Sequence Analysis of a Cryptic Plasmid pKW2124 from Weissella cibaria KLC140 and Construction of a Surface Display Vector

  • Plasmid isolation of kimchi-derived Weissella cibaria KLC140 revealed six different plasmids. The smallest plasmid, pKW2124, was DNA sequenced and characterized, showing 2,126 bp with a GC content of 36.39% and five putative open reading frames (ORFs). In silico analysis of these ORFs showed ORF1 encodes a putative replication protein similar to rolling circular replication proteins from other lactic acid bacteria. However, a single-stranded intermediate was not detected when S1 nuclease was treated, suggesting it may follow theta replication. Interestingly, the replication initiation site of this plasmid is 100% identical to other plasmids from lactic acid bacteria, suggesting it may function for replication initiation. To construct a surface layer expression vector, pTSLGFP, slpA encoding the surface layer protein from Lactobacillus acidophilus was PCR amplified and fused with the gfp gene, forming a SLGFP fused gene. The plasmid pKW2124 was cloned into the XbaI site of pUC19, forming an Weissella-E. coli shuttle vector pKUW22. NheI-linearized pTSLGFP was ligated into pKUWCAT containing pKUW22 and the chloramphenicol acetyltransferase gene from pEK104, resulting in an 8.6 kb pKWCSLGFP surface layer expression vector. After transformation of this vector into W. cibaria KLC140, a GFP fluorescence signal was detected on the surface of the transformant, substantiating production of SLGFP fused protein and its secretion. This is the first report for construction of a Weissella surface layer expression vector, which may be useful for surface layer production of beneficial proteins in Weissella.

Brucella abortus 국내 분리주의 Heat Shock Protein 암호 groE 유전자의 염기서열 분석과 발현 (Sequence analysis and expression of groE gene encoding heat shock proteins of Brucella abortus isolates)

  • GroE that is a heat shock protein composed of GroEL and GroES is known as an immunodominant target of both the humoral and cellular immune responses in bovine brucellosis. This study was carried out to characterize groE gene encoding heat shock proteins of B. abortus isolated in Korea and to evaluate the immunogenicity of the GroE protein expressed in E. coli system. In PCR the specific signals with the size of 2,077 bp were detected in five strains isolated from the mammary lymphnodes of the dairy cattle that were serologically positive and the reference strains. In comparison of the sequences of nucleotides and amino acids among the strains, GroES showed 100% identity in both sequences. GroEL was evaluated 99.0~99.9% in nucleotides and 98.0~100% homology in amino acids. The groE gene including groES and groEL was inserted into pET29a vector and constructed pET29a-GroE recombinant plasmids. The inserted groE was confirmed by digestion with Nco1 and EcoR1 endonucleases and nucleotide sequencing. E. coli BL (DE3) was transformed with pET29a-GroE, named as E. coli BL (DE3)/pET29a-GroE. In SDS-PAGE, it was evident that the recombinant plasmid effectively expressed the polypeptides for GroES (10 kDa) and GroEL (60 kDa) in 0.5, 1 and 2 hours after IPTG induction. The immuno-reactivity of the expressed proteins were proved in mouse inoculation and Western blot analysis.

발육중인 생쥐 하악 과두에서 연골 및 골의 특이 유전자 발현 (Expression of mRNAs characteristic of cartilage and bone in the developing mandibular condyle of mice)

  • 하악과두 연골이 발생하고 분화하는 과정에서 나타내는 특성을 규명하기 위하여, 발생 16, 18일과 출생 1일, 5일, 10일, 20일 및 30일 후의 ICR생쥐의 하악과두를 형태학적으로 분석하고, 생쥐 I형, II형, X형 교원질, Indian hedgehog (IHH) 및 BMP-4 등의 mRNA 발현을 in-situ hybridization 방법으로 연구하였다. 1. 생쥐 I형 및 II형 교원질 mRNA는 하악과두의 발생 및 성장과정에서 모두 발현되었다. I형 교원질 mRNA는 휴지층과 증식층의 상부에서 관찰된 반면 II형 교원질은 휴지층과 증식층 그리고 비대연골층의 상부에서 관찰되었다. 2. 하악과두 연골은 성장에 따라 비대연골층이 계속 증가하는 소견을 보였으며, 비대 연골층의 세포들은 특징적으로 X형 교원질 mRNA의 발현을 보였다. 3. BMP-4 mRNA는 하악과두 연골 원기와 골화중인 하악골체에서 모두 발현되었다. 4. IHH mRNA는 하악과두의 발생과정에서 증식 연골층의 하부와 비대연골층의 상부에서 선택적으로 관찰되었다.

