국내 갈매나무과 13종에 대한 부위별 물 추출물의 면역 증진 활성 (Immune-enhancing Activity of Water Extracts for Each Part of 13 Species (Rhamnaceae) in Korea)

  • 본 연구에서는 국내 자생하고 있는 갈매나무과 식물 13종에 대한 부위별(잎, 가지, 열매) 물 추출물의 NO 생성능을 통하여 면역증진 활성을 측정하여 기능성을 검토하였다. 망개나무, 헛개나무, 까마귀베개, 산황나무, 갈매나무, 참갈매나무, 상동나무 등에서 50% 이상 NO 생성능을 보여줌으로써 대식세포 활성화를 검증하였다. 또한 PCR 전기영동을 통하여 면역증진과 관련된 cytokine인 iNOS, COX-2, IL-1β, IL-6, TNF-α의 발현을 확인하였다. 본 연구를 통해 갈매나무과 분류군들의 부위별 NO 생성능, mRNA 발현 등과 같은 면역 활성에 대한 과학적 근거를 제시할 수 있었고, 이는 면역증진 관련 소재 발굴 및 제품 개발에 기초자료로 활용할 수 있을 것으로 판단된다.

UV-B 조사에 따른 버섯 추출물의 항염증 및 항알레르기 활성 (Anti-inflammatory and Anti-allergic Effects of Lentinula edodes Extract by UVIrradiation)

  • 본 연구에서는 표고버섯(Lentinula edodes)을 자외선 조사를 통한 ergocaciferol (비타민 D2) 함량을 증진시킨 추출물을 이용하여 염증 및 알레르기 반응에 미치는 효과를 확인하였다. 표고버섯 추출물의 항염증 및 항알레르기 효능은 LPS로 활성화된 대식세포(RAW 264.7)와 PMA와 A23187에 의해 활성화된 비만세포(RBL-2H3)로부터 분비 또는 발현되는 TNF-α, IL-6, IL-1β, IL-4와 같은 cytokine과 histamine 분비량을 측정하여 관찰하였다. LPS에 의해 활성화된 대식세포에서 자외선 조사 표고버섯 추출물 처리에 의해 pro-inflammatory cytokine인 TNF-α와 IL-6의 분비량을 ELISA 방법으로 측정하였을 때 현저히 감소함을 확인하였고 mRNA 발현 또한 감소됨을 확인하였다. 비만세포에 자외선 조사 표고버섯 추출물과 PMA, A23187을 함께 처리한 경우 비만세포의 탈과립에 의해 분비되는 histamine의 양이 유의적으로 감소함을 확인하였고 IL-4의 발현양 또한 감소함을 확인할 수 있었다. 이상의 결과는 자외선 조사에 의해 비타민 D2 함량을 증진시킨 표고버섯 추출물이 염증과 알레르기 반응의 cytokine의 발현을 저해하는 것으로 보아 염증 및 알레르기 질환의 예방과 치료에 효과적으로 이용될 수 있을 것으로 사료된다.

유육종증환자의 기관지폐포세척액내 Interleukin에 대한 연구 (Interleukin Levels in the Bronchoalveolar Lavage Fluid of Patients with Pulmonary Sarcoidosis)

