Serological Identification of Potato Viruses in Korea (감자 바이러스의 혈청학적 동정에 관한 연구)

  • A total of 230 apparently healthy looking potato stocks and 80 potato stocks with symptoms of virus infection were collected from various seed potato farms in Korea and the incidence of potato virus X (PVX), potato virus S (PVS), potato virus M (PVM) and potato virus Y (PVY) was determined by serological microprecipitin tests. Results obtained are as follows. 1. Serological microprecipitin test retreated the presence of PVX, PVS, PVM and PVY in a number of potato stocks grown for the production of seed potatoes in Korea. 2. The occurrence of potato virus M is reported here for the first time in Korea with experimental evidence. 3. Practically $100\%$ (290 stocks, of the apparently healthy looking potato stocks were demonstrated to be infected with both PVX and PVS. The infection percentages of potato stocks with combination of PVX, PVS, PVM and PVY were as follows. PVX+PVS+PVM:$10.3\%$, PVX+PVS+PVY:$4.5\%$, PVX+PVS+PVM+PVY:$1.03\%$ 4. Irish Cobbler and Shimabara, which are the two major potato varieties in Korea, appear to be symptomless carriers of PVX and PVS. However, when these varieties were infected additionally with PVY, usually severe symptoms resulted. 5. Serological microprecipitin technique appears to be highly suitable for early, quick and reliable diagnosis of PVX, PVS PVM and PVY. It is particularly suited for large scale testing of seed potato stocks for the presence of viruses mentioned above.

Genetic diversity and population structure in five Inner Mongolia cashmere goat populations using whole-genome genotyping

  • Objective: As a charismatic species, cashmere goats have rich genetic resources. In the Inner Mongolia Autonomous Region, there are three cashmere goat varieties named and approved by the state. These goats are renowned for their high cashmere production and superior cashmere quality. Therefore, it is vitally important to protect their genetic resources as they will serve as breeding material for developing new varieties in the future. Methods: Three breeds including Inner Mongolia cashmere goats (IMCG), Hanshan White cashmere goats (HS), and Ujimqin white cashmere goats (WZMQ) were studied. IMCG were of three types: Aerbas (AEBS), Erlangshan (ELS), and Alashan (ALS). Nine DNA samples were collected for each population, and they were genomically re-sequenced to obtain high-depth data. The genetic diversity parameters of each population were estimated to determine selection intensity. Principal component analysis, phylogenetic tree construction and genetic differentiation parameter estimation were performed to determine genetic relationships among populations. Results: Samples from the 45 individuals from the five goat populations were sequenced, and 30,601,671 raw single nucleotide polymorphisms (SNPs) obtained. Then, variant calling was conducted using the reference genome, and 17,214,526 SNPs were retained after quality control. Individual sequencing depth of individuals ranged from 21.13× to 46.18×, with an average of 28.5×. In the AEBS, locus polymorphism (79.28) and expected heterozygosity (0.2554) proportions were the lowest, and the homologous consistency ratio (0.1021) and average inbreeding coefficient (0.1348) were the highest, indicating that this population had strong selection intensity. Conversely, ALS and WZMQ selection intensity was relatively low. Genetic distance between HS and the other four populations was relatively high, and genetic exchange existed among the other four populations. Conclusion: The Inner Mongolia cashmere goat (AEBS type) population has a relatively high selection intensity and a low genetic diversity. The IMCG (ALS type) and WZMQ populations had relatively low selection intensity and high genetic diversity. The genetic distance between HS and the other four populations was relatively high, with a moderate degree of differentiation. Overall, these genetic variations provide a solid foundation for resource identification of Inner Mongolia Autonomous Region cashmere goats in the future.

Quality Characters and Identification of Breeding Materials in Malting Barley (맥주보리 품질 관련 형질과 고품질 육종소재 탐색)

  • Germplasms and breeding lines of malting barley at National Yeongnam Agricultural Experiment Station (NYAES) were evaluated for malting barley quality improvement. Among the numerous malting barley quality parameters, the mean of protein content was 12.3% between 10.7% to 14.0% and range of the $\beta$-glucan content was from 3.5% to 5.8%, the mean was 4.6 percent. The length of acrospire, non-germination rate, friability, the speed of filtration, extract yield, kolbach index, diastatic power were significantly different between the individual varieties, however the other traits were not significantly different. The results of correlation analysis among 15 quality parameters showed significant positive correlation between crude protein content and malt protein content. However, other quality parameters such as sugar content, fiablity, extract yield, and kolbach index had negative correlation with crude protein content. Therefore, crude protein content could be one of the major factors that deteriorate quality. The varieties of Viva, Nishino-chikara, Kinukei 9, Kinukei 12, Sacheon 6 and Jinyangbori that showed over the 80% in extract yield and the higher diastatic power, will be used by crossing parents for improve the quality of malting barley.

