Background: The insulin receptor substrate-1 (IRS-1) is the primary docking molecule for the insulin-like growth factor I receptor (IGF-IR), and is required for activation of the phosphatidylinositol 3'-kinase (PI3K) pathway. IRS-1 activation of the (PI3K) pathway regulates IGF-mediated survival, enhancement of cellular motility and apoptosis. Therefore, we attempted to ascertain whether IRS-1 genetic variations affect an individual's risk for non-small cell lung cancer (NSCLC). Methods: Two-hundred and eighteen subjects, either diagnosed with NSCLC or control subjects, were matched by age, gender and smoking status. Genomic DNA from each subject was amplified by PCR and analyzed according to the restriction fragment length polymorphism (RFLP) profile to detect the IRS-1 G972R polymorphism. Results: The frequencies of each polymorphic variation, in the control population, were as follows: GG=103 (94.5%) and GR=6 (5.5%); for the NSCLC subjects, the genotypic frequencies were as follows: GG=106 (97.2%) and GR=3 (2.8%). We could not demonstrate statistically significant differences in the genotypic distribution between the NSCLC and the control subjects (p=0.499, Fisher's Exact test). The relative risk of NSCLC, associated with the IRS-1 G972R polymorphic variation, was 1.028 (95% CI; 0.63~9.90). In addition, we found no differences between polymorphic variants with regard to the histological subtype of NSCLC. Conclusion: We did not observe any noteworthy differences in the frequency of the IRS-1 G972R polymorphism in NSCLC patients, compared to control subjects. These results suggest suggesting that, in our study population, the IRS-1 G972R polymorphism does may not appear to be associated with an increased risk of NSCLC.
BACKGROUND/OBJECTIVES: The anti-diabetic activity of pear through inhibition of ${\alpha}-glucosidase$ has been demonstrated. However, little has been reported about the effect of pear on insulin signaling pathway in obesity. The aims of this study are to establish pear pomace 50% ethanol extract (PPE)-induced improvement of insulin sensitivity and characterize its action mechanism in 3T3-L1 cells and high-fat diet (HFD)-fed C57BL/6 mice. MATERIALS/METHODS: Lipid accumulation, monocyte chemoattractant protein-1 (MCP-1) secretion and glucose uptake were measure in 3T3-L1 cells. Mice were fed HFD (60% kcal from fat) and orally ingested PPE once daily for 8 weeks and body weight, homeostasis model assessment of insulin resistance (HOMA-IR), and serum lipids were measured. The expression of proteins involved in insulin signaling pathway was evaluated by western blot assay in 3T3-L1 cells and adipose tissue of mice. RESULTS: In 3T3-L1 cells, without affecting cell viability and lipid accumulation, PPE inhibited MCP-1 secretion, improved glucose uptake, and increased protein expression of phosphorylated insulin receptor substrate 1 [p-IRS-1, ($Tyr^{632})$)], p-Akt, and glucose transporter type 4 (GLUT4). Additionally, in HFD-fed mice, PPE reduced body weight, HOMA-IR, and serum lipids including triglyceride and LDL-cholesterol. Furthermore, in adipose tissue, PPE up-regulated GLUT4 expression and expression ratio of p-IRS-1 ($Tyr^{632})/IRS$, whereas, down-regulated p-IRS-1 ($Ser^{307})/IRS$. CONCLUSIONS: Our results collectively show that PPE improves glucose uptake in 3T3-L1 cells and insulin sensitivity in mice fed a HFD through stimulation of the insulin signaling pathway. Furthermore, PPE-induced improvement of insulin sensitivity was not accompanied with lipid accumulation.
The insulin-mediated Ras/mitogen-activated protein (MAP) kinase cascade was examined in SK-N-BE(2) and PC12 cells, which can and cannot produce reactive oxygen species (ROS), respectively. Tyrosine phosphorylation of the insulin receptor and insulin receptor substrate 1 (IRS-1) was much lower in SK-N-BE(2) cells than in PC12 cells when the cells were treated with insulin. The insulin-mediated interaction of IRS-1 with Grb2 was observed in PC12 but not in SK-N-BE(2) cells. Moreover, the activity of extracellular-signal-related kinase (ERK) was much lower in SK-N-BE(2) than in PC12 cells when the cells were treated with insulin. Application of exogenous $H_2O_2$ caused increased tyrosine phosphorylation and Grb2 binding to IRS-1 in SK-N-BE(2) cells, while exposure to an $H_2O_2$ scavenger (N-acetylcysteine) or to a phophatidylinositol-3 kinase inhibitor (wortmannin), and expression of a dominant negative Rac1, decreased the activation of ERK in insulin-stimulated PC12 cells. These results indicate that the transient increase of ROS is needed to activate ERK in insulin-mediated signaling and that an inability to generate ROS is the reason for the insulin insensitivity of SK-N-BE(2) cells.
