Immunogenecity of Low Molecular Weight Immunogens in Laying Hens (닭에서 저급 분자량 항원의 면역원성)

  • The immunogenecity of low molecular weight ($MW{\le}30,000$) immunogens in laying hens was investigated. Immunogens were insulin derivatives and ${\beta}$-lactoglobulin(${\beta}-Lg$). Insulin derivatives were reduced- carboxymethylated(RCM) insulin, and RCM-A and RCM-B chain of insulin. The yolk antibodies against RCM-A chain of insulin appeared after the first immunization. The yolk antibodies against RCM-B chain of insulin were elicited 5 weeks after the third booster injection. Although the anti-RCM-B chain yolk antibodies recognized native insulin, the anti-RCM-A chain yolk antibodies didn't native insulin. The anti-RCM insulin yolk antibodies were induced after the second booster injection and showed cross-reactivities with native insulin. On the other hand, ${\beta}-Lg$ showed stronger immunogenecity than insulin derivatives. The $anti-{\beta}-Lg$ yolk antibodies were produced after the second booster injection and the peak titer was reached 3 weeks after the third booster injection.

  • IgY (egg yolk immunoglobulin) against Helicobacter pylori was produced by immunizing hen with some Helicobacter pylori antigens. H. pylori whole cell, whole cell lysates, partially purified urease and p54 protein, which showed high antigenicity in mice, were used as immunogens. Four hens were immunized with these immunogens three times. IgY was purified from immunized egg yolk with polyethylene glycol (M.W. 8000) and its anti-H. pylori titer was determined by enzyme linked immunosorbent assay (ELISA). The anti-H. pylori titer reached peak at 8 weeks and was maintained over 20 weeks. H. pylori cells were agglutinated with these purified IgY and the specificity of these purified IgY was detected with immunoblotting.

  • To identify the immunogens of a PRT strain of Infectious Hematopoietci Necrosis Virus (IHNV) isolated from cultrued fish in Korea (Park et al, 1993). a panel of 4 monoclonal antibodies (MAbs) against IHNV-PRT strain and two polyclonal antisera from rainbow trout survived IHN disease were prepared. Proteins of purified IHNV-PRT strain were analysed on 10% SDS-PAGE and transferred onto NC paper and were incubated with the antibody solutions. With the polyclonal antibodies, four bands ($M_1$, $M_2$, G and 90Kd) were detected and the band density was in the order of $M_2$ > 90Kd > $M_1$ > G. However, with the MAbs, only two bands(G and 90Kd) were detected. The origin of 90Kd protein was not clear but maybe cell. All the results represented that among the five proteins of IHNV-PRT strain (Park et al., 1993), $M_2$, $M_1$ and G proteins were immunogens and $M_2$ protein was the strongest one.

Control of swine respiratory disease using egg yolk antibodies I.   • Swine respiratory diseases have induced severe economic losses in swine industry worldwide. Several methods have been developed and applied to prevent and control the disease. However, those are still problematic in swine industry. Recently, the use of egg yolk antibodies with several advantages was introduced and applied to control diseases in animal as well as human. As the first step of the use of egg yolk antibodies in the control of the swine respiratory diseases, we investigated the immunogens of the causative agensts of the diseases and immune response in egg yolk of hens immunized with them. Bacterial antigens prepared from Bordetella bronchiseptica, Pasteurella multocida 3A and 4D, and Actinobacillus pleuropneumaniae serotype 2 and 5 were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, Western blot and toxicity test in mice. The antigens were injected into laying hens in order to produce antibodies against them in egg yolk. After chickens were immunized three times in 2 weeks interval, the profile of antibody production was examined by ELISA. The production of antibody in egg yolk was started in 2 weeks after the first injection, reached peak in 6-8 weeks and maintained until 12 weeks. Of two adjuvants used in this study, ISA70 was more effective than aluminum hydroxide gel in enhancing immunogenecity, laying rates and safety in hens. These results suggested that egg yolk antibodies could be a good source for production of antibodies specific to pathogenic bacteria inducing respiratory diseases of swine.

Protective efficacy of attenuated Salmonella Typhimurium strain expressing BLS, Omp19, PrpA, or SOD of Brucella abortus in goats

  • Background: Attenuated Salmonella strain can be used as a vector to transport immunogens to the host antigen-binding sites. Objectives: The study aimed to determine the protective efficacy of attenuated Salmonella strain expressing highly conserved Brucella immunogens in goats. Methods: Goats were vaccinated with Salmonella vector expressing individually lipoprotein outer-membrane protein 19 (Omp19), Brucella lumazine synthase (BLS), proline racemase subunit A (PrpA), Cu/Zn superoxide dismutase (SOD) at 5 × 109 CFU/mL and challenge of all groups was done at 6 weeks after vaccination. Results: Among these vaccines inoculated at 5 × 109 CFU/mL in 1 mL, Omp19 or SOD showed significantly higher serum immunoglobulin G titers at (2, 4, and 6) weeks post-vaccination, compared to the vector control. Interferon-γ production in response to individual antigens was significantly higher in SOD, Omp19, PrpA, and BLS individual groups, compared to that in the vector control (all p < 0.05). Brucella colonization rate at 8 weeks post-challenge showed that most vaccine-treated groups exhibited significantly increased protection by demonstrating reduced numbers of Brucella in tissues collected from vaccinated groups. Real-time polymerase chain reaction revealed that Brucella antigen expression levels were reduced in the spleen, kidney, and parotid lymph node of vaccinated goats, compared to the non-vaccinated goats. Besides, treatment with vaccine expressing individual antigens ameliorated brucellosis-related histopathological lesions. Conclusions: These results delineated that BLS, Omp19, PrpA, and SOD proteins achieved a definite level of protection, indicating that Salmonella Typhimurium successfully delivered Brucella antigens, and that individual vaccines could differentially elicit an antigen-specific immune response.

