Expression of IGF-1 and Its Receptor Genes in the Oocytes and Preimplantation Embryos in Mouse (생쥐 난자와 착상전 초기배아에서 IGF-1과 IGF-1 수용체 유전자 발현)

  • Insulin-like growth factors (IGF-1 and IGF-2) play an important regulatory role in premplantation embryonic development. To study the role of IGF-1 during premplantation embryonic development in mouse, the presence of mRNA transcripts for IGF-1 and IGF-lR in the oocytes and preimplantation embryos was examined. In this study, the transcripts of IGF-1 was detected in oocytes using primers for IGF-1. The PCR products were identified by Msp I restriction enzyme digest. We revealed that the transcripts of IGF-1 and IGF-1R were presented in the oocytes and preimplantation embryos. The highest mRNA levels in GV stage oocytes were decreased at 4- or 8-cell stage and then reincreased upto blastocyst. The presence of IGF-1 and IGF-lR in GV-oocytes suggests that the transcripts in the early stage embryos were derived from maternal genome. Additionally, the presence of IGF-1 and IGF-lR in the oocytes and preimplantation embryos suggests that IGF-1 plays an autocrine role during preimplantation embryonic development through IGF-lR as a signalling pathway.

Studies on the Production of Transgenic Rabbits with Growth Hormone Receptor and IGF-1 Receptor Genes (성장관련 유전자를 이용한 형질전환토끼의 생산에 관한 연구)

  • Transgenic rabbits were produced by DNA microinjection using growth hormone receptor (GHR) and IGF-1 receptor (IGF-1R) genes fused to metallothionein(MT) promoter. The overall efficiencies for production of transgenic rabbits were 3.2% and 3.1% for GHR and IGF-lR genes, respectively. Founder rabbits transmitted transgenes to their progenies through medelian fashion. Growth rate of GHR and IGF-lR transgenic rabbits was significantly faster than that of non-transgenic rabbits. Transgenic rabbits grew large. (25% and 15% increase in body weight of GHR and IGF-lR transgenic rabbits, respectively) than non-transgenic rabbits and organ weight of transgenic rabbits increased, suggesting that GHR and IGF-1R genes affects growth rates in transgenic rabbits.

The association between serum IGF-1 and neonatal growth and disease in a NICU (신생아집중치료실 입원아의 혈청 IGF-1과 성장 및 질병 사이의 관련성)

  • Purpose : The objective of this study was to establish the serum IGF-1 level in newborn infants, and investigate its association with growth and diseases. Methods : In a retrospective study, serum IGF-1 levels were measured for newborn infants admitted to NICU at Kyungpook University Hospital from March 2007 to July 2007. Birth data, disease history, and hospital course were obtained from medical records. Results : Of 52 blood samples obtained at birth, serum IGF-l levels in 30 preterm infants ($31.6{\pm}27.3$ ng/mL) were lower than in 22 full-term infants ($53.4{\pm}40.0$ ng/mL; P<0.05). In sick full-term infants, serum IGF-1 levels ($46.0{\pm}40.2$ ng/mL) were lower than in healthy full-term infants ($64.1{\pm}39.5$ ng/mL; P<0.05). In preterm infants, there were no differences in IGF-1 levels between healthy ($33.2{\pm}23.3$ ng/mL) and sick infants ($30.6{\pm}30.4$ ng/mL); however, IGF-1 levels in both sick and healthy preterm infants were lower than in healthy full-term infants. Among infants admitted after 8 days of life, serum IGF-1 levels were higher in infants who gained weight ($70.8{\pm}36.2$ ng/mL) than in infants who lost weight ($13.3{\pm}19.9$ ng/mL; P<0.01); however IGF-1 levels showed no difference between gender or method of delivery. Conclusion : The study showed lower IGF-l levels in preterm infants than in full-term infants. Additionally, the IGF-l level in infants with weight loss was lower than in infants with weight gain. These results indicate that serum IGF-1 is associated with gestational age and postnatal growth.

