Genetic Studies of Oenothera odorata Populations in Korea Based on Isozyme Analysis

  • The genetic variation in Korean evening primorse (Oeothera odorata L.) populations was examiend to estimate the level of allozyme variation within populatons using starch gel electrophoresis. 7 of 13 loci (Adh, Est-1, Est-2, Mdh-2, Pgd-2, Pgm-1, and Idh) revealed (Ps=43.2%) were polymorphic. The mean number of alleles per locus (A) and polymorphic locus (Ap) for populations were 1.64 and 2.46, respectively. The effective number of alleles (Aep) within populations relatively was low ranging from 1.08 to 1.22 with a mean of 1.14. Within populations, the mean number of allele per polymorphic loci (Ap) was 2.46, the mean number of alleles per locus (A) was 1.64, and the mean genetic diversity was 0.093. About 2.7% of the total allozyme diversity resided among populations (Mean GST=0.0274). FIS, a measure of the deviation from random mating within 13 populations, was relative low (mean FIS=0.03636). The indirect estimate of gene flow, based on the mean GST, was high (Nm=8.88). Estimates of gene flow were consistent with low levels of genetic differentiation among populations.

Change in Proteomic Profiles of Genetically Modified 1,3-Propanediol-Producing Recombinant E. coli

  • The recombinant E. coli $\Delta$6 mutant (galR, glpK, gldA, IdhA, lacI, tpiA) was used to produce 1,3-propanediol (PD) from glucose. The 1,3-PD production increased with feedback control of the glucose concentration using fed-batch fermentation. The maximum 1,3-PD concentration produced was 43 g/l after 60 h of fermentation. Glycerol production was minimized when controlling the glucose concentration at less than 1 g/l. The expression levels of seven enzymes related to the 1,3-PD production metabolism were compared during the cell growth phase and 1,3-PD production phase, and their expression levels all increased during 1,3-PD production, with the exception of alcohol dehydrogenase.

Bioinformatics Interpretation of Exome Sequencing: Blood Cancer

  • We had analyzed 10 exome sequencing data and single nucleotide polymorphism chips for blood cancer provided by the PGM21 (The National Project for Personalized Genomic Medicine) Award program. We had removed sample G06 because the pair is not correct and G10 because of possible contamination. In-house software somatic copy-number and heterozygosity alteration estimation (SCHALE) was used to detect one loss of heterozygosity region in G05. We had discovered 27 functionally important mutations. Network and pathway analyses gave us clues that NPM1, GATA2, and CEBPA were major driver genes. By comparing with previous somatic mutation profiles, we had concluded that the provided data originated from acute myeloid leukemia. Protein structure modeling showed that somatic mutations in IDH2, RASGEF1B, and MSH4 can affect protein structures.

  • Intraspecific and interspecific isozyme variations and their relationship of 16 local populations in 3 species of Phyllostachys, that is, P. bambusoides, P. nigra var. henonis and P. pubescens were investigated by multi-variate analysis. Leaf isozymes of Phyllostachys such as 6-PGD (6-phosphogluconate dehydrogenase), MDH (malate dehydrogenase), PGI (phosphoglucoisomerase), PRX (peroxidase), PGM (phosphoglutamase), IDH (isocitrate dehydrogenase) showed electrophoretic variations in the number of zymotypes (7, 6, 6, 9, 3 and 5, respectively). In the cluster analysis on the isozymic characteristics, 16 populations were classified into 3 species at the euclid genetic distance of 2.041. P. nigra var. henonis and P. bambusoides were clustered first at 2.813 and then P. pubescens at 3.001. So far, 3 local types of intraspecific ariation were found in P. nigra var. henonis and P. bambusoides.

  • A horizontal starch gel electrophoresis for enzyme proteins extracted from three Korean species and one Chinese species of Semisulcospira was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different of buffer systems. Two loci from each enzyme of GAPDH, GOT, ICDH, IDH and PEP(VL); three loci from each of three enzymes, EST, PEP(LGG) and PGDH; and five loci from GPI were observed. Most of the loci in three pleurocerid species employed showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-2, PEP(LGG-3) and PGDH-1 loci in Korean S. libertina and PEP(LGG-3), PGM-1 and PGM-2 loci in Chinese S. libertina showed polymorphic banding patterns. Three Korean Semisulcospira species including S. libertina were more closely clustered in a dendrogram within the range of genetic identity values of 0.818-0.936, and these clusters were lineated with Chinese S. libertina at the value of 0.621.

Effect of IOA and Media on Production of Somatic Hybrid by the Asymmetric Protoplast Fusion Between Three Solanum Species

  • In this study, we surveyed the influence of IOA (iodoacetamide) and media upon protoplast fusion for the efficient production of the somatic hybrid among S. sisymbriifolium and other Solanum species (S. intergrifolium and S. toxicarium). Regardless of a breed, as the IOA concentration increases, the cell division tends to decrease. As the influence of the media on the colony formation, we could get 5 colonies from the fusion of S. sisymbriifolium and IOA-treated S. integrifolium protoplast, but none was observed in other fusions. As a result of analyzing their IDH isozyme, we found a somatic hybrid in 2 objects.