가시오갈피 물 추출물이 간세포에서 포도당 이용 대사에 미치는 영향 (Effects of Acanthopanax senticosus Water Extract on Glucose-Regulating Mechanisms in HepG2 Cells)

  • 본 연구에서는 가시오갈피 물 추출물(ASW)를 이용하여 아직 시도된 바가 없는 HepG2 세포 내 포도당 유입과정 및 glucokinase(GK) 활성을 통한 포도당 이용대사 실험을 수행하였다. 포도당의 세포 내 유입은 GLUT2의 transcription factor들 중 하나인 $HNF-1{\alpha}$의 활성화로 GLUT2의 유전자 발현이 증가하여 이루어지는 것을 확인하였다. GK 활성 측정 결과 ASW가 GK를 활성화하여 포도당의 인산화에 영향을 주는 것을 확인하였고 AMP-activated protein kinase의 인산화 증가로 glycolysis에 관여하는 효소인 GK의 단백질 발현은 증가하고, gluconeogenesis에 관여하는 phosphoenolpyruvate carboxykinase의 단백질 발현은 감소하는 것을 확인하였다. 그리고 인산화된 포도당이 glycogen으로 전환 저장되는 메커니즘을 pPI3k-pAkt-pGSK-$3{\beta}$의 단계별 단백질 발현을 확인함으로써 검증하였으며, glycogen 함량 측정을 통해 확인하였다. 본 연구를 통해 ASW가 다양한 메커니즘에 작용하여 당뇨의 예방 및 개선에 활용할 수 있는 잠재적 소재임을 확인하였고, 이는 ASW가 천연 기능성 소재로서의 개발가치가 높음을 시사한다.

결핵균 항원 자극에 의한 결핵성 흉수 림프구의 IFN-${\gamma}$ mRNA 발현 (IFN-${\gamma}$mRNA Expression in Tuberculous Pleural Lymphocytes After in vitro Stimulation with M. tuberculosis Antigens)

  • 연구배경 : IFN-${\gamma}$는 결핵균에 대한 숙주의 면역학적 방어기전에서 핵심적인 역할을 한다. 그러므로 결핵균 항원들이 IFN-${\gamma}$ 유전자 발현에 미치는 영향을 알아보는 것은 결핵균에 대한 숙주의 방어기전을 밝히고 이를 이용한 백신의 개발에 이용될 수 있을 것이다. 방 법 : 결핵성 흉막염 환자의 흉수에서 얻은 림프구 배양액에 결핵균(H37Rv), PPD, Ag85B, man-LAM, ara-LAM을 첨가하여 자극한 후 림프구의 IFN-${\gamma}$ mRNA의 발현 정도를 역전사 중합효소연쇄반응을 이용하여 비교하였다. 결 과 : 1) 결핵균(H37Rv)이 결핵성 흉수 림프구의 IFN-${\gamma}$ mRNA의 발현을 증가시켰다. 2) 결핵균 항원 중 PPD와 Ag85B는 결핵성 흉수 림프구의 IFN-${\gamma}$ mRNA의 발현을 증가시켰지만 man-LAM은 결핵성 흉수 림프구의 IFN-${\gamma}$ mRNA의 발현을 억제시켰다. 3) LAM 중에서 man-LAM은 용량이 증가함에 따라 결핵성 흉수 림프구의 IFN-${\gamma}$ mRNA의 발현의 억제 정도가 증가하였지만 ara-LAM의 경우 이와 같은 현상이 관찰되지 않았다. 결 론 : 결핵성 흉수 림프구의IFN-${\gamma}$ mRNA의 발현은 PPD와 Ag85B의 자극에 의해 항진되지만 man-LAM의 자극에 의해서는 억제되었다.

