Experimental Studies on the Inhibitory Effect of Immediate-Type Allergic Reaction of Tongku-tang (통규탕의 즉각형 알레르기 반응 억제 효과에 관한 실험적 연구)

  • This report describes an inhibitory effect of Tongku-tang(TKT) on mast cell-mediated immediate-type allergic reactions. TKT is an Oriental herbal prescription, which has been successfully applied for the treatment of allergic disorders, mainly skin anaphylactic diseases in eastern medicine. TKT has concentration-dependently inhibited the ear swelling response induced by intradermal injection of non-specific mast cell degranulator compound 48/80 in mice. TKT also inhibited mast cell-dependent passive cutaneous anaphylaxis activated by dinitrophenyl (DNP)-IgE antibody in rats. I studied the effect of TKT on the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. TKT did not inhibit significantly the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. However, TKT inhibited both TNF-α and IL-1β secretion induced by phorbol 12-myristate 13-acetate and A23187 respectively. These results provide evidence that TKT may be beneficial in the treatment of immediate-type allergic reaction.

Inhibitory Effect of Immediate-Type Allergic Reaction of Poncirus trifoliata L. by Anal Therapy (肛腸療法에 의한 枳實의 卽刻型 알레르기 反應 抑制 效果)

  • Anal therapy is another way of taking medicine It is a traditional pathway but not available in common situation. Nevertheless. It has many benefit and usefulness, it has not treated so much. The dried immature fruit of Poncirus trifoliata L(Ji-Sil) is widely used to treat urticaria, itch and indigestion in traditional Korean Medicine. So this study was carried out to examine the effect of an aqueous extract from immature fruit of Poncirus trifoliate L. on immediate-type allergic reaction by anal administration. Anal administration of Poncirus trifoliata L. showed a marked inhibition rate in systemic immediate-type allergic reaction with a dose of 0.1 - 1 g/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Poncirus tnjoliata L. (0.1 -1 g/kg) significant1y reduced plasma histamine contents induced by compound 48/80. Anal administration of Poncirus trifoliata L. (10 g/kg) also inhibited to $44.6\%$ (P<0.01) local allergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. These results provide evidence that anal therapy of Poncirus trifoliata L. may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.

Inhibitory Effects of Ulmus parvifolia and Liriope platyphylla Wang et Tang on Histamine Release from Rat Peritoneal Mast Cells

  • Twenty-four different natural food materials extracted with 80% methanol were used to investigate the inhibition of cell-mediated immediate type allergic reactions induced by compound 48/80 in rat peritoneal mast cells (RPMCs). Nine 80% methanol extracts screened at a concentration of $10\;{\mu}g/mL$ inhibited histamine release from RPMCs induced by compound 48/80. Of these, two materials (Ulmus parvifolia and Liriope platyphylla Wang et Tang) were extracted and fractionated into four different solvent types (chloroform, ethylacetate, butanol, and water), and the fractions with major anti-allergic effects were assessed. The chloroform fraction of U. parvifolia (UP) at $5\;{\mu}g/mL$ and the ethylacetate fraction of L. platyphylla Wang et Tang (LPWT) at $1\;{\mu}g/mL$ showed the greatest inhibition of histamine release induced by compound 48/80. The chloroform fraction of UP and the ethylacetate fraction of LPWT in combination showed a greater inhibition of histamine release than either fraction alone. The cAMP levels in RPMCs treated with UP and LPWT were significantly greater than in cells treated with compound 48/80 alone. Our studies suggest that extracts from UP and LPWT may alleviate immediate type hypersensitivity reactions through the increase of cAMP levels in the mast cells.

  • Ro09-0198, a cyclic peptide isolated from culture filtrates of Streptoverticillium griseove-rticillatum, induced platelet aggregation and serotonin release simultaneously. LDH release was not observed. Addition of peptide to rat platelet, loaded with $Ca^{2+}$ chelator quin-2, caused immediate rise in cytosolic free $Ca^{2+}$. Liposomal membrane containing phosphatidylethanolamine was damaged by peptide and released $^{45}Ca$ dose dependently.

