Effect of Incubation Period, Temperature and pH on Mycelial Growth of Cylindrocarpon destructans (Zinssm.) Scholten Causing Root-rot of Ginseng (배양기간, 온도, pH가 인삼 근부병균 Cylindrocarpon destructans (Zinssm.) Scholten의 균사생육에 미치는 영향)

  • Cylindvocarpon destmtalns isolate CY-92-01, pathogen of root-rot of Panax ginseng showed t the maximum mycelial growth on the Czapek solution agar among the thirteen kinds of media. Five isolates (Isolate CY-92-01, CY-92-03, CY-92-07, CY-94-01, CY-94-02) of C. destructan from various growth stages of p. ginseng recovered from several geographical sites also showed maximum growth in the Czapek-Dox broth compared with potato dextrose broth and V-8 juice broth. Rapid growth rate was maintained until 12 days after inoculation on the Czapek-Dox broth and mycelial weight was somewhat constant until 20 days. After 30 days of incubation, the mycelial weight began to decrease. The fungal growth occurred from 5$^{\circ}C$ to $25^{\circ}C$ and optimum temperature for growth was 2$0^{\circ}C$. Mycelial weight orderly decreased at 15, 25, 10, and 5$^{\circ}C$. Quantitative measurement was impossible at 5$^{\circ}C$. No fungal growth was occurred at the temperature higher than 3$0^{\circ}C$. Growth was observed at all tested pH ranges from 2.8 to 8.0. Optimum pH for growth was 4.0~5.0 followed by pH 3.3~3.5 and 5.4~6.0. The least growth occurred at pH 2.8.

  • The experiment were carried out to establish the optimal condition of bulblet size, low temperature treatment and time of incubation in acclimatization on stem emergence of Lilium oriental hybrids. The large size bulblet was better than middle and small size bulblets in percentage of stem emergence and weight of bulblets. The highest percentage of stem emergence was 95% in large size bulblet 'Acapulco' but small size bulblet of 'Casablanca' was no stem emergence. Low temperature treatment(5℃)for breaking dormancy was needed at least more than 9weeks. Weight of bulblets and percentage of stem emergence was not good in 5℃ treatment for 6 weeks. Microbial Transglutaminase Modifies Gel Properties of Porcine Collagen

  • We studied the gel properties of porcine collagen with microbial transglutaminase (MTGase) as a catalyst. A creep meter was used to measure the mechanical properties of gel. The results showed samples with high concentration of MTGase gelled faster than those with a low concentration of MTGase. The gel strength increased with incubation time and the peaks of breaking strength for 0.1, 0.2 and 0.5% MTGase were obtained at 40, 20 and 10 min incubation time, respectively. According to SDS-PAGE, the MTGase was successfully created a collagen polymer with an increase in molecular weight, whereas no change in formation was shown without MTGase. The sample with 0.5% MTGase began to polymerize after 10 or 20 min incubation at $50^{\circ}C$, and complete polymerization occurred after 40-60 min incubation. Scanning electron microscopic analysis revealed that the gel of porcine collagen in the presence of MTGase produced an extremely well cross-linked network. The differential scanning calorimetric analysis showed the peak thermal transition of porcine collagen gel was at $36^{\circ}C$, and that with MTGase no peak was detected during heating from 20 to $120^{\circ}C$. The melting point of porcine collagen gel could be controlled by MTGase concentration, incubation temperature and protein concentration. Knowledge of the structural and physicochemical properties of porcine collagen gel catalyzed with MTGase could facilitate their use in food products.

Nitrogen Mineralization and Dynamics in the Forest Soil (삼림토양의 질소 무기화와 무기질소의 동태)

  • Mineral nitrogen dynamics and net mineralization of nitrogen in oak(quercus accutissima) and pine(pinus rigida) forest soils were studied. Nitrogen mineralization was determined over 8-week period by incubation method at laboratory. Initial water content of incubating soils was adjusted by applying suction(30mmhg), and lossof water during incubation was recovered with deionized water using syringe at every 3 or 4days. Temperature of incubator was maintained with 35+0.3c during the incubation period. Content of organic matter, total nitrogen, nh4-n and no3-n in soils in oak stand were significantly highter than those in pine stand. soil ph was lower in pine stand than in oak stand. initial nh4-n and no3-n of soils used in incubation experiment were 12.6 ug/g and 6.5 ug/g for oak stand, and 5.3ug/g and 5.1 ug/g for pine stand, respectively. Production of nh4-n increased from the beginning st both stands, and showed a peak at 5th week in oak stand(28.5 ug/g) and 6th week in pine stand(16.7 ug/g), and then decreased. intial no3-n of soils in oak(6.5 ug/g) and pine(5.1ug/g)stands, increased to 36.2 ug/g in soils of oak stand(5th week) and 13.4 ug/g in pine stand(4th week), respectively. The low values of no3-n of the field soil in the growing season compared with those of incubating soils at both stands indicate that considerable amount of nh4-n and no3-n produced in soils of oak and pine stands during two-months incubation were 59.7 and 141.6mg/kg soil, and 51.9 and 41.2mg/kg soil, respectively.

