Functionalized Poplar Powder as a Support Material for Immobilization of Enoate Reductase and a Cofactor Regeneration System

  • In this study, functionalized poplar powder (FPP) was used as a support material for the immobilization of enoate reductase (ER) and glucose-6-phosphate dehydrogenase (GDH) by covalent binding. Under optimal conditions, the immobilization efficiency of ER-FPP and GDH-FPP was 95.1% and 84.7%, and the activity recovery of ER and GDH was 47.5% and 37.8%, respectively. Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) analysis indicated that FPP was a suitable carrier for enzyme immobilization. ER-FPP and GDH-FPP exhibit excellent thermal stabilities and superior reusability. Especially, ER-FPP and GDH-FPP enable the continuous conversion of 4-(4-Methoxyphenyl)-3-buten-2-one with $NAD^+$ recycling. While the immobilization strategies established here were simple and inexpensive, they exploited a new method for the immobilization and application of ER and its cofactor recycling system.

Protective effects of Korean Red Ginseng against sub-acute immobilization stress-induced testicular damage in experimental rats

  • Background: Excessive stress causes varied physiological and psychological disorders including male reproductive problems. Here, we attempted to investigate the protective effects of Korean Red Ginseng (Panax ginseng Meyer; KRG) against sub-acute immobilization stress-induced testicular damage in experimental rats. Methods: Male rats (age, 4 wk; weight, 60-70 g) were divided into four groups (n = 8 in each group): normal control group, immobilization control group, immobilization group treated with 100 mg/kg of KRG daily, and immobilization group treated with 200 mg/kg of KRG daily. Normal control and immobilization control groups received vehicle only. KRG (100 mg/kg and 200 mg/kg) was mixed in the standard diet powder and fed daily for 6 mo. Parameters such as organ weight, blood chemistry, sperm kinematic values, and expression levels of testicular-related molecules were measured using commercially available kits, Western blotting, and reverse transcription polymerase chain reaction. Results: Data revealed that KRG restored the altered testis and epididymis weight in immobilization stress-induced rats significantly (p < 0.05). Further, KRG ameliorated the altered blood chemistry and sperm kinematic values when compared with the immobilization control group and attenuated the altered expression levels of spermatogenesis-related proteins (nectin-2, cAMP responsive element binding protein 1, and inhibin-${\alpha}$), sex hormone receptors (androgen receptor, luteinizing hormone receptor, and follicle-stimulating hormone receptor), and antioxidant-related enzymes (glutathione S-transferase m5, peroxiredoxin-4, and glutathione peroxidase 4) significantly in the testes of immobilization stress-induced rats. Conclusion: KRG protected immobilization stress-induced testicular damage and fertility factors in rats, thereby indicating its potential in the treatment of stress-related male sterility.

Effect of Ginsenoside Re on Depression- and Anxiety-Like Behaviors and Cognition Memory Deficit Induced by Repeated Immobilization in Rats

  • In this study, we assessed the effects of ginsenoside Re (GRe) administration on repeated immobilization stress-induced behavioral alterations using the forced swimming test (FST), the elevated plus maze (EPM), and the active avoidance conditioning test (AAT). Additionally, we examined the effect of GRe on the central adrenergic system by observing changes in neuronal tyrosine hydroxylase (TH) immunoreactivity and brain-derived neurotrophic factor (BDNF) mRNA expression in the rat brain. Male rats received 10, 20, or 50 mg/kg GRe (i.p.) 30 min before daily exposures to repeated immobilization stress (2 h/day) for 10 days. Activation of the hypothalamic-pituitary-adrenal (HPA) axis in response to repeated immobilization was confirmed by measuring serum levels of corticosterone (CORT) and the expression of corticotrophin-releasing factor (CRF) in the hypothalamus. Repeated immobilization stress increased immobility in the FST and reduced open-arm exploration in the EPM test. It also increased the probability of escape failures in the AAT test, indicating a reduced avoidance response. Daily administration of GRe during the repeated immobilization stress period significantly inhibited the stress-induced behavioral deficits in these behavioral tests. Administration of GRe also significantly blocked the increase in TH expression in the locus coeruleus (LC) and the decrease in BDNF mRNA expression in the hippocampus. Taken together, these findings indicate that administration of GRe prior to immobilization stress significantly improved helpless behaviors and cognitive impairment, possibly through modulating the central noradrenergic system in rats. These findings suggest that GRe may be a useful agent for treating complex symptoms of depression, anxiety, and cognitive impairment.

청심포자음(淸心蒲子飮)의 구속 스트레스 및 절개 통증에 대한 항스트레스 작용과 진통효과 (Anti-stress and Anti-nociceptive Effects of the Aqueous Extracts of Chungsimyeonja-eum against Immobilization Stress and Incision Pain in Rats)

  • The aqueous extract of Chungsimyeonja-eum has been used to treat palpitation, anemia, chronic fatigue, hypertension, and stroke. It also possesses various pharmacological effects including hypotensive, blood circulating, sedative, anti-inflammatory, diuretic, and anti-stress activities. In the present results, the latency time of plantar test was decreased in the immobilization stress and incusion pain-induction group. However the paw withdrawal latency values were increased in the immobilization stress and incision pain-induction groups after treatment with the aqueous extract of Chungsimyeonja-eum. The c-Fos expression in the paraventricular nucleus (PVN), and 5-hydroxytryptamine (5-HT) and tryptophan hydroxylase (TPH) in the dorsal raphe were enhanced in the immobilization stress and incision pain-induction groups. However, the treatment with the aqueous extract of Chungsimyeonja-eum suppressed the immobilization stress and incision pain-induced increase of c-Fos, 5-HT, and TPH expressions. Here in this study, we have demonstrated the protective effects of Chungsimyeonja-eum on immobilization and incision pain-induced stress. The present study revealed that Chungsimyeonja-eum treatment diminishes immobilization and pain stress.

