The Vegetations of Hansan and Chubong Islets (한산도와 추봉도의 식생)

  • The vegetations of Hansan and Chubong islets in Tongyoungsi were investigated from April, 1997 to December, 1998 and a synthesis table was constructed. Based on the synthesis table, the vegetation was classfied into 6 communties: Pinus thunbergii community, Pinus densiflora community, Machilus thunbergii community, Chamaecyparis obtusa afforestation, Cryptomeria japonica afforestation, Miscanthus sinensis var. purpurascens community, and 3 subcommunities: Pinus thunbergii typical subcommunity, Quercus variabilis subcommunity, Carpinus coreana subcommunity. The vegetation of Pinus densiflora community around Jeseungdang area is well preserved. However the islet requirs the ecological restoration, and we suggest that the rare species such a Crypsinus hastatus, old trees of Machilus thunbergii and Pinus thunbergii, and the habitat for Machilus thunbergii and Camellia japonica must be preserved.

  • 꼬리명주나비(서ericinus montela) 조혈기관의 분포 및 미세구조를 광학현미경과 투과전자현미경을 이용하여 관찰하였다. 꼬리명주나비의 조혈기관은 흉부 제 2 · 3체절의 날개발생부위(imaginal wing disc)에 부착되었거나 인접해 있었다 각각의 조혈기관은 비세포성 기저 막으로 둘러싸인 여러 개의 조혈섬으로 구성되어 있었으며, 조혈 섬들은 compact islet(치밀하게 배열된 조혈섬)과 loose islet(느슨하게 배열된 조혈섬)으로 구분되었다. 혈구의 분화는 loose islet 내에서 진행되었으며, 시원세포로부터 원시혈구. 부정형혈구, 과립혈구, 소구혈구가 독자적 경로를 통해 분화되었다.

Viability and Functions of Alginate-microencapsulated Islets Isolated from Neonatal Pigs

  • Patients with Type I diabetes mellitus have been treated with porcine insulin for several decades and pigs have recently been deemed an ideal source of microencapsulated islet cells for clinical xenotransplantation. In this study, neonatal pigs were anesthetized and sacrificed prior to a pancreatectomy. Islet cells were isolated from pancreas via collagenase digestion. Islet cells were separated and collected by hand under microscopic guidance. These cells were suspended in 1.4% sodium alginate solution and encapsulated by dropping them into 1.1% calcium chloride solution and in which the round gel in size was 250-400 ${\mu}m$ in diameter. Viability of the microencapsulated islet cells cultured in medium at $37^{\circ}C$ was assessed by MTT assay. Furthermore, insulin released in response to glucose challenge was investigated using an enzyme-linked immunosorbent assay. Secretion of insulin was low in response to the basal glucose solution (4.4 mM) in medium and was significantly higher in response to the high glucose solution (16.7 mM). The viability of microencapsulated islet cells did not differ significantly over a period of 7 days; that is, the increasing pattern of insulin concentration in the culture medium after glucose stimulation interval day was similar throughout the 7 days cultivation. In summary, experimental evidences indicated that the effects of alginate-microencapsulation prolonged survival of the neonatal porcine islets in vitro cultures and the insulin response to glucose of the islets was maintained.