The Impact of ESG Performance on Corporate Value of Chinese Companies

  • This study examined the influence of ESG performance on the firm value of Chinese equipment manufacturing companies over the period from 2011 to 2020. The analysis indicated that while ESG performance exerted a negative influence on firm value, this impact was not statistically significant. These findings substantiate the Institutional Difference Hypothesis (IDH), which posits that country-specfic institutional variations sifnificantly shape the strategic decision-making of organizations. When disaggregating the three components of ESG, variations in the results were observed both for overall ESG performance and for each individual dimension. Of the three dimension elements, the environmental (E) and governance (G) facets had a negative bearing, whereas the social (S) facet had a positive influence. Notably, only the governance dimension (G) demonstrated a statistically significant influence. These outcomes affirm the institutional difference hypothesis, illustrating divergent results across distint ESG dimensions.

Imaging-Based Versus Pathologic Survival Stratifications of Diffuse Glioma According to the 2021 WHO Classification System

  • Objective: Imaging-based survival stratification of patients with gliomas is important for their management, and the 2021 WHO classification system must be clinically tested. The aim of this study was to compare integrative imaging- and pathology-based methods for survival stratification of patients with diffuse glioma. Materials and Methods: This study included diffuse glioma cases from The Cancer Genome Atlas (training set: 141 patients) and Asan Medical Center (validation set: 131 patients). Two neuroradiologists analyzed presurgical CT and MRI to assign gliomas to five imaging-based risk subgroups (1 to 5) according to well-known imaging phenotypes (e.g., T2/FLAIR mismatch) and recategorized them into three imaging-based risk groups, according to the 2021 WHO classification: group 1 (corresponding to risk subgroup 1, indicating oligodendroglioma, isocitrate dehydrogenase [IDH]-mutant, and 1p19q-codeleted), group 2 (risk subgroups 2 and 3, indicating astrocytoma, IDH-mutant), and group 3 (risk subgroups 4 and 5, indicating glioblastoma, IDHwt). The progression-free survival (PFS) and overall survival (OS) were estimated for each imaging risk group, subgroup, and pathological diagnosis. Time-dependent area-under-the receiver operating characteristic analysis (AUC) was used to compare the performance between imaging-based and pathology-based survival model. Results: Both OS and PFS were stratified according to the five imaging-based risk subgroups (P < 0.001) and three imaging-based risk groups (P < 0.001). The three imaging-based groups showed high performance in predicting PFS at one-year (AUC, 0.787) and five-years (AUC, 0.823), which was similar to that of the pathology-based prediction of PFS (AUC of 0.785 and 0.837). Combined with clinical predictors, the performance of the imaging-based survival model for 1- and 3-year PFS (AUC 0.813 and 0.921) was similar to that of the pathology-based survival model (AUC 0.839 and 0.889). Conclusion: Imaging-based survival stratification according to the 2021 WHO classification demonstrated a performance similar to that of pathology-based survival stratification, especially in predicting PFS.

Prediction of Prognosis in Glioblastoma Using Radiomics Features of Dynamic Contrast-Enhanced MRI

  • Objective: To develop a radiomics risk score based on dynamic contrast-enhanced (DCE) MRI for prognosis prediction in patients with glioblastoma. Materials and Methods: One hundred and fifty patients (92 male [61.3%]; mean age ± standard deviation, 60.5 ± 13.5 years) with glioblastoma who underwent preoperative MRI were enrolled in the study. Six hundred and forty-two radiomic features were extracted from volume transfer constant (Ktrans), fractional volume of vascular plasma space (Vp), and fractional volume of extravascular extracellular space (Ve) maps of DCE MRI, wherein the regions of interest were based on both T1-weighted contrast-enhancing areas and non-enhancing T2 hyperintense areas. Using feature selection algorithms, salient radiomic features were selected from the 642 features. Next, a radiomics risk score was developed using a weighted combination of the selected features in the discovery set (n = 105); the risk score was validated in the validation set (n = 45) by investigating the difference in prognosis between the "radiomics risk score" groups. Finally, multivariable Cox regression analysis for progression-free survival was performed using the radiomics risk score and clinical variables as covariates. Results: 16 radiomic features obtained from non-enhancing T2 hyperintense areas were selected among the 642 features identified. The radiomics risk score was used to stratify high- and low-risk groups in both the discovery and validation sets (both p < 0.001 by the log-rank test). The radiomics risk score and presence of isocitrate dehydrogenase (IDH) mutation showed independent associations with progression-free survival in opposite directions (hazard ratio, 3.56; p = 0.004 and hazard ratio, 0.34; p = 0.022, respectively). Conclusion: We developed and validated the "radiomics risk score" from the features of DCE MRI based on non-enhancing T2 hyperintense areas for risk stratification of patients with glioblastoma. It was associated with progression-free survival independently of IDH mutation status.

Disruptions of Two Apparent rho-Independent Transcription Terminator Structures do not help in Enhancing the Expression of aceK in E. coli

  • Two apparent rho-independent transcription terminator structures within the coding sequence of aceK have been destroyed to access their roles in the differential expression between aceA and aceK in the glyoxylate bypass operon of E. coli. The effect of mutations on the expression of aceK was evaluated in two different ways: one by maxicell labeling and the other by lacZ fusion gene construction. The maxicell labeling experiment with the mutant operon clones has failed, like that of the wild type operon clone, to visibly show isocitrate dehrogenase (IDH) kinase/phosphatase, the product of aceK, on the autoradiogram of a protein gel. When the same mutations were introduced into an aceK::lacZ fusion gene to quantitatively evaluate the mutational effect, the activity of ${\beta}-galactosidase$ in neither of the mutant versions of the fusion gene was elevated significantly enough to explain the degree of polarity observed in this region. Thus, we conclude that neither of these intragenic, apparent rho-independent transcription terminator structures, which have long been suspected as a major determinant in the down regulation of aceK, really act as a premature transcriptional terminator.
