제대혈에서 아디포넥틴, 렙틴, 인슐린, IGF-I, IGFBP-3와 신생아의 신체계측과의 상관관계 (The relationship between adiponectin, leptin, insulin, insulin-like growth factor and IGF binding protein-3 in cord blood and neonatal anthropometric parameters)

  • 목 적 : 건강한 만삭아의 제대혈에서 아디포넥틴, 렙틴, 인슐린, IGF-I, IGFBP-3를 측정하여 출생체중, 신장, 지방량과의 관계를 알아보고자 하였다. 또한 적정 체중아 그룹에서 이 호르몬들의 출생후 1개월 동안 변화에 대해서 살펴보기로 하였다. 방 법 : 임신과 관련된 합병증이 없었던 산모에서 태어난 200명의 건강한 만삭아(남아 109명, 여아 91명)를 대상으로 하여 제대혈을 채취하여 혈장과 혈청을 분리하였고 출생체중, 출생신장, 머리둘레, 가슴둘레, 피부두께를 측정하였으며 폰더랄지수를 계산하였다. 대상 신생아들을 출생체중에 따라 AGA (n=132), SGA (n=29), LGA (n=39)의 세 그룹으로 나누었다. 적정체중아 중 15명에서 생후 3일, 7일, 30일에 신생아의 혈액을 채취하여 제대혈과 같은 방법으로 분리하였고 체중과 신장을 측정하였다. 결 과 : 아디포넥틴과 인슐린, IGF-I은 AGA, LGA군보다 SGA군에서 더 낮았다. 렙틴은 AGA, SGA군보다 LGA군에서 더 높았다. 아디포넥틴과 렙틴, 인슐린, IGF-I, IGFBP-3는 출생체중과 피부두께의 합과 양의 상관관계에 있었다. 출생 시 신장과 양의 상관관계를 보인 호르몬은 아디포넥틴, 렙틴, IGF-I이었다. 아디포넥틴은 렙틴과 양의 상관관계를 보였으나 인슐린, IGF-I, IGFBP-3와는 상관관계가 없었다. IGF-I은 남아보다 여아에서 더 높은 수치를 보였다. 출생 후 1개월 동안 렙틴은 생후 7일까지는 생리적 체중감소와 더불어 감소하다가 그 후 증가하였고 IGF-I 또한 생후 3일에 감소하다가 1개월 후 급격한 증가를 보였다. 결 론 : 아디포넥틴과 렙틴, 인슐린, IGF-I, IGFBP-3 모두 태아의 성장에 중요한 역할을 하며, 아디포넥틴은 인슐린, IGF-I 축과는 다른 기전으로 태아의 성장을 조절한다고 생각할 수 있었다. IGF-I은 남아에서보다 여아에서 더 높은 수치를 보여서 IGF-I의 성별간의 차이가 자궁 내에서도 존재하였다. 생후 1개월 동안의 성장에 다른 호르몬보다 IGF-I 이 더 중요한 역할을 한다는 사실을 알 수 있었다.

비만 청소년에서 Insulin, Insulin-like Growth Factor-I, Insulin-like Growth Factor Binding Protein-3에 대한 연구 (Serum Insulin, Insulin-like Growth Factor-I and Insulin-like Growth Factor Binding Protein-3 Levels in Obese Adolescents)