Isolation and Identification of Volatile Compounds from Red Wine Manufactured with Vitis vinifera grapes (Vitis vinifera 적포도주 휘발성분의 분리 및 동정)

  • In order to investigate some chemical components, higher alcohols, ethyl acetate and volatile compounds of Vitis vinifera red wines were analyzed by HPLC, GC and GC-MS. During the process of ripening for the content of wine and acids were much changed, particurally the content of tartaric acid was decreased largely. Content of total phenolics and phenol flavonoid in wines which manufactured with Malbec varieties as 470mg/L and 245mg/L respectively, was higher than those in Cabernet sauvignon (310mg/L, 135mg/L) and Cabernet franc (425mg/L, 125mg/L) wines, and nonflavonoid was higher in Cabernet franc wine(300mg/L) than in others. Content of acetaldehyde was higher in Malbec wine (33mg/L) than in Cabernet sauvignon (26mg/L) and Cabernet franc (28mg/L) wines. Amount of methyl alcohol, propanol and isoamyl alcohol were higher in Cabernet sauvignon wine than in others, and isobutanol was more in Malbec wine(64mg/L), ethyl acetate was more in Cabernet franc wine as 35mg/L than in Malbec(28mg/L) and in Cabernet sauvignon (23mg/L) wines. Volatile compounds were isolated about 87~91 varieties from concentrates of three red wines by GC, and thirty-five compounds including terpine-4-ol were identified by GC-MS.

A Taxonomic Study of Asarum (Aristolochiaceae) in Korea (한국산 족도리풀속(Asarum, 쥐방울덩굴과)의 분류학전 연구)

  • Taxonomic treatment and the identification key for 6 species and 4 varieties from the genus Asarum (Aristolochiaceae) in Korea were presented on the basis of the morphological analyses. Recently the taxonomy of the genus Asarum in Korea is controversial in the definition of species and the establishment of variation range. Our morphological studies supported that the species A. patens, A. misandrum and A. versicolor should be recognized as independent species by the unique morphological characters such as calyx lobes, stylar protuberance and leaf variegation. Second, A. sieboldii var. cornutum, A. koreanum, A. maculatum and A. sonunsanense, regarded as species or variety by different scholars, showed a close relationship with A. sieboldii by the similar calyx characters. Thus, new combinations, such as A. sieboldii for. cornutum, A. sieboldii for. koreanum, A. sieboldii for. maculatum and A. sieboldii for. sonunsanense, are proposed. And A. heterotropoides var. seoulense and A. heterotropoides var. mandshuricum also had a close relationship, thus, new combinations, A. mandshuricum for. seoulense, A. mandshuricum for. mandshuricum, are proposed. Furthermore, it is appropriate that A. heterotropoides var. heterotropoides, only distributed in Japan, is revised into A. heterotropoides as independent species by the unique character from the 2 varieties above. Consequently, the genus Asarum in Korea is classified into 3 species and 7 forma.

Identification of volatile flavor compounds in Jeju citrus fruits (제주감귤류의 휘발성 향기성분의 확인)