  • In an attempt to understand the possible effects of temperature and X-irradiation on the activities of surfactant in rabbits, the pulmonary surfactant from the rabbit was subjected to the incubation at $37^{\circ}C$ and X-irradiation with 900r in vitro, and activities of surfactant were measured at 10, 30, 60, and 90 minutes. Tension-area diagram of the lung extract was recorded automatically by the modified Langmuir-wilhelmy balance with a synchronized recording system designed in this Department. A comparison was made with the normal and the following results were obtained. 1) The maximal surface tension, minimal surface tension, width of the tension area diagram at the surface area of 40% and stability index of the normal rabbit lung extract were $31.6{\pm}3.11\;dynes/cm,\;8.2{\pm}0.56\;dynes/cm,\;21.4{\pm}4.40\;dynes/cm\;and\;1.12{\pm}0.22$,respectively. 2) In the $37^{\circ}C$ incubation group, maximal surface tension was similar to the normal value, while minimal surface tension was significantly lower and stability infer was markedly higher than the normal. 3) In the group where X-irradiation of 900r in vitro was applied, maximal surface tension did not differ greatly with the normal or the $37^{\circ}C$ incubation group. The minimal surface tension was significantly lower than the normal but comparing with the $37^{\circ}C$ incubation group, some decrease in minimal surface tension was noted. The width of the tension·area diagram at 40% and stability index in the irradiated group were significantly higher than the normal but a tendency of increase was noted comparing with the $37^{\circ}C$ incubation group.

Estimation of N Mineralization Potential and N Mineralization Rate of Organic Amendments as Affected by C:N Ratio and Temperature in Paddy Soil

  • Understanding N mineralization dynamics in soil is essential for efficient nutrient management. An anaerobic incubation experiment was conducted to examine N mineralization potential and N mineralization rate of the organic amendments with different C:N ratio in paddy soil. Inorganic N in the soil sample was measured periodically under three temperature conditions ($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$) for 90 days. N mineralization was accelerated as the temperature rises by approximately $10%^{\circ}C^{-1}$ in average. Negative correlation ($R^2=0.707$) was observed between soil inorganic N and C:N ratio, while total organic carbon extract ($R^2=0.947$) and microbial biomass C ($R^2=0.824$) in the soil were positively related to C:N ratio. Single exponential model was applied for quantitative evaluation of N mineralization process. Model parameter for N mineralization rate, k, increased in proportion to temperature. N mineralization potential, $N_p$, was very different depending on C:N ratio of organic input. $N_p$ value decreased as C:N ratio increased, ranged from $74.3mg\;kg^{-1}$ in a low C:N ratio (12.0 in hairy vetch) to $15.1mg\;kg^{-1}$ in a high C:N ratio (78.2 in rice straw). This result indicated that the amount of inorganic N available for crop uptake can be predicted by temperature and C:N ratio of organic amendment. Consequently, it is suggested that the amount of organic fertilizer application in paddy soil would be determined based on temperature observations and C:N ratio, which represent the decomposition characteristics of organic amendments.

A Study on the Measurement Method for Benthic Nutrient Flux in Freshwater Sediments (담수 퇴적물의 영양염 용출 측정 방법에 관한 고찰)

  • Accurate measurement of benthic nutrient fluxes (BNF) is a prerequisite for evaluating the effect of sediments on nutrient cycle in the surface water. The intact sediment cores were collected in July 2015 at the midstream of Nakdong River. We identified pre-incubation time (6, 12, 24 hr), dissolved oxygen concentration (90, 70, 50% saturation), diffusive boundary layer thickness (0, 0.6-0.8, 1.2-1.4 mm), and incubation temperature (10, 17, 20, $25^{\circ}C$) as the most important control factors, and measured the BNF fluctuation with the variation of these factors using the laboratory sediment core incubation method. Since the chemical composition, redox condition, hydrodynamic regimes and microbial activities at the sediment-water interface were changed as a result of the alteration of control factors, sediment core incubation should be conducted under as close to the natural conditions of study site as possible, in order to produce the results similar to actual values. Relative percentage differences between two replicates were below 20% in most control factors, which showed satisfactory precision for strict compliance with the experimental conditions and procedures. In the further studies, we will compare the results of core incubation with those of in situ measurements to confirm the accuracy of the sediment core incubation method.