  • 목 적 : 소아 비만은 전세계적으로 증가하고 있는 추세이며, 신체적으로나 정신적으로 장애를 일으킨다. 특히 고혈압, 관상동맥질환, 당뇨병 등의 성인병이 문제가 되고 있다. 이에 비만 청소년에서 혈중 인슐린, 인슐린저항지수, 유리 지방산, IGF-I, IGFBP-3를 측정하였고, 심혈관 질환으로 진행할 수 있는 위험인자로 유용한지 알아보기 위해 본 연구를 실시하였다. 방 법 : 64명의 중등도 이상의 비만 청소년를 대상군으로 하였고, 같은 연령의 정상 청소년 20명을 대조군으로 하였다. 신장, 체중을 이용하여 비만도와 체질량지수를 산출하였고, 안정된 상태에서 수축기, 이완기 혈압을 측정하였다. 10시간 이상 금식 후 총 콜레스테롤, 중성지방, HDL 콜레스테롤, LDL 콜레스테롤, AST, ALT, 혈당, 인슐린, 유리지방산, IGF-I, IGFBP-3를 측정하였다. 결 과 : 인슐린은 비만군에서 $15.6{\pm}7.0{\mu}IU/mL$로 정상군보다 유의하게 높았고(P<0.01), 인슐린저항지수도 비만군에서 $64.6{\pm}31.6$으로 정상군보다 유의하게 높았다(P<0.01). IGF-I은 비만군에서 $498.1{\pm}122.2ng/mL$로 정상군보다 유의하게 높았고(P<0.05), IGFBP-3도 비만군에서 $3,777{\pm}4,721ng/mL$로 정상군보다 유의하게 높았다(P<0.05). 인슐린은 체질량지수(r=0.394), 비만도(r=0.304)와 양의 상관관계를 나타냈다. IGFBP-3는 비만도(r=0.419), 이완기 혈압(r=0.264), 체질량지수(r=0.247)와 양의 상관관계를 보였다. 인슐린저항지수는 체질량지수(r=0.595), 혈중 중성지방(r=0.515), 비만도(0.469)와 양의 상관관계를 나타냈다. 결 론 : 비만 청소년에서 혈중 인슐린, 인슐린저항지수, IGF-I, IGFBP-3가 증가하였다. 청소년 비만에서 이들 검사는 심혈관질환으로의 진행을 예측하는데 유용하리라 생각한다.

Conjugated Linoleic Acid에 의한 대장암 세포 증식 억제 기전 연구 (Study of the Mechanism for the Growth Inhibitory Effects of Conjugated Linoleic Acid on Caco-2 Colon Cancer Cells)

  • Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of linoleic acid (LA) and exhibits anticarcinogenic activity in a variety of animal models. We have previously observed that CLA inhibited the growth of Caco-2 cells, a human colon adenocarcinoma cell line. The present study was performed to determine whether the growth inhibitory effect of CLA is related to change in secretion of IGF- II and/or IGF-binding proteins (IGFBPs) that have been shown to regulate Caco-2 cell proliferation by an autocrine mechanism. Cells were incubated in serum-free medium with various concentrations of CLA or linoleic acid (LA). Immunoblot analysis of 24-hours, serum-free, conditioned medium using a monoclonal anti-IGF-IIantibody revealed that Caco-2 cells secreted both mature 6,500 Mr and higher Mr forms of pro IGF-II. The levels of pro IGF-II and mature IGF-IIwere decreased by 43 $\pm$ 2% and 53 $\pm$ 6%, respectively by treatment with 50 $\mu$ M CLA. LA slightly increased pro IGF- II levels. Results from Northern blot analysis showed that CLA decreased IGF-II mRNA levels at 50 $\mu$ M concentration suggesting that CLA regulation of IGF-II protein expression occurs partly at the transcriptional level. Ligand blot analysis of conditioned media using 1251-IGF-II revealed that CLA slightly decreased IGFBP-2 levels and increased IGFBP-4 levels. We confirmed our previous results that CLA inhibited cell growth in a dose-dependent manner but LA slightly increased cell growth. Exogenous IGF-II mitigated the growth inhibitory effect of CLA. These results indicate that the growth inhibitory effect of CLA may be at least in part mediated by decreasing IGF-II and IGFBP-2 secretion and increasing IGFBP-4 secretion in Caco-2 cells.

Association Analysis between Insulin-like Growth Factor Binding Protein 3 (IGFBP3) Polymorphisms and Carcass Traits in Cattle

  • The insulin-like growth factor binding protein 3 (IGFBP3) has been investigated as a candidate gene for growth promoting effects in beef cattle and a modulator of IGF bioactivity. Previously, we have reported twenty two sequence variants discovered in Korean native cattle (Hanwoo). In this study, we examined the association between gene-specific polymorphisms of IGFBP3 and cold carcass weight (CW) and marbling score (MS) among Korean native cattle. Among twenty two polymorphisms, four common polymorphic sites (-854G>C, -100G>A, +421G>T and +3863C>A) were genotyped in our beef cattle (n = 437). Statistical analysis revealed that one common polymorphism in the promoter region (-854G>C) showed putative associations with MS (p = 0.03). IGFBP3 variation/haplotype information analyzed in this study will provide valuable information into strategies for the production of a commercial line of beef cattle.

