Water quality prediction of inflow of the Yongdam Dam basin and its reservoir using SWAT and CE-QUAL-W2 models in series to climate change scenarios (SWAT 및 CE-QUAL-W2 모델을 연계 활용한 기후변화 시나리오에 따른 용담댐 유입수 및 호내 수질 변화 예측)

  • This paper analyzes the impact of two climate change scenarios on flow rate and water quality of the Yongdam Dam and its basin using CE-QUAL-W2 and SWAT, respectively. Under RCP 4.5 and RCP 8.5 scenarios by IPCC, simulations were performed for 2016~2095, and the results were rearranged into three separate periods; 2016~2035, 2036~2065 and 2066~2095. Also, the result of each year was divided as dry season (May~Oct) and wet season (Nov~Apr) to account for rainfall effect. For total simulation period, arithmetic average of flow rate and TSS (Total Suspended Solid) and TP (Total Phosphorus) were greater for RCP 4.5 than those of RCP 8.5, whereas TN (Total Nitrogen) showed contrary results. However, when averaged within three periods and rainfall conditions the tendencies were different from each other. As the scenarios went on, the number of rainfall days has decreased and the rainfall intensities have increased. These resulted in waste load discharge from the basin being decreased during the dry period and it being increased in the wet period. The results of SWAT model were used as boundary conditions of CE-QUAL-W2 model to predict water level and water quality changes in the Yongdam Dam. TSS and TP tend to increase during summer periods when rainfalls are higher, while TN shows the opposite pattern due to its weak absorption to particulate materials. Therefore, the climate change impact must be carefully analyzed when temporal and spatial conditions of study area are considered, and water quantity and water quality management alternatives must be case specific.

Association with Kinetic Characteristics of sperm in Duroc Boar and the Zygote Arrest 1 gene Polymorphism (g.2540T>C) (Zygote arrest 1 유전자 변이(g.2540T>C)와 두록 정액의 운동학적 특성과의 연관성 분석)

  • The sperm quality is determined by the kinetic characteristics and acrosome integrity of the sperm. In the previous studies, analysis of semen quality had large errors because those experiments by using microscope had been conducted by people. In recent years, the molecular biological methods have been newly developed to complement the previous techniques. The ZAR1 gene is known to be a gene that affects early embryonic development in vertebrates, but there is no study of the association with semen. In this study, we analyzed the association between the kinetic characteristics and ZAR1 single nucleotide polymorphism (SNP) genotype. To detect the SNPs, we performed sequencing using genomic DNA from the whole bloods of Duroc pigs. We identified an SNP in the ZAR1 gene g.2540T>C. ZAR1 SNP genotypeing in 105 pigs revealed that the major and minor alleles were T and C, respectively. After we analyzed the association between the kinetic characteristics of sperm and the ZAR1 SNP genotype, we found a significant association in MOT (p<0.01), VSL (p<0.05) of the kinetic characteristics in the Duroc boars. It was confirmed that the boars with T allele were lower in MOT and VSL than C allele. Therefore, pigs with C allele are judged to be better at the MOT and VSL of semen. Based on these results, ZAR1 SNP genotyping may be a useful molecular biomarker to improve semen quality by applying molecular breeding technology.

Evaluation of the Storage Characteristics of Kangjung Added with Gromwell Extracts (지치 추출물 첨가에 의한 강정의 저장성 연장 효과)

  • This study examined the effects of gromwell (Lithospermum erythrorhizon) extracts on the storage characteristics of Kangjung. At an increase of $0{\sim}0.25%$, there was no significant difference in the moisture content of wild and cultivated gromwell extract added to Kangjung, whereas there was a significant difference when increased to 0.5%. When compared, the occurrence of lipid oxidation among the controlled gromwell extract group stored at $60^{\circ}C$, resulted in peroxide values exceeding 40 meq/kg which is a maximum value for Hankwa within 12 hours of storage. However, at an increase of 0.25%, the cultivated gromwell extract added group reached limits within 16 hours. Furthermore, at a 0.5% increase in cultivated gromwell and 0.25% in wild gromwell, extract added groups exceeded this limit within 20 hours of storage. Gromwell added Kangjung, a fried food, showed efficiency in suppressing oxidation of fat, more so in the wild gromwell than the cultivated gromwell. In addition, efficiency in suppressing oxidation of fat increased at the 0.5% level than the 0.25% level. In conclusion, it is believed that an unused pigment resource called Lithospermum erythrorhizon, normally reserved for medicinal use, can be distributed as a safe food additive. In addition, as Lithospermum erythrorhizon is a natural antioxidant in fried foods, we believe this satisfies the demand for natural food.

