The Effect of KangwhalSokdan-tang(Jianghuoxuduan-tang) Administration on Muscle Regeneration in Atrophied Rat Muscle Induced by Hindlimb Suspension (후지체공(後肢滯空) 유발 흰쥐의 위축 근육에 대한 강활속단탕(羌活續斷湯)의 근 재생효과)

  • Objectives : This experimental study was designed to investigate the effect of KangwhalSokdan-tang(Jianghuoxuduan-tang) on the muscle regeneration of atrophied rat muscle by hindlimb suspension. Materials and methods : In this study, Sprague-Dawley rats weighing about 250g were subjected to hindlimb suspension and divided into total four groups: Normal group(n=6), Control group(n=6), Hindlimb non-treatment group(n=6), Hindlimb treatment group(n=6). Experiments were seperately tried two times. The first trial was studied by the following two groups; The first was normal group(n=6). The second was group(n=18) for hindlimb suspension during 2 weeks (control I group). The second trial after 2 weeks hindlimb suspension was studied by the following three groups; The third group(n=6) was expired immediately after 2 weeks hindlimb suspension. The forth group(n=6) was given free activity during 2 weeks after 2 weeks hindlimb suspension. The fifth group(n=6) was administrated of KST during 2 weeks after 2 weeks hindlimb suspension. In order to investigate degree of muscle atrophy, body weight and gastrocnemius muscle mass were compared. To analyze muscle regeneration factors(expression of IGF-1, Myogenin, MyoD), Western blot was used. Results : The results were analyzed by statistical process as follows, 1. In body weight, all hindlimb suspension groups were lower than normal group, but tendency of increase was shown in KST group compared to non-treatment group after 2 weeks hindlimb suspension. 2. In gastrocnemius muscle mass, KST group on both side was significantly higher than non-treatment group after 2 weeks hindlimb suspension. 3. In case of IGF-I, Type I of KST group was significantly increased than non-treatment group, but Type II was not shown significance. 4. There was no significantly difference in Myogenin. 5. In MyoD, Type I of KST group was significantly increased than control group, and Type II of KST group was significantly increased than non-treatment group. Conclusions : In summary, this study demonstrates that KST administration has an effect to prevent muscle atrophy and contribute muscle regeneration and proliferation. And also it is suggested that IGF-I and MyoD is major factors of myogenesis expression to KST adminstration after hindlimb suspension.

The Effects of Rehmannia glutinosa on the Protein Expression Related to the Angiogenesis, Cell Survival and Inflammation (생지황(生地黃)이 혈관신생, 세포생존 및 염증관련 단백질발현에 미치는 영향)

  • Objective : Angiogenesis induced by hypoxia and inflammation are an essential process of solid tumors and psoriasis. We researched the HIF-1 ${\alpha}$ (hypoxia inducible factor 1 alpha), VEGF(Vascular Endothelial Growth Factor), survival related PI3K-Akt, and inflammation related COX-2 protein expressions to get the information of the mechanism and effects of Rehmannia glutinosa in HepG2 and HaCaT cell lines. Method : To investigate the roles of the Rehmannia glutinosa extract, we performed MTS assay and western blots using HaCaT cells and HepG2 cells. HaCaT cells and HepG2 cells were treated with $50{\mu}g/ml$ and $100{\mu}g/ml$ Rehmannia glutinosa extracts. After 4hrs, HaCaT cells were treated with IGF-II protein for 24hrs and HepG2 cells were treated with $CoCl_2$. Results : 1. We could ohserve that the reduction of the protein level of HIT-1 ${\alpha}$ induced by IGF-II in HaCaT cells. 2. We Could ohserve that the decreased PI3K-Akt and COX-2 expression level by Rehmannia glutinosa extracts treated in HaCaT cells independently ith ERK1/2. 3. We could observe that the reduction of the protein level of HIF-1 ${\alpha}$ induced by $CoCl_2$ in HepG2 cells. Conclusion : These results suggest that Rehmannia glutinosa extracts contributes to the anti-survival pathway and anti-inflammatory activities. Also, we could assume that Rehmannia glutinosa act as anti-inflanmmatory or anti-hypoxia agents via reduction of COX-2 and HIF-1 ${\alpha}$.

A Study on the Growth Effects of Ecklonia cava By-product (감태 가공부산물을 이용한 성장효과에 관한 연구)

  • We investigated the beneficial effects of Ecklonia cava by-product (ECB), a residual product obtained after polyphenol extraction from Ecklonia cava, on normal rats. Male Sprague-Dawley rats were divided into the following three groups: Control group (NC), received basal diet; ECB 0.1 group, received basal diet supplemented with 0.1% ECB for 21 days; ECB 0.5 group, received basal diet supplemented with 0.5% ECB for 21 days. The productivity, serum immunoglobulin level, expression levels of muscle-related genes, and cecal microflora were measured in all the treatment groups to evaluate the potential use of ECB as a feed additive. The ECB 0.1 group exhibited enhanced expression of Myod, Myog, and Igf1 genes, which increased the body weight of rats. Additionally, treatment with ECB increased the cecal Lactobacillus spp. counts and the serum immunoglobulin G levels in rats. Thus, ECB can effectively increase body weight and can be a potential feed additive for improving growth.