Molecular and Circulatory Expression of Insulin Growth Factors in Indian Females with Advanced Cervical Cancer

  • Background: Recent studies have demonstrated an association between insulin growth factor (IGF) and insulin growth factor binding protein-3 (IGFBP-III) serum levels and increased risk for various cancers. However, little information is available on clinical implications of the IGF system in Indian patients with cervical cancer. This study explored associations by analyzing their expression profiles in cervical cancer cases. Materials and Methods: Totals of 50 patients with advanced cervical cancer and 40 healthy controls were enrolled. Human papillomavirus (HPV) and cervical biopsy sample were obtained from all participating women. Circulatory levels were estimated by ELISA and the tissue expression was assessed using RT-PCR and Western blotting. Results: Levels of IGF-I and II showed significant increase whereas IGFBP-III showed significant decline in all patients as compared to controls. Spearman correlation analysis between IGFs and HPV status showed significant correlations. Conclusions: We demonstrated elevated circulating levels and tissue expression of IGF-I and IGF-II in advancer cancer cervix patients, as compared with controls, with a converse trend being apparent for IGFBP-III. In future, associations of the IGF system and clinical outcome of cervical cancer patients in post treatment samples might point to significance in disease mapping as a prognostic marker after validation with a larger patient series.

Effect of Dietary Energy Level on Nutrient Utilization, Insulin-like Growth Factor-I and Insulin-like Growth Factor Binding Protein-3 in Plasma, Liver and Longissimus dorsi Muscle in Growing-finishing Pigs Using Soybean Oil as an Energy Source

  • Two experiments were carried out to study the effects of dietary energy level on nutrient digestion, nitrogen (N) utilization, growth performance, insulin-like growth factor-I (IGF-I), and insulin-like growth factor binding protein-3 (IGFBP-3) in plasma, liver and longissimus dorsi muscle in growing-finishing pigs. In experiment 1 (Exp 1), 15 castrated male pigs (Duroc${\times}$Landrace${\times}$Large White) (Body weight, BW, 55.6${\pm}$1.8 kg) were divided into three groups and fed rations containing 13.33, 14.87 and 17.35 MJ digestible energy (DE)/kg as treatments I, II and III, respectively, using soybean oil as an energy source. The experiment lasted 8 days and faecal and urinary samples were collected during the last 3 days. The results showed that the digestibility of dry matter (DM), energy and N was increased from treatments I to III (p<0.01). N-retention and N-retention rate were not influenced by dietary DE level (p>0.05). In experiment 2 (Exp 2), 36 female pigs (Duroc${\times}$Landrace${\times}$Large White) (BW 41.5${\pm}$3.8 kg) were divided into three groups. The pigs were fed with the same three rations used in Exp 1 for 60 days. At the end of Exp 2, eight pigs were selected from each group for blood sampling and 4 pigs for slaughter trial. The results indicated that average daily feed intake (ADFI) and N-intake were significantly decreased (p<0.01), and DE intake (p<0.01) and average daily gain (ADG) (p<0.05) were increased. IGF-I and IGFBP-3 in plasma were increased (p<0.05). No significant differences in IGF-I and IGFBP-3 in liver and longissimus dorsi muscle were found between different treatments. It was concluded that higher dietary DE level improved nutrient digestibility, ADG and feed/gain ratio when soybean oil was used as an energy source in the ration of growing-finishing pigs. No significant differences were found in Nretention and IGF-I and IGFBP-3 in liver and longissimus dorsi muscle between different treatments.

Prader-Willi syndrome: a single center's experience in Korea

  • Purpose: Prader-Willi syndrome (PWS) is a complex genetic disorder that results from the lack of paternally expressed genes in the chromosome 15q11-q13 region. This study was performed to delineate the clinical features of PWS infants and toddlers and the effects of two-year growth hormone (GH) treatment according to gender and age at the start of treatment. Methods: The clinical characteristics and the results of the GH treatment were reviewed retrospectively for 30 PWS patients diagnosed by molecular genetic testing and clinical manifestations. Results: The mean age at diagnosis with PWS was 13.7 months (2-47 months of age). All patients showed the characteristics of facial dysmorphism, including brown hair and almond-shaped eyes. Most patients showed developmental delays/mental retardation (93.3%), cryptorchidism (75%), feeding problems in infancy (73.3%), and neonatal or infantile hypotonia (66.7%). Among 30 patients, 14 PWS infants and toddlers had been treated with GH for more than two years. Two years of GH treatment resulted in an improvement in head circumference-standard deviation score (HC-SDS), body weight-SDS, insulin-like growth factor-1 (IGF-1) SDS, IGF binding protein-3 (IGFBP-3) SDS, lean body mass, and bone mineral content, especially in IGFBP-3 SDS and motor development in PWS patients younger than two years of age. There was significant increase in IGF-1 SDS and IGFBP-3 SDS among male PWS patients after GH treatment. Conclusion: Our study showed increases in IGFBP-3 SDS and an improvement in motor development among individuals under two years of age after GH treatment, and significant difference in IGF-1 SDS and IGFBP-3 SDS by gender.

