  • 본 논문에서는 새로운 방식으로 1차 상관면역 함수를 설계하는 방법을 제시하고 이 방법을 이용하여 1차 상관면역 함수 개수의 하한값을 구한다. 이 값은 1차 상관면역 함수의 개수에 대한 기존의 제시된 결과를 크게 개선한 값이다.

면역세포 활성에 대한 Substance P의 영향 (Effects of Substance P on the Activities of Immune Cell)

  • The neuropeptide substance P(SP) has been recognized to modulate immune systems, with close proximity between peptidergic sensory nerve endings and immune cells. These include the macrophage and neutrophil activation, IL-2 production in T cell, augmentation of Ig synthesis, mast cell degranulation, $PGE_2$ and collagenase secretion in synoviocytes. In this study I examined SP-induced various biological activities such as antimicrobial action, cytokine production, and mast cell degranulation in the presence or absence of other inflammatory cell activators. Antimicrobial studies showed that undifferentiated HL-60 cells were not affected by SP. However, SP significantly enhanced antimicrobial action of TPA-treated or dbcAMP-treated HL-60 cells which had been differentiated into PMN or macrophage/monocyte. I could not find synergistic relationship between SP and LPS in parallel experiments of the above. SP did not induce IL-l production from murine macrophage cell line RAW264.7 whether costimulated with LPS or not. Mast cell degranulation was occured only when stimulated with high dose ($10^{-5}M$) of SP and the degree of this activation was slightly reduced by simultaneous application of $MIP-1{\alpha}$. In addition, CGRP which is known to be a common coexisting neuropeptide with SP within specific fibers did not augment the function of SP on mast cell degranulation. These results suggest that immunoregulatory activities of SP could be mediated through direct upregulation of various functions of immune cells and also upregulation of responsiveness of immune cells to other immune activators.

Restoration of Declined Immune Responses and Hyperlipidemia by Rubus occidenalis in Diet-Induced Obese Mice

  • Hyperlipidemia, which is closely associated with a fatty diet and aging, is commonly observed in the western and aged society. Therefore, a novel therapeutic approach for this disease is critical, and an immunological view has been suggested as a novel strategy, because hyperlipidemia is closely associated with inflammation and immune dysfunction. In this study, the effects of an aqueous extract of Rubus occidentalis (RO) in obese mice were investigated using immunological indexes. The mice were fed a high-fat diet (HFD) to induce hyperlipidemia, which was confirmed by biochemical analysis and examination of the mouse physiology. Two different doses of RO and rosuvastatin, a cholesterol synthesis inhibitor used as a control, were orally administered. Disturbances in immune cellularity as well as lymphocyte proliferation and cytokine production were significantly normalized by oral administration of RO, which also decreased the elevated serum tumor necrosis factor $(TNF)-{\alpha}$ level and total cholesterol. The specific immune-related actions of RO comprised considerable improvement in cytotoxic T cell killing functions and regulation of antibody production to within the normal range. The immunological evidence confirms the significant cholesterol-lowering effect of RO, suggesting its potential as a novel therapeutic agent for hyperlipidemia and associated immune decline.

As a Modulator, Multitasking Roles of SIRT1 in Respiratory Diseases

  • As far the current severe coronavirus disease 2019 (COVID-19), respiratory disease is still the biggest threat to human health. In addition, infectious respiratory diseases are particularly prominent. In addition to killing and clearing the infection pathogen directly, regulating the immune responses against the pathogens is also an important therapeutic modality. Sirtuins belong to NAD+-dependent class III histone deacetylases. Among 7 types of sirtuins, silent information regulator type-1 (SIRT1) played a multitasking role in modulating a wide range of physiological processes, including oxidative stress, inflammation, cell apoptosis, autophagy, antibacterial and antiviral functions. It showed a critical effect in regulating immune responses by deacetylation modification, especially through high-mobility group box 1 (HMGB1), a core molecule regulating the immune system. SIRT1 was associated with many respiratory diseases, including COVID-19 infection, bacterial pneumonia, tuberculosis, and so on. Here, we reviewed the latest research progress regarding the effects of SIRT1 on immune system in respiratory diseases. First, the structure and catalytic characteristics of SIRT1 were introduced. Next, the roles of SIRT1, and the mechanisms underlying the immune regulatory effect through HMGB1, as well as the specific activators/inhibitors of SIRT1, were elaborated. Finally, the multitasking roles of SIRT1 in several respiratory diseases were discussed separately. Taken together, this review implied that SIRT1 could serve as a promising specific therapeutic target for the treatment of respiratory diseases.