Effect of Bovine Colostral Whey Fraction containing Insulin-like Growth Factor on Cell Proliferation (젖소 초유 중의 Insulin-like Growth Factor-1 함유 분획이 세포 성장에 미치는 영향)

  • Insulin-like growth factor-I (IGF-I) rich fraction, which was obtained molecules ranged between 30 kDa and 1 kDa, was fractionated by ultrafiltration from bovine colostral whey with 30 kDa and 1 kDa membrane. IGF-I included in fractionated IGF-I rich fraction was confirmed by SDS-PAGE and western blotting and then the quantity of IGF-I was measured by ELISA. IGF-I concentration in IGF-I rich fraction was 10ng/mg protein. Effect of IGF-I rich fraction on in vitro proliferation of several cells was tested. IEC-6 cell proliferation rate was increased 60%. 53%, 30%, and 20% at l0ng, 1ng, 0.1ng and IGF-I of IGF-I, respectively, compared to control group which was not supplemented by IGF-I rich fraction. IGF-I rich fraction stimulated in vitro proliferation of IEC-6 cell in a dose dependent manner by increasing cell number. Detroit 551 cell proliferation was enhanced 56% and 26% at 10ng and 1ng level of IGF-I, respectively, compared to control group. EL-4 cell and L6 cell proliferation was increased 53% and 46% at 10ng of IGF-I, respectively, compared to control group.

Effect of velvet antler on the function of adipocytes(3T3-L1) and its association with IGF-1 (녹용(鹿茸)이 지방세포(脂肪細胞)(3T3-L1)와 성장인자(成長因子)에 미치는 영향(影響))

  • Objectives: These experimental studies were designed to investigate the effects of antler's extracts on the expression of leptin, IGF-l and IGF-1 receptor on cultured 3T3-L1 cells. Methods: A mouse adipoblast cell line, 3T3-L1, was cultured with or without a differentiation medium containing Isobutylmethylxanthin, Dexametasone and Insulin before adding extracts of antler of various concentratio(10, 50, ]00, 200${\mu}g/ml$). The expression of leptin and IGF-1 receptor was measured by western blot assay, and expression of IGF-1 was determined by F ACS analysis. Results and Conclusion: The 3T3-L1 cells' differentiation did not show a significant induction by extracts of antler. The expression of leptin was significantly decreased depend on antler's concentration. The expression of IGF-1 showed a slightly increase by extracts of antler, whereas that of IGF-receptor showed a tendency to increase. The total amount of intracellular triglyceride showed a tendency to diminish as the concentration of antler's extract increase.

Effect of YGF251 on Secretion Of IGF-1 in Human Blood (YGF251 투여에 의한 인체내 혈중의 IGF-1 분비량 변화 효과)

  • For stimulating the in vivo secretion of IGF-1 (insulin-like growth factor-1) which is well known to promote the various physiological actions in human body, the natural herbal extract, YGF251 (young growth factor 251), was developed and evaluated for its effect as IGF-1 secretagogue in this study. The clinical study was peformed as double blind test, and 31 adult female and male volunteers between the age of 40 and 70 were investigated for their changes of concentration of IGF-1 , insulin level, weight, blood pressure, and liver and kidney functions. As the result of paired sample test on the change of the concentration of IGF-1, in YGF251 treated group, it was 245.6 ng/mL before dosing. The concentration of IGF-1 was increased to 269.3 ng/mL after a month and to 275.6 ng/mL after two months, and both were statistically significant (p〈0.05). While in control group, the concentration of IGF-1 was 280.0 ng/mL before dosing, but decreased to 239.2 ng/mL after a month and to 230.2 ng/mL after two months, and both were also statistically significant (p〈0.05). In YGF251 treated group, the concentration of insulin in blood increased about 2 times after a month dosing as an average level, but in control group, it showed a decrease of 36% compared with before dosing. And there were little changes regarding to the measured weight and blood pressure. Various measured data in order to observe the alteration in liver and kidney functions by the administration of YGF251 showed a little change within measuring error range.

Luteolin Inhibits Proliferation Induced by IGF-1 Pathway Dependent ERα in Human Breast Cancer MCF-7 Cells

  • The growth of many breast tumors is stimulated by IGF-1, which activates signal transduction pathways inducing cell proliferation. $ER{\alpha}$ is important in this process. The aim of the study was to investigate relationships in vitro among inhibitory effects of luteolin on the growth of MCF-7 cells, IGF-1 pathway and $ER{\alpha}$. Our results showed that luteolin could effectively block IGF-l-stimulated MCF-7 cell proliferation in a dose- and time-dependent manner and block cell cycle progression and induce apoptosis evidenced by the flow cytometric detection of sub-G1DNA content. Luteolin markedly decreased IGF-l-dependent IGF-IR and Akt phosphorylation without affecting Erk1/2 phosphorylation. Further experiments pointed out that $ER{\alpha}$ was directly involved in IGF-l induced cell growth inhibitory effects of luteolin, which significantly decreased $ER{\alpha}$ expression. Knockdown of $ER{\alpha}$ in MCF-7 cells by an $ER{\alpha}$-specific siRNA decreased the IGF-l induced cell growth inhibitory effects of luteolin. $ER{\alpha}$ is thus a possible target of luteolin. These findings indicate that the inhibitory effect of luteolin on the growth of MCF-7 cells is via inhibiting IGF-l mediated PI3K-Akt pathway dependent of $ER{\alpha}$ expression.