IRS-1C PAN의 수치화상데이터를 사용하여 수치표고모델을 생성하는 기법을 연구하였다. 번들조정기법을 적용하여 라인별 외부표정요소를 결정하였으며, 그 결과 위성의 자세와 위치 모두에 있어서 1차다항식이 가장 유효하였다. 수치표고모델과 정사투영화상을 생성한 후, 연구 대상영역내의 상이한 표고를 가지고 있는 16점의 평가점을 선정하여, 연구결과의 지도좌표와 비교해본 결과, 표고오차의 RMSE가 $\pm{16.66m}$인 수치표고 모델을 생성할 수 있었다.
In this study, occurrence of canine brucellosis was surveyed in kennels, indoor dogs and stray dogs in Korea, and infrequent restriction site-polymerase chain reaction (IRS-PCR) was applied to analyze DNA polymorphism of Brucella canis (B. canis) isolates. Among a total of 501 dogs tested, B. canis antibodies by both rapid screening agglutination with 2-mercaptoethanol (2-ME RSAT) and immunochromatographic assay were detected in only 14.1% of kennel dogs. There were no seropositive cases in indoor dogs and stray dogs. DNA polymorphism was observed in 16 B. canis isolates by the IRS-PCR. Sixteen isolates were tested with primers, PsalA, PsalC, PsalG and PsalT, and different primers produced different DNA patterns. In regard to the IRS-PCR pattern of 16 isolates, 9 (56.3%) belonged to the IRS-PCR type I. The remaining 7 were differentiated as type II, III and IV. An application of the primer PsalC provided discrimination between B. canis isolated in 2005 and others.
벼 기계이앙을 위한 어린모 육묘에 이앙 가능한 최단기 육묘일수와 맷트형성기간을 구명하기 위하여 육묘온도와 깔판재료를 달리하여 그 효과를 조사하였던 바 다음과 같은 결과를 얻었다. 1. 육모온도가 높을수록, 또 육묘기간이 길어질수록 어린모의 맷트형성이 양호하였으며, Metalaxyl(25% 수화제, 1,000 배액) 종자침종처리는 무처리에 비하여 맷트형성 기간이 단축되었다. 2. Metalaxyl 침종처리는 무처리에 비하여 어린모의 초장신장 효과가 있었고, 경엽중과 근중도 무거웠으며, 입고병과 뜸묘의 발생을 방지하였다. 3. 어린모의 맷트형성 촉진 보조재로서 흡수지깔판(키친타올 1컵)의 효과가 인정되었으며, 'Metalaxyl 침종+비닐+흡수지'처리는 다른 처리에 비하여 맷트형성 기간을 1~2일 단축시켰다. 4. 어린모의 최소 육묘일수는 파종 후 5일이였다. 육묘조건은 종자를 Metalaxyl 1,000배액에 24시간 침종한 후 비닐+흡수지 깔판에 파종하여 30/2$0^{\circ}C$(획 / 야) 온도에서 육묘한다. 5. 일반농가 비닐하우스에서 어린모 육묘시(중부지방)에는 5월의 하우스내 온도를 야간 11~16$^{\circ}C$, 획간 28~32$^{\circ}C$ 정도로 볼 때, 산파상자를 사용할 경우에는 'Metalaxyl 침종+비닐깔판'처리로, 또 어린모 전용 육묘상자를 사용할 경우에는 'Metalaxyl 침종'처리로 육묘하면 파종 후 6일에 안전 이앙이 가능한 것으로 판단된다.