Assessment of Dietitian's Nutritional Quality Management for School Food Service (학교급식 영양사의 영양적 품질관리 수행도 평가)

  • The purpose of this study was to evaluate school dieticians' performance of nutritional quality control, and further to establish effective and objective standards of nutritional quality control. Data for this study came from 200 school dieticians' responses in the Chungbuk area. The total quality management (TQM)-based questionnaire was structured. The questionnaire consisted of the following four fields (1) performance of nutritional quality control, (2) performance of stepwise food production to maximize nutrient preservation rate, (3) management of documents and records related to nutritional quality control, and (4) other relating matters. The items of the questionnaire were measured on a five-point Likert scale which ranged from 'strongly agree' to 'strongly disagree'. First, the analysis indicated that school dieticians performed 'least' on human resource management', 'mediocre' on nutritional quality control, and best on 'leadership'. Second, the analysis on performance of stepwise food production to maximize nutrient preservation rate showed that dieticians considerably endeavored to maximize nutrients of cooked food, but it was found out that the most of nutrient destruction can be caused by heating during cooking. Third, the result showed that the systematic use of documents and records for nutritional quality control was not sufficiently accomplished, especially in the production phase of food. In addition, the measure by the Pearson correlation coefficient indicated that there was a significant relationship between performance of nutritional quality control and performance of stepwise food production to maximize nutrient preservation rate, and between performance of nutritional quality control and management of documents and records related to nutritional quality control. Finally, the findings of this study suggest that more effort should be exerted to carefully establish TQM-based standards for the improvement of nutritionary quality.

A Study on the Service Quality Improvement by Kano Model & Weighted Potential Customer Satisfaction Index (Kano 모델 및 가중 PCSI를 통한 서비스품질 개선에 관한 연구)

  • The Banking industry is expanding rapidly. To keep the competitive advantages, participating companies concentrate their resource to provide the distinguishable services by increasing the service quality. This study is to find that how three kinds of service quality(process, output, and service environment) affect on the customer satisfaction. In this paper, WPCSI (Weighted Potential Customer Satisfaction Index) was developed using Kano model and PCSI. Kano's model of service quality classification was used to improve customer satisfaction, customer satisfaction index was calculated. Customer satisfaction index was calculated using the existing potential for improving customer satisfaction index (PCSI Index) to complement the limitations of the weighted potential improve customer satisfaction index (WPCSI) were used. Analysis using PCSI improve the quality of service levels may be useful in assessing. However, this figure is a marginal degree of importance on customers and quality characteristics have been overlooked but has its problems. A service provided to customers with some important differences depending on the interpretation of the scope for improvement is to be classified. In other words, the level of customer satisfaction and the satisfaction of the current difference between the comparison factor for the company to provide information about the priority of the improvement was not significant. Companies are also considered important that the customer does not consider the uniform quality of service provided can be fallible. In this study, the weighted potential to improve it improve customer satisfaction index (WPCSI) proposed a new customer satisfaction index. This is for customers to recognize the importance of quality characteristics by weighting factors, to identify practical and improved priority to provide more useful information than has been. Weighted potentially improve customer satisfaction index (WPCSI) presented in this study by the customers aware of the importance of considering the quality factor is an exponent. The results, 'Employees' working ability', 'provided the desired service level', 'staff to handle this task quickly enough' to the customer of the factors had significant effects on satisfaction are met. On the other hand 'aggressiveness on the product description of employees', 'service environment as a whole, beautiful enough to' meet and shows no significant difference between satisfaction. But 'aggressiveness on the product description of employees' and reverse (逆) were attributable to the quality. Small dogs and overly aggressive products that encourage the customer dissatisfaction that can result in widening should be careful because the quality factor can be said. As a result, WPCSI is more effect to find critical factors which can affect customer satisfaction than PCSI. After that, we discuss effects and advantages of customer satisfaction using WPCSI. This study, along with these positive aspects, the limitations are implied. First, this study directly to the bank so that I could visit any other way for customers, utilizing the Internet or mobile to take advantage of the respondents were excluded from the analysis. Second, in survey questionnaires can help improve understanding of the measures will be taken. In addition to the survey targeted mainly focused on Seoul, according to a sample, so sampling can cause problems is the viscosity revealed intends.

