RBL-2H3 비만세포와 OVA/alum 감작 생쥐에서 청심보혈탕(淸心補血湯)의 항알레르기 효과 (Anti-allergic Effects of Chungshimbohyeoltang in RBL-2H3 Mast Cells and OVA/alum Sensitized Mice)

  • Objectives The suppressive effect of CSBHT has been mysterious. Thus, the present study is designed to investigate the suppressive effect and its mechanism. Methods To investigate the anti-allergy effect from ChungShimBoHyeolTang(CSBHT), RBL-2H3 cell was used and examined by Real-Time PCR, and IL-4 and IL-13 from RBL-2H3 was examined by ELIS. In addition, GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos, c-Jun, NF-${\kappa}B$ p65 transcription factors of RBL-2H3 mast cell were examined by Western Blotting. Also, OVA/alum-sensitized mice were orally administrated CSBHT and serum OVA-specific IgE production, IL-4, and IL-13 production in splenocytes supernatant were examined. Results As a result of treating with CSBHT extract, RBL-2H3 mast cells significantly suppressed the IL-4 and IL-13 mRNA expression and IL-4 and IL-13 production. Western blot analysis of transcription factors involving IL-4 and IL-13 expression also revealed a prominent decreases of mast cell's specific transcription factors including GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos, and NF-${\kappa}B$ p65. Also, examining the mice, administration of CSBHT suppressed the amount of OVA-specific IgE in OVA/alum-sensitized mice and IL-4 and IL-13 production in splenocytes supernatant. Conclusions The study suggested that the anti-allergic activities of CSBHT suppresses IL-4 and IL-13 production from the Th2 cytokines by suppressing transcription factors as GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos and NF-${\kappa}B$ p65 in mast cells.

청상보하탕의 기관지 천식 환자에 대한 임상적 효과 (Clinical Effects of Chuongsangboha-tang in Asthmatic Patients)

  • Background: Nowadays asthma is considered to be an inflammatory disease characterized by airway hyperresponsiveness and pulmonary eosinophilia. Production of cytokines by bronchial epithelial cells may contribution to the local accumulation of inflammatory cells in patients with bronchial asthma. In many recent studies molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of asthma. Objectives: We aimed to identify the clinical effects of Chuongsangboha-tang and the effects of Chuongsangboha-tang on serum cytokines in asthmatic patients. Materials and Methods: The subjects consisted of 36 patients with asthma who had been treated with Chuongsangboha-tang for four weeks. Chuongsangboha-tang is an herbal decoction which has been used of the traditional therapeutic agent of asthma. PFT, Quality of Life Questionnaire for Adult Korean Asthmatics (QLQAKA), blood eosinophil, serum IgE, Serum IL-4, IL-5, IFN- were checked before and 4 weeks after treatments. Results: Treatment with Chuongsangboha-tang for four weeks resulted in significant increase in FEV1.0%, PEFR%, QLQAKA. The blood eosinophil, serum IgE, IL-4 and IL-5 in asthmatic patients increased significantly compared to the normal control group, while the serum IFN-decreased significantly. Conclusions: This study shows that Chuongsangboha-tang has effects on improvement of pulmonary function and quality of life in asthmatic patients. Obviously, further research concerning this is still necessary.

Inhibitory effect of Taemyeongcheong on allergic reactions

  • Taemyeongcheong (TMC) is Korean traditional extracted drink with various health ingredients. TMC has been used to treat hepatic damage, obesity, gastritis, and colitis. However, the role of TMC on allergic reaction has not been studied yet. In this study, we investigated the anti-allergic effects of TMC against a compound 48/80-induced systemic anaphylactic reaction and IgE-mediated passive cutaneous anaphylaxis (PCA). TMC significantly inhibited the compound 48/80-induced systemic anaphylactic reaction and IgE-mediated PCA reaction. Furthermore, TMC reduced the levels of interleukin (IL)-1β, IL-4, IL-13, and vascular endothelial growth factor in the serum of mice under PCA reaction. Taken together, these results suggest that TMC can play a useful role as an anti-allergic agent.

