등 마사지가 동종 조혈모세포이식 환자의 면역반응, 신체증상 및 정서상태에 미치는 효과 (Effects of Back Massage on Immune Response, Symptom Distress and Mood State of Patients Undergoing Allogeneic Hematopoietic Stem Cell Transplantation)

  • Purpose: To examine the effect of back massage on immune response, symptom distress, and mood state of patients undergoing allogeneic hematopoietic stem cell transplantation (allogeneic HSCT). Methods: Subjects were thirty-seven patients undergoing sibling allogeneic HSCT (including 16 in the experimental group and 21 in the control group). Experimental subjects participated in an intervention group of back massage for 10 minutes, once a day and 5 times a week, from one week prior to the HSCT to the third week after the HSCT or a control group. A non-equivalent pretest-posttest design was used. t-test and Repeated measures ANOVA were used to examine group differences by using SAS. Results: No significant group differences were found in Immune response (CD4+, CD8+,CD19+, CD56+) and symptom distress. The experimental group had significantly less mood state (anxiety, confusion) than the control group. Conclusion: The back massage for the patients undergoing allogeneic HSCT may be effective in altering the anxiety and confusion during hematopoietic stem cell transplantation. However, this study did not provide evidence in improving immune response and symptom distress.

아연 섭취 수준이 고지방 식이로 유도된 비만쥐의 면역 기능에 미치는 영향 (Effects of Zn Intake on Immune Responses in High Fat Diet-induced Obese Rats)

  • The purpose of this study was to investigate the effect of zinc intake on immune responses in high fat diet induced obese rats. The immune status was assessed by the measurements of immunoglobulins (IgG, A, and M) production by SRBC(sheep red blood cell) with mitogen in vivo. The delayed type hypersensitivity(DTH) response was also measured as an index of cell mediated immunity. The re sults are summarized as follows : 1) There were no significant differences in the feed intake, weight gain and feed efficiency ratio of obese rats by the different dietary zinc levels. 2) White blood cell (WBC) counts were significantly affected as the dietary zinc levels decreased. The capacity of Ig M production in obese rats was significantly higher in normal zinc group than that of low and high zinc group. Cell mediated immune response evaluated by means of DTH testing has also been found to be highly impaired by zinc deficiency and overload. From these observations, it was suggested that adequate levels of zinc may promote the immune function of obese individuals. The relationship and its functional role of the zinc in obesity remains to be further studied.

Ex vivo Boosted Immune Cell Therapy for Canine Hepatic Disease

  • A 12-year-old male American Cocker Spaniel was diagnosed with a type of chronic hepatits (CH) called cholangioheaptits. Routine supportive medication was administered to the patient, and ex vivo boosted immune cell (EBI-C) therapy was used for the treatment. A histopathologic examination of the liver 19 months later revealed that the cholangiohepatitis had progressed to cholangiocarcinoma. The medication and immune cell therapy was maintained. Two months after the new diagnosis, the patient's state worsened, and the dog died 635 days after the first visit. EBI-C therapy is a type of immunotherapy, where immune cells are isolated from the patient's peripheral blood mononuclear cells, expanded ex vivo, and then infused into the patient intravenously every two weeks. EBI-Cs (mean: 2.78 × 108 cells) were obtained 38 times and infused every two weeks. Most EBI-C were T-lymphocytes (99.24% of total EBI cells). T-lymphocytes produce large interferon (IFN)-γ, and IFN-γ inhibits liver fibrosis in dogs with CH. Moreover, in bile duct cancer, an increase in T-lymphocytes correlates with decreasing tumor invasion and metastasis. Thus, we propose that EBI-C therapy is applicable as a new supportive therapy for canine liver disease if other treatments like drug medication, surgery, or radiation are unavailable.

