An Experimental Study on the Anti-inflammatory Effect of Taeeumjowui-tang (Taiyintiaowei-tang) in Allergic Late Inflammation (알레르기 후기 반응 염증 억제 효과에 관한 태음조위탕(太陰調胃湯)의 실험적 연구)

  • Objective : Allergic disease, a very common chronic illness that affects all ages of patients, has been well characterized as an IgE-dependent immunologic response. Recently, interest has grown about the late inflammatory reaction as well as the early one characterized by IgE and mast cells in allergic disease. The purpose of this study was to find the anti-inflammatory effect of Taeeumjowui-tang (Taiyintiaowei-tang) in allergic reaction. Methods : The experiment was performed using Raw 264.7 cells pretreated with Taeeumjowui-tang contents extracts. The results were measured for different concentrations of Taeeumjowui-tang extracts (100, 200, $300{\mu}g$/ml): the toxicity and proliferation of cells by MTT analysis, LPS-induced NO production using Griess reagent and IL-6/ TNF-${\alpha}$ production, which is a significant criteria for diagnosing allergic reaction. Results : No toxicity of Taeeumjowui-tang (100, 200, $300{\mu}g$/ml) on Raw 264.7 cells was found after 24 hours incubation. LPS-induced NO production was reduced after treatment with Taeeumjowui-tang (100, 200, $300{\mu}g$/ml)(P<0.001). IL-6 decreased only at $100{\mu}g$/ml(P<0.05). TNF-${\alpha}$ production decreased only at $300{\mu}g$/ml, but still statistically insignificant. There was no relationship between any components of Taeeumjowui-tang alone and inhibition of NO production. Conclusions : These data suggest that Taeeumjowui-tang has anti-inflammatory effects in allergic reaction.

Analysis of Recent Clinical Studies to Establish Korean Herbal Medicine Clinical Trial Guidelines for the Common Cold (감기 임상시험 가이드라인 제정을 위한 최신 임상시험 연구 분석)

  • Objectives: The aim of this study was to help develop a guideline for the common cold. We searched recent clinical studies of the common cold in Western medicine and reviewed their objectives, inclusion and exclusion criteria, primary outcome, secondary outcome, and assessment tools to establish evidenced-based guideline.Methods: We searched electronic databases (Cochrane Library, MEDLINE, EMBASE) to identify eligible randomized controlled trials (RCTs) about the common cold for the last 10 years. We included 29 RCTs and showed their research summary via their objectives, participants, interventions, control, treatment duration, and results. We also analyzed the definition of the common cold presented in the article, inclusion and exclusion criteria, primary and secondary outcomes, and assessment tools.Results: We reported the aforementioned areas in detail. At first, the definition of the common cold was confused across the articles. Second, herbal medication clinical trials for the common cold have been extensively studied recently. Third, the eligibility criteria frequently included the Jackson Symptom score. Fourth, validated assessment tools (i.e., the Wisconsin Upper Respiratory Symptom Survey-21) have only been used in a few recent studies.Conclusions: Our research will be helpful to establish Korean herbal medicine clinical trial guidelines for the common cold.

Preliminary Study for Development of Pattern Identification Tool of Chronic Cough (만성기침 변증도구 개발을 위한 기초연구)

  • Objectives : The aim of this study was to develop a standard tool of pattern identification for chronic cough, which will be applied to clinical research. Methods : The items and structures of the instrument were based on reviews of textbooks and published literature. The advisor committee on this study consisted of 11 Korean respiratory internal medicine professors, one clinical fellow, and five clinicians with 10 years of experience. The questionnaire, which includes the signs and symptoms of chronic cough, was studied by the delphi method. Delphi examination was carried out via email, through evaluating the importance of symptoms included in each pattern. Results : We divided the pattern identification of chronic cough into five patterns: Wind-Cold, Phlegm-Turbidity, Liver-Fire, Lung Deficiency, and Kidney Yang Deficiency. By the Delphi method and a score evaluation, 38 items were chosen for pattern identification of chronic cough. Conclusions : Through this study, we created a Korean instrument for the pattern identification tool for chronic cough. We expect to apply this tool to subsequent research as its validity and reliability are further confirmed.