MITOGENIC EFFECTS OF NICOTINE TO HUMAN PERIODONTAL LIGAMENT(PDL) CELLS IN VITRO (In Vitro에서 니코틴이 치주인대세포에 미치는 세포분열효과에 대한 연구)

  • Nicotine is one of the major components of cigarette smoking which causes various systemic and local diseases to human body. Mitogenic effects of nicotine to systemic disease are interesting factors in the results of cellular Proliferation especially to vascular and pulmonary tissue or cells. The study of local effects concerns with destruction of tissue and delayed healing rate after various surgical treatment. Platelet-Derived Growth factor(PDGF) and Insulin-like growth factor(IGF) are blown as major mitogens to human PDL cells. The purpose of this study was to investgate the mitogenic effects of nicotine to human PDL cells. We studied the expression of PDGF-$\alpha$ receptor, PDGF-$\beta$receptor, and IGF-l receptor mRNA from the nicotine treated human PDL cells by northern analysis. The experimental groups were divided into different serum($1\%,\;10\%$) and nicotine (100ng/m1,1000ng/m1) concentrations and each group was studied by time course. The results of this study showed upregulation of PDGF-${\alpha},\;{\beta}$ receptor and IGF-l receptor mRNA at 100ng/ml nicotine concentration and $10\%$ serum group to the time course. These results suggest that physiologically attainable nicotine concentrations may stimulate the mitogenic gene synthesis to human PDL cells in vitro.

IGF-1 from Adipose-Derived Mesenchymal Stem Cells Promotes Radioresistance of Breast Cancer Cells

  • Purpose: The aim of this study was to investigate effects of adipose-derived mesenchymal stem cells (AMSCs) on radioresistance of breast cancer cells. Materials and Methods: MTT assays were used to detect any influence of AMSC supernatants on proliferation of breast cancer cells; cell migration assays were used to determine the effect of breast cancer cells on the recruitment of AMSCs; the cell survival fraction post-irradiation was assessed by clonogenic survival assay; ${\gamma}$-H2AX foci number post-irradiation was determined via fluorescence microscopy; and expression of IGF-1R was detected by Western blotting. Results: AMSC supernatants promoted proliferation and radioresistance of breast cancer cells. Breast cancer cells could recruit AMSCs, especially after irradiation. IGF-1 derived from AMSCs might be responsible for the radioresistance of breast cancer cells. Conclusions: Our results suggest that AMSCs in the tumor microenvironment may affect the outcome of radiotherapy for breast cancer in vitro.

Effects of Feeding Betaine on Performance and Hormonal Secretion in Laying Hens

  • The effects of dietary betaine on performance, blood compositions, hepatic amino acid concentrations and hormonal secretions were examined in laying hens. Egg production was significantly higher in birds fed the 16.5 % protein diet compared to those fed 14.5 % protein diet(p<0.05), whereas dietary supplementation of betaine did not show any significant effect. The high level of protein and betaine supplementation significantly improved egg weight, egg mass and feed conversion(p<0.05), while eggshell breaking strength, eggshell thickness and Haugh unit were not influenced by betaine and dietary protein levels. Supplemental betaine did not affect serum total protein, albumin and BUN concentration. However, uric acid concentration significantly increased in 600 ppm betaine-fed groups(p<0.05). Concentrations of most hepatic amino acid were influenced by increased protein feeding and dietary betaine supplementation. Hormone studies recorded significantly higher serum and hepatocyte IGF-I concentration in 600 and 1,200 ppm betaine treatments(p<0.05) compared to those of control group. IGF-I mRNA gene expression of hepatocytes revealed statistically correlated increase in 600 and 1,200 ppm betaine-fed groups compared to the controls(p<0.05). Serum IGFBP-3 concentration was significantly elevated in 600 ppm betaine treatments. However, the secretion of IGFBP-1 in hepatocyte of laying hens fed with 600 and 1,200 ppm of betaine showed a significant decrease compared to the control group(p<0.05). Results of these study show that dietary betaine supplementation affects protein and hormone metabolism in laying hens.