Exosomal Communication Between the Tumor Microenvironment and Innate Immunity and Its Therapeutic Application

  • Exosomes, which are well-known nanoscale extracellular vesicles, are multifunctional biomaterials derived from endosomes and perform various functions. The exosome is a critical material in cell-cell communication. In addition, it regulates the pathophysiological conditions of the tumor microenvironment in particular. In the tumor microenvironment, exosomes play a controversial role in supporting or killing cancer by conveying biomaterials derived from parent cells. Innate immunity is a crucial component of the host defense mechanism, as it prevents foreign substances, such as viruses and other microbes and tumorigenesis from invading the body. Early in the tumorigenesis process, the innate immunity explicitly recognizes the tumor via Ags and educates the adaptive immunity to eliminate it. Recent studies have revealed that exosomes regulate immunity in the tumor microenvironment. Tumor-derived exosomes regulate immunity against tumor progression and metastasis. Furthermore, tumor-derived exosomes regulate polarization, differentiation, proliferation, and activation of innate immune cells. Exosomes produced from innate immune cells can inhibit or support tumor progression and metastasis via immune cell activation and direct cancer inhibition. In this study, we investigated current knowledge regarding the communication between tumor-derived exosomes and innate immune cell-derived exosomes (from macrophages, dendritic cells, NK cells, and neutrophils) in the tumor microenvironment. In addition, we discussed the potential development of exosomal immunotherapy using native or engineered exosomes against cancer.

면역세포의 cytokine 유리에 미치는 substance P의 영향 (Effects of Substance P on the Release of Cytokines from Immune Cell Lines)

  • The neuropeptide substance P(SP) has been implicated in the mediation of inflammation and immune-mediated disease such as arthritis. Recently, it was reported that SP was markedly increased around the blood vessels in inflamed gingiva as well as in close association with the inflammatory cell infiltrate. These results support that SP may contribute to the pathophysiology of neuronal inflammation in human periodontal tissues. SP may regulate inflammatory/immune responses by stimulating the proliferation of human T cells, differentiation and antibody-secreting potential of B cells, macrophage respiratory burst, connective tissue proliferation, and the secretion of cytokines from monocytes and T cells. Here, I studied potential role of SP as a costimulatory chemical signal in inflammatory/immune responses, by determining the released proinflammatory cytokines such as $MIP-1{\alpha}$, $IL-1{\beta}$, and IL-6 from culture supernatants of homogeneous immune cell lines. Serum free cell supernatants were concentrated with TCA precipitation, fractionated with SDS-PAGE, and subjected into western blot analysis. Among 15 cell lines tested, macrophage/monocyte cell line RAW264.7 and WRl9m.1 showed the highest level of induction of $MIP-1{\alpha}$ when stimulated with LPS. Discrete IL-6 bands with multiple forms of molecular mass were detected from supernatants of B cell lines A20(32kDa), Daudi(32, 35kDa), and SKW6.4(29kDa), which were expressed constitutively. $IL-1{\beta}$ could not be detected by the method of western blot analysis from supernatants of all cell lines tested except RAW264.7, WRl9m.1, and erythroid cell line K562 which showed the least amount of $IL-{\beta}$ secretion. SP $10^{-9}M$ with suboptimal dose of LPS treatment showed synergistic induction of $MIP-1{\alpha}$ release from RAW264.7 or WR19m.1, and also IL-6 release from A20, but this synergism is not the case in costimulation of RAW264.7 or WRl9m.1 with SP $10^{-9}M$ and TPA. Although treatment of T cell line CTLL-R8 with SP $10^{-7}M$ or PHA+TPA induced modest level of $MIP-1{\alpha}$ secretion, synergism was not observed when they are applied together. These findings all together suggest the possibility of a regulatory role of SP in inflammatory/immune reaction through differential modulation of bioactivities of other chemical cosignals.

마우스에 있어서 Cimetidine, Ranitidine 및 Famotidine이 면역반응에 미치는 영향 (The Effect of Cimetidine, Ranitidine and Famotidine on the Immune Response in ICR Mice)

  • Experiments were performed on mice to investigate the influences of cimetidine, ranitidine and famotidine on the immune response. Immune response were evaluated by antibody, Arthus reaction (Arthus), delayed type hypersensitivity (DTH), rosette forming cell (RFC), phagocyte activity and whit( blood cell (WBC) in mice, sensitized and challenged with sheep red blood cells (SRBC). The weight of liver, spleen and thymus were measured. Following results obtained in this experiment. 1) The administration of cimetidine as compared to normal group significantly decreased Arthus, Hemagglutinin titer (HA), RFC, DTH, WBC and phagocyte activity, but increased the activity of serum albumin. 2) The administration of ranitidine as compared to normal group decreased RFC and HA. 3) The administration of Famotidine as compared to normal group decreased DTH and RFC, and significantly decreased HA, Arthus and serum protein. 4) The administration of ranitidine and famotidine decreased more humoral immune response than cellular immune response, but the administration of cimetidine significantly decreased humoral and cellular immune response, WBC and phagocyte activity.

