효모를 Na-alginate에 고정화한 후 연속반응기를 이용한 glucose 발효로 에탄올을 생산하였다. 그 결과 고정화 효모의 활성화 시간은 20~25시간이었다. 연속발효에서 고정화효모의 온도안정성은 30~$37^{\circ}C$였으며 pH 안정성은 pH 4.0~pH 8.0, 최적 희석속도는 $0.2h^[-1}$ 이었고 에탄올생산 최적 당농도는 15%였다. 최적조건에서 에탄올수율은 0.23, 생산된 에탄올 농도는 33.90g/l 그리고 에탄올 생산성은 7.12g/$l\cdot h$로 각각 나타났다.
The lipase from Mucor racemosus NRRL 3631 was partially purified by fractional precipitation using 60% ammonium sulfate, which resulted in a 8.33-fold purification. The partially purified lipase was then immobilized using different immobilization techniques: physical adsorption, ionic binding, and entrapment. Entrapment in a 4% agar proved to be the most suitable technique (82% yield), as the immobilized lipase was more stable at acidic and alkaline pHs than the free enzyme, plus 100% of the original activity was retained owing to the thermal stability of the immobilized enzyme after heat treatment for 60 min at $45^{\circ}C$. The calculated half-lives (472.5, 433.12, and 268.5 min at 50, 55, and $60^{\circ}C$, respectively) and the activation energy (9.85 kcal/mol) for the immobilized enzyme were higher than those for the free enzyme. Under the selected conditions, the immobilized enzyme had a higher $K_m$ (11.11 mM) and lower $V_{max}$ (105.26 U/mg protein) when compared with the free enzyme (8.33 mM and 125.0 U/mg protein, respectively). The operational stability of the biocatalyst was tested for both the hydrolysis of triglycerides and esterification of fatty acids with glycerol. After 4 cycles, the immobilized lipase retained approximately 50% and 80% of its original activity in the hydrolysis and esterification reactions, respectively.
Jack bean urease was immobilized on gelatin beads with the help of glutaraldehyde. The optimum immobilization (67.6%) was obtained at 30mg/ml gelatin concentration, 0.5 mg/bead enzyme protein concentration, 1 % glutaraldehyde and at $4^{\circ}C$ incubation temperature. The $t_{1/2}$ of immobilized urease was approximately 90 days at $4^{\circ}C$ compared with $t_{1/2}$ of 20 days for the soluble urease, under identical condition. The apparent optimum pH shifted from 7.3 to 8.0 when the urease was immobilized. The optimum stability temperature of immobilized urease was found to be $60^{\circ}C$ while that of soluble urease was $45^{\circ}C$. Time-dependent thermal inactivation studies showed monophasic kinetics for soluble urease and immobilized urease at $70^{\circ}C$, respectively. The immobilized urease beads stored at $4^{\circ}C$ showed practically no leaching over a period of 30 days. Here we are presenting an easy and economical way of immobilizing urease on the gelatin beads making it suitable for various applications.
Since carbohydrates are major mediators in cell-to-cell adhesion and communication, the development of specific and strong binders against them could generate promising therapeutics. As the first step towards that goal, sugar molecules have to be immobilized to be used as an affinity matrix. The amino functionality in sugar is the most active nucleophile for the immobilization, if the amino group is available. An alternative and general method is to use the hydroxyl group as a direct nucleophile, but the quantitation of immobilized hydroxyl groups is not easily done. To overcome this limitation, we have developed a method to immobilize various isomers of monosaccharides with p-nitrophenyl groups to the beads by using their hydroxyl groups. It was found that the amount of immobilized sugar was independent of the structure of the sugar, but was dependent on the number of hydroxyl groups. We also developed a sensitive method to quantify the amount of immobilized sugar at the picomolar scale by utilizing commercially available glycosidases to release a sensitive reporter molecule, p-nitrophenol, and detect it by HPLC. This new technique would allow a facile quantitation method for immobilized sugar molecules, which could be used as the affinity matrix to develop strong binders against biologically important sugars.
A thermostable trehalose synthase (TtTSase) from Thermus thermophilus HJ6 was immobilized on chitosan activated with glutaraldehyde. The yield of immobilization was evaluated as 39.68%. The optimum pH of the immobilized enzyme was similar to that of the free enzyme. However, the optimal temperature ranges were shifted by about $4^{\circ}C$ owing to better thermal stability after immobilization. The half-life of heat inactivation for free and immobilized enzymes was 5.7 and 6.3 days at $70^{\circ}C$, respectively, thus showing a lager thermostability of the immobilized enzyme. When tested in batch reaction, the immobilized enzyme retained its relative activity of 53% after 30 reuses of reaction within 12 days, and still retained 82% of its initial activity even after 150 days at $4^{\circ}C$. A packed-bed bioreactor with immobilized enzyme showed a maximum yield of 56% trehalose from 100 mM maltose in a continuous recycling system (bed volume: 10 ml) under conditions of pH 7.0 and $70^{\circ}C$.

The adhesion of activated and normal platelets to fibrinogen requires the receptor binding site of GPIIb/IIIa. These recognition sites exists in the A ${\alpha}$ chain(RGDS at 572-575 and RGDF at 95-98) and the carboxy-terminal of ${\gamma}$ chain (HHLGGAKQAGDV at 400-411) of fibrinogen. In this study, we developed RGDF-immobilized surface to detect the unctional state of platelet. RGDF-immobilized surface was prepared on the glass using photolithographic technology. Platelet adhesion to RGDF-immobilized surface was observed by staining platelets with mepacrine using a fluorescence microscope using mepacrine. Using the RGDF peptide of fragment E, we observed that the platelets pretreated with PGE1 interacted incompletely with RGDF-immobilized surface, whereas ADP activated platelets interacted with the surface extensively. These results show that the distinct selectivity of RGDF-immobilized micro-patterned surface can be used to detect the unctional state of platelets.