Expression of SDF-$1{\alpha}$ and leptin, and their effect on expression of angiogenic factors in mouse ovaries

  • Objective: Ovarian angiogenesis plays an important role in folliculogenesis. However, little is known about the expression of angiogenic factors during follicular development according to female age. Stromal cell derived factor-$1{\alpha}$ (SDF-$1{\alpha}$) plays a role in granulosa cell survival and embryo quality as an angiogenic chemokine. Leptin is also involved in folliculogenesis and angiogenesis. This study examined expression of SDF-$1{\alpha}$ and leptin, and their effects on the expression of angiogenic factors in the ovary during follicular development according to female age. Methods: Ovaries were collected from C57BL mice of two age groups (6-9 weeks and 24-26 weeks) at 6, 12, 24, and 48 hours after 5 IU pregnant mare's serum gonadotropin (PMSG) injection. The expression of ovarian SDF-$1{\alpha}$ and leptin mRNA was evaluated by RT-PCR. In the organ culture experiment, the ovaries were cultured in transwell permeable supports with Waymouth's medium treated with various doses of SDF-$1{\alpha}$(50-200 ng/mL) or leptin (0.01-1 ${\mu}g$/mL) for 7 days. Then, mRNA expression of vascular endothelial growth factor (VEGF), endothelial nitric oxide synthase (eNOS), and visfatin were examined in the cultured ovaries. Results: Expression of SDF-$1{\alpha}$ and leptin in the ovary was significantly lower in the aged mouse group compared to the young mouse group ($p$ <0.05). Expression of these two factors increased with follicular development after PMSG administration. SDF-$1{\alpha}$ treatment stimulated visfatin expression in a dose-dependent manner, while leptin treatment significantly increased eNOS expression. Conclusion: These results suggest that decrease of ovarian SDF-$1{\alpha}$ and leptin expression may be associated with aging-related reduction of ovarian function. SDF-$1{\alpha}$ and leptin may play a role in follicular development by regulating the expression of angiogenic factors in mouse ovaries.

Diode laser surgery in the treatment of oral proliferative verrucous leukoplakia associated with HPV-16 infection

  • Background: Proliferative verrucous leukoplakia (PVL) is an oral potentially malignant disorder, characterized by multifocal expression, progressive clinical evolution, and a high rate of malignant transformation. Evidence-based information regarding optimal PVL management is lacking, due to the paucity of data. The present report describes a case of PVL associated with HPV-16 infection and epithelial dysplasia treated by diode laser surgery, and the outcome of disease clinical remission over a 2-year follow-up period. Case report: A 61-year-old Caucasian male with oral verrucous hyperkeratosis presented for diagnosis. The lesions were localized on the maxillary gingiva and palatal alveolar ridge. Multiple biopsy specimens have been taken by mapping the keratotic lesion area. Microscopic examination was compatible with a diagnosis of PVL with focal mild dysplasia, localized in the right maxillary gingiva. Polymerase chain reaction (PCR) was done for human papillomavirus (HPV) detection which revealed presence of HPV DNA, and the genotype revealed HPV 16 in the sample. The PVL in the right gingival area was treated on an outpatient basis by excision with a diode laser. This approach resulted in good clinical response and decreased morbidity over a 2-year follow-up period. Conclusions: This case illustrates the benefit of a conservative approach by diode laser treatment than wide surgical excision for management of the PVL lesions associated with mild dysplasia and HPV-16 infection.

  • Purpose : Cyclosporine A (CsA) is a versatile immunosuppresive agent used to prevent graft rejection syndrome and treat autoimmune disease. One of the major side effects associated with CsA is the abnormal gingival hyperplasia. The purpose of this study was to investigate the relationship between the mRNA expression of the MMP-1, TIMP-1, and $TGF-{\beta}_1$ and the concentration of CsA in cultured human gingival keratinocytes. Materials & Methods : Gingival keratocytes were obtained from gingival tissues of 4 healthy donors. The cultured gingival keratocytes were incubated with increasing concentrations of CsA (0-2000 ng/ml) for 24 hours and the expression of MMP-1, TIMP-1, and $TGF-{\beta}_1$ were determined by reverse transcription-polymerase chain reaction (RT-PCR). Results : The expressions of MMP-1 and $TGF-{\beta}_1$ were not significantly different according to the concentrations of CsA. The expression of TIMP-1 was significantly increased at the CsA concentration of 500 ng/ml. Conclusion : We concluded that the gingival hyperplasia induced by CsA was more related with TIMP-1 than MMP-1 or $TGF-{\beta}_1$ on gingival collagen metabolism in patients treated with CsA.