The Regulation of Insulin-Like Growth (IGF) Factors and IGF Binding Proteins by High Glucose in Mesangial Cells

  • It has been reported that glomerulosclerosis mediated by the dysfunction of mesangial cells and insulin-like growth factors (IGFs) are associated with the development of diabetic nephropathy. However, it is not yet known the effect of high glucose on IGF-I, -II secretion, IGF-I receptor, and IGFBPs expression in the mesangial cells. Thus, this study was conducted to examine the effect of high glucose on IGF system and its involvement of protein kinase C (PKC) and oxidative stress in mesangial cells. In this study, high glucose (25 mM) increased IGF-I and IGF-II secretion and mRNA expression (P<0.05), which was blocked by PKC inhibitor (staurosporine, 10/sup -8/ M) and antioxidant (N-acetyl cystein, 10/sup -5/ M). High glucose decreased IGFBP-1 and -2 expression but increased IGFBP-5 expression. These alteration of IGFBPs by high glucose was also prevented by staurosporine and NAC, suggesting the role of PKC and oxidative stress. Indeed, high glucose increased PKC activity. Furthermore, high glucose-induced increase of lipid peroxide (LPO) formation was blocked by PKC inhibitors. In conclusion, high glucose alters IGF system via PKC-oxidative pathways in mesangial cells.

  • Pregnancy-associated plasma protein-A (PAPP-A) is known as an important biomarker for fetal abnormality during first trimester and has a pivotal role in follicle development and corpus luteum formation. And also, it is being revealed that an expression of PAPP-A in various cells and tissues such as cancer and lesion area. PAPP-A is the major IGF binding protein-4 (IGFBP-4) protease. Cleavage of IGFBP-4 results in loss of binding affinity for IGF, causing increased IGF bioavailability for proliferation, survival, and migration. Additionally, PAPP-A can be used as a promising therapeutic target for healthy longevity. Despite growing interest, almost nothing is known about how PAPP-A expression is regulated in any tissue. This review will focus on what is currently known about the zinc metalloproteinase, PAPP-A, and its role in cells and tissues. PAPP-A is expressed in proliferating cells such as fetus in uterus, granulosa cells in follicle, dermis in wound, cancer cells, and Sertoli cells in testis. They have common characteristics of proliferation faster than normal cells with stimulating IGFs action and inhibiting IGFBPs. The PAPP-A functions and expression studies in livestock have not yet been conducted much. Further studies are needed to use PAPP-A as a marker for healthy longevity in animal science.

Role of the insulin-like growth factor system in gonad sexual maturation in Pacific oyster Crassostrea gigas

  • Background: The IGF system plays important roles in controlling growth, development, reproduction, and aging of organisms. Methods: To estimate maturation of the Pacific oyster Crassostrea gigas, we investigated the expression of insulin-like growth factor (IGF) system components and sex-specific genes. To determine the role of the IGF system in the growth and spawning period of female and male oysters, we examined mRNA expression levels of the C. gigas insulin receptor-related receptor (CIR), IGF binding protein complex acid labile subunit (IGFBP_ALS), and molluscan insulin-related peptide (MIP), as well as those of vitellogenin (Vg) and receptor-type guanylate cyclase (Gyc76C) in gonads of C. gigas collected between April and October, when sex can be determined visually in this species. Results: We found that MIP, IGFBP_ALS, and CIR mRNA expression levels were dependent on sex and month and were greater in males than in females. CIR and Vg mRNA expression levels were very similar among females, whereas IGF system components and Gyc76C were very similarly expressed among males. The highest expression values were observed in May, when oysters are mature; CIR and Vg mRNA expression levels were highest in females, and those of MIP, IGFBP_ALS, CIR, and Gyc76C were highest in males. Interestingly, we observed a 1:1 proportion of females to males during this period. Conclusion: Our results suggest that IGF system components, as well as Vg and Gyc76C, are associated with sexual maturation in C. gigas.
