Study on the Changes in Saponins from Ginseng Callus by Tissue Culture -Part 1. Comparison of Saponins from Callus Tissue and from the Root of Ginseng Plant- (조직배양(組織培養)에 의한 인삼성분(人蔘成分)의 변화(變化) -제1보(第一報) Callus와 인삼성분(人蔘成分)의 비교(比較)-)

  • To study on the changes in saponins from callus mass by tissue culture, the callus was derived from the petiole of Korean Ginseng (Panax Ginseng C.A. Meyer) and cultivated on Murashige and Skoog's agar medium supplemented with 2.4-dichlorophenoxyacetic acid and kinetin for 8 months. Then, well-grown callus was analyzed for its components estimation. The results obtained are as follows: (1) When saponins isolated from callus mass were chromatographed on a silca gel plate, and determined by the thinchrograph TFG-10, the ratio of Rb, c to Rg(f) in saponins was 2.16 to 1 and Rb, c, d to Re, g (f) was 1 to 1.63, while in the case of saponins from the root of Panax Ginseng grown by soil culture, the ratio of Rb, c to Rg(f) was 1.03 to 1 and the ratio of Rb, c,d to Re, g(f) was 1 to 1.17. (2) Sapogenins were obtained from the hydrolysates of saponins, and determined by thinchrograph TFG-10. The ratio of panaxadiol to panaxatriol in sapogenins from callus saponins was 2.66 to 1, while the ratio of panaxadiol to panaxatriol in sapogenins from ginseng root saponins was 1.86 to 1. From the results above mentioned, we concluded that the relative contents of sapogenins in saponins from callus mass by tissue culture were different from those in saponins from ginseng root by soil culture.

Dead Lactobacillus plantarum Stimulates and Skews Immune Responses toward T helper 1 and 17 Polarizations in RAW 264.7 Cells and Mouse Splenocytes

  • This study was undertaken to evaluate the immunomodulatory effect of dead nano-sized Lactobacillus plantarum (nLp) in RAW 264.7 cells and murine primary splenocytes. nLp is a dead, shrunken, processed form of L. plantarum nF1 isolated from kimchi (a traditional Korean fermented cabbage) and is less than 1 μm in size. It was found that nLp treatment stimulated nitric oxide (NO) production more in RAW 264.7 macrophages than pure live L. plantarum (pLp), and that the stimulatory properties were probably largely derived from its cell wall. In addition, nLp induced murine splenocyte proliferation more so than pLp; in particular, a high dose of nLp (1.0 × 1011 CFU/ml) stimulated proliferation as much as lipopolysaccharide at 2 μg/ml. Moreover, according to our cytokine profile results in splenocytes, nLp treatment promoted Th1 (TNF-α, IL-12 p70) responses rather than Th2 (IL-4, IL-5) responses and also increased Th17 (IL-6, IL-17A) responses. Thus, nLp stimulated NO release in RAW 264.7 cells and induced splenocyte proliferation more so than pLp and stimulated Th1 and Th17 cytokine production. These findings suggested that dead nLp has potential use as a functional food ingredient to improve the immune response, and especially as a means of inducing Th1/Th17 immune responses.

Effect of Dietary Antimicrobials on Immune Status in Broiler Chickens

  • This study evaluated the effects of dietary anticoccidial drugs plus antibiotic growth promoters (AGPs) on parameters of immunity in commercial broiler chickens. Day-old chicks were raised on used litter from a farm with endemic gangrenous dermatitis to simulate natural pathogen exposure and provided with diets containing decoquinate (DECX) or monensin (COBN) as anticoccidials plus bacitracin methylene disalicylate and roxarsone as AGPs. As a negative control, the chickens were fed with a non-supplemented diet. Immune parameters examined were concanavalin A (ConA)-stimulated spleen cell proliferation, intestine intraepithelial lymphocyte (IEL) and spleen cell subpopulations, and cytokine/chemokine mRNA levels in IELs and spleen cells. ConA-induced proliferation was decreased at 14 d post-hatch in DECX-treated chickens, and increased at 25 and 43 d in COBN-treated animals, compared with untreated controls. In DECX-treated birds, increased percentages of $MHC2^+$ and $CD4^+$ IELS were detected at 14 d, but decreased percentages of these cells were seen at 43 d, compared with untreated controls, while increased $TCR2^+$ IELs were evident at the latter time. Dietary COBN was associated with decreased fractions of $MHC2^+$ and $CD4^+$ IELs and reduced percentages of $MHC2^+$, $BU1^+$, and $TCR1^+$ spleen cells compared with controls. The levels of transcripts for interleukin-4 (IL-4), IL-6, IL-17F, IL-13, CXCLi2, interferon-${\gamma}$ (IFN-${\gamma}$), and transforming growth factor${\beta}$4 were elevated in IELs, and those for IL-13, IL-17D, CXCLi2, and IFN-${\gamma}$ were increased in spleen cells, of DECX- and/or COBN-treated chickens compared with untreated controls. By contrast, IL-2 and IL-12 mRNAs in IELs, and IL-4, IL-12, and IL-17F transcripts in spleen cells, were decreased in DECX- and/or COBN-treated chickens compared with controls. These results suggest that DECX or COBN, in combination with bacitracin and roxarsone, modulate the development of the chicken post-hatch immune system.

