Sphingolipids in neuroinflammation: a potential target for diagnosis and therapy

  • Sphingolipids are ubiquitous building blocks of eukaryotic cell membranes that function as signaling molecules for regulating a diverse range of cellular processes, including cell proliferation, growth, survival, immune-cell trafficking, vascular and epithelial integrity, and inflammation. Recently, several studies have highlighted the pivotal role of sphingolipids in neuroinflammatory regulation. Sphingolipids have multiple functions, including induction of the expression of various inflammatory mediators and regulation of neuroinflammation by directly effecting the cells of the central nervous system. Accumulating evidence points to sphingolipid engagement in neuroinflammatory disorders, including Alzheimer's and Parkinson's diseases. Abnormal sphingolipid alterations, which involves an increase in ceramide and a decrease in sphingosine kinase, are observed during neuroinflammatory disease. These trends are observed early during disease development, and thus highlight the potential of sphingolipids as a new therapeutic and diagnostic target for neuroinflammatory diseases.

면역조혈계 및 재생조직의 방사선 손상에 대한 생약복합물(HIM-I)의 방호 효과 (Effect of a Herb Mixture (HIM-I) on the Protection of the Hematopoietic-Immune System and Self-renewal Tissues against Radiation Damage)

  • 방사선 생체손상의 방어 및 회복에 높은 효과를 나타내는 생약조합을 도출하고자 하였다. 이전의 연구에서 6가지 기존 보기${\cdot}$보혈 한약처방제의 방어효과를 검색하여, 재생조직 방어에는 사물탕 및 보중익기탕이, 조혈계 방어에는 보중익기탕 및 사물탕이 높은 효과를 나타내는 것으로 보고된바 있다. 본 연구에서 기존 6가지 처방제의 면역 세포 활성화 효과를 검색한 결과 모두 높은 효과를 나타내지 않았다. 따라서, 처방제의 구성 생약 각각의 효과검증 결과를 바탕으로 재생조직 방어, 조혈계 방어, 면역조혈계 회복 증진 효과를 동시에 높게 나타내는 새로운 생약조합을 도출하였다. 즉, 당귀, 천궁, 백작약을 동일 무게 비율로 혼합하여 열수 추출한 것을 새로운 생약복합물 HIM-I으로 개발하였으며 그 효과를 검정하였다. 본 생약복합물 HIM-I은 면역 세포 활성화에서는 상기한 한약처방제보다 월등하게 높은 효과를 보였으며, 조혈계 촉진효과도 높은 것으로 나타났다. 또한, 재생조직 및 면역조혈계의 방어 측면에서도 사물탕 및 보중익기탕과 같은 높은 효과를 보였다. 한편, 시험관 내에서 높은 항산화 효과를 나타내는 것으로 보아, 이 항산화 작용이 방사선 손상에 대한 방어효과의 작용기전 중의 하나로 생각되었다. 이상의 결과로 보아, 생약복합물 HIM-I은 재생조직방어, 조혈계 방어, 면역조혈계 회복 증진 효과를 동시에 나타냄으로써 방사선뿐만 아니라 여러 가지 원인으로부터 생체를 방호하고 회복시키는데 유용할 것으로 생각된다.

Health monitoring of a historical monument in Jordan based on ambient vibration test

  • This paper summarizes the experimental vibration-based structural health monitoring study on a historical monument in Jordan. In this work, and within the framework of the European Commission funded project "wide-Range Non-Intrusive Devices Toward Conservation of Historical Monuments in the Mediterranean Area", a seven and a half century old minaret located in Ajloun (73 km north of the capital Amman) is studied. Because of their cultural value, touristic importance and the desire to preserve them for the future, only non-destructive tests were allowed for the experimental investigation of such heritage structures. Therefore, after dimensional measurements and determination of the current state of damage in the selected monument, ambient vibration tests are conducted to measure the accelerations at strategic locations of the system. Output-only modal identification technique is applied to extract the modal parameters such as natural frequencies and mode shapes. A Non-linear version of SAP 2000 computer program is used to develop a three-dimensional finite element model of the minaret. The developed numerical model is then updated according to the modal parameters obtained experimentally by the ambient-vibration test-results and the measured characteristics of old stone and deteriorated mortar. Moreover, a parametric identification method using the N4Sid state space model is employed to model the dynamic behavior of the minaret and to build up a robust, immune and noise tolerant model.

위암 치료에 대한 한방 임상 가이드라인 (Clinical Practice Guidelines of Korean Medicine for Gastric Cancer)

  • Objectives : The purpose of this study was to present the clinical guidelines for gastric cancer. Methods : The literature on Western and Oriental medical treatment of gastric cancer were collected, analyzed and summarized from the Google Scholar, KIOM OASIS and PUBMED from 1993 to 2014. Results : Gastric cancer is the second most common cancer in Korea. In recent studies, applying integrative oriental and western medicine can suppress the tumor, improve the survival, the immune system, and the quality of life in gastric cancer. But there isn't still a unified protocol for gastric cancer treatment, so we have difficulty in clinical application. This study will be helpful for understanding and building systems for integrative gastric cancer treatment. Conclusion : Further studies on integrative gastric cancer treatment are needed to improve the survival of gastric cancer patients and build the clinical practice guidelines of gastric cancer.