  • The Pancreatic islet are the clusters of endocrine cells scattered through out the exocrine pancreas. Transplantation of a sufficient pancreatic islets can normalize blood glucose level so that may prevent devastating complications of type I diabetes(IDDM) and other side effects of the IDDM. Recently, there are several approaches to transplant sufficient pancreatic islet, and it was comprised in increase or regeneration of the endogenous $\beta$-cell mass from donor's pancreas, but relatively few studies have been devoted to the morphological characters of the isolated and 3 day cultured pancreatic islets. We investigated morphological pattern of intracellular structure of isolated and 3 day cultured pancreatic islets. The morphological characters of the pancreatic islets were observed by scanning electron microscope and transmission electron microscope, and insulin distribution of the each islets were observed by transmission electron microscope, and were labeled with insulin antibody. Intracellular structures including nuclei, mitochondria, RER, Golgi complex and many secretory granules were normally appeared in the isolated pancreatic islets which was extracted immediately dornor's pancreas, however, There is a significant morphological changes between the 3 day cultured pancreatic islets and isolated islets. 3 day cultured pancreatic islet's $\beta$-cells had normal nuclei but increased cytoplasm mass and RER and developed Golgi complex. Insulin secretory granules were decreased in numbers rather than isolated pancreatic islet. In this study, the pattern of intracellular structure variation was examined during pancreatic islet culture. Most distinct features are variation of the insulin secretory granules, and developed RER, and dilated golgi complex. Therefore, we suggested that the various change of the morphological characters on cultured pancreatic islets were responsible for the function(biosynthesis and secretion of insulin) and growth. These results were also cultured islets have greater ability to recover and maintain normoglycemia than isolated islet transplantation.

Cytologic Findings of Pancreatic Islet Cell Tumor with Lymph Node Metastasis (림프절 전이를 동반한 췌소도세포종 1예의 세침흡인 생검소견)

  • Islet cell neoplasms (ICNs) of pancreas are uncommon, and the cytologic features of ICN are not well delineated. We report a case of islet cell tumor with lymph node metastasis, describing the cytologic, histologic, and immunohistochemical findings. A 40-year-old woman was admitted due to upper gastrointestinal bleeding of 2 days' duration. Computed tomography of the abdomen showed a diffusely infiltrating bulky mass in the body and tail of the pancreas. The fine needle aspirate showed moderate to high cellularity, monotonous cell population, single ceil predominance over small cell clusters, and eccentrically located nuclei. Although the definite diagnosis of ICN on the cytologic basis is difficult, the cytomorphologic features are sufficiently distinctive to suggest the diagnosis.

Effect of the Deep Sea Water on the Blood Glucose and the Langerhans' Islet in the STZ-induced type I Diabetic Mice (해양심층수 섭취가 STZ로 유발된 제 1형 당뇨 생쥐의 혈당치 및 췌도에 미치는 영향)

  • Effects of deep sea water on blood glucose and the Langerhans' islet in STZ-induced type I diabetic mice were investigated. Results showed that diabetic symptoms in STZ-induced diabetic mice improve with a supplement of deep sea water. That is, the level of blood glucose was lower, the area of the Langerhan' islet was wider, and the number of pancreatic $\beta$-cells was larger in the diabetic mice supplied with deep sea water than in the diabetic mice supplied with tap water. Such effects might be related to the high level of Mg$^{2+}$ in the deep sea water.

  • FasL, perforin, $TNF{\alpha}$, IL-1 and NO have been considered as effector molecule(s) leading to ${\beta}$-cell death in autoimmune diabetes. However, the real culprit(s) of ${\beta}$-cell destruction have long been elusive despite intense investigation. Previously we have suggested $IFN{\gamma}/TNF{\alpha}$ synergism as the final effector molecules in autoimmune diabetes of NOD mice. A combination of $IFN{\gamma}$ and $TNF{\alpha}$ but neither cytokine alone, induced classical caspase-dependent apoptosis in murine insulinoma and pancreatic islet cells. $IFN{\gamma}$ treatment conferred susceptibility to $TNF{\alpha}$-induced apoptosis on otherwise resistant murine insulinoma cells by STAT1 activation followed by IRF-1 induction. Here we report that $IFN{\gamma}/TNF{\alpha}$ synergism induces apoptosis of human pancreatic islet cells. We also observed STAT1 activation followed by IRF-1 induction by $IFN{\gamma}$ treatment in human islet cells. Taken together, we suggest that $IFN{\gamma}/TNF{\alpha}$ synergism could be involved in human islet cell death in type 1 diabetes, similar to murine type 1 diabetes.

