Peculiar Fatigue Fracture Behavior of Ultrasonic Nanocrystal Surface Modified SCM435 (초음파 나노 결정 표면개질된 SCM435의 특이한 피로 파괴 특성)

  • Using the fatigue limit (∆σunsm) and residual stress (σr) of the UNSM smooth specimen, the harmless maximum crack depth (ahlm) according to the crack aspect ratio (As) was evaluated. In addition, the relationship between the minimum crack depth (aNDI1, aNDI2) detectable by non-destructive inspection(NDI), the crack depth (a25, a50) that reduces the fatigue limit by 25% and 50%, and ahlm were evaluated. The harmless crack condition was determined by the deepest crack point (point A). Since ahlm is larger than a25 and a50, a25 and a50 can secure the safety and reliability of steel via UNSM. Because aNDI1 and aNDI2 are larger than a25 and a50, cracks in a25 and a50 cannot be detected by non-destructive testing. Therefore it is necessary to apply more precise NDI.

Differential responses of peroxidases in sweetpotato suspension-cultured cells to cadmium treatment

  • As cultured plant cells can grow in high oxidative stress conditions, they form an excellent system to study antioxidant mechanisms and the mass production of antioxidants. Oxidative stress is a major cause of damage in plants exposed to various types of environmental stress, including heavy metals, such as cadmium (Cd). Heavy metal accumulation can interfere with many cell functions and plant growth. To evaluate the contribution of oxidative stress to Cd-induced toxicity, cultured sweetpotato (Ipomoea batatas) cells were treated with increasing concentrations of Cd (0, 10, 25, and 50 μM) and cultured further. Cell growth was significantly inhibited by 25 and 50 μM of Cd, and the total protein content increased with 50 μM of Cd. Additionally, the activity of peroxidase (POD) and ascorbate peroxidase (APX), antioxidant enzymes that remove hydrogen peroxide (a reactive oxygen species), increased in the cells after treatment with 50 μM of Cd. The expression analysis of POD, APX, and peroxiredoxin (PRX) isolated from sweetpotato cultured cells in a previous study revealed the differential expression of POD in response to Cd. In this study, the expression levels of several acidic POD (swpa2, swpa3, and swpa4) and basal POD (swpb1, swpb2, and swpb3) genes were increased in Cd-treated cultured cells. These results indicate that Cd-mediated oxidative stress is closely linked to improved POD-mediated antioxidant defense capacity in sweetpotato suspension-cultured cells.

  • Sulphur-hexafluoride has extensively been used as an insulating gas of electric power equipment in substation for its high dielectric strength, non-toixity and non-flammable properties. But it is very expensive and is liquidizable under the condition of low temperature and high pressure. And it is very sensible to the locally high electric field strength. To resolve these problems, we studied 50% FOV and V-t characteristics of $SF_6-N_2$ mixtures under positive and negative lightning impulses using a 25mm rod-plane gap. 50% FOV of $SF_6-N_2$ 50% mixture was about 85% of pure $SF_6$ and it was known that $SF_6-N_2$ mixtures can be used as an economic substitution insulating gas for $SF_6$.

Purification and Characterization of Xylanase II from Trichoderma koningii ATCC 26113 (Trichoderma koningii ATCC 26113으로부터 Xylanase II의 순수분리 및 특성)

  • A 1, 4-.betha.-D-xylanase, designated as xylanase II, was purified from the culture filtrate of Trichoderma koningii ATCC 251131 by column chromatography on Sephadex G-75, SP-Sephadex C-50, DEAE-Sephadex A-50 and Sephadex G-50 with an overall yield of 6.97%. It has a molecular weight of 21.000 and an isoelectric point of 9.4. The enzyme activity is optimal at pH 5.0 and at a temperature of 50.deg.C. Xylanase II is stable up to 50.deg.C, while 40 and 90% of its activity are lost after the incubation for 30 and 60 min at 60.deg.C. The enzyme degrades xylan with relatively high activity, as well as carboxymethylcellulose and Avicel. Its $K_{m}$ values for oat-spelt xylan, larchwood xylan and Avicel are 7.48, 1.98 and 13.33 mg/ml, respectively. The hydrolysis products of oat-spelt xylan by xylanase II are xylose, xylobiose, xylotriose and arabinoxylotriose, while the reaction products of larchwood xylan are xylose, xylobiose, xylotriose and small amount of higher oligomers. The action paterns of the enzyme demonstrate that xylanase II is endo-enzyme.

  • Trunk implantation method of phosphamidon 50% Lq.is commonly applied to control the pine needle gall midge (Thecodiplosis laponensts Uchida et Inouye) Since trunk implantaion is normally practiced during the late Spring, it is often difficult to accommodate necessay labor in mral area. As an alternative, aerial spraylng of less toxic ~nsecticide was designed. Usage of less toxic insecticide in the aenal control of pine needle gall midge can reduce the damage to forest ecosystem. The buproferin, one of the insect growth regulator, was selected at d~fferent rate of dilutions and the treatments effects ruere evaluated at different date and time When 50 t~mes diluted solution of buprofezin 40% SC was treated on different period, there was no significant difference in dficacies. the control efficacies of buproferin 40% SC was measured by occurrences (%) of gall formation of the pine needle gall midge using a ultra low volume (ULV) sprayer and the dilutions of 10X 30X and 50X of buprofez~n 40% SC gave efficacies. 72.4. 57.6 and 8.4, respectively.