흰쥐 발목고정으로 유발된 비복근 위축에서 보조약물 투여가 미치는 효과 (Effect of Adjuvant Administration on Gastrocnemius Atrophy Induced by Ankle Joint Fixation in Rats)

  • Purpose : In this study we tested the hypothesis that eucommia ulmoides (EU) extract would improve disuse-induced muscle atrophy following hindlimb immobilization. Method : The hindlimb immobilization was performed with casting tape to keep the left ankle joint in a fully extended position. The rats in EU treated group were orally administrated with eucommia ulmoides water extract. After 2 weeks of immobilization, all animals were sacrificed, and the whole gastrocnemius muscles were dissected from both legs. The morphology of right and left gastrocnemius muscles in both EU and control groups were assessed by hematoxylin and eosin staining. Results : Eucommia ulmoides extract represented the significant protective effects against the reductions of the gastrocnemius muscles weight and average cross section area to compared with Control group. Conclusion : Eucommia ulmoides has protective effects against immobilization induced muscle atrophy.

Purification and Immobilization of Cyclodextrin glucanotransferase from recombinant Bacillus subtilis

  • 정제, 농축된 free CGTase 는 pH 6.0 - 7.0의 범위에서 안정하였으며, $70^{\circ}C$정도의 고온에서도 안정한 특성을 가지고 있었으며, CGTase의 고정화를 위한 방법으로서 CNBr-activated sepharose 4B에 의한 고정화가 가장 높은 효율올 나타내는 것으로 결정되었다. 고정화의 최적조건은 $30^{\circ}C$, 60rpm, pH 6.0의 phosphate buffer 하에서 9 시간 동안 고정화하는 것이었고, CNBr-activated sepharose 4B를 이용한 고정화의 경우 실험에 사용된 다른 모든 이온교환 수지에서 얻어진 효율에 비해 월등한 효과를 보였다.

  • Coater runnability of the paper coating and final product properties can be affected by the immobilization of coating color and the dewatering into the base paper. During the dewatering, the rheological properties and solids content of a coating are dramatically changed. For the purpose of obtaining better coater runnability and high quality of coater paper many papermakers are trying to improve the water retention of paper coatings by using some additives such as thickener and co-binder. In this study, we tried to investigate the rheological properties and the immobilization point of coatings with immobilization cell during the dewatering of coatings.

  • Immobilization behavior of methyl tert-butyl ether (MTBE) by various cyclodextrins(CDs) was studied to investigate the feasibility of MTBE removal using cyclodexrins. Even though MTBE has relatively low hydrophobicity and higher polarity compared to other organics, it was effectively immobilized by CDs. The immobilization isotherms was shown as a type of Freundlich isotherms, and the immobilization capacity of -CDs was the largest among natural COs. The initial apparent association constant for MTBE-CD complex follows the order : gamma = beta > methyl-beta > hydroxypropyl beta > alpha. These differences of the constants are related to the size of MTBE and CDs. The size of beta-CD and gamma-CD is large to encapsulate MTBE molecule into the cavity, which that of alpha-CB is too small to encapsulate MTBE.

Microarray Analysis of Gene Expression by Ginseng Water Extracts in a Mouse Adrenal Cortex after Immobilization Stress

  • To investigate the effects of repeated immobilization-stress challenge on the the hypothalamic-pituitary-adrenal axis, the genomic transcriptome in the adrenal cortex of immobilization-stressed mouse was analyzed by using a cDNA microarray. Mice were subjected to immobilization stress for 2 h per day for 5 consecutive d. With a 4.0-fold cutoff of arbitrary criteria, the expression levels of 168 out of 41,174 genes were significantly modulated in the adrenal cortex by stress when comparing the control and experimental groups. These genes were related to apoptosis, cell cycle, immune response, inflammatory responses, and signal transduction, and thus may be used as potential targets for the development of therapeutics for chronic stress or depression. Six significant genes among these were selected for real time polymerase chain reaction analysis to confirm the change of their expression levels. The gene for phospho 1 was also further investigated because its expression showed the greatest fold-change.

Polydopamine-coated chitosan hydrogels for enzyme immobilization

  • To address inherent weaknesses such as low mechanical strength and limited enzyme loading capacity in conventional chitosan or alginate beads, an additional step involving the exchange of anionic surfactants with hydroxide ions was employed to prepare porous chitosan hydrogel capsules for enzyme immobilization. Consequently, excellent thermal stability and long-term storage stability were confirmed. Furthermore, coating the porous chitosan hydrogel capsules with polydopamine not only improved mechanical stability but also exhibited remarkable enzyme immobilization efficiency (97.6% for M1-D0.5). Additionally, it was demonstrated that the scope of application for chitosan hydrogel beads, prepared using conventional methods, could be further expanded by introducing an additional step of polydopamine coating. The enzyme immobilization matrix developed in this study can be selectively applied to suit specific purposes and is expected to be utilized as a support for the adsorption or covalent binding of various substances.