Effects of proteinate complex zinc on growth performance, hepatic and splenic trace elements concentrations, antioxidative function and immune functions in weaned piglets

  • Objective: To assess the effects of proteinate complex zinc (PC-Zn) on growth performance, antioxidative function, trace element concentrations and immune function in weaned piglets. Methods: Three hundred newly weaned barrows ($Duroc{\times}Landrace{\times}Yorkshire$), 28 days of age, were randomly allotted to 3 dietary groups of 5 replicate pens per group for 4 weeks of feeding. Experimental diets were: i) zinc deficient diet (ZnD, 24 mg/kg Zn supplementation from $ZnSO_4$), ii) inorganic Zn diet supplemented with 120 mg/kg of Zn from Zn sulfate ($ZnSO_4$), and iii) organic Zn diet supplemented with 120 mg/kg of Zn from PC-Zn. The body weight of pigs were recorded at the beginning, at the middle and at the end of the experiment, and the amount of feed supplied each day was recorded. Five barrows from each dietary treatment group were selected to be anesthetized and euthanized at the end of the trial to determine the Zn, Cu, Fe, and Mn concentrations, the hepatic metallothionein content, the levels of methane dicarboxylic aldehyde (MDA), Mn, and Cu/Zn superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) in the spleen, the levels of interleukin (IL)-2, IL-4, IL-10, interferon $(IFN)-{\gamma}$, $CD3^+$, $CD4^+$, and $CD8^+$ T lymphocyte. Results: The accumulation of Zn in the spleen, levels of SOD, GSH-Px, IL-4, IL-10, the proportions of $CD3^+$ and $CD4^+$ T lymphocyte, and the ratio of $CD4^+/CD8^+$ T lymphocyte were increased by organic Zn supplementation compared to ZnD, while the levels of MDA, $IFN-{\gamma}$, and proportion of $CD8^+$ T lymphocyte were lowered. Conclusion: These findings indicate that Zn can improve the antioxidant potential and immune functions of weaned piglets.

식이지방과 토마토가 BALB/c 마우스의 면역활성에 미치는 영향 (Effect of Dietary Fats and Tomato on the Immune Functions of BALB/c Mice)

  • 본 연구는 식이지방(우지, 잇꽃유)을 급여한 마우스에게 토마토가 면역활성에 미치는 영향을 평가하기 위하여 수행하였다. AIN-76 diet에 포화지방(beef tallow)과 불포화지방(safflower oil)을 각각 5%씩 중량비로 혼합한 식이를 대조군으로 하였고, 실험군은 토마토(수분함량, 95%)를 고려하여 건물기준으로 0.5%를 첨가한 실험식이를 제조하여 BALB/c 마우스에게 10주 동안 급여하였다. 지연성 과민반응검사는 4, 7, 10주에, 항체생산 세포수측 정은 7, 10주에 실시하였다. 혈액은 심장천자(heart puncture)로 채취하였으며 저장된 혈청은 양적혈구(SRBC)에 대한 적혈구응집소가측정(AGG test)에 이용하였다. 각각의 장기는 적출하여 칭량하였고, DTH test후 적출한 비장을 대상으로 세포조직학적 검사를 실시하였다. 토마토군은 대조군에 비해 4주와 10주의 비장계수(spleen index)와 흉선계수(thymus index)가 통계적으로 높았다(p<0.05). 양적혈구에 대한 지연성 과민반응검사에서 토마토군은 4주, 7주, 10주 모두 24시간의 반응값이 대조군 보다 통계적으로 유의하게 높았다(p<0.05). 토마토군은 3시간은 7주, 24시간은 4주, 48시간은 4주와 10주에서 대조군에 비해 통계적인 유의성이 있었다(p<0.05). 항체생산 세포수는 식이급여 7주에 토마토군이 대조군보다 통계적인 차이가 나타난 반면 10주는 차이가 없었다. 양적혈구에 대한 응집소가는 식이의 급여기간이 길어질수록 감소하였는데, 토마토군이 대조군에 비해서 특히 10주의 응집소가는 유의성이 있었다. 임파구(Iymphocyte)는 토마토군이 대조군보다 다소 증가하였고, 비장조직 검사결과 토마토군은 백색수(white pulp)가 심하게 증식되어 활발한 면역반응을 보였다.