Improvement of Bronchial Immune Hypersensitivity Reaction by Extracts from Chrysanthemum morifolium and Scutellaria baicalensis

  • This study aimed to discover functional materials derived from resources, which can improve the troublesome symptoms of a bronchus by improving bronchial hyperresponsiveness as air pollution in Korea caused by fine dust and yellow dust is getting worse. Of natural resources grown naturally in Korea, Chrysanthemum morifolium(CM), and Scutellaria baicalensis(SB) have been used as a safe raw material for drinking or medicine for a long time, and it has been found that a combination of CS73 can improve bronchial health function in experimental animal models. Analysis of serum of animal models with asthma induced by ovalbumin (chicken egg albumin) and analysis of cytokine production in BALF (Bronchoalveolar lavage fluid) showed that inflammatory indices IL-1β, IL-4, IL-5, IL-6, IL-10, IL-13, IL-17A, IL-17F, and IL-17E were significantly reduced and that respective production of IL-2 and IFN-γ was significantly increased in the group taking extracts from CS73 when compared with the control group. These results suggested that the combination of CS73 could be used as a natural treatment for asthma. In addition, in the animal models that the combination of CS73 significantly decreased the respective production of IgE, histamine and TSLP when compared with the control group. In experimental models, the ratio of Chrysanthemum morifolium to Scutellaria baicalensis of 7 : 3 had more excellent effect than other combined experimental groups, which suggests that the above combination can be developed as a natural treatment for asthma and is valuable as a pharmaceutical composition with an effect of improving bronchial health, capable of contributing to the public health threatened by fine dust.

Crown and Root Rot of Greenhouse Tomato Caused by Fusarium oxysporum f. sp. radicis-lycopersici in Korea

  • Forty(40) isolates of Fusarium oxysporum isolated from wilting tomato plants at Buyeo of Korea in 1997 were inoculated to four tomato cultivars (Ponderosa, Okitsu 3, Walter, and Zuiken) to examine pathogenic reactions. Isolation rates of F. oxysporum f. sp. lycopersici (FOL) races 1 and 2, and F. oxysporum f. sp. radicis-lycopersici(FORL) were 3.5%, 24.5%, and 57.5%, respectively. Mycelial growth on potato-dextrose agar at different temperature for the three pathogens was $26^{\circ}$. In the pathogenicity tests, however, the range of optimum temperature for disease development for FORL was between 15 and $20^{\circ}$, while that for races 1 and 2 of FOL were specifically pathogenic to tomato only. This suggests that host ranges of FORL and FOL differ significantly.

  • In this study, we examined transmission efficiency and expression level of the transgenes in the transgenic mice. The transgenic lines secreting a considerable amount of human lactoferrin(LF) thrombopoietin(TPO), interleukin-10(IL-10) into their milk were subjected to access the inheritance and maintenance of transgenic phenotype. They were bred through three generations. The transmission frequency for each generations(F9, F10, F11) of 3 lines was 38.03±10.43%(13/35), 48.33±3.76%(19/39) and 31.83±8.88%(9/28) in the LF line, 51.33±18.98%(20/38), 63.70±35.71%(12/20) and 29.57± 15.05%(8/26) in the TPO line, 38.27±17.74%(15/37), 47.47±29.88%(14/28) and 50.87±5.85%(14/28) in the IL-10 line, respectively. Ginsenoside F2 attenuates chronic-binge ethanol-induced liver injury by increasing regulatory T cells and decreasing Th17 cells

  • Background: Recently, beneficial roles of ginsenoside F2 (GF2), a minor constituent of Panax ginseng, have been demonstrated in diverse inflammatory diseases. However, its roles in alcoholic liver inflammation and injury have not been clearly understood. Here, we investigated the underlying mechanism by which GF2 ameliorated alcoholic liver injury. Methods: To induce alcoholic liver injury, C57BL/6J wild type (WT) or interleukin (IL)-10 knockout (KO) mice were orally administered with ethanol (3 g/kg) or ethanol-containing GF2 (50 mg/kg) for 2 wk. Liver injury and infiltration of macrophages and neutrophils were evaluated by serum biochemistry and immunohistochemistry, respectively. The changes of hepatic immune cells were assessed by flow cytometry and polymerase chain reaction analysis. In vitro differentiation of naïve T cells was performed. Results: GF2 treatment significantly attenuated alcoholic liver injury, in which infiltrations of inflammatory macrophages and neutrophils were decreased. Moreover, the frequencies of Foxp3+ regulatory T cells (Tregs) increased but IL-17-producing T (Th17) cells decreased in GF2-treated mice compared to controls. Furthermore, the mRNA expression of IL-10 and Foxp3 was significantly increased, whereas IL-17 mRNA expression was suppressed in GF2-treated mice. However, these beneficial roles of GF2 were not observed in GF2-treated IL-10 KO mice, suggesting a critical role of IL-10. Similarly, GF2 treatment suppressed differentiation of naïve T cells into Th17 cells by inhibiting RORgt expression and stimulating Foxp3 expression. Conclusion: The present study suggests that GF2 treatment attenuates alcoholic liver injury by increasing IL-10 expression and Tregs and decreasing IL-17 expression and Th17 cells.