Effects of Incubation and Thawing Temperature on Frozen-thawed Stallion Epididymal Spermatozoa (말의 정소상체 정자의 동결 후 해동 온도 및 Incubation의 효과)

  • Cryopreservation of epididymal spermatozoa offers a potential tool for rescuing genetic material from males of genetically elite populations. Castration, catastrophic injury, sudden death or any other event that makes semen collection or mating impossible may prematurely terminate a stallion reproduction. Stallion epididymal spermatozoa vary widely in the loss of progressive motility, acrosomal integrity, and viability during freezing and thawing. The objective of this work was to investigate the effect of (1) freezing package types on cryopreservation efficiency, (2) thawing temperatures (37, 56 or $70^{\circ}C$) on Computer Assisted Sperm Analysis (CASA) parameters and (3) post-thawing incubation time (0, 1, 2 or 4h) on castrated stallion epididymis. Post-thawed sperm motility ranged between 59.69% and 64.28% ($56^{\circ}C$ and $37^{\circ}C$) in various thawing temperatures. When stallion epididymis sperm was frozen, straw was better than freezing tube on VCL (Velocity of Curvilinear Line) and VAP (Velocity of Average Path) parameter. Higher percentage of motility was observed at $37^{\circ}C$ thawing temperature even though no significant difference was observed among various temperatures. The motility, VCL, ALH (Amplitude of Lateral Head displacement), VAP, BCF (Beat-Cross Frequency) and STR (Straightness index) parameter of post-thawed sperm were significantly decreased by increasing the incubation time for all thawing temperatures. The present study showed that type of freezing package (Straw vs. Freezing tube) was not significantly different on cryopreservation efficiency. Furthermore, stallion epididymal spermatozoa frozen-thawed at $37^{\circ}C$ for 1 min resulted the highest proportion of motility and velocity movement. In addition, motility and viability of frozen-thawed stallion epididymal spermatozoa were also decreased by incubation.

In-situ Analysis of Temperatures Effect on Electromigration-induced Diffusion Element in Eutectic SnPb Solder Line (공정조성 SnPb 솔더 라인의 온도에 따른 Electromigration 확산원소의 In-situ 분석)

  • In-situ observation of electromigration in thin film pattern of 63Sn-37Pb solder was performed using a scanning electron microscope system. The 63Sn-37Pb solder had the incubation stage of electromigration for edge movement when the current density of $6.0{\times}10^{4}A/cm^2$ was applied the temperature between $90^{\circ}C\;and\;110^{\circ}C$. The major diffusion elements due to electromigration were Pb and Sn at temperatures of $90-110^{\circ}C\;and\;25-50^{\circ}C$, respectively, while no major diffusion of any element due to electromigration was detected when the test temperature was $70^{\circ}C$. The reason was that both the elements of Sn and Pb were migrated simultaneously under such a stress condition. The existence of the incubation stage was observed due to Pb migration before Sn migration at $90-110^{\circ}C$. Electromigration behavior of 63Sn-37Pb solder had an incubation time in common for edge drift and void nucleation, which seemed to be related the lifetime of flip chip solder bump. Diffusivity with $Z^*$(effective charges number) of Pb and Sn were strongly affect the electromigration-induced major diffusion element in SnPb solder by temperature, respectively.

The Effect of Shading Treatment on the Uniform Primordium Formation and Development of Pleurotus ostreatus during Incubation (배양기간 중 차광처리가 느타리 발이 균일도 및 생육에 미치는 영향)

  • The study was conducted to investigate the effect of shading treatment on the uniform primordium formation and development of fruit body during incubation of major cultivated oyster mushrooms (Pleurotus ostreatus). Oyster mushrooms cultivars Suhan No. 1 and Gonji No. 7 were incubated while shading during different times in the incubation room wherein temperature of $20^{\circ}C$ was maintained. Substrate temperature during incubation was increased to $24{\sim}25^{\circ}C$ in 11~12 days for Suhan No. 1 and to $25{\sim}26^{\circ}C$ in 11~12 days for Gonji No. 7 regardless of shading times. The $CO_2$ generation was the highest as 9~11% for Suhan No. 1 and 9~10% for Gonji No. 7 at the time of temperature increase the highest, while $O_2$contents became decreased, thus constructed good environment for the growth of hyphae. Therefore, ratio of ununiformal primordia formation was reduced by 20% as against control for Suhan No. 1 and by 13% for Gonji No. 7 when shading was applied from $10^{th}$ day of inoculation with no difference in fruit body yield.