  • Objectives : Ryodoraku, which is a physiological function test using electric current, is closely related to skin sympathetic tone. Pulse analysis is known to reflect cardiovascular reactivity. Refer to the previous researches Ryodoraku and pulse analysis have value as tools for diagnosing respiratory diseases. In this study we examined the diagnostic values of Ryodoraku and pulse analysis for respiratory disease patients. Methods : For this study. we conducted Ryodoraku and pulse analysis on 114 people, including 83 respiratory disease outpatients and 31 volunteers who did not have any respiratory symptoms or disease history. The respiratory patients were divided into three subgroups according to their symptoms: rhinorrhea group, cough-sputum group and wheezing-dyspnea group. Then we compared the disease groups with the control group. Results : When all experimental groups were compared with the control group, mean Ryodoraku was significantly lower. Mean H2, mean H3 and mean H6 were significantly lower in the rhinorrhea group (P<0.05), all the test results of Ryodoraku were evidently lower in the cough-sputum group (P<0.01), and most results of Ryodoraku were evidently lower in the wheezing-dyspnea group except H1 (P<0.01). Compared with the control group on pulse analysis, mean YP+/YP- was significantly lower in the wheezing-dyspnea group (P<0.05). Conclusion : Ryodoraku and pulse analysis were found to have a high value as quantitative diagnosis tools reflecting individuals' weakness and firmness. Nevertheless, more research is needed to find the further values.

  • Objectives: This study aimed to evaluate any clinical research regarding asthma published by the Korean Oriental Medical Society and its sub-societies for future rigorous clinical research. Methods: Every article relevant to asthma was initially obtained from journals in the Korean Oriental Medical Society and its sub-societies by electronic search at journal web sites or manual search. Journals were limited to those registered by the Korea Research Foundation. From initial findings, two independent reviewers selected clinical articles and these articles were further analyzed separately into predefined criteria according to prospective and retrospective studies. For prospective studies, quality assessment was also conducted. Results: From 99 articles initially obtained, 19 were finally analyzed. 14 were prospective whereas 5 were retrospective. Among the prospective articles, there were no randomized controlled trials and the non-randomized studies had no control groups, with several problems of quality regarding pre-calculated study size and unbiased assessment. None of the retrospective studies described asthma diagnosis objectively and outcome measures were either non-relevant to asthma or non-validated. Conclusions: Well-designed randomized controlled studies for asthma are needed and more rigorous non-randomized controlled studies should be conducted.

Effects of Arctium lappa Linne on the DNFB-induced allergic dermatitis (우방자(牛蒡子)의 아토피 피부염에 대한 효과)

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease which is characterized by severe pruritis, erythema, edema, effusion and scabs. The aim of this study was to understand effects of Arctium lappa Linne, which is well known for its efficacy for various skin diseases, on atopic dermatitis Methods : We conducted this experiment using the DNFB-induced NC/Nga mice. After two weeks of applying DNFB to NC/Nga mice, severe symptoms of atopic dermatitis occurred. We divided the mice into three different groups: a control group which was given no treatment at all, a group treated with dexamethasone (1mg/kg), and another group treated with Arctium lappa Linne (300mg/kg). After one week of treatment, results were recorded according to their improvement on skin, itching behavior, IL-4, and INF-$\gamma$ measurement, which is a significant criterion for diagnosing atopic dermatitis. Results : Itching behavior showed significant improvement in the Arctium lappa Linne group. However Arctium lappa Linne failed to reduce IL-4 and INF-$\gamma$ count. Also there was no satisfying improvement on AD-like skin lesions on the rostral back of the NC/Nga mice treated with Arctium lappa Linne. Conclusions : Arctium lappa Linne showed improvement in itching behavior in NC/Nga mice induced by DNFB. Arctium lappa Linne showed neither significant improvement on skin lesions nor in IL-4 and INF-$\gamma$ measurement.