STUDIES ON THE EXTRACTION OF SEAWEED PROTEINS 4. Precipitation Conditions and Nutritional Evaluation of Isolated Seaweed proteins (해조단백질 추출에 관한 연구 4. 추출단백질의 심전조건 및 영양적 평가)

  • For the effective utilization of diverse and abundant resource of seaweeds in Korea as a food protein supplment, extraction conditions of water, salt, and alkali soluble proteins were investigated in previous work(Ryu and Lee, 1977: Lee et al., 1977: Lee et al., 1978). The present study as a part of the serial work was thus aimed to find the conditions of isolation and purification of extracted proteins, and to evaluate the nutritional quality of the isolated seaweed proteins in terms of amino acid composition, chemical score, protein score, modified essential amino acid index(MEAAI), and in vitro digestibility presented as pepsin-pancreatin digest residue index (PPDRI). As for the isolation of extracted proteins, TCA treatment was more effective for the proteins from rhodophyceae and Chlorophyceae while the precipitation at isoelectric point was more desirable for Phaeophyceae proteins. In amino acid composition, water soluble protein fraction was superior to the other fractions in Porphyra suborbiculata whereas both water and alkali soluble fractions seemed to bo more benefitial for Enteromorpha linza and Ulva pertusa; the extraction with alcohol-alkali mixed solvent for Undaria pinnatifida and Sargassum fulvellum. Glutamic acid and aspartic acid content was particularly high in all protein fractions examined. The total amino acid content of Porphyra suborbiculata and Enteromorpha linza was almost equivalent to that of dried whole egg although the essential amino acid content was lower. A comparative analysis was made on the inedexes between raw seaweed powder and isolated protein. Chemical score of Porphyra suborbiculata and Ulva pertusa was approximately 35 and 56 in cafes of raw powder and isolated protein respectively while only 10 to 16 for raw powder of Undaria pinnatifida and Sargassum fulvellum and 30 to 35 for their isolated proteins. Protein score of all isolated proteins was in the range of 63 to 73 which indicates that isolated protein would be mere valuable than the fern of raw seaweed powder. Digestibility by means of PPDRI was found to be extremely low in case of raw powder but it could be doubled in case of isolated protein yielding 67 to 70 for Porphyra suborbiculata and Ulva pertusa.

Feed Value, Growth and Dry Matter Yield of High Yielding Rice(Oryza sativa L.) for the Twice-cutting System (2회 예취 재배법을 이용한 초다수 벼(Oryza sativa L.) 계통의 생육·수량 및 사료가치 평가)

  • The four rice(Oryza sativa L.) lines, previously demonstrated to have vigorous herbage production at the vegetative growth stage, were investigated for their growth traits, dry matter yield and feed value using the twice-cutting system that 1st-cut is harvested at panicle formation stage and 2nd-cut at the yellow ripen stage. The twice-cutting system is regarded as an effective method in the production of forage rice by escaping lodging and insect damages caused by the overgrowth and typhoon around the heading stage.The feed value of the twice-cut rice plant of there lines were compared with the control of normal, cutting at the yellow ripen stage. The relative feed value(RFV), the total digestible nutrient(TDN) and the crude protein(CP) content of the twice-cut straw were higher the control in every lines. However, the dry matter yield (DMY), represented by the summed forage production of 1st and 2nd cut-rice plant, were lower the control, and the DMY of straws were increase but grains were decrease for twice-cutting system. In addition, TDN yield showed the similar tendency with DMY.Recently, as one of the breeding aims the researches for improving the digestibility of rice straw have been required. In this regard, twice-cutting system is recognized with the fact that it has the effect which raises the digestibility of the straw. Especially, the 2nd cut of IR73111-B-R-15-3-1 line was similar to the control at the TDN yield of straw, and would be used as a breeding material for product the roughage of good quality.