Reduction of Injection Dose in 18F-FDG Fusion PET (PET-CT 검사에서 18F-FDG 투여량 감소에 대한 고찰)

  • Purpose With the recent rise of social issue regarding radiation exposure, attention to medical radiation use has been placed under a great spotlight. During PET-CT examination, generally about 40% more of $^{18}F$-FDG is used than EANM recommendation. While maintaining the diagnostic test result, we hope to find optimal injection dose to minimize the $^{18}F$-FDG in patients by utilizing the latest PET-CT scanner which is equiped with the newest technology. Materials and Methods During this experiment, the Biograph Truepoint 40 (siemens, USA) installed in 2007 and mCT 64 (siemens, USA) installed in 2011 were used and evaluated NECR (noise-equivalent counting rate) by using a scatter phantom. For the image quality evaluation of each scanner, we injected 3.7, 4.44 and 5.18 MBq/kg of $^{18}F$-FDG in NEMA IEC Body Phantom and also evaluated SNR between two scanners by using the data acquired at 60, 70, 80, 90, 100, 110 and 120 sec per bed. For the clinical evaluation, actual data of patients who were injected $^{18}F$-FDG 3.7, 4.44, 5.18 MBq/kg were used to compare SNR and draw a final result. Results As a result, mCT 64 peak NECR value was 1.65e+005, which is 10% higher than Turepoint 40. SNR values using the IEC body phantom was 17.9%, 17.4% and 17.1% higher in $^{18}F$-FDG 3.7 MBq/kg, 4.44 MBq/kg and 5.18 MBq/kg. In clinical patients, SNR values of the image mCT 64 was 16.5, which is 25% higher than Turepoint 40 scanner. Conclusion To draw a conclusion from the test result of this experiment, the same quality of SNR could be attained even with 10% reduced injection dose, if when the duration is extended by 10 sec/bed. This optimal result was possible due to enhanced equipment. The NECR (one of the equipment's performance assessment criteria for the scanner) increased by 10% and the SNR (one of the image quality assessment criteria) also increased by 17.5%. Therefore, we can expect to reduce the injection dose without deterioration of image quality. In consequence, it will also help to decrease the patient's anxiety of the radiation exposure.

Relationship of Academic Stress, Sociality and Self-Directed Learning in Nursing Students (일 대학 간호대학생의 학업스트레스, 사회성 및 자기주도적 학습간의 관계)

  • The purpose of this study is to investigate the levels of academic stress, sociality, and self-directed learning in nursing students, and to examine the relationship between the variables. Data were collected using a self-administered questionnaire from 208 students in the first and second years of a nursing program. This study was performed from December 5 to December 10, 2015. The data were analyzed with SPSS WIN 18.0 software (SPSS Inc., Chicago, IL, USA). The results were as follows. The mean score of academic stress was $3.20{\pm}0.53$, that of sociality was $2.91{\pm}0.25$, and that of self-directed learning was $3.33{\pm}0.33$. There was a negative correlation between academic stress and self-directed learning (r=-0.16, p=0.024) and a positive correlation between sociality and self-directed learning (r=0.17, p=0.015). Higher academic stress correlated with lower self-directed learning, while higher sociality correlated with higher self-directed learning. The results indicate that the levels of academic stress, sociality, and self-directed learning in nursing students can influence the quality of nursing education. Thus, there is a need to develop programs to reduce academic stress and to help improve sociality and self-directed learning for nursing students.

Influence of Parameters on Adhesion Strength on TiN Film by using R.F. Plasma Assisted Chemical Vapor Deposition (PACVD로 증착된 TiN 박막의 밀착성에 관하여)

  • In this study, TiN film was deposited onto steel by R.F.-PACVD in order to investigate the influence of parameters on the adhesion strength between film and substrate. Experimental results showed that adhesion strength by SAT is different from by optical microscopy. Adhesion strength is increased when the deposition temperature increases and is influenced by R.F. power and electrode distance. Especially heat treatment on the substrate has influenced over the adhesion strength, so it showed the 22 Newtons in adhesion strength by SAT and adhesion strength is decreased when deposition thickeness is thick and hardness is high. Also if the film is thick and high hardness simultaneous, the film was delaminated seriously.
