Mucosal Immunity Related to FOXP3+ Regulatory T Cells, Th17 Cells and Cytokines in Pediatric Inflammatory Bowel Disease

  • Background: We aimed to investigate mucosal immunity related to forkhead box P3 ($FOXP3^+$) regulatory T (Treg) cells, T helper 17 (Th17) cells and cytokines in pediatric inflammatory bowel disease (IBD). Methods: Mucosal tissues from terminal ileum and colon and serum samples were collected from twelve children with IBD and seven control children. Immunohistochemical staining was done using anti-human FOXP3 and anti-$ROR{\gamma}t$ antibodies. Serum levels of cytokines were analyzed using a multiplex assay covering interleukin $(IL)-1{\beta}$, IL-4, IL-6, IL-10, IL-17A/F, IL-21, IL-22, IL-23, IL-25, IL-31, IL-33, interferon $(IFN)-{\gamma}$, soluble CD40L, and tumor necrosis factor-${\alpha}$. Results: $FOXP3^+$ Treg cells in the lamina propria (LP) of terminal ileum of patients with Crohn's disease were significantly (P < 0.05) higher than those in the healthy controls. $ROR{\gamma}t^+$ T cells of terminal ileum tended to be higher in Crohn's disease than those in the control. In the multiplex assay, serum concentrations (pg/mL) of IL-4 ($9.6{\pm}1.5$ vs. $12.7{\pm}3.0$), IL-21 ($14.9{\pm}1.5$ vs. $26.4{\pm}9.1$), IL-33 ($14.3{\pm}0.9$ vs. $19.1{\pm}5.3$), and $IFN-{\gamma}$ ($15.2{\pm}5.9$ vs. $50.2{\pm}42.4$) were significantly lower in Crohn's disease than those in the control group. However, serum concentration of IL-6 ($119.1{\pm}79.6$ vs. $52.9{\pm}39.1$) was higher in Crohn's disease than that in the control. Serum concentrations of IL-17A ($64.2{\pm}17.2$ vs. $28.3{\pm}10.0$) and IL-22 ($37.5{\pm}8.8$ vs. $27.2{\pm}3.7$) were significantly higher in ulcerative colitis than those in Crohn's disease. Conclusion: Mucosal immunity analysis showed increased $FOXP3^+$ T reg cells in the LP with Crohn's disease while Th17 cell polarizing and signature cytokines were decreased in the serum samples of Crohn's disease but increased in ulcerative colitis.

The Influence of Dietary Characteristics on the Milk Quantity and Quality of Riverine Buffaloes: Estimate of the Energy/Protein Requirements, for a Medium-high Production, in the First Ninety Days of Lactation

  • The data used came from two trials undertaken under the same climatic conditions (spring-summer). In both trials pluriparious buffaloes were utilized similar in weight, body condition score, and milk production from the previous year. From the first trial the data used was from the sub-period 23-88 DIM provided by seven animals fed ad libitum with diet A (6.69 MJ/kg DM; 158.30 g/kg of crude protein) with a forage/concentrate ratio of 48/52. From the second trial the data used was from the sub-period 33-90 DIM provided by seven animals fed ad libitum with diet B (6.63 MJ/kg DM; 179.50 g/kg of crude protein) and by seven animals fed ad libitum with diet C (5.99 MJ/kg DM; 155.40 g/kg of crude protein), each of the diets had the same forage/concentrate ratio (53/47). A significant difference was found in milk production between group B and C (13.08 vs. 11.56 kg/d, p<0.05), an intermediate production (12.10 kg/d) was noted in group A. A significant difference was found between fat (76.58 vs. 69.24 g/kg, p<0.05), protein (46.14 vs. 43.16 g/kg, p<0.05) and casein (39.94 vs. 34.98 g/kg, p<0.05) of the milk of group B with respect to group A. The milk of group C gave fat values (71.80 g/kg), protein (45.52 g/kg) and casein (39.06 g/kg) statistically equal to those of group B. The milk of groups B and C, in respect to the milk of group A, gave values of $K_{20}$ (1.77, 1.82 vs. 3.68 min, p<0.05), statistically lower and values of $A_{30}$ (48.28, 47.27 vs. 40.64 mm, p<0.05) statistically higher. Two simple linear regressions were calculated where the independent variable (x) was the daily standardized milk production, the dependent variable (y) or the daily intake of net energy or crude protein. Equation 1) NE (MJ/d) = 74.4049+2.8308${\times}$kg of normalized milk; equation 2) CP (kg/d) = 1.4507+0.1085${\times}$kg of normalized milk, both the equations were significant (p<0.05) with determination coefficients of 0.58 and 0.50 respectively. For a production of normalized milk that varies from 9 to 13 kg, the respective energy-protein concentrations fluctuate from 6.09 to 6.78 MJ/kg DM and from 148.00 to 174.46 g/kg DM.