We propose short packet communication in an underlay cognitive radio network assisted by an intelligent reflecting surface (IRS) composed of multiple reconfigurable reflectors. This scheme, called the IRS protocol, operates in only one time slot (TS) using the IRS. The IRS adjusts its phases to give zero received cumulative phase at the secondary destination, thereby enhancing the end-to-end signal-to-noise ratio. The transmitting power of the secondary source is optimized to simultaneously satisfy the multi-interference constraints, hardware limitations, and performance improvement. Simulation and analysis results of the average block error rates (BLERs) show that the performance can be enhanced by installing more reconfigurable reflectors, increasing the blocklength, lowering the number of required primary receivers, or sending fewer information bits. Moreover, the proposed IRS protocol always outperforms underlay relaying protocols using two TSs for data transmission, and achieves the best average BLER at identical transmission distances between the secondary source and secondary destination. The theoretical analyses are confirmed by Monte Carlo simulations.

As a part of the "Dust, Ice, and Gas In Time" (DIGIT) key program on Herschel, we observed GSS30-IRS1, a Class I protostar located in Ophiuchus (d =125 pc), with Herschel/Photodetector Array Camera and Spectrometer (PACS). More than 70 lines were detected within a wavelength range from 50 ${\mu}m$ to 200 ${\mu}m$: CO lines from J = 14-13 to 41-40, several $H_2O$ lines of Eup = 100 K to 1500 K, 16 transitions of OH rotational lines, and two atomic [O I] lines at 63 and 145 ${\mu}m$. The [C II] line, known as a tracer of externally heated gas by the interstellar radiation field, is also detected at 158 ${\mu}m$. All lines, except [O I] and [C II], are detected only at the central spaxel of $9^{\prime\prime}.4{\times}9^{\prime\prime}.4$. The [O I] emission is extended along a NE-SW orientation, which is consistent with the known outflow direction, while the [C II] line is detected over all spaxels. One possible explanation of the detection of the [C II] line and no correlation of its spatial distribution with any other molecular emission is the existence of the enhanced ISRF nearby GSS30-IRS1. One interesting feature of GSS30-IRS1 is that the continuum emission is extended beyond the point-spread function (PSF), unlike the molecular line emission, indicative of significant external heating. The best-fit continuum model of GSS30-IRS1 with the physical structure including flared disk, envelope, and outflow shows that the internal luminosity is 11 $L_{\odot}$, and the region is also externally heated by a radiation field enhanced by a factor of 25 compared to the local standard interstellar field.

Phosphoserine phosphatase (PSPH) is one of the key enzymes of the L-serine synthesis pathway. PSPH is reported to affect the progression and survival of several cancers in an L-serine synthesis-independent manner, but the mechanism remains elusive. We demonstrate that PSPH promotes lung cancer progression through a noncanonical L-serine-independent pathway. PSPH was significantly associated with the prognosis of lung cancer patients and regulated the invasion and colony formation of lung cancer cells. Interestingly, L-serine had no effect on the altered invasion and colony formation by PSPH. Upon measuring the phosphatase activity of PSPH on a serine-phosphorylated peptide, we found that PSPH dephosphorylated phospho-serine in peptide sequences. To identify the target proteins of PSPH, we analyzed the protein phosphorylation profile and the PSPH-interacting protein profile using proteomic analyses and found one putative target protein, IRS-1. Immunoprecipitation and immunoblot assays validated a specific interaction between PSPH and IRS-1 and the dephosphorylation of phospho-IRS-1 by PSPH in lung cancer cells. We suggest that the specific interaction and dephosphorylation activity of PSPH have novel therapeutic potential for lung cancer treatment, while the metabolic activity of PSPH, as a therapeutic target, is controversial.

Ocean Color Monitor (OCM) onboard the Indian Remote Sensing Satellite IRS-P4 has been used to retrieve chlorophyll concentration in the Bay of Bengal and the Arabian Sea using a bio-optical algorithm. Cloud masking and atmospheric corrections have been performed before applying mapping function to derive chlorophyll concentration from IRS-P4 OCM data. We have retrieved chlorophyll concentration from OCM, and MODIS during the summer and winter season along the eastern and western coast of India at every 1 degree latitude at increasing distance (25, 50, 100, 150 and 200km) away from the coast as well as near river mouths for the period 2000-2003. We have also studied spatial and temporal dynamics of monthly MODIS Aqua (for period July 2002-April 2004). The seasonal dynamics of chlorophyll concentration over the Bay of Bengal and the Arabian Sea have been discussed using OCM and MODIS for both the coastal region and the open sea.