Effect of Plant Extract(YGF) on Inducing IGF-1 Secretion (식물추출물(YGF)의 lGF-1 분비촉진에 미치는 영향)

  • The extracts of a lot of (different) plants were studied for inducing the secretion of growth hormone and IGF-1. In the cell test, it was found that the concentration of rat growth hormone was 88 - 99 ng/mL in a certain plant extract treatment group which was at least 6 times higher than the amount in control group. The concentration of IGF-1 in blood was 1,140ng/mL at 8 hours after the oral administration of YGF, which was remarkably much higher than that in control group. This meant that YGF maintained the level of IGF-1 secretion higher in the treated group. In addition, the increase of 6% in bone length was shown for the long-term oral administration of YGF. Thus, YGF seemed to affect significantly for inducing the secretion of IGF-1 which was essential for the physical growth and anti-aging.

Suppressed Cell Proliferation and Differentiation Following an Over-expression of Myostatin is Associated with Inhibited Expression of Insulin-like Growth Factor II and Myogenin in Rat L6 Myoblasts

  • Myostatin (MSTN) and insulin-like growth factors (IGFs) are a known inhibitor and stimulators of proliferation and differentiation of muscle cells, respectively. The present study was performed to investigate the relationship of MSTN-induced growth inhibition to expression of the IGF system components and myogenin, a muscle cell-specific transcription factor, in rat L6 myoblasts. The L6 cells transfected with a green fluorescent protein-MSTN plasmid expression construct had a 47% less cell number than mock-transfected cells after 3-d serum-free culture, accompanied by delayed differentiation which was suggested by inhibited aggregation of cells. Moreover, cells transfected with the expression construct had decreased expression of IGF-II and myogenin genes, but not IGF-I or its receptor genes, as examined by reverse transcription-polymerase chain reaction. The reduced mitosis of the L6 cells transfected with the MSTN-expression construct increased following an addition of either IGF-I or IGF-II to the culture medium, but not to the level of mock-transfected cells. By contrast, myogenin gene expression in these cells increased after the addition of either IGF to the level of mock-transfected cells. Collectively, these results suggest that the inhibitory effect of MSTN on L6 cell proliferation and differentiation is likely to be partly mediated by serially suppressed expression of IGF-II and myogenin genes, not IGF-I gene.

Evaluation of circulating IGF-I and IGFBP-3 as biomarkers for tumors in dogs

  • Background: Serum-based parameters are considered non-invasive biomarkers for cancer detection. In human studies, insulin-like growth factor-I and II (IGF-I and IGF-II) and insulin-like growth factor binding protein-3 (IGFBP-3) are useful as diagnostic or prognostic markers and potential therapeutic targets. Objectives: This study examined the diagnostic utility of circulating IGF-I, IGF-II, and IGFBP-3 levels in healthy dogs and dogs with tumors. Methods: The serum concentrations of these biomarkers in 86 dogs with tumors were compared with those in 30 healthy dogs using an enzyme-linked immunosorbent assay (ELISA). Results: The ELISA results showed no difference between healthy dogs and dogs with tumors in the serum IGF-II concentrations. On the other hand, there was a significant difference in the circulating IGF-I and IGFBP-3 levels between healthy dogs and dogs with tumors. The concentrations of serum IGF-I (median [interquartile range], 103.4 [59.5-175] ng/mL) in dogs with epithelial tumors were higher than those (58.4 ng/mL [43.5-79.9]) in healthy dogs. Thus, the concentrations of serum IGFBP-3 (43.4 ng/mL [33.2-57.2]) in dogs with malignant mesenchymal tumors were lower than those (60.8 ng/mL [47.6-70.5]) in healthy dogs. Conclusions: The serum IGF-I and IGFBP-3 levels can be used as diagnostic biomarkers in dogs with tumors.