Artemisia fukudo essential oil attenuates LPS-induced inflammation by suppressing NF-${\kappa}B$ and MAPK activation in RAW 264.7 cells

  • In the present study, the chemical constituents of Artemisia fukudo essential oil (AFE) were investigated using GC-MS. The major constituents were ${\alpha}$-thujone (40.28%), ${\beta}$-thujone (12.69%), camphor (6.95%) and caryophyllene (6.01%). We also examined the effects of AFE on the production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-IL-$1{\beta}$ (IL-$1{\beta}$), and IL-6 in lipopolysaccharide (LPS)-activated RAW 264.7 cells. Western blotting and RT-PCR analyses indicated that AFE has potent dose-dependent inhibitory effects on pro-inflammatory cytokines and mediators. We investigated the mechanism by which AFE inhibits NO and $PGE_2$ by examining the level of nuclear factor-${\kappa}B$ (NF-${\kappa}B$: p50 and p65) activation within the mitogen-activated protein kinase (MAPK: ERK, JNK and p38) pathway, which is an inflammation induced signal pathway in RAW 264.7 cells. AFE inhibited LPS-induced ERK, JNK and p38 phosphorylation. Furthermore, AFE inhibited the LPS-induced phosphorylation and degradation of $I{\kappa}B-{\alpha}$, which is required for the nuclear translocations of the p50 and p65 NF-${\kappa}B$ subunits in RAW 264.7 cells. Our results suggest that AFE might exert an anti-inflammatory effect by inhibiting the expression of pro-inflammatory cytokines. Such an effect is mediated by a blocking of NF-${\kappa}B$ activation which consequently inhibits the generation of inflammatory mediators in RAW 264.7 cells. AFE may be useful for treating inflammatory diseases.

A Study on Factors Influencing Medical Treatment Decision-Making for Overactive Bladders in Female Patients over 40 - Data from Clinical Trial Participants - (40세 이상 과민성방광 여성 환자의 의료이용 결정요인에 관한 연구 - 임상시험 참여자를 중심으로 -)

  • Purpose: The aim of this study is to determine associated factors affecting the utilization of Medical Care for an overactive bladder. Methods: Total 114 women with overactive bladder participated in this study. Patients were categorized in two groups, the patients with treatment experiences for an overactive bladder(n=40) and without treatment experiences for an overactive bladder(n=74). Patients completed Overactive Bladder Severity Score, Beck Depression Inventory, King's Health Questionnaire, and questionnaires on treatment experience. The results were analysed using SPSS for Windows 12.0k. All statistical tests were two-sided with a p-value < 0.05 being significant. Results: Patients with treatment experience had a greater score on KHQ, OABSS, BDI and had longer symptom duration period than those without treatment experience. And only Impact of Life(P=0.004), Role Limitation(P=0.004), Social Limitation(P=0.007), Emotion(P=0.035), Sleep/Energy(P=0.001), Incontinence Severity Measures(P=0.012) among KHQ domains were statistically significant difference between two groups. Conclusions: The most influencing factor associated with utilization of Medical Service in overactive bladder is the impact of the patient's quality of life.

Effect of Licochalcone A on the Production of Cytokines in LPS-Activated RAW264.7 Macrophage Cells (Licochalcone A가 대식세포주의 사이토카인 생성에 미치는 영향)

  • Licochalcone A is a chalcone isolated from the roots of Glycyrrhiza inflate. In this study, we examined the effects of licochalonce A on the production of cytokines in LPS-activated macrophages. Licochalcone A inhibited the secretion of proinflammatory cytokines such as IL-1$\beta$, IL-6, and TNF-$\alpha$. The reduced secretion of proinflammatory cytokines is related to the differences in the mRNA expression of IL-1$\beta$, IL-6, and TNF-$\alpha$. Moreover, licochalcone A inhibited the mRNA expression of IL-12p40, IL-18, and IL-23p19. To investigate its mechanism, we performed gel shift assay. Licochalcone A reduced nuclear NF-${\kappa}B$ binding activity in LPS-activated RAW264.7 cells. Taken together, these results suggest that licochalcone A has anti-inflammatory effects in LPS-activated macrophages and its mechanism could be through the down-regulation of binding to the ${\kappa}B$ site.

