions to the distal portions of the small intestine. They were increased to the middle portions but thereafter decreased, and no cells were found in the terminal portions. Somatostatin-immunoreactive cells were restricted to the small intestine and these cells were decreased toward to distal portions of the small intestine. Gas/CCK-immunoreactive cells were detected in the pylorus and small intestine. They were most predominant in the pylorus and the proximal portions of the small intestine but thereafter decreased toward to the distal regions. Serotonin-immunoreactive cells were observed throughout the alimentary tract. They were most predominant in the pylorus and proximal portions of the small intestine but a few cells were observed in the large intestine. BPP-immunoreactive cells were restricted to the distal portions of the small intestine with rare frequency. No bombesin-immunoreactive cells were found in this study.
The distribution and relative frequency of insulin-immunoreactive cells in the pancreas was studied during developmental stages (fetus, neonate, 1-month-old, 6-month-old and adult) of the Korean native goat by immunohistochemical methods. The different distribution and relative frequency of glucagon-immunoreactive cells in the pancreas of the Korean native goat was observed during development. Insulin-immunoreactive cells were detected in the exocrine and endocrine portions (pancreatic islets) of the all ages, and in the duct of the 6-month-old. The relative frequencies of these cells were increased in the pancreatic islets with ages but decreased in the exocrine portions. Generally, they were distributed in the interacinar spaces or central zone of the pancreatic islets in all ages. However, the distributions and relative frequencies in the pancreatic islets of the neonate Korean native goat were divided into three patterns : 1) located in the inner zone with numerous frequencies, 2) the peripheral zone of the pancreatic islet with moderate frequencies and 3) the peripheral zone of the pancreatic islet with a few frequencies patterns.
A 2-year-old female Maltese dog was admitted with a history of pyometra and resulting peritonitis and septicemia. Uterine specimen sampled by ovariohysterectomy was processed routinely for histopathological observation. Grossly, the uterine mucosa was covered with necrotic debris and on the cut surface, lesion extended into the uterine wall. Microscopically, severe necrosis was observed throughout thickened mucosa, submucosa, and wall of uterus. Tumorous lesions composed of anaplastic cells with bizarre nuclei or tubular structures of cuboidal to short columnar cells were infrequently observed around the necrotic lesions and muscular layer far from necrotic areas. Immunohistochemically, central necrotic area with ambiguous cell and tissue structures, peri-necrotic tumor lesions, and muscular layer were strongly positive for cytokeratin. Since huge necrosis of adenocarcinoma lesions in this case made it difficult to diagnose, immunohistochemical results enable to diagnose as a severe necrotizing adenocarcinoma. Thus, histopathological and immunohistochemical findings in this case may serve as an important knowledge to diagnose uterine adenocarcinoma with huge necrosis in the veterinary field.
Gastrointestinal stromal tumors (GISTs) are the most common primary mesenchymal tumors of the digestive tract. They have been commonly observed in adults but have been rarely described in children. They arise typically from the intestinal wall and rarely in the mesentery, omentum, or retroperitoneum. GISTs originate from the interstitial cell of Cajal and are characterized by overexpression of the receptor tyrosine kinase c-kit. Up to 94% of these tumors express the CD117 on immunohistochemical stain. Surgery is the main modality of treatment for primary resectable GIST. Completely resectable GIST with low risk has excellent prognosis after primary surgical intervention, with over 90% of the 5-year survival. We report a case of 10-year-old girl presenting with an upper gastrointestinal bleeding caused by gastrointestinal stromal tumor.

This experiment was performed to investigate the effect of Sinjigolpy-tang on the diabetic rats induced by STZ. After experimental diabets was induced by 55mg/kg of STZ injection, we administered Sinjigolpy-tang extract for 14 days after STZ injection. Glucagon and insulin granules in Langerhans islets were stained by use of immunohistochemical(ABC) method and observed the relative amount of the each granules in Langerhans islet by light microscope and image analysis system. Area % of insulin granules in Langerhans islets in Sinjigolpy-tang increased and showed the statistically significant difference with the control group at 14th day. Area % of glucagon granules in Langerhans islets in Sinjigolpy-tang decreased and showed the statistically significant difference with the control group at 7th and 14th day. It can be inferred that Sinjigolpy-tang has a control effect on glucagon and insulin granules in Langerhans islets of diabetic rats induced by STZ.