보중익기탕가미방의 면역기능 증진 효과 (Immune Function-enhancing Effects of Bojungikkitanggami-bang)

  • The immune system acts to protect the host from infectious agents that exist in the environment and from other noxious insults. The immune system has two functional divisions: the innate and the acquired. Both components involve various factors such as cytokines. A number of methodologies exist to assess aspects of immune function. There are large inter-individual variations in many immune functions even among the healthy. Genetics, age, gender, smoking habits, habitual levels of exercise, alcohol consumption, diet, stage in the female menstrual cycle, stress, history of infections and vaccinations, and early life experiences are likely to be important contributors to the observed variation. While it is clear that individuals with immune responses significantly below 'normal' are more susceptible to infectious agents and exhibit increased infectious morbidity and mortality, it is not clear how the variation in immune function among healthy individuals relates to variation in susceptibility to infection. Oriental medicine is an important factor contributing to immune competence. The author investigated the immune enhancement effects of Bojungikkitanggami-bang (BITB). The forced swimming test (FST) has been used as a screening model for new immune enhancement agents. In the present study, the author investigated the effects of BITB on FST and blood biochemical parameters related to fatigue, glucose (Glc); blood urea nitrogen (BUN); lactate dehydrogenase (LDH); creatinine; and total protein (TP). The author found that BITB (1 g/kg) significantly reduced the immobility time in the FST compared to the control. In addition, the contents of Glc, LDH, BUN, TP in the blood serum were increased in BITB (1g/kg)-fed group. Also, the author investigated the effects of BITB on the production of cytokines in human T-cell line, MOLT-4 cells. BITB (1 mg/ml) significantly increased the interferon (IFN)-vproduction compared with media control (about 2.2-fold for IFN-γ) at 24 h. However, BITB has not affect the production of IL-2 and IL-4. In addition, BITB increased the protein expression level of IFN-γ in MOLT-4 cells. Thus, BITB may have therapeutic value in generating or enhancing immune function in a clinical setting.

전통적인 한방 처방 경옥고의 면역 증강 효과 (Immune-enhancing effects of a traditional herbal prescription, Kyung-Ok-Ko)

  • Objectives : A traditional herbal prescription, Kyung-Ok-Ko (KOK), has long been used in oriental medicine as an invigorant for age-related diseases, such as amnesia and stroke. However, the beneficial value of KOK for immune responses is largely unknown. Based on the above mentioned effects of KOK, other previous reports, and its use in traditional medicine, we hypothesized that KOK displays beneficial effects against methotrexate (MTX)-induced immune suppression. Methods : We investigated the effects of KOK (0.6 g/kg/day, oral (p.o.)) on deteriorated immunity caused by MTX (2 mg/kg/day, p.o.) in an immune suppression mouse model. MTX was fed to mice once a day for 7 days. After the immune responses of the mice deteriorated by MTX treatment, KOK in water was fed to the mice once a day for 14 days. We then measured the expression levels of various cytokines, such as T helper cell (Th1, Th2) cytokines, and the number of immune cells, such as spleen T cells, B cells, and macrophages. Results : The data showed that MTX decreased Th1 profiles (interferon $(IFN)-{\gamma}$, interleukin (IL)-2, IL-12) and the number of immune cells, and increased Th2 profiles (IL-4, IL-5, IL-13), which were normalized significantly by post-administration of KOK. However, there was no significant difference in body-weight gain between MTX- and KOK-treated mice. Conclusion : These results indicate that KOK has immune-enhancing functions and reduces immunotoxicity of MTX, suggesting that supplementation with KOK will improve immune responses clinically and be useful for the prevention of immune-related diseases.