Effects of Fermented Soybean on Body Weight, Body Fat and Serum Lipid in Obese Women (대두발효물 섭취가 비만 여성의 혈청 지질 농도와 체중 및 체지방 감소에 미치는 영향)

  • Objectives: The purpose of this study was to examine the effects of fermented soybean on body weight, body fat, serum lipid profiles in obese women, especially specific to menopausal woman. Methods: Sixty healthy obese volunteers who visited ${\bigcirc}{\bigcirc}$ University Oriental Hospital from May 20th, 2014 to September 25th, 2014 took part in clinical trial. They divided into 2 groups, 30 volunteers allocated to fermented soybean and other 30 to placebo group. Body weight, BMI, waist and hip ratio, serum lipid were measured 3 times, and fat percentage, leptin, adiponectin were evaluated 2 times. Results: All 60 volunteers completed 12-week trial. 5 men were excluded, and 2 women against the clinical decision rule were excluded. In the end, 53 women were studied as clinical subjects. After 12 weeks intervention, there was no effects in comparison of group by time interaction. Without considering time interaction, there was a significant difference in triglyceride level between soybean group and placebo group (p=0.044). Treatment group were dividing by age 40, a group in age 40 or over 40, and other group aged below 40. There was a significant difference in group by time interaction of total cholesterol level, and without considering time interaction, there was a significant change in waist-hip ratio between groups. Conclusions: There were no effects on weight and body fat decrease in 12-week trial using fermented soybean as a supplement. But there were significant differences in triglyceride change between the treatment and placebo groups, also cholesterol and waist and hip ratio in soybean group divided by age 40. It seems that fermented soybean is effected on improving serum lipid profiles.

Association between polymorphisms in Interleukin-17 receptor A gene and childhood IgA nephropathy (IgA 신병증 환자에서 Interleukin-17 수용체 A 유전자의 단일염기다형성 연관성 연구)

  • Purpose : Interleukin-17 (IL-17) is produced by activated CD4+T cells and exhibits pleiotropic biological activity on various cell types. IL-17 was reported to be involved in the immunoregulatory response in IgA nephropathy (IgAN). Our aim was to investigate the association between single-nucleotide polymorphisms (SNPs) in IL-17 receptor A (IL-17RA) gene and childhood IgAN. Methods : We analyzed the SNPs in the IL-17RA in 156 children with biopsy-proven IgAN and 245 healthy controls. We divided the IgAN patients into 2 groups and compared them with respect to proteinuria (${\leq}4$ and >$4mg/m^2/h$, ${\leq}40$ and >$40mg/m^2/h$, respectively) and the presence of pathological levels of biomarkers of diseases such as interstitial fibrosis, tubular atrophy, or global sclerosis. Results : No difference was observed between the SNP genotypes rs2895332, rs1468488, and rs4819553 between IgAN patients and control subjects. In addition, no significant difference was observed between allele frequency of SNPs rs2895 332, rs1468488, and rs4819553 between patients in the early and advanced stage of the disease. However, significant difference was observed between the genotype of SNP rs2895332 between patients with proteinuria (>$4mg/m^2/h$) and those without proteinuria (codominant model OR 0.36, 95% CI 0.19-.66, P <0.001; dominant model OR 0.35, 95% CI 0.17-.69 P =0.002; recessive model OR 0.12, 95% CI 0.01-.06 P =0.025). Conclusion : Our results indicate that the SNP in IL-17RA (rs2895332) may be related to the development of proteinuria in IgAN patients.