A Study on the Airtightness Performance of New Han-ok Bedrooms (신한옥 침실 공간의 기밀성능 평가 연구)

  • The purpose of this study is to evaluate the airtightness performance of New Han-ok and to supply fundamental data for standards establishment. Air leakage testings were accomplished by means of blower door test in 26 bedrooms of 16 Han-oks located in Jeonnam happy villages. Followings are results. 1) Air change per hour at 50 Pa(ACH50) is located on 8.42~78.38. 2) No correlation between ACH50 and volumes, floor area, above grade surface area. 3) The more wood structural elements are exposed, attached spaces, wooden sliding and casement windows, the less airtightness performance. 4) An Airtightness with ACH50/20(NL, Normalized leakage) is located on 0.42~3.92 and building leakage class following F(4%), G(11%, sufficiently leaky, No need mechanical ventilation), H(4%, Need of cost-effective tightening), I(31%), J(50%) by a single-story house the normalized leakage of ASHRAE.

Design Verification of Cabin Pressurization System by Flight Test of T-50 Advanced Trainer (T-50 비행시험을 통한 조종실 여압시스템의 설계검증)

  • The cabin pressurization system response should be consistent with the design limits such as the cabin pressure schedule, the pressure regulation tolerance, the maximum rate of pressure change during normal and abnormal operation and the maximum cabin air inflow rate change. In this paper, the results of pressure loss analysis and flight test for cabin pressurization system of T-50 advanced trainer are introduced. The pressure tolerance at unpressurized condition using calculated exit area of pressurization components through pressure loss analysis is predicted. Pressurization components of D company are selected and the predicted pressure tolerance is in good agreement with flight test results. Finally, T-50 pressurization system is verified by some flight tests of T-50 advanced trainer to comply with various pressurization design criteria of MIL-E-18927.

Design and Fabrication of X-Band 50 W Pulsed SSPA Using Pulse Modulation and Power Supply Switching Method (펄스 변조 및 전원 스위칭 방법을 혼용한 X-대역 50 W Pulsed SSPA 설계 및 제작)

  • In this paper, a X-band 50 W pulsed solid state power amplifier(SSPA) is designed and fabricated for radar systems. The SSPA consists of a driver amplifier, a high power amplifier, and a pulse modulator. The high power stage employes four 25 W GaAs FET to deliver 50 W at X-band. To meet the stringent target specification for the SSPA, we used a new hybrid pulse switching method, which combine the advantage of pulse modulation and bias switching method. The fabricated SSPA shows a power gain of 44.2 dB, an output power of 50 W over a 1.12 GHz bandwidth. Also, pulse droop < 1 dB meet the design goals and a rise/fall time is less than 12.45 ns. Fabricated X-band pulsed SSPA size is compact with overall size of $150{\times}105{\times}30\;mm^3$.

The CsSTE50 Adaptor Protein in Mitogen-Activated Protein Kinase Cascades Is Essential for Pepper Anthracnose Disease of Colletotrichum scovillei

  • Anthracnose, caused by the ascomycete fungus Colletotrichum scovillei, is a destructive disease in pepper. The fungus germinates and develops an infection structure called an appressorium on the plant surface. Several signaling cascades, including cAMP-mediated signaling and mitogen-activated protein kinase (MAPK) cascades, are involved in fungal development and pathogenicity in plant pathogenic fungi, but this has not been well studied in the fruit-infecting fungus C. scovillei. Ste50 is an adaptor protein interacting with multiple upstream components to activate the MAPK cascades. Here, we characterized the CsSTE50 gene of C. scovillei, a homolog of Magnaporthe oryzae MST50 that functions in MAPK cascades, by gene knockout. The knockout mutant ΔCsste50 had pleiotropic phenotypes in development and pathogenicity. Compared with the wild-type, the mutants grew faster and produced more conidia on regular agar but were more sensitive to osmotic stress. On artificial and plant surfaces, the conidia of the mutant showed significantly reduced germination and failed to form appressoria. The mutant was completely non-pathogenic on pepper fruits with or without wounds, indicating that pre-penetration and invasive growth were both defective in the mutant. Our results show that the adaptor protein CsSTE50 plays a role in vegetative growth, conidiation, germination, appressorium formation, and pathogenicity in C. scovillei.

An Analysis of Effects on Job Seeking Activities of Unemployed in Fifties in Korea: Based on The Resilience (한국 50대 실업자의 구직활동 영향요인에 관한 분석: 회복탄력성을 중심으로)

  • The purpose of this study is to improve the reemployment rate of unemployed job seekers aged 50 to 59 by supporting the foundation of our society and enhancing their value of life. We analyzed the impact of resilience, a psychological factor, on job seeking of unemployed people in their fifties. Subjects were visited and provided employment services in the Employment Welfare Plus Center in the Seoul Metropolitan Area. We devised a questionnaire consisting of three variables of general characteristics, resilience, and job search. Also, the resilience variables comprised sub-variables of control, positiveness, and sociality, and the job search variables involved sub-variables of job preparation, attitude, and willness. Study results the resilience of unemployed persons aged 50 to 59 is above average, and sub-variables of positiveness are highest compared to other sub-variables. Second, there is no significant gender difference in resilience for job search activities. Third, resilience and its sub-variables, such as control and affirmation, affect job-seeking activities. Therefore, this study suggests strengthening the resilience of job seekers in their fifties as well as providing psychological counseling and job training for job-seeking activities.