  • 연구배경: 유육종증은 전신을 침범하는 만성 육아종성 질환으로 단핵구성 폐포염이 선행되어 육아종형성을 유도하며, 여러 종류의 cytokine들이 육아종성 염증과 그에 따른 섬유화의명태생리가전에 관여한다고 알려져 있다. 특히 최근에는 IL-6와 IL-8이 유육종증에서 광범위하게 염증반응을 진행시키는 것으로 보고되고 있고, 반면에 IL-10은 단핵구/대식세포와 T림프구의 기능을 억제하여 IL-1, IL-6, TNF-$\alpha$, IFN-$\gamma$ 등의 분비를 감소시키므로써 육아종성 염증반응을 억제한다고 보고되고 있다. 본 연구에서는 유육종증환자들의 폐포세척액내에서 IL-6, IL-8, IL-10의 농도를 측정하고 유육종증의 활성화를 나타내는 임상적 지표들과 비교해 봄으로써 이러한 lnterleukin들이 유육종증의 병태생리기전에 관여하는지 알아보고자 하였다. 대상 및 방법: 폐조직검사를 통해 조직학적으로 확진된 유육종증환자 14명과 건강한 정상대조군 6명을 대상으로 기관지폐포세척술을 시행하였다. 유육종증환자들은 중요 장기(심장, 중추신경, 후방 포도막등)의 침범여부, 고칼숨혈증, 심각한 뼈기능감소 및 질환의 진행여부에 따라 활동성(n=7)과 비활동성(n=7)으로 분류하였고 회수한 폐포세척액을 동일한 조건에서 10배로 농축한 후 ELISA방법을 이용하여 IL-6, IL-8, IL-10을 측정하여 이 Interleukin들의 상호관계와 유육종증의 활성도를 비교하였다. 결 과: 기관지폐포세척액내 IL-6농도는 특히 활동성 유육종증환자군에서 비활동성 유육종증환자군 및 정상대조군보다 유의하게 높았다(각각 p<0.05). IL-8 농도는 활동성 유육종증환자군에서 비활동성 유육종증환자군 및 정상대조군보다 높은 경향을 보였으나 통계적 유의성은 없었고 IL-10농도는 특히 활동성 유육종증환자군에서 정상대조군 및 비활동성 유육종증환자군보다 낮은 경향을 보였으나 통계적 유의성은 없었다. 활동성 유육종증환자군의 기관지폐포세척액내 IL-6농도는 기관지폐포세척액내 IL-8 농도 및 CD4/CD8 비(比)와 서로 의미 있는 상관관계를 보였다(p<0.05). 결 론: 활동성 유육종증환자군의 기관지폐포세척액내에서 IL-6농도가 비활동성 유육종증환자군과 정상대조군에서 보다 유의하게 높았고 이러한 IL-6는 기관지폐포세척액내 CD4/CD8 비(比) 및 IL-8농도와도 상관관계를 보여, 주로 IL-6이 유육종증의 진행과정과 활성화에 종요한 역할을 하고 있는 것으로 생각된다. 또한 세포성 면역반응을 억제할 수 있는 cytokine으로 생각되는 IL-10의 농도가 유육종증환자의 폐내에서 증가되지 않은 것도 이 질환의 발병이나 진행에 기여할 것으로 추정되며 앞으로 유육종증의 치료에 IL-6의 단일클론성 항체나 IL-10이 이용될 수도 있을 것으로 생각된다.

Increased Cytokine and Nitric Oxide Levels in Serum of Dogs Experimentally Infected with Rangelia vitalii

  • This study aimed to measure the levels of interferon-gamma (IFN-${\gamma}$), tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin 1 (IL-1), interleukin 6 (IL-6), and nitrite/nitrate ($NO_x$) in serum of dogs experimentally infected with Rangelia vitalii. Twelve female mongrel dogs were divided into 2 groups; group A (uninfected controls) composed by healthy dogs (n=5) and group B consisting of dogs inoculated with R. vitalii (n=7). Animals were monitored by blood smear examinations, which showed intraerythrocytic forms of the parasite on day 5 post-infection (PI). Blood samples were collected through the jugular vein on days 0, 10, and 20 PI to determine the serum levels of IFN-${\gamma}$, TNF-${\alpha}$, IL-1, IL-6, and $NO_x$. Cytokines were assessed by ELISA quantitative sandwich technique, and $NO_x$ was measured by the modified Griess method. Cytokine levels (IFN-${\gamma}$, TNF-${\alpha}$, IL-1, and IL-6) were increased (P<0.01) in serum of infected animals. Serum levels of $NO_x$ were also increased on days 10 PI (P<0.01) and 20 PI (P<0.05) in infected animals. Therefore, the infection with R. vitalii causes an increase in proinflammatory cytokines and nitric oxide content. These alterations may be associated with host immune protection against the parasite.

국내 상업용 간장과 재래식 간장 유래 다당에 의한 대식세포 활성화 (Macrophage Activation by Polysaccharides from Korean's Commercial and Traditional Soy Sauces)

  • 본 연구에서는 일본식으로 제조된 상업용 간장과 우리나라 전통방식으로 제조된 재래식 간장으로부터 다당을 분리하여 RAW 264.7 대식세포주를 이용한 면역 증진활성을 비교하였다. 간장 유래 다당, CSP-0 및 KTSP-0는 RAW 264.7 세포주에 대해 모든 농도에서 세포 독성을 나타내지 않았다. 또한 재래식 간장 유래 다당인 KTSP-0는 상업용 간장 유래 다당 CSP-0보다 대식세포에 의한 NO 및 ROS의 생산을 농도의 존적으로 증가시켰으며, KTSP-0를 $1,000{\mu}g/m{\ell}$ 처리하였을 시 가장 높은 생산능을 나타내었다. 간장 유래 다당의 RAW 264.7 세포로부터 면역반응에 중요한 cytokine인 IL-6와 TNF-${\alpha}$의 cytokine mRNA의 발현량과 해당 단백질의 생산을 각각 real-time PCR과 ELISA로 확인한 결과, CSP-0는 IL-6와 TNF-${\alpha}$ mRNA의 발현 및 생산에 유의적인 영향을 미치지 않았지만, KTSP-0는 농도의존적으로 IL-6와 TNF-${\alpha}$ mRNA 발현 및 생산을 증가시키는 것을 확인하였으며, $1,000{\mu}g/m{\ell}$ 처리 시 최대 활성을 보였다. 한편, 대식세포의 탐식작용에서 중요한 역할을 하는 Fc 수용체의 발현 증가를 RT-PCR로 확인한 결과, CSP-0는 FcR I, II의 발현에 모두 영향을 미치지 않았지만, KTSP-0는 FcR I의 발현을 선택적으로 증가시킴이 확인되어 항원에 결합한 IgG와 강하게 결합하여 탐식작용을 촉진시킬 것으로 예상되었다. 본 연구를 통해 상업용 간장에서 분리한 다당, CSP-0보다 우리나라 전통 재래식 간장에서 분리한 다당, KTSP-0가 대식세포를 활성화하여 높은 면역 증진 효과를 나타내는 것으로 결론 지을 수 있었다.