Effect of Snail Extract on Bone Growth in Vitro and in Vivo (달팽이 추출물이 골 성장에 미치는 in Vitro 및 in Vivo 영향)

  • This study investigated the effect of snail extract on the growth parameters of old female rats (27 weeks). Rats were administered orally with snail extract at a dose of 100 mg/kg, 200 mg/kg, chondroitin sulfate 10 mg/kg and 0.9% saline (control) for 8 weeks. Bone mineral density (BMD) and serum concentrations of insulin-like growth factor 1 (IGF-1) and insulinlike growth factor-binding protein 3 (IGFBP-3) were significantly higher in rats exposed to snail extract for 8 weeks. MG-63 cells (human osteoblast-like cells) were treated with snail extract for 48 h. Their differentiation and proliferation was investigated with Western blot and morphological changes observed via immunofluorescence staining of ${\beta}-catenin$. Treatment with snail extract significantly increased the levels of growth factors including ${\beta}-catenin$ and IGF-1. The snail extract affected osteoblast formation. Morphological changes in MG-63 cells were observed via immunofluorescence staining. Treatment with snail extract increased the expression of ${\beta}-catenin$ in MG-63 cells. Results suggest that the treatment of MG-63 cells with snail extract increased the longitudinal growth and growth factor levels. Snail extract may be pharmacologically effective in osteogenic differentiation in vitro and represents a potential therapeutic agent for bone formation.

Branched-chain Amino Acids Reverse the Growth of Intrauterine Growth Retardation Rats in a Malnutrition Model

  • This experiment was conducted to determine the effect of dietary supplementation with BCAA (branched-chain amino acids: leucine, isoleucine and valine) on improving the growth of rats in a malnutritional IUGR (Intrauterine Growth Retardation) model, which was established by feeding restriction. In the experimental treatment, rats were fed purified diets supplemented with BCAA (mixed) during the whole gestation period, while arginine and alanine supplementation were set as the positive and negative control group, respectively. The results showed that, compared to the effect of alanine, BCAA reversed IUGR by increasing the fetus weights by 18.4% and placental weights by 18.0% while fetal numbers were statistically increased. Analysis of gene and protein expression revealed that BCAA treatment increased embryonic liver IGF-I expression; the uterus expressed higher levels of estrogen receptor-$\alpha$ (ER-$\alpha$) and progesterone receptor (PR), and the placenta expressed higher levels of IGF-II. Amino acid analysis of dam plasma revealed that BCAA supplementation effectively enhanced the plasma BCAA levels caused by the feed restriction. BCAA also enhanced the embryonic liver gluconeogenesis by augmenting the expression of two key enzymes, namely fructose-1,6-biphosphatase (FBP) and phosphoenolpyruvate carboxykinase (PEPCK). In conclusion, supplementation of BCAA increased litter size, embryonic weight and litter embryonic weight by improving the dam uterus and placental functions as well as increasing gluconeogenesis in the embryonic liver, which further provided energy to enhance the embryonic growth.

Effects of Kochiae Fructus Extracts on the Expression of Angiogeneis and Inflammation Related Proteins (지부자(地膚子)의 신생혈관 및 염증매개 단백질 발현에 미치는 영향)

  • Hypoxia induced angiogenesis and inflammation are essential processes for metastasis and progress of solid tumors. We examined the anti-angiogenic and inflammation related activity of Kochiae Fructus (KF) extract. To investigate the roles of the KF extract, we performed MTS assay, western blots using HaCaT cells and $HepG_2$ cells. The results are as follows. The protein level of $HIF-1{\alpha}$ was reduced when induced by $CoCl_2$ in $HepG_2$ cells treated with KF extract and induced by IGF-11 in HaCaT cells treated with KF extract. KF extract reduced the mRNA level of VEGF in HaCaT cells and KF extract reduced the protein level of iNOS in HaCaT cells. These results suggest that KF extract contributes to the anti-angiogenic and anti-inflammatory activities and also we could assume that KF extract act as antioxidant or anti-inflammatory agents via reduction of $HIF-1{\alpha}$.

Effect of NDM Hair Tonic on Hair Growth Promotion in C57BL6 Mice (C57BL6 생쥐에서 NDM 헤어토닉의 모발성장 촉진 효과)

  • NDM hair tonic is composed of several plant extracts which are known to be used in oriental medicine. This study was carried out to investigate the effect of NDM hair tonic on hair regrowth in an alopecia model of C57BL6 mice. Hair of six-weeks old mice were trimmed by electric clippers and electric shavers, so as not to damage the skin. The next day, mice without visible scraches were selected, randomized and separated in groups of 6 mice. There were four experimental groups including saline (negative control), 50% ethanol (vehicle control), 3% minoxidil (MXD), and NDM tonic. The test compounds were topically treated with 0.15 $m\ell$ per mouse per day for 2 weeks. The hair regrowth was determined photographically and histologically and the quantity of endocrine factors, IGF-1 and TGF-$\beta$, in the skin of mice was measured by PCR. No clinical signs were found in all animals. The topical treatment of NDM tonic for 2 weeks to dorsal skin accelerated hair regrowth faster than either the controls or MXD treatment. The NDM tonic treatment also promoted hair follicle development and elongation as compared with the controls or MXD treatment. Both NDM tonic and MXD treatment significantly increased the expression of IGF-1 and TGF-$\beta$ in the skin of C57BL6 mice as compared with the controls (p<0.05). These results suggest that NDM tonic has a hair growth activity and be useful for the treatment of baldness or alopecia.