Studise on the Effects of Gami-Shengmnayuipung-tang on Acnes (가미승마위풍탕이 면포(Acne)에 미치는 실험적 연구)

  • Objective: This study was carried out to investigate the effects of Gami-Shengmayuipung-tang on acne. Methods : The effects of Gami-Shengmayuipung-tang (SYTSRG) on acne were measured by the $5\alpha-reductase$ inhibition, the sterilizing power for Propionibactrium acnes, the cytotoxicity of human monocyte, the inhibition for prostaglandins$(PGE_2)$, interleukins $(IL-l\beta)$ and $TNF-\alpha$ in inflammation, and the size of the hamtster ear sebaceous gland related to P. acnes. Results: On the $5\alpha-reductase$ inhibition of SYTSRG in vitro, an undiluted solution of SYTSRG showed $80\%$ inhibition on $5\alpha-reductase$ and 1/10 diluted solution of SYTSRG showed $40\%$ inhibition on $5\alpha-reductase$. On the sterilizing power for Propionibactrium acnes related acne, an undiluted solution and 1/10 diluted solution of SYTSRG formed $12\beta{\AE}$ clear zone diameters. SYTSRG did not show cytotoxicity of human monocyte. Concentrations of $0.01\%\;and\;0.04\%\;and\;0.08\%$ of SYTSRG inhibited the production of prostaglandins $(PGE_2)$ in the human monocyte stimulated with P. acnes LPS. $0.08\%$ and less of SYTSRG inhibited the production of interleukins $(IL-l\beta)$ in the human monocyte stimulated with P. acnes LPS. Concentrations of $0.04\%,\;0.08\%\;and\;0.12\%$ of SYTSRG inhibited the production of $TNF-\alpha$ in the human monocyte stimulated with P. acnes LPS. As the antiandrogenic compound, SYTSRG was used in hamster ears with topical application. SYTSRG diminished the size of the hamster ear sebaceous gland in males, but not in females. Conclusion: The present data suggest that SYTSRG may affect the primary stage of inflammation of acne.

  • To determine if initial infection with Mycobacterium tuberculosis changes the balance of cytokines between T cells and macrophages, we evaluated interferon (IFN)-${\gamma}$), interleukin-12 (IL)-12, and tumor necrosis factor (TNF)-${\alpha}$ productions by peripheral blood mononuclear cells (PBMC) from 15 untreated active pulmonary tuberculosis (TB) patients and 12 healthy tuberculin reactors (HTR). Freshly isolated PBMC were stimulated with Triton X-100 solubilized protein (TSP), 30-kDa or purified protein derivatives (PPD) antigen for 6, 18 and 96 hours. IL-12 p40 production by antigen-stimulated PBMC from TB patients was significantly decreased compared with that in HTR. In addition, IFN-${\gamma}$ production was significantly depressed in TB patients than that in HTR at a 96-hr stimulation. However, TNF-${\alpha}$ production was significantly higher in antigen-stimulated PBMC from TB than that of HTR. A pronounced increase in IFN-${\gamma}$ protein followed neutralization of IL-10 in early TB patients. However, neutralization of TNF-${\alpha}$ did not significantly alter IFN-${\gamma}$ induction in PBMC from TB patients. There were no significantly differences in the cytokine productions among three proteins, TSP, 30-kDa or PPD antigen. These results indicate that development of TB may be strongly associated with dysregulated productions of IL-12, IFN-${\gamma}$ and TNF-${\alpha}$, during the initial immune responses to M. tuberculosis. Further understanding of operative cytokine networks during human immune cell responses to protein antigens of M. tuberculosis may improve strategies for vaccine development.

Modulatory Activity of CpG Oligonucleotides from Bifidobacterium longum on Immune Cells

  • The purpose of this study was to characterize and investigate the immune activity of CpG oligodeoxynucleotides (ODNs) from Bifidobacterium longum. Bacterial CpG motifs have attracted considerable interests because of their immunomodulatory activities. Genomic DNA from B. longum was prepared and amplified for 4 different 180-188-mer double-stranded ODNs (BLODN1-BLODN4). When immune cells (RAW 264.7 murine macrophages and JAWS II dendritic cells) with these ODNs were treated, BLODN4 induced the highest immune activity. To assess the effectiveness of the CpG sequences within BLODN4, single-stranded 40-mer ODNs containing CpG sequences (sBLODN4-1, sBLODN4-2) were synthesized. sBLODN4-1 induced higher level of cytokines such as interleukin (IL)-12p40 and tumor necrosis factor (TNF)-$\alpha$ by macrophage and IL-6 and TNF-$\alpha$ by dendritic cells than did sBLODN4-2. The results suggest that CpG ODNs-enriched components of B. longum might be useful as an immunomodulatory functional food ingredient.