Neuroprotective effects of consuming bovine colostrum after focal brain ischemia/reperfusion injury in rat model

  • To investigate the neuroprotective effects of bovine colostrums (BC), we evaluate the ability of consuming BC after focal brain ischemia/reperfusion injury rat model to reduce serum cytokine levels and infarct volume, and improve neurological outcome. Sprague-Dawley rats were randomly divided into 4 groups; one sham operation and three experimental groups. In the experimental groups, MCA occlusion (2 h) and subsequent reperfusion (O/R) were induced with regional cerebral blood flow monitoring. One hour after MCAO/R and once daily during the experiment, the experimental group received BC while the other groups received 0.9% saline or low fat milk (LFM) orally. Seven days later, serum pro-inflammatory cytokine (IL-$1{\beta}$, IL-6, and TNF-${\alpha}$) and anti-inflammatory cytokine (IL-10) levels were assessed. Also, the infarct volume was assessed by using a computerized image analysis system. Behavioral function was also assessed using a modified neurologic severity score and corner turn test during the experiment. Rats receiving BC after focal brain I/R showed a significant reduction (-26%/-22%) in infarct volume compared to LFM/saline rats, respectively (P < 0.05). Serum IL-$1{\beta}$, IL-6, and TNF-${\alpha}$ levels were decreased significantly in rats receiving BC compared to LFM/saline rats (P < 0.05). In behavioral tests, daily BC intake showed consistent and significant improvement of neurological deficits for 7 days after MCAO/R. BC ingestion after focal brain ischemia/reperfusion injury may prevent brain injury by reducing serum pro-inflammatory cytokine levels and brain infarct volume in a rat model.

Induction of Immune Responses by Two Recombinant Proteins of Brucella abortus, Outer Membrane Proteins 2b Porin and Cu/Zn Superoxide Dismutase, in Mouse Model

  • The diagnosis of Brucella abortus is mainly based on serological methods using antibody against LPS, which has diagnostic problems. Therefore, to solve this problem, we evaluated two proteins of B. abortus, Cu/Zn superoxide dismutase (SodC) and outer membrane proteins 2b porin (Omp2b). The genes were cloned and expressed in a pMAL system, and the recombinant proteins, rOmp2b and rSodC, were purified as fusion forms with maltose-binding protein. The identity of the proteins was confirmed by SDS-PAGE and Western blot analysis with sera of mice infected with B. abortus. Production of cytokines and nitric oxide (NO) was investigated in RAW 264.7 cells and mouse splenocytes after stimulation with the proteins. Moreover, cellular and humoral immune responses were investigated in BALB/c mice after immunization with the proteins. TNF-${\alpha}$, IL-6, and NO were significantly inducible in RAW 264.7 cells. Splenocytes of naive mice produced IFN-${\gamma}$ and IL-4 significantly by stimulation. Moreover, number of IgG, IFN-${\gamma}$, and IL-4 producing cells were increased in immunized mice with the two proteins. Production of IgG and IgM with rOmp2b was higher than those with rSodC in immunized mice. These results suggest that the two recombinant proteins of B. abortus may be potential LPS-free proteins for diagnosis.

