The Roles of IPP Professor for Successful Operations of IPP System : Based on KOREATECH Case Study (장기현장실습 제도의 성공적인 운영을 IPP 전담교수의 역할 : 한국기술교육대학교 사례를 중심으로)

  • In this paper, we suggest the roles of IPP (Industry Professional Practice) professor that are necessary for efficient and systematic operation and management of IPP system, which has been implemented by KOREATECH recently under the goal of nurturing industry-oriented talents with practical skills, based on the analysis of actual operational practices of advanced overseas universities and the IPP experiences for 2 years. The roles of IPP professor are largely divided into the following three: IPP manager; student career counselor; educator of IPP educational program. During the initial stage of IPP system, the role of IPP manager is most significant. However, as the system is systematically managed and operated, the role of IPP manager gradually decreases, and the role of career counselor becomes more important. Since IPP system is an educational program that gives credits, IPP professor should also perform the role of educator, including designing experiential learning curriculum and evaluation on educational goal accomplishment.

8-60hIPP5m-Induced G2/M Cell Cycle Arrest Involves Activation of ATM/p53/p21cip1/waf1 Pathways and Delayed Cyclin B1 Nuclear Translocation

  • Protein phosphatase 1 (PP1) is a major serine/threonine phosphatase that controls gene expression and cell cycle progression. The active mutant IPP5 ($8-60hIPP5^m$), the latest member of the inhibitory molecules for PP1, has been shown to inhibit the growth of human cervix carcinoma cells (HeLa). In order to elucidate the underlying mechanisms, the present study assessed overexpression of $8-60hIPP5^m$ in HeLa cells. Flow cytometric and biochemical analyses showed that overexpression of $8-60hIPP5^m$ induced G2/M-phase arrest, which was accompanied by the upregulation of cyclin B1 and phosphorylation of G2/M-phase proteins ATM, p53, $p21^{cip1/waf1}$ and Cdc2, suggesting that $8-60hIPP5^m$ induces G2/M arrest through activation of the ATM/p53/$p21^{cip1/waf1}$/Cdc2/cyclin B1 pathways. We further showed that overexpression of $8-60hIPP5^m$ led to delayed nuclear translocation of cyclin B1. $8-60hIPP5^m$ also could translocate to the nucleus in G2/M phase and interact with $pp1{\alpha}$ and Cdc2 as demonstrated by co-precipitation assay. Taken together, our data demonstrate a novel role for $8-60hIPP5^m$ in regulation of cell cycle in HeLa cells, possibly contributing to the development of new therapeutic strategies for cervix carcinoma.

Sensitization of Cervical Carcinoma Cells to Paclitaxel by an IPP5 Active Mutant

  • Paclitaxel is one of the best anticancer agents that has been isolated from plants, but its major disadvantage is its dose-limiting toxicity. In this study, we obtained evidence that the active mutant IPP5 ($8-60hIPP5^m$), the latest member of the inhibitory molecules for protein phosphatase 1, sensitizes human cervix carcinoma cells HeLa more efficiently to the therapeutic effects of paclitaxel. The combination of $8-60hIPP5^m$ with paclitaxel augmented anticancer effects as compared to paclitaxel alone as evidenced by reduced DNA synthesis and increased cytotoxicity in HeLa cells. Furthermore, our results revealed that $8-60hIPP5^m$ enhances paclitaxel-induced G2/M arrest and apoptosis, and augments paclitaxel-induced activation of caspases and release of cytochrome C. Evaluation of signaling pathways indicated that this synergism was in part related to downregulation of NF-${\kappa}B$ activation and serine/threonine kinase Akt pathways. We noted that $8-60hIPP5^m$ downregulated the paclitaxel-induced NF-${\kappa}B$ activation, $I{\kappa}B{\alpha}$ degradation, PI3-K activity and phosphorylation of the serine/threonine kinase Akt, a survival signal which in many instances is regulated by NF-${\kappa}B$. Together, our observations indicate that paclitaxel in combination with $8-60hIPP5^m$ may provide a therapeutic advantage for the treatment of human cervical carcinoma.

Influence of a Co-op Program on Participants' Academic Performance, Employment and Job Performance: Focusing on the Case of KOREATECH

  • KOREATECH has operated a Co-op program called 'Industry Professional Practice (IPP)' that allows students to work full-time in relevant industries for at least four months since 2012, and also developed a systematic performance evaluation model on the basis of Kirkpatrick's four-stage assessment model. This study aims to share KOREATECH's Levels 3 and 4 evaluation results which are clearly what practitioners and academic investigators of cooperative education want to know the most. For this purpose, a questionnaire survey was conducted for KOREATECH graduates and their superiors to examine influence of IPP experiences on employment and job performance. A total of 730 alumni who graduated within last 5 years participated in the survey, with 213 (29.2%) having IPP experiences, and 162 superiors rated their subordinates on seven performance criteria. The analysis results were mixed. On the one hand, employment rate of IPP participants was 89.7%, compared to 86.8% of alumni without field experiences. Participants of the IPP program were more satisfied with their salary and felt less unfair about their career and promotion opportunities than alumni without field experiences. On the other, superiors rated their subordinates without IPP experiences slightly more positively than IPP participants in terms of job performance. These contrasting results are judged to show strengths and weaknesses of the IPP program at the same time. The limitations of the study and future research directions are discussed at the last part.

