Studies on the High Protein Mutants of Rice (수도 고단백 돌연변이계통에 관한 연구)

  • Several high protein mutant lines(M4 plant generation, 1974) obtained from X-ray irradiated Jinheung variety were examined at three different locations for their agronomic characters, protein and grain yields. On the other hand, high protein-short culmed-early maturity mutant line No. 398 (M$_{10}$ plant generation, 1974) induced from Hokwang was crossed back to its mother to investigate the gene(s) controlling protein and its pleiotropic relation to other mutated characters. Although variation of protein percent of mutant lines from Jinheung was comparatively large depending on year and location, most of the high protein mutant lines had higher protein yield per unit area than the mother variety and their grain yields were equal to or better than the mother, being resistant to both leaf and neck blast. They were several days earlier-maturing and had shorter-culm except one mutant line. The culm length and heading date of F$_1$ between high protein mutant 398 and its mother Hokwang were intermediate. Accurate assessment of segregation of culm length and heading date in F$_2$ generation and protein percent in F$_3$ seeds will be conducted in 1975.

Selection of Yeast Mutant Strain with High RNA Content and Its High Cell-Density Fed-Batch Culture. (고함량 RNA 효모 변이주의 선별 및 고농도세포 유가배양)

  • To obtain a yeast mutant with high RNA content and high growth rate, Saccharomyces cerevisiae MTY62 was mutated with ethylmethane sulfonate. Among the selected mutants that were sensitive to the high concentration of KCl, M40-10 strain was finally selected due to its rapid cell growth and high RNA content in the tube and baffled-flask cultures. In the batch culture of M40-10 mutant, the maximum specific growth rate ($\mu_{max}$) of $0.38 h^{-1}$ , RNA concentration of 3210 mg-RNA/1, and RNA content of 183 mg-RNA/g-DCW were obtained, which were 23%, 15%, and 12% increased levels, respectively, compared to those of MTY62 parent strain. The intermittent fed-batch culture of M40-10 strain resulted in the maximum cell concentration of 35.6 g-DCW/1, RNA concentration of 5677 mg/1, and RNA content of 160 mg-RNA/g-DCW. Through the constant fed-batch culture, the maximum cell concentration of 46.4 g-DCW/1, RNA concentration of 6270 mg-RNA/1, and RNA content of 135 mg-RNA/g-DCW were obtained. At the 20 h culture time in the fed-batch cultures of M40-10 strain, the cell and RNA concentrations were increased by 30% and 10%, respectively, over the parent strain MTY62. In addition, it was also found that the accumulated RNA within the mutant cell was not degraded until the end of fed-batch cultivation, indicating that the M40-10 cell is a mutant with weak acidic RNase activity.y.

Plasma Concentrations of Clozapine and its Metabolites and FMO3 Variations in Korean Schizophrenic Patients (정신분열병 환자에서 Clozapine과 그 대사물들의 혈장농도 및 FMO3 유전자 변이)

  • Objective : The relationship between the total daily dose of clozapine given and the plasma concentrations of clozapine and its metabolites(N-desmethylclozapine and clozapine N-oxide) and the effect of Glu158Lys (wild-type : Glu, 'H' ; variant : Lys, 'h') and Glu308Gly(wild-type : Glu, 'D' ; variant : Lys, 'd') variation in FMO3 gene on plasma concentrations of clozapine and its metabolites was studied in schizophrenic patients. Methods : Trough plasma concentrations of clozapine and its metabolites were measured in 34 schizophrenic patients receiving clozapine. The genetic variation of 'h' and 'd' in FMO3 were analyzed in 21 among 34 patients. Results : A linear relationship between the total daily dose of clozapine given(mg/kg body weight per day) and the plasma concentrations(nM) of clozapine was revealed by regression analysis(p<0.001) in the 23 patients receiving a constant daily dose of clozapine for 8 days. The plasma molar concentration ratios of clozapine N-oxide/clozapine in 8 subjects with 'hh' or 'Hh' alleles were not different from those in 6 subjects with 'HH' alleles and the plasma molar concentration ratios in 6 subjects with 'dd' or 'Dd' alleles were not different from those in 8 subjects with 'DD' alleles. Conclusion : The effect of Glu158Lys and Glu308Gly variation in FMO3 gene on clozapine metabolism could not be shown.

  • Variation of plant characters and correlation anlaysis of its in Bupleurum falcatum, medicinal plant, were investigated to find useful selection characters and to obtain fundamental data for breeding. The variation was generally high. In plants having high height and many branch, stem thickness was high and leaf and root weight were great. The group of short height and many branch showing low frequency was higher than that of medium height and branch showing high frequency. And the correlation between top and root characters were positively significant The major top characters correlated with root yield were stem thick-ness, branch number, node number, node position attached 1st branch(NPFB), and leaf area.

The Development of a Learning Program for Enhancing the Skills of Control Variables and the Effects of Its Applications (변인 통제 능력을 강화하기 위한 수업 프로그램의 개발 및 적용 효과 분석)

  • The main purpose of this study was to develop a teaching program, especially designed to improve the skills of control variables. The secondary purpose was to investigate the effect of the program on enhancing students' scientific reasoning and understanding. The program was designed based on the 3-step learning model: i.e. students recognize the necessity of controlling the variables (step 1), perform their own experiments (step 2), and reflect on their variables control process (step 3). The program included 9 topics of increasing difficulty. In results, Lawson's SRT scores increased in both experimental and control groups after application of the program, but the difference was not statistically significant. After the application, there was an increase in type A and type B which implied that students' skills of control variables was improved. In addition, responses of students in the experimental group to the open-ended items showed that it was challenging for them to think scientifically and critically when controling variables, but they ended up feeling proud of their achievement after the program.