탁리소독음(托裏消毒飮) 합(合) 황련해독탕(黃連解毒湯)의 아토피피부염에 대한 실험적 연구 (Effect of Taklisodok-um and Hwangryunhaedok-tang on Atopic Dermatitis)

  • Objective : This Experimental study was done to investigate the Effect of Taklisodok-um and Hwangryunhaedok-tang(TH) on Atopic Dermatitis. Methods : We assessed effects of TH on the IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo, on the IL-4, IL-5, CCR3 in the skin tissues of ear and dorsum with NC/Nga mice. And we assessed effects of TH on the COX-2, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 with RAW 264.7 cell. Results and Conclusion : 1. IgE, IL-4, IL-5, IL-6, IgM, IgGl levels in the serum of TH treated NC/Nga mouse group were decreased compared to the untreated control mice. IFN-r showed a increase in the experimental group compared to the untreated control group. The spleen weight of TH treated NC/Nga mice was decreased compared to the untreated control group. 2. mRNA expression levels of IL-4, IL-5 and CCR3 in the skin tissues of TH treated NC/Nga mice were decreased, and expression levels of IL-6 in the skin tissues of TH treated NC/Nga mice were decreased compared to the untreated control group. IFN-${\gamma}$ mRNA expression levels were increased compared to the untreated control group. 3. Judged from that IL-$1{\beta}$, TNF-${\alpha}$, IL-6 express of gene, effect of inflammatory Cytokines revelation were decreased compared to the untreated control group. 4. Depend on the strength of TH, inflammatory RAW 264.7 in the serum of TH were inhibited compared to the untreated control serum that leaded a COX-2 activity model. 5. Histological observation of the ear and skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of TH treated NC/Nga mice were highly reduced compared to the untreated control group.

Serial Changes in Mannose-Binding Lectin in Patients with Sepsis

  • Background: Mannose-binding lectin (MBL) deficiency leads to increased susceptibility to infection. We investigated whether serial changes in MBL levels are associated with the prognosis of patients diagnosed with septic shock, and correlated with cytokine levels. Methods: We enrolled 131 patients with septic shock in the study. We analyzed the serum samples for MBL and cytokine levels at baseline and 7 days later. Samples on day 7 were available in 73 patients. Results: We divided the patients with septic shock into four groups according to serum MBL levels (< $1.3{\mu}g/mL$ or ${\geq}1.3{\mu}g/mL$) on days 1 and 7. Patients with low MBL levels on day 1 and high MBL levels on day 7 showed a favorable prognosis for 28-day survival (odds ratio, 1.96, 95% confidence interval, 1.10-2.87; p=0.087). The high MBL group on day 7 showed a significant decrease in monocyte chemoattractant protein 1, interleukin (IL)-$1{\beta}$, IL-6, IL-8, interferon-${\gamma}$, and granulocyte macrophage colony-stimulating factor levels compared with the low MBL group on day 7. Conclusion: The increase in MBL levels of patients with septic shock may suggest a favorable prognosis and attenuate pro-inflammatory and anti-inflammatory responses.

면역세포의 cytokine 유리에 미치는 substance P의 영향 (Effects of Substance P on the Release of Cytokines from Immune Cell Lines)

  • The neuropeptide substance P(SP) has been implicated in the mediation of inflammation and immune-mediated disease such as arthritis. Recently, it was reported that SP was markedly increased around the blood vessels in inflamed gingiva as well as in close association with the inflammatory cell infiltrate. These results support that SP may contribute to the pathophysiology of neuronal inflammation in human periodontal tissues. SP may regulate inflammatory/immune responses by stimulating the proliferation of human T cells, differentiation and antibody-secreting potential of B cells, macrophage respiratory burst, connective tissue proliferation, and the secretion of cytokines from monocytes and T cells. Here, I studied potential role of SP as a costimulatory chemical signal in inflammatory/immune responses, by determining the released proinflammatory cytokines such as $MIP-1{\alpha}$, $IL-1{\beta}$, and IL-6 from culture supernatants of homogeneous immune cell lines. Serum free cell supernatants were concentrated with TCA precipitation, fractionated with SDS-PAGE, and subjected into western blot analysis. Among 15 cell lines tested, macrophage/monocyte cell line RAW264.7 and WRl9m.1 showed the highest level of induction of $MIP-1{\alpha}$ when stimulated with LPS. Discrete IL-6 bands with multiple forms of molecular mass were detected from supernatants of B cell lines A20(32kDa), Daudi(32, 35kDa), and SKW6.4(29kDa), which were expressed constitutively. $IL-1{\beta}$ could not be detected by the method of western blot analysis from supernatants of all cell lines tested except RAW264.7, WRl9m.1, and erythroid cell line K562 which showed the least amount of $IL-{\beta}$ secretion. SP $10^{-9}M$ with suboptimal dose of LPS treatment showed synergistic induction of $MIP-1{\alpha}$ release from RAW264.7 or WR19m.1, and also IL-6 release from A20, but this synergism is not the case in costimulation of RAW264.7 or WRl9m.1 with SP $10^{-9}M$ and TPA. Although treatment of T cell line CTLL-R8 with SP $10^{-7}M$ or PHA+TPA induced modest level of $MIP-1{\alpha}$ secretion, synergism was not observed when they are applied together. These findings all together suggest the possibility of a regulatory role of SP in inflammatory/immune reaction through differential modulation of bioactivities of other chemical cosignals.