  • 연구배경 : 종양괴사인자(tumor necrosis factor ; TNF)는 다양한 생물학적 기능을 가지고 있는 바, 그 중 생체 외에서 증명된 뚜렷한 항암 효과로 말미암아 최근 항암 유전자요법의 중요한 대상으로 관심을 모으고 있다. 현재 유전자 이입의 기술적 문제로 생체 외에서 암세포에 유전자 이입을 시행한 후 이를 다시 환자의 생체내로 이식하는 방법이 연구의 주종을 이루고 있다. 그러나 저자들의 과거의 연구를 포함한 여러 연구에서 TNF가 이입된 암세포는 TNF에 대해 내성을 보이는 것으로 증명되었고 이에는 새로이 방어 단백질을 합성하는 것이 관여할 것이라는 시사가 있었다. 이 획득내성의 기전을 밝히는 것이 종양생물학의 이해를 넓히고 보다 효과적인 항암 유전자요법을 개발하기위한 매우 중요한 과제로 생각된다.

내탁천금산(內托千金散)이 DNCB로 유발된 알레르기성 접촉피부염에 미치는 영향 (The effects of Naetakchunkeum-san on the Allergic Contact Dermatitis induced by DNCB)

  • Background and Objectives : Allergic Contact Dermatitis is the disease affected by industrialization. The more industrialization advanced, the more materials that could induce the allergic contact dermatitis have been increased. Therefore in oriental medicine, various studies have been performed. The objective of this study is to investigate the effects of Naetakchunkeum-san on the Allergic Contact Dermatitis induced by 2,4-dinitro-chlorobezene(DNCB). Meterial and Methods : Twenty eight mice were divided into four groups ; normal, control, experimental group A and B. Control and experimental group were induced allergic contact dermatitis by DNCB. Experimental group A was orally administered the Naetakchunkeum-san and experimental group B was orally administered the prednisolone. In this study, ear thickness measurement, observation auricle microphotograph, Myeloperoxidase(MPO) activity measurement, Reverse transcription-polymerase chain reaction(RT-PCR) analysis of the mRNA level of $TNF-\alpha$, $IL-1\beta$, $INF-\gamma$ were performed on these four groups. In addition, the effect of Naetakchunkeum-san on cell viability and the effect of Naetakchunkeum-san on the compound 48/80-induced histamine release from HMC and RPMC were measured, Results : 1. In contact hypersensitivity assay, experimental group A and B showed decreased ear thickness compared with control group, 2. In experimental group A, pathological lesion of dermatitis were alleviated. In addition, the numbers of infiltrated cells were reduced, and cleft was not shown compared with control group, In experimental group B, similar results were shown. 3. There was a significant increase in MPO activity in control group compared with normal group, Experimental group A and B significantly inhibited the increase in MPO activity compared with control group. 4, The level of expression of $TNF-\alpha$, $IL-1\beta$, $INF-\gamma$ in experimental group A and B were significantly lower than those in control group. As the internal control, cyclophilin mRNA was also reverse-transcribed and amplified. 5, In MTT assay, there were no statistically significant differences in 100 ${\mu}g/ml$, 200 ${\mu}g/ml$, 500 ${\mu}g/ml$, 1000 ${\mu}g/ml$ Naetakchunkeum-san treated group from 0 ${\mu}g/ml$ Naetakchunkeum-san treated group as determined by the Tukey test. 6. Naetakchunkeum-san dose-dependently inhibited the compound 48/80-induced histamine release from both HMC and RPMC. Conclusions : According to above experiments, Naetakchunkeum-san may be applied to allergic contact dermatitis.