A Case Study of Asian Colleges's Co-op Program for Successful Operations of IPP Program (장기현장실습 제도의 성공적인 운영을 위한 아시안 대학의 코업 프로그램 사례분석)

  • This paper presents several case studies of asian colleges's co-op program for successful operation of KOREATECH IPP(Industry Professional Practice) program. The IPP program is a new educational model that combines academic study and industrial work through university-industry cooperation. The IPP experience will lead the university graduates to improve their employment competencies and will be a key to solve the problems of the labor mismatch between university and industry. We will discuss and analyze the strengths and the weaknesses of the Co-op programs of the Kyoto sangyo university, the Hong Kong polytechnic university and the suranaree university in thailand. Based on these case studies, we summarized several benchmarking points for the successful operation of IPP program.

An exploration of factors affecting the Satisfaction of long-term on-site training: Focusing on IPP of K University (장기현장실습 프로그램의 만족도 영향요인 탐색: K대학 IPP 프로그램을 중심으로)

  • The purpose of this study was to investigate the factors affecting the satisfaction of the program for the quality improvement of the long-term on-site training operated by the university. The survey questionnaire data of 393 students who participated in the IPP program of K university were used to analyze the data and the results were as follows: First, factors affecting the satisfaction of long-term on-site training were corporation factors and university factors. Specifically, job contents, coaching, job scope, center support were found to have a significant effect in that order. Second, the factors affecting the satisfaction of long-term on-site training were different according to group characteristics(IPP participation types, major types, corporation types). It is necessary to intensively manage the common and important corporation factors, and it is necessary to closely examine each factor derived from group characteristics, and to support and strive at university level according to group characteristics in student counseling and matching.

Influences of Student's Major Satisfaction on Job-seeking Stress - Focusing on Differences of IPP Experienced, Nonexperienced Groups - (대학생의 전공만족도가 취업스트레스에 미치는 영향 - IPP형 장기현장실습 경험 유무 집단 간 차이를 중심으로 -)

  • This study empirical analysed how student's major satisfaction affected job-seeking stress. It also analysed the mean differences of major satisfaction and job-seeking stress between two groups(IPP experienced, non-experienced groups). 200 data were collected from the field survey questionnaires administered to a quota sample of university students who had IPP experience or non-experience. For hypothesis testing, the IBM SPSS Statistics 24.0 was used. As a result of empirical research analysis, first, it was found that major satisfaction had a negative effect on job-seeking stress. Among major satisfaction sub categories, subject satisfaction had negative effects on cognitive and physical stress, and relationship satisfaction also had negative effects on cognitive and physical stress. On the other hand, general satisfaction had a negative effect only on emotional stress. Second, there was a significant difference between major satisfaction and job-seeking stress. Major satisfaction of IPP experience group was high, and the job-seeking stress was low. Third, in the group of students who practiced IPP, subject satisfaction had a negative effect on emotional and physical stress, whereas subject satisfaction in the group of students who did not practice IPP had no significant effect on job-seekingt stress. In the case of relationship satisfaction, only non-practice groups had a negative effect on cognitive stress and physical stress. In the case of general satisfaction, only the IPP group showed negative effects on emotional stress. The results of this study supported the preceding studies that major satisfaction had an effect on job-seeking stress, and the study was meaningful in that it confirmed that there were differences between the IPP experienced, non-experienced groups. Based on this results, academic and practical implications were suggested.

Metabolic engineering for isoprenoids production in Escherichia coli

  • Isopentenyl diphosphate (IPP) is the common, five-carbon building block in the biosynthesis of all isoprenoids. IPP in Escherichia coli is synthesized through the non-mevalonate pathway. The first reaction of IPP biosynthesis in E. coli is the formation of 1-deoxy-D-xylulose-5-phosphate(DXP), catalyzed by DXP synthase and encoded by dxs. The second reaction in the pathway is the reduction of DXP to 2-C-methyl-D-erythritol-4-phosphate, catalyzed by DXP reductoismerase and encoded by dxr. To determine if one of more of the reactions in the non-mevalonate pathway controlled flux to IPP, dxs and dxr were placed on several expression vectors under the control of three different promoters and transformed into three E. coli strains ($DH5{\alpha}$, XL1-Blue, and JM101) that had been engineered to produce lycopene, a kind of isoprenoids. Lycopene production was improved significantly in strains transformed with the dex expression vectors. At arabinose concentrations between 0 and 1.33 mM, cells expressiong both dxs and from $P_{BAD}$ on a midium-copy plasmid produced 1.4 -2.0 times more lycopene than cells expressing dxs only. However, at higher arabinose concentrations lycopene production in cell expressing both dxs and dxr was lower than in cells expression dxs only. A comparison of the three E. coli strains trasfomed with the arabinose-inducible dxs on a medium-copy plasmid revealed that lycopene production was highest in XL1-Blue.

At arabinose concentrations between 0 and 1.33 mM, cells expressing both dxs and dxr from PBAD on a medium-copy plasmid produced 1.4 - 2.0 times more lycopene than cells expressing dxs only. However, at higher arabinose concentrations lycopene production in cells expressing both dxs and dxr was lower than in cells expressing dxs only. A comparison of the three E. coli strains transformed with the arabinose-inducible dxs on a medium-copy plamid revealed that lycopene production was highest in XL1-Blue.