  • Objectives : Traditional medicines isolated from natural products often have positive effects in the prevention and healing of various immune disorders, such as allergy and atopic inflammation. Scrophulariae Radix (SR) been used in oriental medicine used for treatment of acute and chronic inflammatory diseases. Mast cells are known to play important roles in the initiation of allergic reactions. In this study, we investigated the effects of SR ethanol extract on inflammatory responses in IgE-stimulated RBL-2H3 mast cells. Methods : Rat basophilic leukemia RBL-2H3 cells were purchased from Korean Cell Line Bank (KCLB No. 22256). Cell viability was measured by MTT assay. Assays for ${\beta}-Hexosaminidase$ Secretion : RBL-2H3 cells were sensitized with dinitrophenyl-ImmunoglobulinE (DNP IgE). The next antigen DNP-BSA ($25ng/m{\ell}$) was added for 10 minutes and the reaction was terminated after 5 minutes in the ice bath. To determine ${\beta}-Hexosaminidase$ release, supernatants were aliquoted into 96-well plates. Samples were mixed with substrate solution and incubated for 1 h at $37^{\circ}C$. Absorbance was measured with a spectrophotometer at 405 nm. IL-4 and tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) concentrations in cell culture supernatants were measured using enzyme-linked immunosorbent assay (ELISA) kits. Results : The cytotoxicity of SRE in RBL-2H3 cells was less than 5%. SRE inhibited DNP-IgE-imduced degranulation of mast cells in RBL-2H3 cells. Also significantly decreased the levels of inflammatory cytokine, IL-4 and TNF-alpha. In this study, the SRE showed potential anti-allergic and antiinflammatory. Conclusions : These results indicate that SRE could be inhibit the allergic response through suppressing the mast cell activation.

DNCB로 아토피가 유도된 NC/Nga생쥐에서 천연 탄산 온천수의 온도별 항아토피 효과 (The anti-atopic effect of natural carbonated hot spring water on DNCB-induced NC/Nga mice under different temperature)

  • Objectives : The aim of this study is to investigate the effect of natural carbonated hot spring water (NCHW) on DNCB-induced NC/Nga mice, an atopic dermatitis (AD) model. Methods :At first, NC/Nga mice were prepared and induced to have atopic lesion on their back skin by dinitrochlorobenzene (DNCB). They swam for 20 minutes everyday in tanks where the one is filled with $28^{\circ}C$ NCHW, the other is with $25^{\circ}C$ carbonated hot spring water and another is with artificial carbonated hot spring water (ACHW). After 3 weeks, We assessed the skin clinical score and macroscopic appearance, total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and histological changes. Results : There are meaningful results of improving atopic lesion-state by relieving the count of total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and recovering skin clinical score in the group with $28^{\circ}C$ NCHW in comparison with the other groups. Conclusions : The NCHW may have potential as an effective treatment for atopic dermatitis.

봉독 면역요법후의 면역학적 변화에 대한 고찰 -알레르기 질환에 응용 가능성을 중심으로- (Immunological Changes on Allergic Response after Beevenom Immunotherapy)

  • Beevenom immunotherapy(BVIT) in allergic patients is a well-established treatment modality for the prevention of systemic anaphylactic reactions caused by insect stings. BVIT is accompanied by increases in allergen-specific IgG, particularly the IgG4 isotype, which blocks not only IgE-dependent histamine release from basophils but also IgE-mediated antigen presentation to T cells. Inhibition of T cells after BVIT also involves decreased induction of the costimulatory molecule ICOS, which, in turn, seems to be dependent on the presence of IL-10, also associated with the inhibited status of T cells after BVIT. Suppression of T cells by IL-10 is an active process, which depends on the expression and participation of CD28. Immune tolerance in specific allergen immunotherapy might be a consequence of decreased Th2 or increased Th1 response of allergen specific T lymphocytes. BVIT shifted cytokine responses to allergen from a TH-2 to a TH-1 dominant pattern, suggesting direct effects on T cells. Many studies showed that severe side effects due to venom immunotherapy are rare. These results suggest that immunological changes after BVIT may be applied to be therapeutic alternative of general allergic diseases including beevenom allergy.