This study was aimed at investigating the possible effects of phytoceramide (Pcer) on learning and memory and their underlying mechanisms. Phytoceramide was orally administered to ICR mice for 7 days. Memory performances were assessed using the passive avoidance test and Y-maze task. The expressions of phosphorylated cAMP response element binding protein (pCREB), brain-derived neurotrophic factor (BDNF) were measured with immunoblot. The incorporation of 5-bromo-2-deoxyuridine (BrdU) in hippocampal regions was investigated by using immunohistochemical methods. Treatment of Pcer enhanced cognitive performances in the passive avoidance test and Y-maze task. Immunoblotting studies revealed that the phosphorylated CREB and BDNF were significantly increased on hippocampus in the Pcer-treated mice. Immunohistochemical studies showed that the number of immunopositive cells to BrdU was significantly increased in the hippocampal dentate gyrus regions after Pcer-treatment for 7 days. These results suggest that Pcer contribute to enhancing memory and BDNF expression and it could be secondary to the elevation of neurogenesis.
To investigate the regional distribution and relative frequency of the neurotensin-, pancreatic polypeptide(PP)- and gastrin/cholecystokinin(Gas/CCK)-immunoreactive cells in the gastrointestinal tract of the bullfrog(Rana catesbeiana) with developmental stages, group of bullfrogs subdivided into the tadpole with hindlegs, metamorphosed bullfrog with tail, 2 weeks after metamorphosed bullfrog and adult bullfrog, were stained by immunohistochemical methods (PAP methods). Neurotensin-immunoreactive cells were observed from the pylorus of the metamorphosed bullfrog with tail, but these cells were not detected after that periods. PP-immunoreactive cells were detected from the adult bullfrog in the pylorus, duodenum and ileum. These cells were most predominant in the pylorus. Gas/CCK-immunoreactive cells were observed from the adult bullfrog in the pylorus. According to these results, most of immunoreactive cells in the gastrointestinal tract of the bullfrog were appeared after the complete metamorphosed periods, in which the complete differentiation of structure of gastrointestinal tract were occurred, and variable changes of the regional distribution and relative frequency with developmental stages were observed.
Pancreatic endocrine cells containing glucagon, insulin, somatostatin and pancreatic polypeptide were identified in the pancreas of the Korean native goat by using immunohistochemical method. Glucagon immunoreative cells were oval or fusiform in shape and located at the periphery of the pancreatic islets. Insulin immunoreactive cells were round or oval in shape and occupied throughout the pancreatic islets except the small area of the periphery. Somatostatin immunoreative cells were oval and elliptical, and mainly located at the periphery of the pancreatic islets. Some of these cells had a cytoplasmic process. Pancreatic polypeptide immunoreactive cells were elliptical or polyhedral and located at the periphery of the pancratic islets where two or more cells formed a cell cluster. The distribution rates of glucagon, insulin, somatostatin and pancreatic polypeptide immunoreactive cells were 24.4%, 44.3%, 13.2% and 18.1% respectively.
We evaluated the effect of low power GaAsAl laser on re-epithelization in full-thickness excisional wound of rat skin. Two full-thickness excisions were made on the back of the experimental animals. Low power laser applications with 10mW intensity were treated experimental animals twice a day for 7 days. On the seventh postoperative day the quantitative analysis of re-epithelization was performed using immunohistochemical staining for proliferating cell nuclear antigen (PCNA). The majority of PCNA immunoreactive cells was observed at epithelial cells in the margin of full thickness excisional wound. The low power laser treatments significantly increased the number of PCNA immunoreactive cell as compared to that of non treated animal group (p<0.01). The shape of PCNA immunoreactive cell appeared as small dark, round to ovoid structures. Most PCNA immunoreactive cells exhibited a high intensity of staining that contrasted sharply with the surrounding background. In conclusion, these findings suggest that GaAlAs laser treatments effectively enhance the epithelial wound healing by the stimulating cell proliferation. Furthermore, the majority of cell proliferation occurred in the margin of full thickness excisional wound.