Isolation and Structure Identification of Photosensitizer from Perilla frutescens Leaves Which Induces Apoptosis in U937 (들깻잎(Perilla frutescens)으로부터 U937 세포에 apoptosis를 유도하는 광과민성 물질의 분리 및 구조동정)

  • In this study, we tried to separate the photosensitizer that induces apoptosis of leukemia cells (U937) from perilla leaves. Perilla leaves (Perilla frutescens Britt var. japonica Hara) are a popular vegetable in Korea, being rich in vitamins (A and E), GABA, and minerals. Dried perilla leaves were extracted with methanol to separate the photosensitizer by various chromatographic techniques. The structure of the isolated compound (PL9443) was identified by 1D-NMR, 2D-NMR, and FAB-mass spectroscopy. Absorbance of the UV-Vis spectrum was highest at 410 nm and was confirmed by the 330, 410, and 668 nm. PL9443 compound was determined to be pheophorbide, an ethyl ester having a molecular weight of 620. It was identified as a derivative compound of pheophorbide structure when magnesium comes away from a porphyrin ring. Observation of morphological changes in U937 cells following cell death induced by treated PL9443 compound revealed representative phenomena of apoptosis only in light irradiation conditions (apoptotic body, vesicle formation). Results from examining the cytotoxicity of PL9443 substance against U937 cells showed that inhibition rates of the cell growth were 99.9% with the concentration of 0.32 nM PL9443. Also, the caspase-3/7 activity was 99% against U937 cells with the concentration of 0.08 nM of PL9443 substance. The result of the electrophoresis was that a DNA ladder was formed by the PL9443. The PL9443 compound is a promising lead compound as a photosensitizer for photodynamic therapy of cancer.

Study on Characteristics of Community and Ecology of Fishes in the Newly Constructed Gunwi Dam Reservoir (신규로 건설된 군위댐 호내 어류 군집 및 생태적 특성에 관한 연구)

  • To secure water resources, dams are normally constructed on the upper - middle part of streams, and it generates physical disturbances such as habitat alteration and stream fragmentation. Such construction can restrict movement of aquatic organisms, especially for freshwater fish which is one of top predator in aquatic ecosystem, and cause genetic fragmentation and community change. In this study, to investigate impact of habitat alteration after dam construction on freshwater fish, we monitored fish community changes, and compared fish fauna between dam reservoir and inflows. Additionally, movement characteristics and habitat boundaries of four species were identified by radio telemetry method. The study was conducted in the Gunwi Dam which was constructed in December 2010. Radio telemetry was applied to Pungtungia herzi, Zacco platypus (living lotic and lentic), Silurus asotus (lentic preferred species) and Zacco koreanus (lotic preferred species). The number of species was remarkably decreased (4 family, 10 species) comparing with before the dam construction (7 family, 15 species). Specifically, Coreoleuciscus splendidus, Niwaella multifasciata, Liobagrus mediadiposalis, Coreoperca herzi and Odontobutis platycephala that inhabit in the lotic environment were not collected in the study area. A total of 8 species were caught in both the dam reservoir and tributaries except 2 species (C. auratus and S. asotus). Sorenson's similarity between the reservoir and its tributaries was high (0.842). All of the radio tagged species stayed in the reservoir except S. asotus which moved to the tributary. These species mainly utilized the shallow littoral zone as a habitat. These results could be useful as a baseline data for efficient management of fishes in lakes.

Technical Efficiency of Medical Resource Supply and Demand (의료자원 공급, 수요의 성과 효율성에 대한 실증분석)

  • The objective of this study is to observe the efficiency of clinical performance on the supply and demand of medical resources in Korea. For the empirical analysis, we constructed the dataset on age standardized mortality rate, the number of physician, specialist, surgery, medical institution, ratio of general hospitals of 16 provinces in Korea from 2006 to 2013. The panel probability frontier model is employed as an analysis method and considered heteroscedasticity and autocorrelation of the error in panel data. In addition, the demographic and socioeconomic characteristics of the 16 provinces, unemployment rate, elderly population ratio, GRDP per capita, and ratio of hospitals in comparison to the general hospitals are used to find the effect on the technical efficiency of clinical performance on supply and demand of medical resources. The results are as follows. First, for the clinical performance, the supply side of human resources such as doctors and specialists and the demand side factors such as chronic illness clinic per unit population have a significant influence, respectively. Second, the technical efficiency of clinical performance on the supply and demand of medical resources of each input component was 59-70% in terms of clinical efficiency in each region. Third. estimates of technical efficiency of inputs that affect clinical performance showed a slight increase in all regions during the analysis period, but the increase trend decreased slightly. Fourth, the ratio of the elderly population and GRDP per capita have a positive influence on the technical efficiency of clinical performance on the supply and demand of medical resources. The difference of each efficiency by region is due to the regional differences of the input medical resources and the combination of them and the demographic and socioeconomic characteristics of the region. It is understood that the differences in technological efficiency due to the complexity of supply and demand of medical resources, demographic structure and economic difference affecting clinical performance by region are different.