Induction of Immunological Tolerance by Treatment of Ginseng Extract (인삼 엑기스의 경구 면역 관용에 관한 연구)

  • In order to develop new bioactive functions ginseng extract, it was studies whether the ginseng extracts on the induction of immunological tolerance In mice. Oral immunologic tolerance was induced by the secondary exposure of egg albumin + alum following gastrointestinal exposure nth egg albumin In mice, and the effect on anti EA antibody in blood, 7 cell subset in spleen were Investigated. The results obtained were as follows. EA group and EA + GE group was capable of conferring tolerance, contained a profound for 5 weeks experimental but saline group restricted to induce tolerance. GE group did not show the activity of tolerance by the first immunogens exposure, but induced the tolerance by the secondary exposure. And also spleen T cells, CD 8+ and CD 4+ were decreased. These results suggested that ginseng may affect the induction of immunological tolerance, which may be associated proliferative response of CD 4+ and CD 8+ in splenocyte.

  • Hematological studies were conducted in reference to the immune enhancing mechanism of formalin applied to, as an inactivating agent, a formalin inactivaed anthrax vaccines in rabbits. Rabbits were inoculated two types of formalinized anthrax immunogens namely capsular and spore vaccines in addition of formalin saline as a control. From immune rabbits, peripheral blood was collected and subjected to count a total erythrocytes, leukocytes, and pyroninophilic lymphocytes. The experimental results were summarized as followings. At a level of 0.5M 0.5ml formalin with or without the addition to vaccine, a total leukocytes count was increased. Due to the increased lymphocytes, the ratio of neutrophil and lymphocyte was lowered within 4 to 12 hours of the postinoculation. Formalin saline, anthrax spore vaccine and capsular vaccine, without group difference, caused an increased level of pyroninophilic lymphocytes in peripheral blood. Throughout the studies, a possible role of immune enhancement by formlin was disscused and suggested.

Development of an ELISA for the Organophosphorus Insecticide Chlorpyrifos

  • A selective enzyme-linked immunosorbent assay (ELISA) for the insecticide chlorpyrifos was developed. Four haptens for chlorpyrifos were synthesized and two of them were used as immunogens after coupling to keyhole limpet hemocyanin by two differe nt approaches. Rabbits were immunized with either of them and the sera were screened against 4 haptens coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigencoated ELISA was developed, which shows an I50 of 160 ppb with a detection limit of 10 ppb. An antibody-coated ELISA was also developed, which shows an $I_{50}$ of 20 ppb with a detection limit of 0.1 ppb. The antibodies showed negligible cross-reactivity with other organophosphorus pesticides except for insecticides chlorpyrifos-methyl and bromophos-ethyl, which makes these assays suitable for the selective detection of chlorpyrifos.

Production of Recombinant Proteins as Immuno-Analytical Markers of Genetically-Modified Organisms (GMO)

  • Marker proteins of genetically-modified organisms (GMO) and their antibodies were prepared and characterized as major components of an analytical system. We selected two GMO markers, neomycin phosphotransferase II and 5- enolpyruvylshikimate-3-phosphate synthase, and produced them from E. coli employing genetic recombination technology. After purification, their structural conformation and binding affinities to the respective antibodies were characterized. The results showed that the recombinant proteins were identical with commercially obtained reference proteins. We further used them as immunogens to raise polyclonal antibodies capable of discriminating GMO containing protein from non-GMO. Well-characterized marker proteins and antibodies will be valuable as immunoreagents in constructing analytical systems such as biosensors and biochips to measure quantities of GMO.

Development of an ELISA for the Organophosphorus Insecticide Isofenphos

  • A selective enzyme-linked immunosorbent assay (ELISA) for the insecticide isofenphos was developed. Three different analogues (haptens) of isofenphos were synthesized and were coupled to carrier proteins through the pesticide thiophosphate group t o use as immunogens or coating antigens. Rabbits were immunized with one of the haptens coupled to BSA for production of polyclonal antibodies and the sera were screened against each of the other two haptens coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigen-coated ELISA was developed, which showed an I50 of 96 ng/mL with the detection limit of 2 ng/mL. The antibodies showed negligible cross-reactivity with other organophosphorus pesticides and the phenol metabolite of isofenphos, which makes the developed assay suitable for the selective detection of isofenphos. An antibody-coated ELISA was also developed, which showed an I50 of 580 ng/mL with a detection limit of 70 ng/mL.