Polystyrene계 음이온 교환수지인 Diaion PA 412에 Aureobasidium pullulans 기원의 fructosyltransferase를 crude enzyme 상태로 고정화하여 fructo-oligosaccharides의 생산을 검토하였다. 고정화 효소의 최적 반응 pH 및 온도는 각각 pH 5.0, $55^{\circ}C$. 이었고 고정화에 의해 열안정성이 크게 증가하였다. 고정화 효소에 의한 fructo-oligosaccharids 생성의 효소 반응 경향은 free cell 및 soluble enzyme과 거의 유사하여 최종 반응산물 중의 당 조성 이 동일하였다. 고정화 효소를 repeated-batch 방법으로 운전하여 fructo-oligosaccharides 생산공정에 적용해 본 결과 $50^{\circ}C$에서 20일까지 안정성을 유지하였다.
Erwinia rhapontici를 Ca-alginate에 고정화시켜 고정화 세포의 반응특성을 고찰하고, STR, PBR을 이용하여 Palatinose의 생산을 검토하였다. Free cell과 고정화 세포의 반응최적 pH는 5.5-6.0, 반응 최적온도는$30-35^{\circ}C$로 동일하였으나 고정화에 의해 pH 및 온도 범위가 보다 넓어졌고, 이때 Free cell및 고정화 세포의 겉보기 Km갑슨 각각 0.13, 0.28M이었다. STR을 이용한 Palatonose 생산시 고정화 세포의 반감기는 약 380 시간으로 낮았으나, PBR을 통해 30일까지 안정운전이 가능하였다. PBR 운전시의 운전온도 30, $33^{\circ}C$에서 Palatinose수율 및 고정화 세포의 안정성은 거의 동일한 결과를 나타내었으며 이때 PBR생산성은 약 120g/l$\cdot$h이었고, Pilot scale인 50L 까지 성공적으로 Scale up 되었다.
Trametes sp.에서 생산되는 laccase는 CNBr-activated Sepharose-4B(CAS4B)에 고정화 되었고, 염료의 연속적인 분해를 위하여 테스트되었다. Laccase는 CAS4B에 효율적으로 고정화되었고, CAS4B에 고정화 된 laccase는 pH, 열, 단백질 구조적인 안정성 면에서 상당히 증가하였다. CAS4B에 고정화 된 accase의 최적 pH는 5, 온도는 4$0^{\circ}C$로서 free laccase와 비교하여 변화가 없었다 기질로서 Reactive Blue 19를 사용하였을 때 free laccase와 고정화 laccase의 $K_{m}$ ($\mu$mol/mL) 값은 각각 0.34와 2.0이었고,V$_{max}$($\mu$mol/mL.min) 값은 각각 0.12, 0.1이었다. Repeated-batch 반응에서 효소의 안정성과 높은 분해 효율 만족하는 조건은 pH 5, 3$0^{\circ}C$이였다. 또한 HBT에 의한 효소의 불활성은 크게 나타나지 않았다. Packed-bed reactor에서 최적으로 운전되었을 때 100 $\mu$M Reactive Blue 19과 0.1 mM HBT가 존재하는 50 $\mu$M Acid Red 57의 연속적인 분해에서 30시간 후에도 분해 효율이 70%로 유지되었다. 고정화 laccase는 Packed-bed reactor에서 azo 염료의 연속적인 분해를 매우 안정적으로 수행하였다.다.
키토산 올리고당을 효율적으로 생산하기 위하여 고정화 효소 를 이용한 키토산의 효소적 가수분해를 시도하였다. Chitosanase는 Chitopearl계 고정화 담체에 대해서 높은 흡착율로 결합되었다. 키틴에 고정화된 효소는 비록 흡착율은 낮았지 만 그 활성은 가장 높게 나타났다. 키틴 고정화 효소는 유리 효소에 비해 약 90% 이상의 활성을 유지하였다. 고정화 효소의 최적 온도는 60°C로서 유리 효소보다 $15^{\circ}C$ 더 높았으며, 열에 대한 안정성도 유리 효소보다 넓은 온도범위에서 우수하였다 그러나 고정화 효소는 pH에 대해서는 어떠한 뚜렷한 효과도 보이지 않았다. 고정화 효소의 저장 안정성은 유리 효소보다 더 높은 저장온도인 60t에서도 더 안정한 것으로 나타났다. 키틴 고정화 효소에 의한 키토산의 가수분해반응은 반응 3시간까지 급격한 증가를 보이다 그 이후의 반응시간 경과에서도 더 이상 증가를 보이지 않았다. 고정화 효소에 의해 생성된 올리고당의 조성은 효소의 반응시간에 따라 크게 의존하였으며, 2시간의 반 응에서 비교적 고차 올라고당인 COS-4-6의 함량은 약 90% 이상이었다 두 효소에 대한 반용속도상수에서, 고정화 효소는 유리 효소에 비해 낮은 기질친화성과 낮은 반웅속도를 보였지 만, 높은 기질농도에서도 전혀 기질저해반응은 일어나지 않았다. 따라서 키틴 고정화 효소는 유리 효소에 비해 활성의 감소없이 효율적으로 키토산을 가수분해할 수 있었으며, 고차 올리고당의 생성 량도 매우 높았다.