Roles of Endoplasmic Reticulum Stress in Immune Responses

  • The endoplasmic reticulum (ER) is a critical organelle for protein synthesis, folding and modification, and lipid synthesis and calcium storage. Dysregulation of ER functions leads to the accumulation of misfolded- or unfolded-protein in the ER lumen, and this triggers the unfolded protein response (UPR), which restores ER homeostasis. The UPR is characterized by three distinct downstream signaling pathways that promote cell survival or apoptosis depending on the stressor, the intensity and duration of ER stress, and the cell type. Mammalian cells express the UPR transducers IRE1, PERK, and ATF6, which control transcriptional and translational responses to ER stress. Direct links between ER stress and immune responses are also evident, but the mechanisms by which UPR signaling cascades are coordinated with immunity remain unclear. This review discusses recent investigations of the roles of ER stress in immune responses that lead to differentiation, maturation, and cytokine expression in immune cells. Further understanding of how ER stress contributes to the pathogenesis of immune disorders will facilitate the development of novel therapies that target UPR pathways.

Dendritic Cell-based Immunotherapy for Rheumatoid Arthritis: from Bench to Bedside

  • Dendritic cells (DCs) are professional antigen presenting cells, and play an important role in the induction of antigen-specific adaptive immunity. However, some DC populations are involved in immune regulation and immune tolerance. These DC populations are believed to take part in the control of immune exaggeration and immune disorder, and maintain immune homeostasis in the body. Tolerogenic DCs (tolDCs) can be generated in vitro by genetic or pharmacological modification or by controlling the maturation stages of cytokine-derived DCs. These tolDCs have been investigated for the treatment of rheumatoid arthritis (RA) in experimental animal models. In the last decade, several in vitro and in vivo approaches have been translated into clinical trials. As of 2015, three tolDC trials for RA are on the list of ClinicalTrial.gov (www.clinicaltrials.gov). Other trials for RA are in progress and will be listed soon. In this review, we discuss the evolution of tolDC-based immunotherapy for RA and its limitations and future prospects.

  • Backgroud: Immunotherapy using dendritic cells (DC) loaded with tumor antigens may represent a potentially effective method for inducing antitumor immunity. We evaluated the effectiveness of DC-based antitumor immune response in various conditions. Methods: DC were cultured from peripheral blood mononuclear cells (PBMNC) in myelogenous leukemia (ML) and lysates of autologous leukemic cells are used as tumor antigen. The effectiveness of interleukin-12 (IL-12) and CD40L (CD154) on the antigen presenting function of lysates-loaded DC was analyzed by proliferation, cytokine production, and cytotoxicity tests with activated PBMNC (mainly lymphocytes). For generating antigen-loaded DC, direct fusion of DC with ML was studied. Results: Antigen loaded DC induced significantly effective antitumor immune response against autologous leukemic cells. Administration of IL-12 on the DC based antitumor immune response showed higher proliferation activity, IFN-$\gamma$ production, and cytotoxic activity of PBMNC. Also, fused cell has a potent antitumor immune response. Conclusion: We conclude that lysates-loaded DC with IL-12 may be effectively utilized as inducer of antitumor immune reaction in ML and in vivo application with DC-based antitumor immunotherapy or tumor vaccination seems to be feasible.

생쥐에서 전기자극 스트레스에 의한 행동반응과 면역 기능 변화 (Behavioral Response and Immune Alterations by Electric Footshock in Mice)

  • The present experiment was designed to investigate the effects of behavioral, response to immune function in response to electric footshock in mice. Mice were subjected to electric footshock for 3 days(two sessions a day, 11 times of shock for about 31 minutes a session). The humoral immune response was measured using mice immunized with rat RBC. The cell-mediated immune responses were evaluated by contact hypersensitivity to 2, 4-dinitrofluorobenzene(DNFB) and by phytohemagglutin(PHA)-stimulated splenocytes proliferation assay. In stressed group, electric footshock suppressed significantly anti-rat RBC antibody production(p<0.05), but enhanced significantly $T_{48}$ relative to $T_{24}$ in contact hypersenstivry (P<.01) and T-cell proliferation response(P<.05) by PHA stimulation elative to control group. T-cell proliferation response by PHA stimulation was significantly correlated to the movement than the sensitivity and coping behavior in the mouse, in response to the electric footshock. These data supper the importance of behavioral response in stress-induced changes of immune functions.