Engineering implications of the RC building damages after 2011 Van Earthquakes

  • Two destructive earthquakes occurred on October 23 and November 9, 2011 in Van province of Turkey. The damage in residential units shows significant deviation from the expectation of decreasing damage with increasing distance to epicenter. The most damaged settlement Ercis has the same distance to the epicenter with Muradiye, where no damage occurred while relatively less damage observed in Van having half distance. These three cities seem to have resembling soil conditions. If the damages are evaluated: joint failures and insufficient lap splice lengths are observed to be the main causes of the total collapses in RC buildings. Additionally, low concrete strength, reinforcement detailing mistakes, soft story, heavy overhang, pounding and short columns are among other damage reasons. Examples of damages due to non-structural elements are also given. Remarkable points about seismic damages are: collapsed buildings with shear-walls, heavily damaged buildings despite adequate concrete strength due to detailing mistakes, undamaged two-story adobe buildings close to totally collapsed RC ones and undamaged structural system in buildings with heavily damaged non-structural elements. On the contrary of the common belief that buildings with shear-walls are immune to total collapse among civil engineers, collapse of Gedikbulak primary school is a noteworthy example.

Preparation of Alginate/Chitosan Microcapsules and Enteric Coated Granules of Mistletoe Lectin

  • The aqueous extract of European mistletoe (Viscum album, L.) has been used in cancer therapy. The purified mistletoe lectins, main components of mistletoe, have demonstrated cytotoxic and immune-system-stimulating activities. Korean mistletoe (Viscum album L. coloratum), a subspecies of European mistletoe, has also been reported to possess anticancer and immunological activities. A galactose- and N-acetyl-D-galactosamine-specific lectin (Viscum album L. coloratum agglutinin, VCA) with Mr 60 kDa was isolated from Korean mistletoe. Mistletoe preparations have been given subcutaneously due to the low stability of lectin in the gastrointestinal (GI) tract. In the present study, we investigated the possibility of alginate/chitosan microcapsules as a tool for oral delivery of mistletoe lectin. In addition, our strategy has been to develop a system composed of stabilizing cores (granules), which contain mistletoe lectin, extract or powder, coated by a biodegradable polymer wall. Our results indicated that successful incorporation of VCA into alginate/chitosan microcapsules has been achieved and that the alginate/chitosan microcapsule protected the VCA from degradation at acidic pH values. And coating the VCA with polyacrylic polymers, Eudragit, produced outstanding results with ideal release profiles and only minimal losses of cytotoxicity after manufacturing step. The granules prepared with extract or whole plant produced the best results due to the stability in the extract or whole plant during manufacturing process.

Human Milk Oligosaccharides as a Missing Piece in Combating Nutritional Issues during Exclusive Breastfeeding

  • Extensive studies have shown that breast milk is the best source of nutrition for infants, especially during the first six months, because it fulfills almost all of their nutritional needs. Among the many functional building blocks in breast milk, human milk oligosaccharides (HMOs) have been receiving more attention recently. Furthermore, it is the third most common group of compounds in human milk, and studies have demonstrated the health benefits it provides for infants, including improved nutritional status. HMOs were previously known as the 'bifidus factor' due to their 'bifidogenic' or prebiotic effects, which enabled the nourishment of the gastrointestinal microbiota. Healthy gastrointestinal microbiota are intestinal health substrates that increase nutrient absorption and reduce the incidence of diarrhea. In addition, HMOs, directly and indirectly, protect infants against infections and strengthen their immune system, leading to a positive energy balance and promoting normal growth. Non-modifiable factors, such as genetics, and modifiable factors (e.g., maternal health, diet, nutritional status, environment) can influence the HMO profile. This review provides an overview of the current understanding of how HMOs can contribute to the prevention and treatment of nutritional issues during exclusive breastfeeding.

