An Effective Antidandruff Agent-IPBC

  • The purpose of this study was to determine the effect of IPBC(3-lodo-2-propynylbutylcarbamate) on dandruff caused by the anthropophilic fungus Malassezia furfur. The effects of IPBC on dandruff were examined by evaluating (a) the MIC value of IPBC using broth dilution method; (b) the remnant antimicrobial activity of IPBC containing shampoo on skin disc; (c) the antidandruff efficacy of 1.0 % IPBC containing shampoo in double blind clinical trial. To investigate the remnant antimicrobial activity of IPBC against Malassezia furfur, guinea pig-skin disc was washed with antidandruff shampoo and then the diameter of inhibition zone per disc was measured. For clinical trial, thirty healthy volunteers, aged 25-35, participated in 4 week study. At 0, 2, 4 weeks, examinations of scaling, itching on scalp were carried out. The MIC(Minimun Inhibition Concentration) values of IPBC range from 0.10 to 1.00${\mu}g/ml$ and it seems that IPBC is more effective in the MIC values than zinc pyrithione, selenium disulphide, piroctone olamine and comparable to ketoconazole, climbazole. When the remnant antimicrobial activity of IPBC shampoo on skin disc was determined, 0.5% IPBC shampoo and 2.0% Ketoconazole shampoo resulted in similar antimicrobial effect. In addition, 1.0%, 2.0% IPBC shampoo was more effective than 2.0% ketoconazole shampoo. After two and four-weeks of 1.0% IPBC shampoo treatment, there was significant reduction of scaling, itching in test group compared to control group. On the basis of these results, it can be concluded that 1.0% IPBC is more effective than 2.0% Ketoconazole in reducing dandruff. It seems that strong capacity of drug binding to the stratum corneum plays a role in its antidandruff effect since adsorption of active ingredients on scalp is very important factor in reducing dandruff.

Anti-mold Effectiveness of Neonicochid Type Wood Preservative (네오니코치드계 목재보존제의 방곰팡이 효력)

  • To development of the low toxic preservatives, we prepared 29 kinds of chemicals using dinotefuran of low toxic insecticide, IPBC and propiconazol of fungicide, and clove oil has fungicidal activity, but is harmless to humans. It was conducted to investigate the inhibitory effectiveness of mycelial growth and anti-mold effectiveness to treated PDA medium and wood specimen with prepared chemicals. It was not showed anti-mold effectiveness that solvent and emulsifier form preservatives. Therefor, we thought that only four component, dinotefuran, IPBC, propiconazol and clove oil, has anti-mold effectiveness. In the PDA medium test, the preservative which has much IPBC showed high anti-mold effectiveness. By the wood specimen test, it was known that many prepared preservatives which showed good inhibitory effectiveness of mycelial growth in the PDA medium test, have not anti-mold effectiveness. Of all prepared preservatives in this study, only three preservatives containing dinotefuran and IPBC, or adding clove oil appeared the excellent anti-mold effectiveness.

Simultaneous quantitative analysis of four isothiazolinones and 3-iodo-2-propynyl butyl carbamate in hygienic consumer products

  • In this study, extraction, clean-up and instrumental analytical conditions were optimized for identifying and quantifying methyl-, chloromethylbenzi-, octyl- isothiazolinone (MIT, CMIT, BIT, OIT) and 3-iodo-2-propynyl butyl carbamate (IPBC), widely used biocidal active ingredients in human hygiene consumer products. Accuracy of the developed ultrasonic assisted extraction method followed by HPLC/MS analysis for four isothiazolinones and IPBC ranged between 60.4% and 113% in various types of consumer product samples. Method detection limits ranged 0.011-0.034 mg/kg for wet wipes, 0.57-1.5 mg/kg for liquid detergent and 0.58-1.6 mg/kg for powder detergent. Wet wipes, powder and liquid detergents collected from local market in Korea were analyzed to demonstrate the applicability of the developed method. Even after the regulation of those compounds in wet wipes, CMIT, MIT and IPBC were still frequently detected from complementary wet wipes without brand labels which were distributed to customers at local retail shops and restaurants. The maximum observed concentrations of MIT and CMIT found in those complementary wet wipes were 70.2 mg/kg and 11.3 mg/kg, respectively.

Simultaneous detection of five biocides in household products by gas chromatography-mass spectrometry

  • A gas chromatography-mass spectrometric method was developed for determining 5-chloro-2-methyl-4-isothiazolin-3-one (CMIT), 2-methyl-4-isothiazolin-3-one (MIT), 1,2-benzisothiazolin-3-one (BIT), 3-iodo-2-propynyl butyl carbamic acid (IPBC) and benzoic acid (BA) in household products. A 0.5 g sample was placed in a test tube and dissolved with 5 mL water, 5 mL methylene chloride and 1.0 mL methanol. The solution was extracted by ultra-sonication followed by mechanical shaking using the salting out effect. Under the established condition, the lowest quantification limits of all analytes were in the range of 0.04-10 mg/kg and their relative standard deviations were less than 8.0 %. The method was used to analyze 10 household products. As a result of analyzing 10 household products, MIT was detected in the range of 1.2-3.5 mg/kg in 3 of 10 samples, CMIT was detected in the range of 2.6-8.2 mg/kg in 3 of 10 samples, and BA was detected in the range of 5.0-15 mg/kg in 4 of 10 samples. Meanwhile, BIT and IPBC were not detected in any of the products. It has been shown that this method can be used for the simultaneous determination of biocides with various physical and chemical properties in household products.

  • Iodopropynyl butylcarbamate and climbazole as anti-dandruff agents widely used in cosmetics and pharmaceutics were micronized using supercritical fluid. Supercritical carbon dioxide was selected due to relatively low critical temperature and critical pressure. Iodopropynyl butylcarbamate and climbazole were chosen because of their solubility in supercritical carbon dioxide. The rapid expansion of supercritical solution (RESS) experiments involved investigations of the effect of pressure, temperature on particle size and morphology.