  • The volatile flavor compounds in five Jeju citrus fruit varieties (Cheonhyehyang, Hallabong, Jinjihyang, Hwanggeum hyang and Redhyang) were extracted by simultaneous distillation-extraction (SDE) using solvent mixture of n-pentane and diethyl ether (1:1, v/v) and analyzed by using gas chromatography-mass spectrometry (GC-MS). The number of aroma compounds were identified with : 104 (4,939.77 mg/kg) in Cheonhyehyang, 98 (3,286.38 mg/kg) in Hallabong, 105 (3,317.56 mg/kg) in Jinjihyang, 102 (4,293.39 mg/kg) in Hwanggeumhyang, and 108 (4,049.94 mg/kg) in Redhyang. The detected main volatile compounds were; limonene, sabinene, ${\beta}$-myrcene, ${\alpha}$-pinene, ${\beta}$-pinene, linalool, 4-terpineol, ${\alpha}$-terpineol, (E)-${\beta}$-ocimene and ${\gamma}$-terpinene. Among the identified volatiles compounds, ethyl-benzene, nonanol, 1-p-menthen-9-al, (E)-isocarveol, methyl salicylate, ${\alpha}$-terpinen-7-al, perilla alcohol, and ethyl-dodecanoate were detected in Cheonhyehyang. only Furthermore, ${\beta}$-chamigrene and ${\alpha}$-selinene were in Hallabong only; 3-hydroxybutanal, (E)-2-nonenal, isoborneol, octyl acetate, (E)-2-undecenal, ${\beta}$-ylangene and guaia-6,9-diene in Jinjihyang. ${\rho}$-Cymenene, ${\beta}$-thujone, selina-4,11-diene and (E,E)-2,6-farnesol in Hwanggeumhyang only; and ${\rho}$-cymen-8-ol, bornyl acetate, carvacrol, bicycloelemene, ${\alpha}$-cubebene and 7-epi-${\alpha}$-selinene in Redhyang only. This study confirmed the differences in composition and content of volatile aroma components in five varieties of Jeju citrus fruits.

Molecular Authentication and Phylogenetic Relationship of Bupleurum Species by the rDNA-ITS Sequences (rDNA-ITS 염기서열 분석을 통한 시호 종 감별용 유전자 마커 개발 및 유연관계 분석)

  • Objectives : Bupleuri Radix (Siho) is prescribed as the root of different Bupleurum species on the pharmarcopoeia in Korea and China. Moreover, other species and varieties of the genus Bupleurum have been also distributed on the herbal market as Bupleuri Radix. However, due to the morphological similarity and frequent occurrence of intermediate forms, the correct identification of this radix is very difficult. To develop a reliable method for correct identification and improving the quality standards of official Bupleuri Radix, we analyzed sequences of the ribosomal RNA gene and internal transcribed spacer (rDNA-ITS) region. Methods : PCR amplification of rDNA-ITS region was performed using ITS1 and ITS4 primer from 6 Bupleurum species and 1 variety, B. falcatum L. (Siho), an improved breed of B. falcatum L. (Samdo-Siho), B. chinense DC. (Buk-Siho), B. scorzonerifolium Willd. (Nam-Siho), B. longiadiatum Turcz. (Gae-Siho), B. euphorbiodes Nakai (Deungdae-Siho) and B. latissimum Nakai (Seom-Siho), and nucleotide sequence was determined after sub-cloning into the pGEM-Teasy vector. Authentic marker nucleotides were estimated by the analysis of ClastalW using entire rDNA-ITS sequence of three samples per species. Results : In comparative analysis of the rDNA-ITS sequences, we found specific nucleotides to distinguish Korean (B. falcatum L. and its variety) and Chinese official species (B. chinense DC. and B. scorzonerifolium Willd.) from others at positions 411 and 447, and positions 89, 101, 415 and 599, respectively. Futhermore, we also found nucleotide indels (insertion and/or deletion) and substitutions to identify each of different Bupleurum species, 2 positions for B. falcatum L. and its variety, 6 positions for B. chinense DC., 49 positions for B. scorzonerifolium Willd., 8 positions for B. euphorbioides Nakai, 7 positions for B. longiradiatum Nakai and 9 positions for B. latissimum Nakai. These sequence differences at corresponding positions are avaliable nucleotide markers to determine the botanical origins of Bupleuri Radix. Moreover, we confirmed the phylogenetic relationship of B. latissimum Nakai, a Korean endemic speices, among Bupleurum species based on the rDNA-ITS sequence. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by the providing of definitive information that can identify each plant species and distinguish it from unauthentic adulterant Bupleurum species.

A Literature Study on the Korean Acupuncture for Oral, Glottal, Labial and Dental diseases (구설순치(口舌脣齒)질환에 대한 한국 침구서적의 치료법 비교 연구)

