Genetic Stability Analysis of in vitro Regenerated Wolly Grass (Imperata cylindrica 'Rubra') Based on Inter-Simple Sequence Repeats (ISSR) Markers (ISSR 표지에 의한 기내재생 홍띠(Imperata cylindrica 'Rubra')의 유전적 안정성 분석)

  • 지구온난화에 따라 농업부문 신재생에너지의 중요성이 증대되고 있으며, 화본과 식물은 바이오에너지작물의 중요한 소재를 제공하고 있다. 화본과 식물의 기내대량증식연구의 일환으로 홍띠식물의 기내 재생 식물체의 유전적 안정성에 대한 기초자료를 제공할 목적으로 기내배양으로 재분화시킨 홍띠(Imperata cylindrica 'Rubra') 재분화 식물체 중 녹색체 재생식물체를 대상으로 ISSR 표지를 사용하여 유전적 안정성을 조사하였다. 재분화식물체는 MS (Murashige and Skoog, 1962)배지에 생장조절제를 첨가한 배지에서 배양하였다. 생장점 부위를 적출하여 캘러스를 유도하고(0.1 mg/L 2,4-D와 2 mg/L BA), 캘러스 증식(0.1 mg/L 2,4-D와 0.05 mg/L BA), 신초 재분화( 0.01 mg/L NAA와 2 mg/L BA) 후 MS배지에서 식물체를 양성하고 순화시켰다. 배양은 26±2℃, 25 µmol/m2/s, 14h/10h (day/night) 광조건 하에서 실시하였다. 재분화식물체는 홍띠 및 녹색 재분화식물체 2 종류로 나타났는데, 이는 생장점에서는 홍띠가 분화되었음에도 불구하고 생장점 주변조직에서 유래한 녹새체가 분화된 후 우세하게 자라서 녹색재생체가 우점하는 것으로 추정된다. ISSR 분석은 대조구로 모식물체 홍띠를(8개체), 재분화식물체는 녹색체 중, 1년간 노지포장에서 재배중인 녹색체(10개체)와 실험실내 화분에서 재배중인 시료를(10개체) 사용하였다. ISSR 밴드패턴을 비교한 결과, 재분화체는 실내포트 재배식물체 10.3%, 노지1년 재배식물체 8.3%로 대조구의 4.1%보다 유전적 다형성 비율이 2배 이상 높게 나타났다. 또한 재분화식물체들의 유전적 유사도를 평가하고 군집분석을 실시하였다.

Evaluation of Genetic Differentiation of Albizia lucida Populations from Eastern Region of the Indian Sub-continent by ISSR Markers

  • Level and distribution of genetic diversity in seven populations of Albizia lucida Benth. in eastern region of the Indian sub-continent were estimated using ISSR markers. Relatively higher level of genetic diversity within populations was observed in seven populations of A. lucida (mean of 0.38). From the result of AMOVA, majority of genetic diversity was allocated within populations (96.2%) resulting in a moderate degree of population differentiation. The observed distribution pattern of I-SSR variant among the populations was coincided with the typical pattern of long-lived woody tree species. Genetic relationships among the populations, reconstructed by UPGMA method, revealed two genetic groups. The population of Anugul and Bargarh turned out to be the most closely related despite a distance location between them. These formations will be of great value in the development of conservation plans for species exhibiting high levels of genetic differentiation due to fragmentation, such as indication of conservation unit size, which populations should be chosen as priority in conservation plans and which samples should be introduced in areas with a low number of individuals of A. lucida.

Diversity of Macrophomina phaseolina Based on Morphological and Genotypic Characteristics in Iran

  • Fifty two Macrophomina phaseolina isolates were recovered from 24 host plant species through the 14 Iranian provinces. All isolates were confirmed to species using species-specific primers. The colony characteristics of each isolate were recorded, including chlorate phenotype, relative growth rate at $30^{\circ}C$ and $37^{\circ}C$, average size of microsclerotia, and time to microsclerotia formation. The feathery colony phenotype was the most common (63.7%) on the chlorate selective medium and represented the chlorate sensitive phenotype of the Iranian Macrophomina phaseolina population. Meantime, inter simple sequence repeats (ISSR) Markers were used to assess the genetic diversity of the fungus. Unweighted pair-group method using arithmetic means (UPGMA) clustering of data showed that isolates did not clearly differentiate to the specific group according to the host or geographical origins, however, usually the isolates from the same host or the same geographic origin tend to group nearly. Our results did not show a correlation between the genetic diversity based on the ISSR and phenotypic characteristics. Similar to the M. phaseolina populations in the other countries, the Iranian isolates were highly diverse based on the phenotypic and the genotypic characteristics investigated and needs more studies using neutral molecular tools to get a deeper insight into this complex species.

Genetic Diversity and Relationship Analysis of Genus Taraxacum Accessions Collected in Korea

  • Genus Taraxacum has been widely used as a folkloric medicine for treatment of diverse diseases. The genetic diversity and relationship among 32 accessions belonging to five Taraxacum species (T. mongolicum T. coreanum, T. coreanum var. flavescens, T. officinale and T. laevigatum) which collected from field, mountain, island and seaside of Korea were evaluated using ISSR markers. A total of 142 ISSR loci detected in the overall species were all polymorphic loci (100%) and interspecies polymorphisms obtained from Korean native and naturalized species were 98.2% and 94.5%, respectively. The genetic similarity matrix (GSM) among 32 accessions ranged from 0.025 to 0.860 with an average of 0.303. According to the clustering analysis, the Korean native species and naturalized species were divided two major clusters. In addition, the different species were divided into independent groups except for the T. coreanum and T. coreanum var. flavescens, and all the 32 accessions could be classified into 7 categories. The study findings indicate that Taraxacum accessions have a high genetic diversity and the dandelion accessions as breeding materials can be effectively utilized for the improvement of Taraxacum breeding.

Variant Identification in Platanus occidentalis L. Using SNP and ISSR Markers

  • The purpose of this study was to identify the variant of Platanus occidentalis, whose bark looks white, also can be classified as P. occidentalis and to examine its genetic difference from the general P. occidentalis. For the variant identification of P. occidentalis, SNP and ISSR analysis were used in this study. Thirteen samples of P. occidentalis white variant were collected in Cheongju and 24 samples of normal P. occidentalis obtained in Cheongju, Pyongtaek, Ansan, Suwon, Osan and Jincheon area. ITS 1 and ITS 2 sequences of white variants were identical with those of P. occidentalis. We could not find any sequence difference between normal and white P. occidentalis. So we concluded that the white variant belongs to normal P. occidentalis even their bark is white and peeled easily. By ISSR test, 98 amplicons were acquired using 10 primers. P. occidentalis and white P. occidentalis showed different band patterns from the UBC #834. According to the result of Nei (1979)'s genetic distance analysis, the members of white P. occidentalis were grouped more tightly than the members of normal P. occidentalis. The UPGMA dendrogram shows that the variant and P. occidentalis divided widely into two groups. These results show that the phenotype of P. occidentalis white variant is caused by genetic factors rather than by environmental factors.