FFA2 Activation Ameliorates 2,4-Dinitrochlorobenzene-Induced Atopic Dermatitis in Mice

  • Gut microbiota produce dietary metabolites such as short-chain fatty acids, which exhibit anti-inflammatory effects. Free fatty acid receptor 2 (FFA2, formerly known as GPR43) is a specific receptor for short-chain fatty acids, such as acetate that regulates inflammatory responses. However, the therapeutic potential of FFA2 agonists for treatment of atopic dermatitis has not been investigated. We investigated the efficacy of the FFA2 agonist, 4-chloro-α-(1-methylethyl)-N-2-thiazoylylbenzeneacetanilide (4-CMTB), for treatment of atopic dermatitis induced by 2,4-dinitrochlorobenzene (DNCB). Long-term application of DNCB to the ears of mice resulted in significantly increased IgE in the serum, and induced atopic dermatitis-like skin lesions, characterized by mast cell accumulation and skin tissue hypertrophy. Treatment with 4-CMTB (10 mg/kg, i.p.) significantly suppressed DNCB-induced changes in IgE levels, ear skin hypertrophy, and mast cell accumulation. Treatment with 4-CMTB reduced DNCB-induced increases in Th2 cytokine (IL-4 and IL-13) levels in the ears, but did not alter Th1 or Th17 cytokine (IFN-γ and IL-17) levels. Furthermore, 4-CMTB blocked DNCB-induced lymph node enlargement. In conclusion, activation of FFA2 ameliorated DNCB-induced atopic dermatitis, which suggested that FFA2 is a therapeutic target for atopic dermatitis.

마우스 만성천식모델에서 경구면역관용 유도에 의한 알레르기 염증의 예방효과 (The Preventive Effect of Allergic Inflammation by Induction of Oral Tolerance in a Mouse Model of Chronic Asthma)

  • 연구배경 : 경구면역관용 유도는 4주 이내의 급성천식모델에서 알레르기 염증반응을 효과적으로 억제하는 것으로 알려져 있다. 하지만 경구면역관용 유도가 4주 이상 지속되는 만성천식모델에서도 알레르기 염증반응을 효과적으로 억제할 수 있는 지에 대해서는 아직 불확실하다. 본 연구에서는 8주 동안 지속되는 만성천식모델을 이용하여 경구면역관용 유도가 알레르기 염증반응 및 기도재형성에 어떤 영향을 미치는 지를 관찰하였다. 방 법 : 5주된 암컷 BALB/c 마우스에 ovalbumin(OVA)을 복강 내로 투여하여 감작시킨 후, 6주 동안 주 2회 OVA를 반복 흡입시킴으로써 만성천식모델을 만들었다. 경구면역관용은 감작시키기 전에 OVA를 경구 투여함으로써 유도하였으며, 저용량 군(OVA 1 $mg/day{\times}6$일)과 고용량 군(OVA 25 $mg/day{\times}1$일)으로 나누었다. 기도과민성은 농도별 메타콜린 흡입에 따른 enhanced pause (penh) 값의 변화로 평가하였으며, 기관지폐포세척액 내의 염증세포수를 측정하고 IL-13, IFN-${\gamma}$ 농도 및 혈청 내 OVA 특이 IgE, IgG1, IgG2a 농도를 ELISA로 측정하였다. 기도재형성 정도는 폐조직의 PAS 및 Masson's trichrome 염색으로 관찰하였다. 결 과 : 경구면역관용군은 천식군에 비해 penh 값, 기관지폐포세척액 내의 염증세포 수, IL-13 및 IFN-${\gamma}$ 농도, 혈청 내의 OVA 특이 IgE, IgG1 및 IgG2a 농도, 폐조직 술잔세포의 과다형성, 기저막하부의 섬유화 정도가 유의하게 감소되었다 (P<0.05). 결 론 : 알레르기 기관지천식모델에서 경구면역관용 유도는 8주까지 장기적으로 기도과민성, 염증반응 및 기도재 형성을 억제하였으며, 그 기전에는 항원 특이 면역글로불린과 Th1 및 Th2 시토카인들에 대한 억제작용이 중요한 역할을 하는 것으로 추측되었다.