Research on Anti-inflammatory Effects of Forsythiae Fructus (연교(連翹)의 LPS로 유도된 Raw 264.7 Cell에서의 항염증(抗炎症)효과)

  • Objectives : The purpose of this study was to examine the anti-inflammatory effects Forsythiae Fructus on LPS-activated RAW264.7 cells. Methods : Statistical significance was examined through measuring MTT, nitric oxide (NO), TNF $\alpha$, IL 6, NOS2 and COX2 of LPS-activated RAW264.7 cells. Results : In the toxicity experiment of FFE, NO significant toxicity was shown on cells in the concentration ranges of 100, 200 and 300 ${\mu}g/ml$. FFE dose dependently decreases 5.49, 25.41, 33.64 % in LPS induced NO production (p<0.05, p<0.001 and p<0.001), however, FFE don't show significant difference in LPS-induced TNF $\alpha$ and IL 6. Furthermore, FFE showed inhibiting tendency against the revelation of NOS2 and COX2 in LPS-activated RAW264.7 cells, dose-dependently according to concentration. Conclusions : These results mean that FFE is effective for anti-inflammation via inhibition of NOS2 and COX2 expression.

  • Errors in the semiconductor process are generated by a change in the state of the equipment, and errors usually arise when the state of the equipment changes or when parts that make up the equipment have flaws. In this investigation, we anticipated that aging of the mass flow controller in the plasma enhanced chemical vapor deposition SiO2 thin film deposition method caused a minute flow rate shift. In seven cases, fourier transformation infrared film quality analysis of the deposited thin film was used to characterize normal and pathological processes. The plasma condition was monitored using optical emission spectrometry data as the flow rate changed during the procedure. Preprocessing was used to apply the collected OES data to the artificial immune system algorithm, which was then used to process diagnosis. Through comparisons between datasets, the learning algorithm compared classification accuracy and improved the method. It has been confirmed that data characterized as a normal process and abnormal processes with differing flow rates may be discriminated by themselves using the artificial immune system data mining method.

The contribution of the nervous system in the cancer progression

  • Cancer progression is driven by genetic mutations, environmental factors, and intricate interactions within the tumor microenvironment (TME). The TME comprises of diverse cell types, such as cancer cells, immune cells, stromal cells, and neuronal cells. These cells mutually influence each other through various factors, including cytokines, vascular perfusion, and matrix stiffness. In the initial or developmental stage of cancer, neurotrophic factors such as nerve growth factor, brain-derived neurotrophic factor, and glial cell line-derived neurotrophic factor are associated with poor prognosis of various cancers by communicating with cancer cells, immune cells, and peripheral nerves within the TME. Over the past decade, research has been conducted to prevent cancer growth by controlling the activation of neurotrophic factors within tumors, exhibiting a novel attemt in cancer treatment with promising results. More recently, research focusing on controlling cancer growth through regulation of the autonomic nervous system, including the sympathetic and parasympathetic nervous systems, has gained significant attention. Sympathetic signaling predominantly promotes tumor progression, while the role of parasympathetic signaling varies among different cancer types. Neurotransmitters released from these signalings can directly or indirectly affect tumor cells or immune cells within the TME. Additionally, sensory nerve significantly promotes cancer progression. In the advanced stage of cancer, cancer-associated cachexia occurs, characterized by tissue wasting and reduced quality of life. This process involves the pathways via brainstem growth and differentiation factor 15-glial cell line-derived neurotrophic factor receptor alpha-like signaling and hypothalamic proopiomelanocortin neurons. Our review highlights the critical role of neurotrophic factors as well as central nervous system on the progression of cancer, offering promising avenues for targeted therapeutic strategies.

The Roles of Innate Lymphoid Cells in the Development of Asthma

  • Asthma is a common pulmonary disease with several different forms. The most studied form of asthma is the allergic form, which is mainly related to the function of Th2 cells and their production of cytokines (IL-4, IL-5, and IL-13) in association with allergen sensitization and adaptive immunity. Recently, there have been many advances in understanding non-allergic asthma, which seems to be related to environmental factors such as air pollution, infection, or even obesity. Cells of the innate immune system, including macrophages, neutrophils, and natural killer T cells as well as the newly described innate lymphoid cells, are effective producers of a variety of cytokines and seem to play important roles in the development of non-allergic asthma. In this review, we focus on recent findings regarding innate lymphoid cells and their roles in asthma.