백작약 조다당분획에 의한 B 세포 증식의 특성 (Characteristics of B cell proliferation by polysaccharide fraction of Paeonia japonica miyabe)

  • Background : Paeonia japonica Miyabe is a medicinal plant which has been widely used as a component of blood-building decoctions (Chinese medicinal concept : Bu-Xie). The immunopharmacological characteristics of the extract of Paeonia japonica (PJ) were investigated. Methods : The effects of fractions of PJ extract on lymphocyte proliferation were measured by $H^3$-thymidine incorporation assay. The proliferated lymphocyte subsets were analyzed in flow cytometry. The subset cell populations of spleen cells were separated by magnetic cell separation system, and their proliferation by the extract were investigated. The effect of the extract on antibody production was determined in mice challenged with sheep red blood cells (SRBC) using hemolytic plaque forming cell assay. Results : Spleen cells were proliferated by water extract of PJ. Polysaccharide fraction (PJ-P) of the extract was most active in the proliferation. It was found in flow cytometry that the lymphocyte subset proliferated by PJ-P was B cell population. Among the separated subset cell populations, T cell-depleted cell population and macrophage-depleted cell population were most proliferated by PJ-P. However, positively selected populations of B cells and T cells were not proliferated by PJ-P. These results indicate that B cell proliferation by PJ-P may require the assistance of macrophages or T cells. These results suggest that firstly PJ-P may stimulate macrophages or T cells, and then B cells are activated. The number of antibody-secreting cells was increased by administration of PJ-P in mice immunized with SRBC as a T-dependent antigen. Conclusion : These results suggest that macrophages and accessory cells are directly activated by PJ-P and then helper T cells and B cells are indirectly activated. As the results, immune responses might be coordinately improved. In conclusion, PJ-P, a polysaccharide of P. japonica, may be a characteristic immunostimulator, which is analogous to polysaccharides such as lentinan, PSK and ginsan.

Use of δ-Aminolevulinic Acid in Swine Diet: Effect on Growth Performance, Behavioral Characteristics and Hematological/Immune Status in Nursery Pigs

  • Certain amino acids are essential precursors of a variety of important biomolecules in addition to their major function as protein building blocks. ${\delta}$-Aminolevulinic acid (ALA) is synthesized from the condensed form of succinyl-CoA with glycine after decarboxylation catalyzed by ALA synthase. The objective of the study was to determine the effects of ALA supplementation on growth performance, behavioral characteristics and hematological/immune status in nursery pigs. A total of 144 pigs weaned at 21 d of age were allotted to three dietary treatments representing (-) control (w/o antibiotics; NC), (+) control (w/carbadox at 50 mg/kg; PC), and the treatment group with ALA supplementation (0.05%; TA). Each treatment had 6 pens (replicates) with 8 pigs per pen. Pigs were fed phase 1 (21.9% CP, 1.40% Lys) and 2 (20.6% CP, 1.15% Lys) experimental diets for 3 and 2 wks, respectively. Feed intake and weight gain were measured weekly during phase 1 and at the end of phase 2. At the end of phase 2, blood samples were taken and analyzed using an automated hematology analyzer. Skin color and activity of pigs (48 h) from all pens in each treatment were measured at the second week of phase 2. Growth performance was not affected (p>0.05) by the dietary supplementation of ALA during the 5 wk nursery period. Pigs in the TA (6.46) and PC (6.68) had a higher (p<0.05) number of red blood cells ($10^6cell/{\mu}L$) than pigs in the NC (6.15). Pigs in PC (12.16) had a higher (p<0.05) hemoglobin level (g/dL) than pigs in the NC group (11.29) and the TA group (11.47). Pigs in the TA and PC had darker (p<0.05) and less (p<0.05) yellow skin color than pigs in the NC. Pigs in the PC tended (p = 0.081) to be less active than pigs in the other groups. There were no differences in behavioral characteristics between the NC and the TA. The data suggest that ALA supplementation has no adverse effects on growth performance of nursery pigs. Moreover, ALA supplementation increased red blood cell counts which may be beneficial to pigs.

  • Lipids, which along with carbohydrates and proteins are among the most important nutrients for the living organism, have a variety of biological functions that can be applied widely in biomedicine. A fatty acid, the most fundamental biological lipid, may be classified by length of its aliphatic chain, and the short-, medium-, and long-chain fatty acids and each have distinct biological activities with therapeutic relevance. For example, short-chain fatty acids have immune regulatory activities and could be useful against autoimmune disease; medium-chain fatty acids generate ketogenic metabolites and may be used to control seizure; and some metabolites oxidized from long-chain fatty acids could be used to treat metabolic disorders. Glycerolipids play important roles in pathological environments, such as those of cancers or metabolic disorders, and thus are regarded as a potential therapeutic target. Phospholipids represent the main building unit of the plasma membrane of cells, and play key roles in cellular signaling. Due to their physical properties, glycerophospholipids are frequently used as pharmaceutical ingredients, in addition to being potential novel drug targets for treating disease. Sphingolipids, which comprise another component of the plasma membrane, have their own distinct biological functions and have been investigated in nanotechnological applications such as drug delivery systems. Saccharolipids, which are derived from bacteria, have endotoxin effects that stimulate the immune system. Chemically modified saccharolipids might be useful for cancer immunotherapy or as vaccine adjuvants. This review will address the important biological function of several key lipids and offer critical insights into their potential therapeutic applications.