The Effects of Several Herbal Medicine Concentrated Solution on the Hyperglycemic Mice Induced with Streptozotocin (수종의 한약재가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향)

  • Objective : This study has been carried out to understand the effect of several herbal medicine concentrated solution on the hyperglycemic mice Induced with streptozotocin(STZ). Methods : The 60 mg/kg of STZ injection into mice twice by 24 h interval and then 120 mg/kg of STZ injection again 3 days after the earlier injection. Control group was subjected to natural recovery, however, treated groups were fed 0.2 ml of several herbal medicine-concentrated solution (PA (x2, several herbal medicine-concentrated solution 1 group); PB (x4 several herbal medicine-concentrated solution 2 group) daily for 6 weeks. Result : The weight of PA was higher than that of control, but weight of PB was lower than control. The blood level of control increased continuously, reaching to 350mg/dL after 6 weeks, however, PA and PB showed a fast reduction of blood glucose. In blood glucose tolerance test, PA and PB showed better resistance than control. The GOT level in significantly(p<0.05) decreased in PA and PB compared with control group. The BUN and creatinine levels are significantly(p<0.01) decreased in PA compared with control group. Feeding of several herbal medicine-concentrated solution in a concentration of PA had an efficient effect on regeneration or recovery of Langerhans islet and ${\beta}-cell$ damaged by STZ. More Langerhans islet and high insulin-immunohistochemical resistance were observed in PA compared with control, but they were higher in PB than in PA. The number of Langerhans islet ${\beta}-cell$ and Langerhans islet. Conclusions : The result from the six weeks of observation demonstrates that the several herbal medicine concentrated solution have a positive effect of lowering the level of blood sugar and they increased insulin concentration. They have an effect for recovery of pancreas tissue and recovery of kidney, liver function from a diabetes mellitus.

Expression of c-Jun in pancreatic islet $\alpha$-cells of nonobese diabetic(NOD) mice

  • This is the first report of c-Jun protein expression and mRNA in a pancreatic islet in a nonobese diabetic(NOD) state mice. In this experiment NOD mice with insulin-dependent diabetes mellitus type I at age 16 weeks(n=7) just before death(n=4) were used. The control group consist of prediabetic NOD(8 weeks n=7) and ICR(8 weeks n=7 and 16 weeks n=7) mice. c-Jun positive cells in the pancreatic islet of NOD mice were localized in the same positions as a-glucagon producing cells. immunoreactivity was negative in the prediabetic NOD(8 weeks) and ICR(8 weeks and 16 weeks) mice. The number of c-Jun positive cells in mice with severe diabetic state just before death were significantly decreased when compared to NOD(16 weeks) mice. Expression of c-Jun in mRNA level was assessed by RT-PCR method. The levels of mRNA in NOD(16 weeks) mice group were elevated in total pancreatic tissues. The present results suggest that the induction of proto-oncogene protein may be of significance in assessing cell specific injury and may play a functional role between pancretic islet $\alpha$-cells and $\beta$-cells in the diabetic state.

  • The pancreatic endocrine cells of the cat-shark, S. torazame, were studied using immunohistochemical method. Five kinds of endocrine cells (glucagon-, somatostatin-, insulin-, 5-HT-and BPP-immunoreactive cells) identified in this study. The chracteristic findings of the distributions of five immunoreactive cells were as follows. Glucagon-immunoreactive cells were detected as clustering group in the epithelia of the interlobular duct and singly the pancreatic acini, respectively. Insulin -immunoreactive cells were moderately observed in the epithelia of the interlobular duct or in the periphery of the islet. Somatostatin-immunoreactive cells were distributed in single or mass groups in the epithelia of the interlobular duct and the exocrine gland of the pancreas. A very few 5-HTimmunoreactive cells were seen in the periphery of the islet and the acini of the pancreas-BPP-immunoreactive cell was singly located in the periphery of the pancreatic islet, but GAS/CCK-and Chromogranin-immunoreactive cells were not found in this study.