Immediate Effect of Pressure Pain Threshold and Flexibility in Tensor Fascia Latae and Iliotibial Band According to Various Foam Roller Exercise Methods

  • Background: The treatment of pain in the iliotibial band friction syndrome has been difficult to determine, according to studies to date. However, recent studies have suggested that flexibility in the subacute stage of pain in the iliotibial band friction syndrome may help reduce pain. Objective: To investigate the immediate effect on pressure pain threshold and flexibility of the tensor fascia latae and iliotibial band by applying static and dynamic myofascial release foam rolling and self-stretching to adults with shortening iliotibial band. Design: Randomized controlled trial Methods: In this study, 50 subjects who were selected in advance as a randomized controlled trial were randomly allocated using a R Studio program. The included subjects were randomly allocated to three intervention groups. The static self-myofascial release 18 people, dynamic self-myofascial release group 16 people separated the self-stretching group 16 people and conducted a homogeneity check in advance. Before the start of the experiment, after of the experiment, 5 minutes after the end of the experiment, the pressure pain threshold and flexibility change for each part were measured. Results: The results of this study showed that the static self-myofascial release showed a significant difference in the pressure pain threshold in the tensor fascia latae and middle, lower part of the iliotibial band, compared with the other intervention groups (p<.05). In change of flexibility, the static self-myofascial release was significantly different than the other intervention groups (p<.05). Conclusion: The result of this study suggest that static self-myofascial release using foam roller may help to improve the pain and flexibility of the iliotibial band and to apply it as a more discerning intervention.

Inhibitory Effect of Immediate-Type Hypersensitivity of Syzygium aromaticum extract by Anal Therapy (肛腸療法에 의한 丁香의 卽刻型 過敏反應 抑制效果)

  • Cloves are the dried flower buds of Syzygium aromaticum (L.) Mere et Perry (Myrtaceae). They have been successfully used for the management of various allergic disorders by oral administration in Korea. In this study, the author investigated the effect of an aqueous extract of Syzygium aromaticum on immediate-type hypersensitivity by anal administration. Anal administration of Syzygium aromaticum showed a marked inhibition rate in systemic hypersensitivity with a dose of 1 mg/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Syzygium aromaticum significantly reduced plasma histamine contents induced by compound 48/80. Anal administration of Syzygium aromaticum (1 mg/kg) also inhibited to $61.4\%$ (P<0.01) local a1lergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. When Syzygium aromaticum was added, the level of cAMP in peritoneal mast cells transiently and significantly increased about 47-fold at 10 second compared with that of basal cells. These results provide evidence that anal therapy of Syzygium aromaticum may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.

Magnoliae Cortex inhibits immediate-type allergic reactions

  • The effect of aqueous extract of Magnoliae Cortex (Magnoliaceae) (MCAE) on the immediate-type allergic reaction was investigated. MCAE inhibited compound 48/80 induced systemic anaphylactic reaction in rats. MCAE (0.1 and 1 g/kg) also significantly inhibited local immunoglobulin E (lgE)-mediated passive cutaneous anaphylactic (PCA) reaction. MCAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) 1gE. Moreover, MCAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-mediated tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ production. These results indicate that MCAE inhibits immediate-type allergic reaction in vivo and in vitro.

Cichorium Intybus inhibits mast cell-mediated immediate-type allergic reactions

  • We investigated the effect of aqueous extract of Cichorium intybus (CIAE) on mast cell-mediated immediate type allergic reactions. CIAE dose-dependently inhibited systemic anaphylactic reaction induced by compound 48/80 in mice. Especially, CIAE inhibited compound 48/80-induced anaphylactic reaction 100% with the dose of 1000 mg/kg. CIAE 1000 mg/kg also significantly inhibited local anaphylactic reaction activated by anti-dinitrophenyl (DNP) IgE. When mice were pretreated with CIAE at a concentration ranging from 0.1 to 1000 mg/kg, the plasma histamine levels were reduced in a dose-dependent manner. CIAE (1 to 1000 g/ml) dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. These results indicate that CIAE inhibits mast cell-mediated immediate-type allergic reactions.

  • Object : We investigated the effect of Gamiokyaek-tang (GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis and acetic acid-induced vascular permeability in rodents. Also perfonned MTT assay and b-hexosaminidase activity is measured in RBL-2H3. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5 mg/ml didn't have an effect on cell viability and cytotoxicity. b-hexosaminidase release in RBL-2H3 was significantly reduced by 2 and 5 mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity .

  • We investigated the effect of aqueous extract of Terminalia chebula (Combretaceae)(TCAE) on the immediate hypersensitivity reaction in vivo and in vivo. TCAE (0.01 to 1 g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in mice. When TCAE was pretreated at concentrations ranging from 0.01 to 1 g/kg, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) significantly inhibited local immunoglobulin E (IgE)-mediated passive cutaneous anaphylactic reaction. TCAE (0.001 to 1 mg/ml) also dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) IgE. TCAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis $factor-{\alpha}$ production from RPMC. These results indicate that TCAE inhibits immediate hypersensitivity reaction in vivo and in vitro.