Molecular Identification of Sagittaria trifolia and S. aginash Based on Barcode (바코드에 의한 보풀속(Genus Sagittaria L.) 보풀과 벗풀의 동정)

  • Since sulfonylurea (SU) herbicide-resistant Monochoria korsakowii in Seosan reclaimed land in 1998 was reported first, herbicide-resistant weed species and their area of occurrence have been steadily increasing. In recent years, Sagittaria trifolia resistant to SU herbicides has been reported in Gimhae, Gyeongnam province. While collecting S. trifolia for constructing barcode database of major weeds we were suspicious by the continuous variation in the leave morphology of the species. In order to identify the S. trifolia from S. aginash we barcoded the species collected from domestic using ITS and compared the sequence variation with the ITS sequences of S. aginash downloaded from NCBI. As a result, it was found that the plants collected from the domestic did not have any variation among individuals although they showed wide morphological variation. On the other hand, interspecific variation between S. trifolia and S. aginash was 4.6%. Plants that are difficult to identify using morphological characters can be identified quickly and accurately using the barcode technique. Herbicide-resistant weeds may require different management practices depending on the species even in the same genus. Domestic herbicide-resistant weeds are steadily increasing. Therefore, accurate identification of these species must be preceded for effective weed control.

Phylogeny of Scopolia Jacq. s. str. based on ITS sequences (ITS 염기서열에 의한 미치광이풀속의 계통)

  • Internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA were determined for 14 individuals representing eight taxa from Scopolia s. str. and related genera, Anisodus and Atropanthe. We found that the ITS sequences of Korean endemic species, S. parviflora, are significantly different from its allied species, S. japonica. This is contradictory to traditional taxonomic treatments in which those species are regarded as conspecific. S. parviflora exhibited closer relationship to S. carniolica, which is disjunctly distributed in Europe. In spite of substantially high sequence divergence between S. japonica and S. parvlflora/S. carniolica clade, morphological resemblance is evident among the species. Morphological stasis concept (retardation of morphological differentiation or evolution of similar characters among the disjuncts in a similar ecological habitat) was referred to understand this rather unusual evolutionary feature. S. lutescens, another Korean endemic species, shared almost identical ITS sequences with S. parviflora. Lack of diagnostic character distinguishing the taxa suggests that they are conspecific. Anisodus carniolicoides, which was originally described in Scopolia, was grouped with A. luridus and A. tanguticus. The monophyletic Anisodus formed a sister group relationship with a monotypic genus Atropanthe.

The Base Sequence of ITS and Genetic Variation in Sarcodon Aspratus (능이버섯의 ITS염기서열과 유전적 변이)

  • The sequence of ITS (partial 16S ribosomal DNA, complete ITS1, 5.8S ribosomal DNA and ITS2, and partial 28S ribosomal DNA) was analysed by PCR and autosequencing in Sarcodon aspratus. The ITS lenght of S. aspratus was 716 base pair. As this sequence compared with other reports on S. aspratus (ace No AF335110), the sequence variation based on nucleotide deletion and substitution was $1.8\%$. This nucleotide variation rate in same species was very higher than in other species. Also, the sequence varitation rates between this S. aspratus and S. imbricatus, and S. squamus were $8\%\;and\;10\%$, respectively. This results suggested that the high sequence variation of S. aspratus might be caused specific host and inhabitat environment which limited gene flow.

Genetic Variation in Mutants Induced by Gamma Ray in Hypsizigus marmoreus (느티만가닥버섯에서 감마선에 의한 돌연변이체들의 유전적 변이)

  • This research was carried out to evaluate whether gamma ray is a useful tool for breeding new strains of mushrooms. For this research, 5 mutant groups, 20 strains of Hypsizigus marmoreus, 2 strains of Lyophyllum decastes, and 1 strain of Lyophyllum shimeji were used. Monokaryon spores from one variety of H. marmoreus were irradiated with 50~2,000 Gy of gamma ray. The propriety dose was 50~200 Gy for mutagenesis. Mutant monokaryon mycelia crossed each order to become dikaryon mycelia. The internal transcribed spacer (ITS) regions of rDNA were amplified using PCR, and the products were sequenced. The sequences of the ITS regions (16 partial rDNA, complete ITS1, 5.8 rDNA and partial rDNA) were analyzed by PCR, and strains of H. marmoreus, L. decastes, and L. shimeji were auto-sequenced. The lengths of the sequenced ITSs were 1,052~1,143 nucleotides. Genetic matrices were calculated using Nei-Li's genetic distance coefficient based on ITS sequence. The dissimilarities were 0~3.35% in strains of H. Hypsizigus. In addition, a phylogenetic tree was constructed based on ITS sequences using the neighbor-joining (NJ) method. The phylogenetic tree revealed that 23 strains and 5 mutant groups were divided into 12 clusters; the mutant groups fell into different clusters. These results show that mushroom spores were mutated effectively by gamma ray; therefore, gamma ray could be a useful tool for breeding new strains of mushrooms.

  • Simple and rapid screening methods were developed to screen mutant strains of Trigonopsis variabilis ATCC10679 (TW). D-amino acid oxidase (D-AAO) from a mutant strain, T26, showed about 30% higher specific activity against cephalosporin C than from its wild type, TW. D-AAO genes from both TW and T26 strains were cloned and sequenced. There was one nucleotide changed from T to C at 811 position, resulting in an amino acid codon changed from Phe-258 to Ser-258.