Effects of G-Rh2 on mast cell-mediated anaphylaxis via AKT-Nrf2/NF-κB and MAPK-Nrf2/NF-κB pathways

  • Background: The effect of ginsenoside Rh2 (G-Rh2) on mast cell-mediated anaphylaxis remains unclear. Herein, we investigated the effects of G-Rh2 on OVA-induced asthmatic mice and on mast cell-mediated anaphylaxis. Methods: Asthma model was established for evaluating airway changes and ear allergy. RPMCs and RBL-2H3 were used for in vitro experiments. Calcium uptake, histamine release and degranulation were detected. ELISA and Western blot measured cytokine and protein levels, respectively. Results: G-Rh2 inhibited OVA-induced airway remodeling, the production of TNF-α, IL-4, IL-8, IL-1β and the degranulation of mast cells of asthmatic mice. G-Rh2 inhibited the activation of Syk and Lyn in lung tissue of OVA-induced asthmatic mice. G-Rh2 inhibited serum IgE production in OVA induced asthmatic mice. Furthermore, G-Rh2 reduced the ear allergy in IgE-sensitized mice. G-Rh2 decreased the ear thickness. In vitro experiments G-Rh2 significantly reduced calcium uptake and inhibited histamine release and degranulation in RPMCs. In addition, G-Rh2 reduced the production of IL-1β, TNF-α, IL-8, and IL-4 in IgE-sensitized RBL-2H3 cells. Interestingly, G-Rh2 was involved in the FcεRI pathway activation of mast cells and the transduction of the Lyn/Syk signaling pathway. G-Rh2 inhibited PI3K activity in a dose-dependent manner. By blocking the antigen-induced phosphorylation of Lyn, Syk, LAT, PLCγ2, PI3K ERK1/2 and Raf-1 expression, G-Rh2 inhibited the NF-κB, AKT-Nrf2, and p38MAPK-Nrf2 pathways. However, G-Rh2 up-regulated Keap-1 expression. Meanwhile, G-Rh2 reduced the levels of p-AKT, p38MAPK and Nrf2 in RBL-2H3 sensitized IgE cells and inhibited NF-κB signaling pathway activation by activating the AKT-Nrf2 and p38MAPK-Nrf2 pathways. Conclusion: G-Rh2 inhibits mast cell-induced allergic inflammation, which might be mediated by the AKT-Nrf2/NF-kB and p38MAPK-Nrf2/NF-κB signaling pathways.

Inhibitory Effect of Astragaloside I and IV on Passive Cutaneous Anaphylaxis Reaction and Scratching Behaviors in Mice

  • To evaluate the antiallergic effect of the dried root of Astragalus membranaceus Bunge (AM) (Leguminosae), which inhibited the mouse passive cutaneous anaphylaxis (PCA) reaction in a preliminary experiment, its main constituents, astragalosides I and IV, were isolated and their antiallergic effects were investigated. Astragalosides I and IV inhibited the PCA reaction induced by the IgE-antigen complex, and the scratching behaviors induced by compound 48/80. These constituents reduced the protein expressions of $TNF-{\alpha}$ and IL-4 in IgE-induced RBL-2H3 cells. These findings suggest that astragalosides I and IV as well as AM can improve IgE-induced anaphylaxis and scratching behaviors.