  • Background : The varieties of manifestations referred to oral, glottal, labial and dental diseases has been long complicated physicians in the efficient diagnosis and treatment of these diseases. Acupuncture has been widely used in Korea throughout the history and provides an efficient method in the treatment of them Objectives : Establish a distinctive and efficient acupuncture method for the treatment of oral, glottal, labial and dental diseases based in literature research Method : We reviewed four Korean medical literature, "Guide to Swollen Sore Treatment", "Treasured Mirror of Eastern Medicine", "Experiential Prescriptions of Acupuncture and Moxibustion", and "Essential Rhymes on Acupuncture and Moxibustion by Master Sa-am", and analyzed the therapeutic characteristics in the treatment of oral, glottal, labial and dental diseases Result : 1. According to "Guide to Swollen Sore Treatment", we could noted frequent application of blood letting methods in the affected area, then rinse ones mouth with salty water and further application of taro plaster. Also found acupuncture methods utilizing heated mole cricket or silkworm in the sublingual region. Regarding herbal method, Realgar was rubbed in the affected area. Mainly used acupuncture points in the treatment of oral, glottal, labial and dental diseases were GV20, GB20, LU5, and auricular anterior hairline 2. In "Treasured Mirror of Eastern Medicine", blood letting method in the sublingual region and burning needle searing method were the most frequently applied in the treatment of oral, glottal, labial and dental diseases. Moxibustion was also applied in the treatment of labial and dental diseases. Particularly, said to apply 3 moxa cones in ear zones of both sides, when no medicine is effective. This demonstrates how emphasized the importance of moxibustion in this kind of disease. Mainly used acupuncture points were GV16, CV24, LI4, EX-HN12, and EX-HN13 3. In "Experiential Prescriptions of Acupuncture and Moxibustion", no other methods beside wrist. Superstitious methods like applying moxibustion on the tooth picture drawn on the roofing tile need further confirmation. Mainly used acupuncture points were LI4, ST36, and HT7. 4. "Essential Rhymes on Acupuncture and Moxibustion by Master Sa-am", identified the cause of diseases according to visceral pattern identification. Stomach and Spleen in charge of vocal, lingual, and labial disease, Kidney for dental disease, and further scrutinizing identification according to Liver, Heart, Stomach, Lung, and Kidney manifestations. Used supplementation and draining needling methods of self meridians and other correlating meridians. Conclusions : After previous study on stroke and eye diseases, we could also find various efficient methods according to oral, glottal, labial and dental diseases, through literature research of korean medical classics. This study will concurrently result in establishing distinctive therapeutic method characteristic of Korea.

Taxonomical Studies of the Genus Acer in Korea by the Morphological Characteristics of Stipules (탁엽(托葉)의 형태적(形態的) 특징(特徵)에 의한 한국산(韓國産) 단풍나무속(屬)의 분류학적(分類學的) 연구(硏究))

  • Taxonomical studies of the genus Acer in Korea (8 species, 3 varieties and 1 forma) were studied by the morphological characteristics of stipules. The results obtained were summarized as follows; 1) The stipules of the genus Acer were arranged oppositely with 4 - 10 stipules consisting of simple leaf, compound leaf and unifoliate compound leaf. The venation of stipules were cladodromous, basal reticulate, fibrous and palmately netted vein. The trichoma of stipules were glandula hair, glandular pubescent, sericeous, pilous, vinous, velutinous, tomentose, ciliate and lanate. As the morphological characteristics of stipules in the genus Acer established a strong point in the identification of species. 2) The species in the genus Acer in Korea were classified into six groups; (1) ginnala group (A. ginnala), (2) koreanum group (A. koreanum, A. japonicum, A. pseudosieboldianum var. koreanum), (3) mono group (A. mono, A, palmatum var. sanguineum, A. palamtum var. dissectum, A. palmatum var. dissectum f. aosidare), (4) negundo group (A. negundo), (5) palmatum group (A. palmatum), (6) platanoides group (A. platanoides, A. truncatum) by the morphological characteristics of stipules, 3) The stipules of platanoides group and negundo group were the least evolved types among the species in the genus Acer. 4) The stipules in the genus Acer were made the inner buds that were metamorphosed to the protected functional aspect by the evolution of petiols.

A Procedure for Inducing the Occurrence of Rice Seedling Blast in Paddy Field

  • Rice blast caused by the filamentous fungus Magnaporthe oryzae, is arguably the most devastating rice disease worldwide. Development of a high-throughput and reliable field blast resistance evaluation system is essential for resistant germplasm screening, resistance genes identification and resistant varieties breeding. However, the occurrence of rice blast in paddy field is easily affected by various factors, particularly lack of sufficient inoculum, which always leads to the non-uniform occurrence and reduced disease severity. Here, we described a procedure for adequately inducing the occurrence of rice seedling blast in paddy field, which involves pretreatment of diseased straw, initiation of seedling blast for the first batch of spreader population, inducing the occurrence of the second batch of spreader population and test materials. This procedure enables uniform and consistent infection, which facilitates efficient and accurate assessment of seedling blast resistance for diverse rice materials.
